  • Electrochemical Lithium Storage Performance of Molten Salt Derived V2SnC MAX Phase

    • MAX phases are gaining attention as precursors of two-dimensional MXenes that are intensively pursued in applications for electrochemical energy storage. Here, we report the preparation of V2SnC MAX phase by the molten salt method. V2SnC is investigated as a lithium storage anode, showing a high gravimetric capacity of 490 mAh g(-1) and volumetric capacity of 570 mAh cm(-3) as well as superior rate performance of 95 mAh g(-1) (110 mAh cm(-3)) at 50 C, surpassing the ever-reported performance of MAX phase anodes. Supported by operando X-ray diffraction and density functional theory, a charge storage mechanism with dual redox reaction is proposed with a Sn-Li (de)alloying reaction that occurs at the edge sites of V2SnC particles where Sn atoms are exposed to the electrolyte followed by a redox reaction that occurs at V2C layers with Li. This study offers promise of using MAX phases with M-site and A-site elements that are redox active as high-rate lithium storage materials.

  • River morphology redistributes potentially toxic elements in acid mine drainage-impacted river sediments: Evidence, causes, and implications

    • River morphology plays a vital role in the transport of substance within them. However, our understanding of how natural and artificial morphologies redistribute different potentially toxic elements in acid mine drainage (AMD)-contaminated rivers remains poor. In this study, we linked morphological river features and physicochemical sediment characteristics to trace the redistribution of various potentially toxic elements and elucidate their implications for remediating rivers prone to AMD pollution. A dense network of sediment/soil samples was collected from different river morphological units, such as channels, dam reservoirs, pools, floodplain sandbars and wetlands in an AMD-impacted river. The analyses showed that the contaminant levels in channel generally decreased downstream from the headwater mine site, however, local fluctuations in certain areas were observed due to the trapping effect of various dams along the river. The As and Pb concentrations were higher at floodplain sandbars, while river channels exhibited higher Cd and Zn contamination. The concentrations and geochemical fractions of As, Cd, Cu, Pb and Zn in sediment/soil cores from sandbar and river channel also varied. Additionally, structural equation modeling analysis indicated that spatial variations in contaminant distributions were directly affected by physicochemical properties (such as the soil/sediment Fe, Zn, and S concentrations, and pH), which are indirectly affected by river morphology. The diverse morphology of the river redistributed AMDderived contaminants and could be used to identify contamination hotspots. Our analyses suggested that the feasibility and efficiency of previously proposed countermeasures varied for contaminants in different geomorphological units. In river channels, As uptake from sediments by aquatic plants may be less efficient than Cd, Cu, and Zn uptake due to its lower bioavailability. Moreover, vegetation prevented contaminant enriched soil particle erosion more than it aided in the phytoremediation of As- and Pb-contaminated sandbars. Thus the finding of this study provide a theoretical foundation for further studies on the transport and storage of AMDderived contaminants in similar rivers, along with the development of targeted remediation methods.

  • Separable Microneedle Patch to Protect and Deliver DNA Nanovaccines Against COVID-19

    • The successful control of coronavirus disease 2019 (COVID-19) pandemic is not only relying on the development of vaccines, but also depending on the storage, transportation, and administration of vaccines. Ideally, nucleic acid vaccine should be directly delivered to proper immune cells or tissue (such as lymph nodes). However, current developed vaccines are normally treated through intramuscular injection, where immune cells do not normally reside. Meanwhile, current nucleic acid vaccines must be stored in a frozen state that may hinder their application in developing countries. Here, we report a separable microneedle (SMN) patch to deliver polymer encapsulated spike (or nucleocapsid) protein encoding DNA vaccines and immune adjuvant for efficient immunization. Compared with intramuscular injection, SMN patch can deliver nanovaccines into intradermal for inducing potent and durable adaptive immunity. IFN-gamma(+)CD4/8(+) and IL-2(+)CD4/8(+) T cells or virus specific IgG are significantly increased after vaccination. Moreover, in vivo results show the SMN patches can be stored at room temperature for at least 30 days without decreases in immune responses. These features of nanovaccines-laden SMN patch are important for developing advanced COVID-19 vaccines with global accessibility.

