    • © 2018 Aquatic ecosystem sustainability around the globe is facing crucial challenges because of increasing anthropogenic and natural disturbances. In this study, the Tianchi Lake, a typical cold-water lake and a UNESCO/MAB (Man and Biosphere) nature reserve located in high latitude and elevation with the relatively low intensity of human activity was chosen as a system to examine the linkages between climate change and eutrophication. As a part of the UNESCO Bogda Man and Biosphere Reserve, Tianchi Lake has been well preserved for prevention from human intervention, but why has it been infected with eutrophication recent years? Our results show that climate change played a significant role in the eutrophication in the Tianchi Lake. Increased temperature, changed precipitation pattern and wind-induced hydrodynamic fluctuations in the summer season were suggested to make a major contribution to the accelerated eutrophication. The results also showed that the local temperature and precipitation changes were closely linked to the large-scale atmospheric circulation, which opens the door for the method to be applied in other regions without local climatic information. This study suggests that there is an urgent need to take into consideration of climate change adaptation into the conservation and management of cold-water lakes globally.

    • The complexity of economic development and humanitarian crises means that energy science and technology should be involved in actions that address almost every major challenges of ecosystem health and sustainability. Energy is the engine of the world economy and the key to ecosystems' functioning, which also has a great impact on global warming. The energy crisis, environmental pollution, overuse of natural resources, water supply shortages, global climate disruption, and deteriorating ecosystems are major challenges to address in order to achieve the United Nations Sustainable Development Goals (SDGs). In light of the frontiers in energy sciences and disruptive innovation in eco-tech, we recognize the need to review and establish working mechanisms that identify and examine issues that are critical to future sustainable development, to offer advice to decision-makers in different social sectors (public and private), to secure a shared future for mankind, and to achieve shared prosperity and common interests through international communications and collaborations.

  • The role of transnational dimensions in emerging economy 'Technological Innovation Systems' for clean-tech

    • The 'Technological Innovation System' (TIS) framework and its system functions have become a popular analytical tool for the study of clean-tech innovation. There is increasing attention for the role of emerging economies in global clean-tech innovation, but the applicability of TIS to emerging economies cases is not entirely straightforward. A key issue is the limited geographical considerations, in particular transnational dimensions in TIS, whereas earlier perspectives on innovation in emerging economies have stressed the role of such transnational dimensions. This paper elaborates transnational TIS actor-networks and institutions, categorizes these in relation to TIS functions, and describes their potential to induce or block TIS development in emerging economies. We draw on insights from the perspectives of National Learning Systems, International Technology Transfer, and Global Production Networks for this purpose. We conclude that the potential effects of these transnational dimensions may be accurately grasped by the existing list of system functions, lending credence to its further application of the TIS framework on emerging economy case studies. Policy makers in emerging economies should recognize these transnational dimensions and seek to optimize their potential effect on domestic TIS development, taking in to consideration a realistic assessment of its role in the global TIS.     • The epigenetic regulation of glioblastoma stem cell (GSC) function remains poorly understood. Here, the authors compare the chromatin accessibility landscape of GSC cultures from mice and patients and suggest that the epigenome of GSCs is cell lineage-regulated and could predict patient survival. There is ample support for developmental regulation of glioblastoma stem cells. To examine how cell lineage controls glioblastoma stem cell function, we present a cross-species epigenome analysis of mouse and human glioblastoma stem cells. We analyze and compare the chromatin-accessibility landscape of nine mouse glioblastoma stem cell cultures of three defined origins and 60 patient-derived glioblastoma stem cell cultures by assay for transposase-accessible chromatin using sequencing. This separates the mouse cultures according to cell of origin and identifies three human glioblastoma stem cell clusters that show overlapping characteristics with each of the mouse groups, and a distribution along an axis of proneural to mesenchymal phenotypes. The epigenetic-based human glioblastoma stem cell clusters display distinct functional properties and can separate patient survival. Cross-species analyses reveals conserved epigenetic regulation of mouse and human glioblastoma stem cells. We conclude that epigenetic control of glioblastoma stem cells primarily is dictated by developmental origin which impacts clinically relevant glioblastoma stem cell properties and patient survival.

