  • Plasma p-tau181 as an outcome and predictor of multidomain intervention effects: a secondary analysis of a randomised, controlled, dementia prevention trial

    • Background: It is unknown whether multidomain interventions, which might preserve late-life cognition, affect Alzheimer's disease pathology. Previous studies measured cerebrospinal fluid and imaging Alzheimer's disease biomarkers in small subsamples of multidomain trial participants. Newly developed assays enable the measurement of blood-based Alzheimer's disease biomarkers in larger samples. We aimed to assess whether plasma tau phosphorylated at threonine 181 (p-tau181) was able to detect or predict 3-year multidomain intervention effects. Methods: This is a secondary analysis of the randomised, controlled, Multidomain Alzheimer Prevention Trial (MAPT) testing a 3-year multidomain intervention, omega-3 fatty acid supplementation, or both versus placebo, in individuals aged 70 years and older in 13 memory centres in France and Monaco. Plasma p-tau181 was measured in stored blood samples in a subsample of 527 participants on an intention-to-treat basis. Changes in cognitive score were calculated as a composite measure using the average of Z scores for the following tests: Mini Mental State Examination orientation items, Free and Cued Selective Reminding Test (sum of free and total recall scores), category fluency, and Digit Symbol Substitution Test. Intervention effects on 3-year change in p-tau181 concentration were estimated by use of a linear mixed model with centre-specific random intercepts. Findings: Recruitment took place between May 30, 2008, and Feb 24, 2011. Median baseline plasma p-tau181 was 8·8 pg/mL (IQR 6·7–11·9) in the total sample, and significantly higher in older individuals, men, APOE ε4 carriers, and participants with renal dysfunction or a positive PET amyloid scan. During 3-year follow-up, individuals with raised baseline p-tau181 underwent greater cognitive decline (eg, mean difference in 3-year change on the composite cognitive score between control group participants with normal and abnormal baseline levels of p-tau was −0·34 [effect size −0·52; 95% CI −0·61 to 0·07] in the fully adjusted model using a 12·4 pg/mL cutoff for abnormal baseline p-tau181), but there were no intervention effects on change in p-tau181 either in this subgroup or the total population, and no effect on cognitive change in individuals with raised baseline p-tau181 (eg, in the fully adjusted model using the 12·4 pg/mL cutoff for p-tau181 abnormality, the mean difference [95% CI] in this subgroup in 3-year decline on the composite cognitive score between the control group and the multidomain + omega-3 group, the omega-3 group, and the multidomain intervention group, was, respectively: 0·13 [−0·21 to 0·47], 0·03 [−0·30 to 0·36], and 0·10 [−0·26 to 0·46]). Surprisingly, individuals with raised baseline p-tau181 showed a decrease in p-tau181 during follow-up (eg, unadjusted mean [95% CI] 3-year change was −3·01 pg/mL (−4·45 to −1·56) in control group subjects with abnormal baseline p-tau181 [using the 12·4 pg/mL abnormal p-tau cutoff]). Interpretation: Our results support the utility of p-tau181 as a prognostic biomarker, but it did not predict or detect intervention effects in this study. Further investigation of its usefulness as a prevention trial outcome measure is required.

    • We report time-resolved, high-resolution optical spectropolarimetric observations of the young double-lined spectroscopic binary V1878 Ori. Our observations were collected with the ESPaDOnS spectropolarimeter at the Canada-France-Hawaii Telescope through the BinaMIcS large programme. V1878 Ori A and B are partially convective intermediate mass weak-line T Tauri stars on an eccentric and asynchronous orbit. We also acquired X-ray observations at periastron and outside periastron. Using the least-squares deconvolution technique (LSD) to combine information from many spectral lines, we clearly detected circular polarization signals in both components throughout the orbit. We refined the orbital solution for the system and obtained disentangled spectra for the primary and secondary components. The disentangled spectra were then employed to determine atmospheric parameters of the two components using spectrum synthesis. Applying our Zeeman Doppler imaging code to composite Stokes IV LSD profiles, we reconstructed brightness maps and the global magnetic field topologies of the two components. We find that V1878 Ori A and B have strikingly different global magnetic field topologies and mean field strengths. The global magnetic field of the primary is predominantly poloidal and non-axisymmetric (with a mean field strength of 180 G). While the secondary has a mostly toroidal and axisymmetric global field (mean strength of 310 G). These findings confirm that stars with very similar parameters can exhibit radically different global magnetic field characteristics. The analysis of the X-ray data shows no sign of enhanced activity at periastron, suggesting the lack of strong magnetospheric interaction at this epoch.