  • Source and migration of short-chain chlorinated paraffins in the coastal East China Sea using multiproxies of marine organic geochemistry

    • Multiple proxies of terrestrial organic matters (TOM) were introduced to study the migration behaviors of short-chain chlorinated paraffins (SCCPs) in the coastal East China Sea (ECS). The contents of SCCPs in the surface sediment collected from Changjiang (Yangtze) River Delta (CRD) and along the Zhejiang-Fujian coastline ranged from 9.0 to 37.2 ng/g (dry weight, d.w.), displaying a "band-style" distribution trend. Spatial distribution patterns of SCCP congeners presented an increasing trend seaward and southward along the coastline for shorter carbon length (C₁₀ + C₁₁) and lower chlorinated (Cl₅ + Cl₆ + Cl₇) congeners, suggesting a spreading tendency seaward and southward from the CRD and the north of the inner shelf. The significant relationship between ΣSCCPs and total organic carbons (TOC) (r(2) = 0.402, p < 0.05) indicated that the migration of SCCPs in sediments was markedly affected by TOC. The spatial patterns of the TOM proxies of TOC δ(13)C, the contents of ΣC₂₇ + C₂₉ + C₃₁ n-alkanes, terrestrial marine biomarker ratio (TMBR), and terrestrial TOC (T-TOC) were all similar to that of ΣSCCPs. Linear relationships between SCCP contents and both the contents of ΣC₂₇ + C₂₉ + C₃₁ n-alkanes (r(2) = 0.537, p < 0.05) and T-TOC (r(2) = 0.495, p < 0.05) were also observed. The consistence demonstrated that a major portion of sedimentary SCCPs in the coastal ECS should be from the river input of Changjiang River and deposited in the CRD and along the inner shelf of the ECS, but only a minor fraction was transported to the offshore areas.

  • ISIS 449884 Injection Add-On to Metformin in Patients with Type 2 Diabetes: A Randomized, Double-Blind, Placebo-Controlled, Phase II Clinical Study

    • Introduction: ISIS 449884, a 2 '-O-methoxyethyl antisense oligonucleotide that targets the glucagon receptor (GCGR), has demonstrated an ability to reduce hepatic glucose output and lower the blood glucose level. The primary objective of this study was to investigate the safety and efficacy of ISIS 449884 as an add-on to metformin in a population of Chinese patients with type 2 diabetes mellitus (T2DM). Method: This was a multicenter, placebo-controlled (2:1), randomized, double-blind, parallel-enrollment, multiple-dose phase II study in Chinese patients with T2DM. A total of 90 patients who were uncontrolled by stable metformin monotherapy were randomized into three cohorts. Thirty subjects were enrolled in each cohort and received injections of ISIS 449884 (50 mg or 60 mg weekly or 100 mg every other week) or a corresponding volume of placebo (0.25 mL and 0.3 mL weekly or 0.5 mL every other week) subcutaneously in a 2:1 ratio for 16 weeks. Results: The primary efficacy endpoint was analyzed in 88 subjects (ISIS 449884, n = 59; placebo, n = 29). The corrected LS mean change from baseline in glycated hemoglobin (HbA1c) at week 17 in the pooled ISIS 449884 treatment group was - 1.31% (95% CI - 1.66%, - 0.96%), and that in the pooled placebo group was 0.15% (95% CI - 0.37%, 0.66%). The LS mean difference between the two groups was - 1.46% (95% CI - 1.92%, - 1.00%, P < 0.001). Treatment-emergent adverse events (TEAEs) occurred in 53/60 subjects (88.3%) and 25/30 subjects (83.3%) in the pooled ISIS 449884 treatment group and the pooled placebo group, respectively, with similar incidences. Drug-related TEAEs occurred in 41/60 subjects (68.3%) and 9/30 subjects (30.0%), respectively. TEAEs of grade 3 or higher occurred in 5/60 (8.3%) subjects and 2/30 (6.7%) subjects, respectively, and none of them were drug related. Conclusions: The ISIS 449884 injection add-on to metformin significantly reduced HbA1c in patients with T2DM uncontrolled by stable metformin monotherapy and showed an acceptable benefit/risk profile.

