    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Visual Object Tracking challenge VOT2021 is the ninth annual tracker benchmarking activity organized by the VOT initiative. Results of 71 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in recent years. The VOT2021 challenge was composed of four sub-challenges focusing on different tracking domains: (i) VOT-ST2021 challenge focused on short-term tracking in RGB, (ii) VOT-RT2021 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2021 focused on long-term tracking, namely coping with target disappearance and reappearance and (iv) VOT-RGBD2021 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2021 dataset was refreshed, while VOT-RGBD2021 introduces a training dataset and sequestered dataset for winner identification. The source code for most of the trackers, the datasets, the evaluation kit and the results along with the source code for most trackers are publicly available at the challenge website(1).

  • Investigation and optimization on melting performance of a triplex-tube heat storage tank by rotational mechanism

    • Phase change heat storage is the backbone of energy storage technology, but its storage time is affected by the low thermal conductivity of phase change materials. Therefore, the melting performance of a triplex-tube latent heat thermal energy storage unit (T-LHTESU) in a phase change heat storage system is studied in this paper, and the rotation mechanism is applied to the unit. Firstly, a numerical model of the T-LHTESU considering the rotation mechanism is constructed, and the validity of the rotation unit is verified by comparison with experimental data. In this unit, N-eicosane is used as a phase change material for heat exchange. The effects of different rotational speeds on the liquid phase distribution, temperature distribution, flow velocity distribution, total energy storage, and energy storage efficiency of the T-LHTESU are studied. The results show that the melting time of this unit at 0.1 and 1 rpm is 46.98 and 69.35% lower than that of the stationary model, respectively. The total amount of stored heat is decreased by 0.67 and 2.17%, and the heat storage efficiency is increased by 87.34% and 219.19%, respectively. This indicates that the addition of the rotation mechanism greatly increases the heat storage efficiency of the T-LHTESU and reduces its total melting time, while the reduction of the total energy stored in the melting cycle is small. Then it is proved that rotation improves the single heat transfer mechanism of the stationary model and eliminates the thermal deposition caused by natural convection by studying the internal temperature/velocity response of the stationary model and the speed of 0.1 rpm. The related geometric structure of the model is optimized by response surface optimization design based on 0.1 rpm rotation speed. The influence of each variable on the target response is obtained, and compared with the original model, the melting time of the optimized model is reduced by 12.24%. Finally, based on the geometric optimization design, the influence of element physical factors (temperature and material of fin/tube wall) on the related melting properties is studied. This study is helpful to promote the effective use of rotation mechanism in phase change heat storage systems and has a certain guiding role in the structural design.

  • Association of Ultra-Processed Food Intake with Cardiovascular And Respiratory Disease Multimorbidity : A Prospective Cohort Study

    • SCOPE: Evidence suggests a positive association between ultra-processed food (UPF) consumption and the incidence of cardiovascular disease (CVD). We aimed to investigate associations between UPF intake and respiratory disease, CVD, and their multimorbidity in a large prospective cohort.METHODS AND RESULTS: Within the UK Biobank, participants who were free from respiratory disease or CVD at baseline and completed at least two times 24-h dietary records were included in this study. After adjusting for socioeconomic status and lifestyle factors, the hazard ratios (95% confidence interval) for each ten percent increase in UPF were 1.06 (1.04, 1.09) for CVD, 1.04 (1.02, 1.06) for respiratory disease, 1.15 (1.08, 1.22) for CVD mortality, and 1.06 (1.01, 1.12) for their multimorbidity, respectively. In addition, replacing 20% of UPF weight in diet with an equivalent proportion of unprocessed or minimally processed foods was estimated to be associated with 11% lower risk of CVD, 7% lower risk of respiratory disease, 25% lower risk of CVD mortality and 11% lower risk of CVD and respiratory disease multimorbidity.CONCLUSION: In this prospective cohort study, higher consumption of UPF was associated with higher risks of CVD and respiratory disease multimorbidity.     • Discrete Gaussian sampling is one of the important components in lattice-based cryptosystems which are promising candidates for post-quantum cryptographic algorithms. For sufficient security and satisfactory performance, the Knuth-Yao algorithm is an efficient way to implement discrete Gaussian samplers. Nevertheless, most polynomials in lattice-based cryptography have 256 coefficients or more, which suffers from long latency to complete the sample generation. In this paper, the first parallel discrete Gaussian sampler with hierarchical structure is proposed, while keeping statistical distance to the actual distribution. Based on the imbalanced visiting frequency of the probability matrix, a three-stage generation strategy is adopted with hierarchical bit search units (BSUs) that can greatly reduce area consumption of the repeated costly lookup tables. Besides the architecture improvement, a lowest-set-bit scanning scheme is introduced to BSUs. Moreover, the parallelism of our design provides obfuscation ability against side-channel attacks (SCAs). A practical hardware implementation of discrete Gaussian distributions with sigma = 3.33 on the Xilinx Virtex-5 XC5VLX30 FPGA device spends 26.12 ns on average to generate 256 samples, consuming 994 slices. Results have verified its advantages of area efficiency over the state-of-the-arts (SOAs).

