    • Reproducible climate reconstructions of the Common Era (1 CE to present) are key to placing industrial-era warming into the context of natural climatic variability. Here we present a community-sourced database of temperature-sensitive proxy records from the PAGES2k initiative. The database gathers 692 records from 648 locations, including all continental regions and major ocean basins. The records are from trees, ice, sediment, corals, speleothems, documentary evidence, and other archives. They range in length from 50 to 2000 years, with a median of 547 years, while temporal resolution ranges from biweekly to centennial. Nearly half of the proxy time series are significantly correlated with HadCRUT4.2 surface temperature over the period 1850-2014. Global temperature composites show a remarkable degree of coherence between high-and low-resolution archives, with broadly similar patterns across archive types, terrestrial versus marine locations, and screening criteria. The database is suited to investigations of global and regional temperature variability over the Common Era, and is shared in the Linked Paleo Data (LiPD) format, including serializations in Matlab, R and Python. Since the pioneering work of D'Arrigo and Jacoby1-3, as well as Mann et al. 4,5, temperature reconstructions of the Common Era have become a key component of climate assessments6-9. Such reconstructions depend strongly on the composition of the underlying network of climate proxies10, and it is therefore critical for the climate community to have access to a community-vetted, quality-controlled database of temperature-sensitive records stored in a self-describing format. The Past Global Changes (PAGES) 2k consortium, a self-organized, international group of experts, recently assembled such a database, and used it to reconstruct surface temperature over continental-scale regions11 (hereafter, ` PAGES2k-2013'). This data descriptor presents version 2.0.0 of the PAGES2k proxy temperature database (Data Citation 1). It augments the PAGES2k-2013 collection of terrestrial records with marine records assembled by the Ocean2k working group at centennial12 and annual13 time scales. In addition to these previously published data compilations, this version includes substantially more records, extensive new metadata, and validation. Furthermore, the selection criteria for records included in this version are applied more uniformly and transparently across regions, resulting in a more cohesive data product. This data descriptor describes the contents of the database, the criteria for inclusion, and quantifies the relation of each record with instrumental temperature. In addition, the paleotemperature time series are summarized as composites to highlight the most salient decadal-to centennial-scale behaviour of the dataset and check mutual consistency between paleoclimate archives. We provide extensive Matlab code to probe the database-processing, filtering and aggregating it in various ways to investigate temperature variability over the Common Era. The unique approach to data stewardship and code-sharing employed here is designed to enable an unprecedented scale of investigation of the temperature history of the Common Era, by the scientific community and citizen-scientists alike.

    • A multifunctional hybrid gel based on cellulose nanofibrils (CNFs) was developed by grafting on its surface stearyl acrylate (PSA) and gadolinium(III) fluoride nanoparticles (GdF3 NPs) via Cu0-mediated surface-initiated radical polymerization (SET-LRP) while encapsulating antimicrobial peptides in it. GdF3 NPs were first surface-modified with 11-phosphonoundecyl acrylate (PDA) to participate in the SET-LRP and cross-linked the grafted polymer-modified CNF. Several characterizations of the hybrid material (GdF3-PSA-CNF) were carried out, such as Fourier transform infrared (FTIR) spectroscopy, thermogravimetric analysis (TGA), rheology, and microscopic analyses. The grafted PSA and cross-linked GdF3 NPs created sophisticated networks in the CNF-based gel, presenting outstanding rheological properties and promising three-dimensional (3D) printability of this hybrid material (GdF3-PSA-CNF). The nanostructures of GdF3 NPs and their incorporated CNF species were characterized via small-angle X-ray scattering (SAXS). In addition, due to the unique intrinsic property of the GdF3 nanoparticles, properties for magnetic resonance imaging (MRI) of GdF3-PSA-CNF were investigated, showing the potential application as a contrast agent. Finally, the encapsulation of the antimicrobial peptides added another function to the hybrid material, evaluated by an antimicrobial test against methicillin-resistant Staphylococcus aureus (MRSA) in vitro.

    • Analogue type millimetre wave phase shifter based on a dielectric rod waveguide with adjacent MEMS tuneable high-impedance surface is proposed. Applying bias voltage to the MEMS varactors of the high-impedance surface allow controlling its effective impedance and consequently the phase factor of the propagation constant inside the waveguide. The measured phase difference between the phase shifter with adjacent high-impedance surface and phase shifter with low impedance surface is up to 378°. The insertion loss of the high-impedance surface as phase shifting element at 80-90 GHz is 0.5-2.7 dB depending on the distance to the dielectric rod waveguide.