  • Associations of the glycaemic index and the glycaemic load with risk of type 2 diabetes in 127594 people from 20 countries (PURE): a prospective cohort study

    • Background: The association between the glycaemic index and the glycaemic load with type 2 diabetes incidence is controversial. We aimed to evaluate this association in an international cohort with diverse glycaemic index and glycaemic load diets. Methods: The PURE study is a prospective cohort study of 127 594 adults aged 35–70 years from 20 high-income, middle-income, and low-income countries. Diet was assessed at baseline using country-specific validated food frequency questionnaires. The glycaemic index and the glycaemic load were estimated on the basis of the intake of seven categories of carbohydrate-containing foods. Participants were categorised into quintiles of glycaemic index and glycaemic load. The primary outcome was incident type 2 diabetes. Multivariable Cox Frailty models with random intercepts for study centre were used to calculate hazard ratios (HRs). Findings: During a median follow-up of 11·8 years (IQR 9·0–13·0), 7326 (5·7%) incident cases of type 2 diabetes occurred. In multivariable adjusted analyses, a diet with a higher glycaemic index was significantly associated with a higher risk of diabetes (quintile 5 vs quintile 1; HR 1·15 [95% CI 1·03–1·29]). Participants in the highest quintile of the glycaemic load had a higher risk of incident type 2 diabetes compared with those in the lowest quintile (HR 1·21, 95% CI 1·06–1·37). The glycaemic index was more strongly associated with diabetes among individuals with a higher BMI (quintile 5 vs quintile 1; HR 1·23 [95% CI 1·08–1·41]) than those with a lower BMI (quintile 5 vs quintile 1; 1·10 [0·87–1·39]; p interaction=0·030). Interpretation: Diets with a high glycaemic index and a high glycaemic load were associated with a higher risk of incident type 2 diabetes in a multinational cohort spanning five continents. Our findings suggest that consuming low glycaemic index and low glycaemic load diets might prevent the development of type 2 diabetes.   • Genome-wide association study of survival in patients with pancreatic adenocarcinoma

    • BACKGROUND AND OBJECTIVE: Survival of patients with pancreatic adenocarcinoma is limited and few prognostic factors are known. We conducted a two-stage genome-wide association study (GWAS) to identify germline variants associated with survival in patients with pancreatic adenocarcinoma. METHODS: We analysed overall survival in relation to single nucleotide polymorphisms (SNPs) among 1005 patients from two large GWAS datasets, PanScan I and ChinaPC. Cox proportional hazards regression was used in an additive genetic model with adjustment for age, sex, clinical stage and the top four principal components of population stratification. The first stage included 642 cases of European ancestry (PanScan), from which the top SNPs (p≤10(-5)) were advanced to a joint analysis with 363 additional patients from China (ChinaPC). RESULTS: In the first stage of cases of European descent, the top-ranked loci were at chromosomes 11p15.4, 18p11.21 and 1p36.13, tagged by rs12362504 (p=1.63×10(-7)), rs981621 (p=1.65×10(-7)) and rs16861827 (p=3.75×10(-7)), respectively. 131 SNPs with p≤10(-5) were advanced to a joint analysis with cases from the ChinaPC study. In the joint analysis, the top-ranked SNP was rs10500715 (minor allele frequency, 0.37; p=1.72×10(-7)) on chromosome 11p15.4, which is intronic to the SET binding factor 2 (SBF2) gene. The HR (95% CI) for death was 0.74 (0.66 to 0.84) in PanScan I, 0.79 (0.65 to 0.97) in ChinaPC and 0.76 (0.68 to 0.84) in the joint analysis. CONCLUSIONS: Germline genetic variation in the SBF2 locus was associated with overall survival in patients with pancreatic adenocarcinoma of European and Asian ancestry. This association should be investigated in additional large patient cohorts.

    • A novel method for selective determination of Cr(III) and Cr(VI) in environmental water samples was developed based on target-induced fluorescence quenching of glutathione-stabilized gold nanoclusters (GSH-Au NCs). Fluorescent GSH-Au NCs were synthesized by a one-step approach employing GSH as reducing/protecting reagent. It was found that Cr(III) and Cr(VI) showed pH-dependent fluorescence quenching capabilities for GSH-Au NCs, and thus selective determination of Cr(III) and Cr(VI) could be achieved at different pHs. Addition of EDTA was able to effectively eliminate the interferences from other metal ions, leading to a good selectivity for this method. Under optimized conditions, Cr(III) showed a linear range of 25-3800 μg L(-1) and a limit of detection (LOD) of 2.5 μg L(-1). The Cr(VI) ion demonstrated a linear range of 5-500 μg L(-1) and LOD of 0.5 μg L(-1). The run-to-run relative standard deviations (n=5) for Cr(III) and Cr(VI) were 3.9% and 2.8%, respectively. The recoveries of Cr(III) and Cr(VI) in environmental water samples were also satisfactory (76.3-116%). This method, with its simplicity, low cost, high selectivity and sensitivity, could be used as a promising tool for chromium analysis in environmental water samples.