  • Constraints on Magnetic Braking from the G8 Dwarf Stars 61 UMa and tau Cet

    • During the first half of their main-sequence lifetimes, stars rapidly lose angular momentum to their magnetized winds, a process known as magnetic braking. Recent observations suggest a substantial decrease in the magnetic braking efficiency when stars reach a critical value of the Rossby number, the stellar rotation period normalized by the convective overturn timescale. Cooler stars have deeper convection zones with longer overturn times, reaching this critical Rossby number at slower rotation rates. The nature and timing of the transition to weakened magnetic braking have previously been constrained by several solar analogs and two slightly hotter stars. In this Letter, we derive the first direct constraints from stars cooler than the Sun. We present new spectropolarimetry of the old G8 dwarf tau Cet from the Large Binocular Telescope, and we reanalyze a published Zeeman Doppler image of the younger G8 star 61 UMa, yielding the large-scale magnetic field strengths and morphologies. We estimate mass-loss rates using archival X-ray observations and inferences from Ly alpha measurements, and we adopt other stellar properties from asteroseismology and spectral energy distribution fitting. The resulting calculations of the wind braking torque demonstrate that the rate of angular momentum loss drops by a factor of 300 between the ages of these two stars (1.4-9 Gyr), well above theoretical expectations. We summarize the available data to help constrain the value of the critical Rossby number, and we identify a new signature of the long-period detection edge in recent measurements from the Kepler mission.

    • The ability to process optical signals without passing into the electrical domain has always attracted the attention of the research community. Processing photons by photons unfolds new scenarios, in principle allowing for unseen signal processing and computing capabilities. Optical computation can be seen as a large scientific field in which researchers operate, trying to find solutions to their specific needs by different approaches; although the challenges can be substantially different, they are typically addressed using knowledge and technological platforms that are shared across the whole field. This significant know-how can also benefit other scientific communities, providing lateral solutions to their problems, as well as leading to novel applications. The aim of this Roadmap is to provide a broad view of the state-of-the-art in this lively scientific research field and to discuss the advances required to tackle emerging challenges, thanks to contributions authored by experts affiliated to both academic institutions and high-tech industries. The Roadmap is organized so as to put side by side contributions on different aspects of optical processing, aiming to enhance the cross-contamination of ideas between scientists working in three different fields of photonics: optical gates and logical units, high bit-rate signal processing and optical quantum computing. The ultimate intent of this paper is to provide guidance for young scientists as well as providing research-funding institutions and stake holders with a comprehensive overview of perspectives and opportunities offered by this research field.

  • Accretion and magnetism on young eccentric binaries : DQ Tau and AK Sco

    • The accretion and ejection of mass in pre-main-sequence (PMS) stars are key processes in stellar evolution as they shape the stellar angular momentum transport necessary for the stars' stability. Magnetospheric accretion on to classical T Tauri stars and low-mass PMS stars has been widely studied in the single-star case. This process cannot be directly transferred to PMS binary systems, as tidal and gravitation effects, and/or accretion from a circumbinary disc (with variable separation of the components in the case of eccentric orbits) are in place. This work examines the accretion process of two PMS eccentric binaries, DQ Tau and AK Sco, using high-resolution spectropolarimetric time series. We investigate how magnetospheric accretion can be applied to these systems by studying the accretion-related emission lines and the magnetic field of each system. We discover that both systems are showing signs of magnetospheric accretion, despite their slightly different configurations, and the weak magnetic field of AK Sco. Furthermore, the magnetic topology of DQ Tau A shows a change relative to the previous orbital cycle studied: previously dominated by the poloidal component, it is now dominated by the toroidal component. We also report an increase of the component's accretion and the absence of an accretion burst at the apastron, suggesting that the component's magnetic variation might be the cause of the inter-cycle variations of the system's accretion. We conclude on the presence of magnetospheric accretion for both systems, together with gravitational effects, especially for AK Sco, composed of more massive components.