    • Pollution and climate change are among the most challenging issues for countries with developing economies, but we know little about the ecological risks that result when these pressures occur together. We explored direct effects of, and interactions between, environmental pollution and climate change on ecosystem health in the Bohai Sea region of Northern China. We developed an integrated approach to assess ecological risks to this region under four scenarios of climate change. Although ecological risks to the system from pollution alone have been declining, interactions between pollution and climate change have enhanced ecological risks to this coastal/marine ecosystem. Our results suggest that current policies focused strictly on pollution control alone should be changed to take into account the interactive effects of climate change so as to better forecast and manage potential ecological risks.

  • Paired glioblastoma cell cultures of the fluorescent bulk tumor and non-fluorescent tumor margin display differential phenotypes and cell states across patients

    • Glioblastoma is an aggressive and therapy-resistant primary brain tumor with a dismal prognosis. The inevitable recurrence is in almost all patients in contact with the resection cavity, suggesting the local peritumoral area as its origin. Glioblastoma cells of this region have seldom been studied and few authenticated models exist. We have explanted matched tissue samples from the bulk tumor and local tumor edge of 13 glioblastoma patients of which 7 were sustainable beyond passage 6. Each edge culture was more invasive and less self-renewing and tumorigenic compared to its paired bulk culture. Three pairs of edge and bulk cultures were profiled with a combined single nucleus (sn) RNA- and ATAC-sequencing. Transcriptome analysis displayed for all patients a shift towards AC-MES cell states in the edge cultures. Chromatin-accessibility profiles uncovered differential regulatory networks with edge cells being enriched for transcription factor (TF) motifs of invasion, neurons, and immune cells. We propose that edge cells have been epigenetically reprogrammed by their unique interactions with various cell types in the peritumoral region. The fact that glioblastoma edge cells display distinct epigenetic regulation compared to their bulk tumor cells has implications for therapy development that should be targeted to and tested on the relapse-causing glioblastoma edge cells.

  • Epigenetic Insights into GABAergic development in Dravet Syndrome iPSC and Therapeutic Implications

    • Dravet syndrome (DS) is a devastating early onset refractory epilepsy syndrome caused by variants in the SCN1A gene. A disturbed GABAergic interneuron function is implicated in the progression to DS but the underlying developmental and pathophysiological mechanisms remain elusive, in particularly at the chromatin level. In this study, we utilized induced pluripotent stem cells (iPSCs) derived from DS cases and healthy donors to model disease-associated epigenetic abnormalities of GABAergic development. Employing the ATAC-seq technique, we assessed chromatin accessibility at multiple time points (Day 0, Day 19, Day 35, and Day 65) of GABAergic differentiation. Additionally, we elucidated the effects of the commonly used anti-seizure drug valproic acid (VPA) on chromatin accessibility in GABAergic cells. The distinct dynamics in chromatin profile of DS iPSC predicted accelerated early GABAergic development, evident at D19, and diverged further from the pattern in control iPSC with continued differentiation, indicating a disrupted GABAergic maturation. Exposure to VPA at D65 reshaped the chromatin landscape at a variable extent in different iPSC-lines and rescued the observed dysfunctional development in some DS iPSC-GABA. This study provides the first comprehensive investigation on the chromatin landscape of GABAergic differentiation in DS-patient iPSC, offering valuable insights into the epigenetic dysregulations associated with interneuronal dysfunction in DS. Moreover, our detailed analysis of the chromatin changes induced by VPA in iPSC-GABA holds the potential to improve development of personalized and targeted anti-epileptic therapies.

  • Drivers of change in China’s energy-related CO2 emissions

    • CO2 emissions are of global concern because of climate change. China has become the largest CO2 emitter in the world and presently accounts for 30% of global emissions. Here, we analyze the major drivers of energy-related CO2 emissions in China from 1978 when the reform and opening-up policy was launched. We find that 1) there has been a 6-fold increase in energy-related CO2 emissions, which was driven primarily (176%) by economic growth followed by population growth (16%), while the effects of energy intensity (−79%) and carbon intensity (−13%) slowed the growth of carbon emissions over most of this period; 2) energy-related CO2 emissions are positively related to per capita gross domestic product (GDP), population growth rate, carbon intensity, and energy intensity; and 3) a portfolio of command-and-control policies affecting the drivers has altered the total emission trend. However, given the major role of China in global climate change mitigation, significant future reductions in China’s CO2 emissions will require transformation toward low-carbon energy systems.