  • Optimization of melting performance of a heat storage tank under rotation conditions : Based on taguchi design and response surface method

    • The melting performance of a rotating triplex-tube latent heat thermal energy storage unit is studied by numerical simulation method. The Taguchi design and response surface method are applied to its optimized design. Firstly, the Taguchi design method is used to quantitatively reveal the specific effects of fin distribution, fin number, and fin material on the melting performance of the unit. Compared with all-inner tube-fin or all-outer tube-fin structures, the melting time of alternating inside and outside fin structures is shortened by 52.64% and 32.42%, the average heat absorption rate is greatly increased by 105.56% and 47.26%, and the total heat is reduced by 2.64% and 2.17%, respectively. Then, the response surface method is applied to the eight alternating fins obtained by the Taguchi design, and the effects of fin length, width, and rotation angle on the melting time and average heat absorption rate of the unit are studied. Compared with the original structure, the optimal structure reduces the total melting time by 7.37% and increases the average heat absorption rate by 7.23%. The geometric parameters’ interaction with the relevant target response is studied, and the corresponding fluid-structure interaction equation is fitted. Finally, the melt growth phenomenon near the wall is found in the initial melting stage of the optimized model by mechanism analysis.

  • Epigenetic–smoking interaction reveals histologically heterogeneous effects of TRIM27 DNA methylation on overall survival among early-stage NSCLC patients

    • Tripartite motif containing 27 (TRIM27) is highly expressed in lung cancer, including non-small-cell lung cancer (NSCLC). Here, we profiled DNA methylation of lung adenocarcinoma (LUAD) and lung squamous cell carcinoma (LUSC) tumours from 613 early-stage NSCLC patients and evaluated associations between CpG methylation of TRIM27 and overall survival. Significant CpG probes were confirmed in 617 samples from The Cancer Genome Atlas. The methylation of the CpG probe cg05293407TRIM27 was significantly associated with overall survival in patients with LUSC (HR = 1.65, 95% CI: 1.30–2.09, P = 4.52 × 10−5), but not in patients with LUAD (HR = 1.08, 95% CI: 0.87–1.33, P = 0.493). As incidence of LUSC is associated with higher smoking intensity compared to LUAD, we investigated whether smoking intensity impacted on the prognostic effect of cg05293407TRIM27 methylation in NSCLC. LUSC patients had a higher average pack-year of smoking (37.49LUAD vs 54.79LUSC, P = 1.03 × 10−19) and included a higher proportion of current smokers than LUAD patients (28.24%LUAD vs 34.09%LUSC, P = 0.037). cg05293407TRIM27 was significantly associated with overall survival only in NSCLC patients with medium–high pack-year of smoking (HR = 1.58, 95% CI: 1.26–1.96, P = 5.25 × 10−5). We conclude that cg05293407TRIM27 methylation is a potential predictor of LUSC prognosis, and smoking intensity may impact on its prognostic value across the various types of NSCLC.

