  • Detection and localization of early- and late-stage cancers using platelet RNA

    • Cancer patients benefit from early tumor detection since treatment outcomes are more favorable for less advanced cancers. Platelets are involved in cancer progression and are considered a promising biosource for cancer detection, as they alter their RNA content upon local and systemic cues. We show that tumor-educated platelet (TEP) RNA-based blood tests enable the detection of 18 cancer types. With 99% specificity in asymptomatic controls, thromboSeq correctly detected the presence of cancer in two-thirds of 1,096 blood samples from stage I–IV cancer patients and in half of 352 stage I–III tumors. Symptomatic controls, including inflammatory and cardiovascular diseases, and benign tumors had increased false-positive test results with an average specificity of 78%. Moreover, thromboSeq determined the tumor site of origin in five different tumor types correctly in over 80% of the cancer patients. These results highlight the potential properties of TEP-derived RNA panels to supplement current approaches for blood-based cancer screening.

  • Morphological development of a small-scale beach nourishment in a non-tidal area

    • Combining hard and soft costal protection solutions can be an appropriate measure to enhance structure lifetime. This study aims to quantify the morphological evolution of the nourished stretch and adjacent coastlines. A small-scale beach nourishment has been performed in front of a rock revetment at site in Faxe Ladeplads in Zealand, Denmark. The overall objective is to learn more about the dynamics of small-scale nourishments in low energy environments. Monitoring techniques include repeated topographic (Trimble RTK-GPS, drone surveying) and bathymetric (single beam) measurements. To measure the hydrodynamic conditions two surface acceleration buoys deployed at -4 and -7 m water depth were used. Sediment volumes from nourishments of size 70,000 m3 and 20,000 m3 redistributed relatively quickly. The nourished material built up the cross-shore profile and a longshore bar in this area, and distributed sediments in the direction of the dominant littoral drift (SW). Results suggest that the morphological evolution of the nourishment is dependent on local hydrodynamic conditions and local geomorphology. These findings have implications for the main objectives of preventing wave overtopping onto an adjacent coastal road during extreme events and restoring a beach for recreation.

    • Access to local nearshore wave climate conditions on a detailed spatial scale is important for many coastal engineering practices. These include assessing the flood risk of coastal infrastructure, designing coastal protection measures, and estimating sediment transport processes and wave run-up. The present study displays a multi-scale wave modelling approach using the numerical wave model SWAN (Booij et al., 1999) together with field data applied in the southern Baltic Sea. The main objective of the study was to investigate the possibility of employing a single wave model for seamless simulations over several scales in time and space, from offshore to nearshore, including the effects of grid size and resolution. Validation with extensive field data was a crucial part of the study.

De vanligaste postoperativa komplikationerna vid tonsillkirurgi är postoperativt illamående och kräkningar (PONV), smärta, blödning och dehydrering. Tonsillkirurgin utförs idag antingen som dagkirurgiskt ingrepp eller som slutenvårdsingrepp med en vårdtid på ca ett dygn. Därmed sker den postoperativa vården i hemmet. Ett flertal studier har beskrivit patienternas upplevelse av höggradig smärta efter tonsillkirurgi. Obehandlad smärta leder till svårigheter att svälja och därmed otillräckligt dryckes- och matintag, vilket leder till dehydrering, ökad blödningsbenägenhet, sämre läkningsförmåga samt negativa  postoperativa beteendeförändringar hos barnet.Syftet var att i en nationell studie kartlägga farmakologiska rutiner och informationen i samband med tonsilloperation hos barn och ungdomar.

    • This study provides a Miocene-to-recent history of Tasman Leakage (TL), driving surface-to-intermediate waters from the Pacific into the Indian Ocean. TL, in addition to Indonesian ThroughFlow (ITF), constitutes an important part of the Southern Hemisphere Supergyre. Here, we employ deep-sea benthic delta C-13 timeseries from the southwestern Pacific and eastern Indian Oceans to identify the history of Tasman Leakage. The delta C-13 results combined with sedimentary evidence show that an inter-ocean connection south of Australia existed from 7 Ma onward. A southward shift in Westerlies combined with a northward movement of Australia created the oceanic corridor necessary for Tasman Leakage (between Australia and the sub-Antarctic Front) at this time. Furthermore, changes in the northern limb of the Supergyre (ITF) are evident in the sedimentary record on Broken Ridge from similar to 3 to 2 Ma when Banda Sea intermediate waters started originating from the North Pacific.