  • MEMS tunable metamaterials surfaces and their applications

    • Microelectromechanical systems (MEMS) are proposed as a technological solution for fabrication of metamaterials. This enables tunability of metamaterials effective properties and allows using metamaterials in wide range of applications. Low loss of the MEMS devices allows the metamaterials application to be extended to millimeter and submillimeter wave frequencies without compromising on performance. Electronic beam steering by MEMS tunable metamaterials at millimeter wavelength is considered and a prototype of a W band analog tunable phase shifter is demonstrated. The insertion loss of the fabricated MEMS tunable metamaterials surface varies from 0.7 dB to a maximum of 3.5 dB (at a resonance frequency). MEMS varactors have shown reliable and repeatable analog operation over 108 cycles.

    • The study aimed to evaluate the antibacterial efficacy of Lugol's solution 5% and Gentian violet 1% against methicillin-resistant Staphylococcus aureus (MRSA) biofilm in vivo. The bactericidal efficacy for treatment of MRSA-biofilm skin wound infection was tested in a murine model. Luciferase-tagged S. aureus Xen31, a MRSA-strain derived from S. aureus ATCC-3359130, was used for infection. Wounds were made in the skin of mice and infected with MRSA. The mice were treated with Lugol's solution and Gentian violet. Application of the antimicrobial agents started 24 hours post infection and was repeated daily for five-days. The antimicrobial effect on the biofilm bacteria was evaluated by measuring bioluminescence from MRSA daily for seven-days. Lugol's solution and Gentian violet showed a significant reduction in luminescent signals from the first assessment day to all subsequent days (P < .001). Lugol's solution and Gentian violet effectively eradicated MRSA in biofilm in vivo and could be alternatives or in addition to topical antibiotics when MRSA-biofilm wound infection is suspected.

  • FANTOM5 CAGE profiles of human and mouse samples

    • In the FANTOM5 project, transcription initiation events across the human and mouse genomes were mapped at a single base-pair resolution and their frequencies were monitored by CAGE (Cap Analysis of Gene Expression) coupled with single-molecule sequencing. Approximately three thousands of samples, consisting of a variety of primary cells, tissues, cell lines, and time series samples during cell activation and development, were subjected to a uniform pipeline of CAGE data production. The analysis pipeline started by measuring RNA extracts to assess their quality, and continued to CAGE library production by using a robotic or a manual workflow, single molecule sequencing, and computational processing to generate frequencies of transcription initiation. Resulting data represents the consequence of transcriptional regulation in each analyzed state of mammalian cells. Non-overlapping peaks over the CAGE profiles, approximately 200,000 and 150,000 peaks for the human and mouse genomes, were identified and annotated to provide precise location of known promoters as well as novel ones, and to quantify their activities.

    • Trematodes are widespread parasitic flatworms that significantly affect mankind either directly as human parasites, or indirectly via the infection of livestock and the related economic damage. The two most important trematode taxa are the blood flukes Schistosoma and the liver flukes Fasciola, but detection and differentiation of these parasites remains a challenge. Recently, microRNAs (miRNAs) were described from extracellular vesicles (EV) for both parasites secreted into respective hosts. These molecules have been proposed as mediators of parasite-host communication, and potential biomarkers for the detection of parasitic infections from host blood. Our aim here was to study similarities and differences in the miRNA complements of Schistosoma mansoni and Fasciola hepatica, EV-load in particular, to predict their targets and potential functions in the parasite-host interaction. We reanalyzed the known miRNA complements of S. mansoni and F. hepatica and found 16 and 4 previously overlooked, but deeply conserved miRNAs, respectively, further moving their complements closer together. We found distinct miRNA enrichment patterns in EVs both showing high levels of flatworm miRNAs with potential for the detection of an infection from blood. Two miRNAs of the protostome specific MIR-71 and MIR-277 families were highly expressed in EVs and could, therefore, have potential as biomarkers for trematode infection. Curiously, we identified nucleotide differences in the sequence of Mir-277-P2 between S. mansoni and F. hepatica that hold great promise for the distinction of both parasites. To test whether the EV-miRNAs of S. mansoni and F. hepatica could be modulating the expression of host genes, we predicted miRNA targets in 321 human and cattle messenger RNAs that overlapped between both hosts. Of several predicted targets, wnt signaling pathway genes stood out and their suppression likely leads to changes in the glucose concentration in host blood and the reduction of inflammatory and immune responses.
