    • Integrated models including optics, structures, control systems, and disturbances are important design tools for Extremely Large Telescopes (ELTs). An integrated model has keen formulated for the European ELT and it includes telescope structure, main servos, primary mirror segment control system, wind, optics, wavefront sensor, deformable mirror, and an AO reconstructor and controller. There are three model phases: Initialization, execution of a solver to determine time responses, and post-processing. In near future, the model will be applied for performance studies and design trade-offs for the European ELT.

  • An APT investigation of an amorphous Cr-B-C thin film

    • A magnetron sputtered amorphous Cr-B-C thin film was investigated by means of atom probe tomography (APT). The film is constituted of two phases; a Cr-rich phase present as a few nanometer large regions embedded in a Cr-poor phase (tissue phase). The Cr-rich regions form columnar chains oriented parallel to the growth direction of the film. It was found that the Cr-rich regions have a higher B:C ratio than the Cr-poor regions. The composition of the phases was determined as approximately 35Cr-33B-30C and 15Cr-40B-42C (at%), respectively. The results suggest that this type of nanocomposite films has a more complex structure than previously anticipated, which may have an importance for the mechanical and electrical properties.

  • Zoledronic Acid Add-on Therapy for Standard-Risk Ewing Sarcoma Patients in the Ewing 2008R1 Trial

    • PURPOSE: The phase III, open-label, prospective, multicenter, randomized Ewing 2008R1 trial (EudraCT2008-003658-13) was conducted in 12 countries to evaluate the effect of zoledronic acid (ZOL) maintenance therapy compared with no add-on regarding event-free survival (EFS, primary endpoint) and overall survival (OS) in standard-risk Ewing sarcoma (EWS). PATIENTS AND METHODS: Eligible patients had localized EWS with either good histologic response to induction chemotherapy and/or small tumors (<200 mL). Patients received six cycles of VIDE induction and eight cycles of VAI (male) or eight cycles of VAC (female) consolidation. ZOL treatment started parallel to the sixth consolidation cycle. Randomization was stratified by tumor site (pelvis/other). The two-sided adaptive inverse-normal four-stage design (planned sample size 448 patients, significance level 5%, power 80%) was changed after the first interim analysis using the Müller-Schäfer method. RESULTS: Between April 2010 and November 2018, 284 patients were randomized (142 ZOL/142 no add-on). With a median follow-up of 3.9 years, EFS was not significantly different between ZOL and no add-on group in the adaptive design (HR, 0.74; 95% CI, 0.43-1.28, P = 0.27, intention-to-treat). Three-year EFS rates were 84.0% (95% CI, 77.7%-90.8%) for ZOL vs. 81.7% (95% CI, 75.2%-88.8%) for no add-on. Results were similar in the per-protocol collective. OS was not different between groups. The 3-year OS was 92.8% (95% CI, 88.4%-97.5%) for ZOL and 94.6% (95% CI, 90.9%-98.6%) for no add-on. Noticeable more renal, neurologic, and gastrointestinal toxicities were observed for ZOL (P < 0.05). Severe renal toxicities occurred more often in the ZOL arm (P = 0.003). CONCLUSIONS: In patients with standard-risk localized EWS, there is no additional benefit from maintenance treatment with ZOL.

    • With the proliferation of commercial Packaged ERP (P-ERP) systems in today’s enterprises, manyreasons exist to look for alternatives in the quest for innovation, business development, cost, agility anddependency. P-ERP provides a solid and proven business support, an ecosystem of consultancies andintegrators, senior management having gained confidence over the last 20 years, and commerciallybased support and development. This leaves companies with still more expensive P-ERP costs, still lessflexibility, a still harder push to lose possibilities for differentiation, still more homogenised businessprocesses, and absence of flexibility to change suppliers and systems. FOS-ERP offers an answer tomost of these questions, but is facing issues in market penetration. In this chapter, barriers of FOS-ERPare reviewed; proposals are made on how to manage barriers. An approach managing co-existence ofP-ERP and FOS-ERP is suggested. Concluding, FOS-ERP is seen as a strong option for enterprises inthe future, but a clear understanding and distinction must be the offset, barriers needs to be managed,and optimal co-existence will in most cases be the realistic scenario.

    • The interaction between DNA and a series of mono-, bis-, tris-, tetrakis-, and hexakis-intercalating 9-aminoacridines has been studied with flow linear dichroism (LD), circular dichroism (CD), electric orientation relaxation (EOR) techniques, and with viscometry and equilibrium analyses. The orientation of the 9-aminoacridine ligand relative to the average orientation of the DNA bases, measured by LD, shows that with both 9-aminoacridine and the bis(acridines) the in-plane short axes of the acridine ligands are oriented perfectly parallel to the planes of the DNA bases, as expected for classical intercalation, whereas the long axes are found to be significantly tilted. This is supported by the DNA lengthening measured by EOR, which for 9-aminoacridine is 1.5 base-pair units, compared with 1.0 for ethidium bromide. Also in case of the tris(acridines) LD, CD, viscometry, and equilibrium data indicate that all acridine ligands are intercalated. The binding analysis shows an increasing degree of cooperativity in the sequence 9-aminoacridine < bis(acridines) < tris(acridines), and the corresponding binding densities, 4, 8, and 11–14, respectively, are in good agreement with those expected from the nearest-neighbor exclusion principle. The LD and CD measurements show that the tetrakis- and hexakis(acridines), despite long and flexible links, bind to DNA with only three of the acridine ligands intercalated.