    • Background A healthy diet and optimal lifestyle choices are amongst the most important actions for the prevention of cardiometabolic diseases. Despite this, it appears difficult to convince consumers to select more nutritious foods. Furthermore, the development and production of healthier foods do not always lead to economic profits for the agro-food sector. Most dietary recommendations for the general population represent a "one-size-fits-all approach" which does not necessarily ensure that everyone has adequate exposure to health-promoting constituents of foods. Indeed, we now know that individuals show a high variability in responses when exposed to specific nutrients, foods, or diets. Purpose This review aims to highlight our current understanding of inter-individual variability in response to dietary bioactives, based on the integration of findings of the COST Action POSITIVe. We also evaluate opportunities for translation of scientific knowledge on inter-individual variability in response to dietary bioactives, once it becomes available, into practical applications for stakeholders, such as the agro-food industry. The potential impact from such applications will form an important impetus for the food industry to develop and market new high quality and healthy foods for specific groups of consumers in the future. This may contribute to a decrease in the burden of diet-related chronic diseases. Key messages Individual differences in ADME (Absorption, Digestion, Metabolism and Excretion) is believed to underpin much of the inter-individual variation in responses. Recent developments in the area of food metabolome databases and fast improvements in innovative metabotyping technologies hold great promise for improved profiling of dietary intake, exposure to individual ingredients, foods and dietary patterns, as well as our ability to identify individual responsiveness. The food industry needs well-defined population clusters or targets in order to be able to design "personalized products". There are indeed excellent industrial opportunities for foods that modulate gut microbiota, and thereby enable the delivery of food bioactive metabolites. It is currently not clear whether knowledge on individual nutrient needs, based on genetic or metagenomic data, would affect long-term dietary and health behaviours. Data to support the development of dietary recommendations may need to be generated by new n-of-1-based study designs in the future.

    • Metabolic dysfunction-associated steatotic liver disease (MASLD) is a global health concern with no effective and specific drug treatment available. The rs2642438 minor allele in mitochondrial amidoxime-reducing component 1 (MARC1) results in an aminoacidic substitution (p.Ala165Thr) and associates with protection against MASLD. However, the mechanisms behind this protective effect are unknown. In this study, we examined the consequences of this aminoacidic substitution on protein stability and subcellular localization.We overexpressed the human MARC1 A165 (wild-type) or 165T (mutant) invivo in mice and invitro in human hepatoma cells (HepG2 and HuH-7), generated several mutants at position 165 by insitu mutagenesis and then examined protein levels. We also generated HepG2 cells stably overexpressing MARC1 A165 or 165T to test the effect of this substitution on MARC1 subcellular localization.MARC1 165T overexpression resulted in lower protein levels than A165 both invivo and invitro. Similarly, any mutant at position 165 showed lower protein levels compared to the wild-type protein. We showed that the 165T mutant protein is polyubiquitinated and its degradation is accelerated through lysine-48 ubiquitin-mediated proteasomal degradation. We also showed that the 165T substitution does not affect the MARC1 subcellular localization.This study shows that alanine at position 165 in MARC1 is crucial for protein stability, and the threonine substitution at this position leads to a hypomorphic protein variant due to lower protein levels. Our result supports the notion that lowering hepatic MARC1 protein level may be a successful therapeutic strategy for treating MASLD.

  • FemtoMAX - An X-ray beamline for structural dynamics at the short-pulse facility of MAX IV

    • The FemtoMAX beamline facilitates studies of the structural dynamics of materials. Such studies are of fundamental importance for key scientific problems related to programming materials using light, enabling new storage media and new manufacturing techniques, obtaining sustainable energy by mimicking photosynthesis, and gleaning insights into chemical and biological functional dynamics. The FemtoMAX beamline utilizes the MAX IV linear accelerator as an electron source. The photon bursts have a pulse length of 100fs, which is on the timescale of molecular vibrations, and have wavelengths matching interatomic distances (Å). The uniqueness of the beamline has called for special beamline components. This paper presents the beamline design including ultrasensitive X-ray beam-position monitors based on thin Ce:YAG screens, efficient harmonic separators and novel timing tools.The FemtoMAX beamline facilitates studies of the structural dynamics of materials on the femtosecond timescale. The first commissioning results are presented.