    • The interaction between DNA methylation of tripartite motif containing 27 (cg05293407TRIM27) and smoking has previously been identified to reveal histologically heterogeneous effects of TRIM27 DNA methylation on early-stage non-small-cell lung cancer (NSCLC) survival. However, to understand the complex mechanisms underlying NSCLC progression, we searched three-way interactions. A two-phase study was adopted to identify three-way interactions in the form of pack-year of smoking (number of cigarettes smoked per day × number of years smoked) × cg05293407TRIM27 × epigenome-wide DNA methylation CpG probe. Two CpG probes were identified with FDR-q ≤ 0.05 in the discovery phase and P ≤ 0.05 in the validation phase: cg00060500KIAA0226 and cg17479956EXT2. Compared to a prediction model with only clinical information, the model added 42 significant three-way interactions using a looser criterion (discovery: FDR-q ≤ 0.10, validation: P ≤ 0.05) had substantially improved the area under the receiver operating characteristic curve (AUC) of the prognostic prediction model for both 3-year and 5-year survival. Our research identified the complex interaction effects among multiple environment and epigenetic factors, and provided therapeutic target for NSCLC patients.

  • Application and analysis of flip mechanism in the melting process of a triplex-tube latent heat energy storage unit

    • In order to improve the characteristics of uneven melting in the melting process of the horizontal latent heat energy storage system, the triplex-tube latent heat energy storage unit is taken as the research object, and the flip mechanism is applied to its melting process. Numerical simulation is used for the research, and the numerical model is verified by experimental data. The results show that under different dimensionless times, the melting performance of the unit can be significantly improved by a single flip. When the dimensionless time is 0.4576, it is found that the total melting time of the unit is reduced by 16.17 %, the average heat absorption rate is increased by 14.7 %, but the total heat energy absorption is reduced by 3.85 %. The results show that the addition of a flip can effectively shorten the melting time and increase the heat absorption rate, but it has a negative effect on the total heat absorption in one melting cycle. Moreover, through the comparison of dynamic flow rate, dynamic temperature response, and temperature interval, it is shown that the addition of flip effectively reduces the negative influence of the hard-to-melt zone on the melting performance of the unit during the melting process. The flip mechanism reduces the proportion of high-temperature phase change material in the melting process and makes the melting process more uniform.

  • Association of Ultraprocessed Food Consumption With Risk of Dementia : A Prospective Cohort

    • BACKGROUND: There has been a growing body of evidence associating consumption of ultra-processed foods (UPF) with adverse health outcomes including depression, cardiovascular disease, all-cause mortality. However, whether UPF are associated with dementia is unknown. The authors investigated the associations between UPF and dementia incidence in UK biobank.METHODS: We included 72,083 participants (55 years or older) who were free from dementia at baseline and provided at least two times 24-h dietary assessments from the UK Biobank study. Follow-up occurred through March 2021. UPF were defined according to the NOVA classification. Incident all-cause dementia comprising Alzheimer's disease and vascular dementia was ascertained through electronic linkages to hospital and mortality records. Cox proportional hazards were used to estimate the association between the proportion (%) of UPF in the diet and the subsequent risk of dementia. In addition, substitution analysis was used to estimate the risk of dementia when substituting UPF with an equivalent proportion of unprocessed or minimally processed foods.RESULTS: During a total of 717,333 person-years of follow-up (median 10.0 years), 518 participants developed dementia, of which 287 developed Alzheimer's disease and 119 developed vascular dementia. In the fully adjusted model, consumption of UPF was associated with higher risk of dementia (hazard ratio (HR) for 10% increase in UPF: 1.25; 95% confidence interval (CI): 1.14, 1.37), Alzheimer's disease (HR: 1.14; 95% CI: 1.00, 1.30) and vascular dementia (HR: 1.28; 95% CI: 1.06, 1.55), respectively. In addition, replacing 10% of UPF weight in diet with an equivalent proportion of unprocessed or minimally processed foods was estimated to be associated with a 19% lower risk of dementia (HR: 0.81; 95% CI: 0.74, 0.89).CONCLUSIONS: In this prospective cohort study, higher consumption of UPF was associated with higher risk of dementia, while substituting unpr2ocessed or minimally processed foods for UPF was associated lower risk of dementia.
