  • Optical radiative properties of ablating polymers exposed to high-power arc plasmas

    • The radiative properties of polymers exposed to high-intensity radiation are of importance for the numerical simulation of arc-induced ablation. The paper investigates the optical properties of polymethylmethacrylate PMMA and polyamide PA6 films exposed to high-power arc plasmas, which can cause ablation of the material. A four-flux radiative approximation is first used to estimate absorption and scattering coefficients of the tested materials in the ultraviolet (UV) and in the visible (VIS) ranges from spectrophotometric measurements. The temperature-induced variation of the collimated transmissivity of the polymers is also measured from room temperature to the glass temperature of PMMA and the melting temperature of PA6. Furthermore, band-averaged absorption and scattering coefficients of non-ablating and ablating polymers are estimated from the UV to the short-wavelength infrared (SWIR), covering the range of interest for the simulation of arc-induced ablation. These estimates are obtained from collimated transmissivities measured with an additional in situ photometric system that uses a high-power, transient arc plasma to both illuminate the samples and to induce ablation. It is shown that the increase in the bulk temperature of PA6 leads to a strong reversible increase in collimated transmissivity, significantly reducing the absorption and scattering coefficients of the material. A weaker but opposite effect of temperature on the optical properties is found in PMMA. As a consequence, it is suggested that the absorption coefficient of polymers used for arc-induced ablation estimates should not be taken directly from direct collimated transmissivity measurements at room temperature. The band-averaged radiation measurements also show that the layer of products released by ablation of PMMA produces scattering radiation losses mainly in the VIS-SWIR ranges, which are only a small fraction of the total incident arc radiation. In a similar manner, the ablation layer of PA6 leads to weak absorption radiation losses, although mainly in the UV range.

    • Experimental measurements of the spatial distribution of temperature and composition of ablation-controlled arc plasmas are a key to validate the predictions of metal evaporation and polymer ablation models. Thus, high-speed photography and space-resolved spectroscopic measurements have been performed to characterize a stable air arc plasma jet controlled by ablation of a polymer nozzle made of Polyoxymethylene copolymer (POM-C) or polyamide (PA6). The spectroscopic analysis is performed along a plane perpendicular to the arc jet axis for a current of 1.8 kA, corresponding to an estimated current density of similar to 65 A mm(-2). Temperature and partial pressure profiles of the plasma for copper, hydrogen and carbon in the gas mixture are estimated as an inverse optimization problem by using measured side-on radiance spectra and radiative transfer spectral simulations. It is shown that the generated ablation-controlled arc has a complicated, non-uniform gas composition. Thus, the generated arc jet has a thin metallic core with a lower almost constant hydrogen pressure, surrounded by a thicker hydrogen and carbon mantle at partial pressures slightly lower than atmospheric pressure. The separation of hydrogen and carbon in the core is a consequence of demixing of the polymer vapour in the plasma. It is found that the overall shape of the temperature and pressure profiles obtained for the arc plasmas with the POM-C and PA6 nozzles are similar although differ in peak values and width.

  • A new 3 mm band receiver for the Onsala 20 m antenna

    • A new receiver for the Onsala 20 m antenna with the possibility of being equipped with 3 mm and 4 mm bands has been built and the 3 mm channel has been commissioned during the Spring 2014. For single-dish operation, the receiver uses an innovative on-source/off-source optical switch. In combination with additional optical components and within the same optical layout, the switch provides two calibration loads (for the 3 mm and 4 mm channels), sideband rejection measurement, and tuning possibilities. The optical layout of the receiver employs all cold (4 K) offset elliptical mirrors for both channels, whereas the on-off switch employs flat mirrors only. The 3 mm channel employs a sideband separation (2SB) dual polarization receiver with orthomode transducer (OMT), 4-8 GHz intermediate frequency (IF), x? 2pol x? upper and lower sidebands (USB? +? LSB). The cryostat has four optical windows made of high density polyethylene (HDPE) with anti-reflection corrugations, two for the signal and two for each frequency band cold load. The cryostat uses a two-stage cryocooler produced by Sumitomo HI? RDK? 408D2 with anti-vibration suspension of the cold-head to minimize impact of the vibrations on the receiver stability. The local oscillator (LO) system is based on a Gunn oscillator with aphase lock loop (PLL) and four mechanical tuners for broadband operation, providing independently tunable LO power for each polarization. This paper provides a technical description of the receiver and its technology and could be useful for instrumentation engineers and observers using the Onsala 20 m telescope.

    • Delivery of Yop effector proteins by pathogenic Yersinia across the eukaryotic cell membrane requires LcrV, YopB and YopD. These proteins were also required for channel formation in infected erythrocytes and, using different osmolytes, the contact‐dependent haemolysis assay was used to study channel size. Channels associated with LcrV were around 3 nm, whereas the homologous PcrV protein of Pseudomonas aeruginosa induced channels of around 2 nm in diameter. In lipid bilayer membranes, purified LcrV and PcrV induced a stepwise conductance increase of 3 nS and 1 nS, respectively, in 1 M KCl. The regions important for channel size were localized to amino acids 127–195 of LcrV and to amino acids 106–173 of PcrV. The size of the channel correlated with the ability to translocate Yop effectors into host cells. We suggest that LcrV is a size‐determining structural component of the Yop translocon.

  • New fibrin conduit for peripheral nerve repair

    • An ideal substitute to treat a nerve gap has not been found. Initially, silicone conduits were employed. Later, conduits were fabricated from collagen or polyesters carbonates. More recently, it has been shown that a bioresorbable material, poly-3-hydroxybutyrate (PHB), can enhance nerve repair. The present investigation shows the use of fibrin as a conduit to guide nerve regeneration and bridge nerve defects. In this study we prepared and investigated a novel nerve conduit made from fibrin glue. Using a rodent sciatic nerve injury model (10-mm gap), we compared the extent of nerve regeneration through the new fibrin conduits versus established PHB conduits. After 2 and 4 weeks, conduits containing proximal and distal stumps were harvested. We evaluated the initial axon and Schwann cell stimulation using immunohistochemistry. The conduits presented full tissue integration and were completely intact. Axons crossed the gap after 1 month. Immunohistochemistry using the axonal marker PGP 9.5 showed a superior nerve regeneration distance in the fibrin conduit compared with PHB (4.1 mm versus 1.9 mm). Schwann cell intrusion (S100 staining) was similarly enhanced in the fibrin conduits, both from the proximal (4.2 mm versus 2.1 mm) and distal ends (3.2 mm versus 1.7 mm). These findings suggest an advantage of the new fibrin conduit for the important initial phase of peripheral nerve regeneration. The use of fibrin glue as a conduit is a step toward a usable graft to bridge peripheral nerve lesions. This might be clinically interesting, given the widespread acceptance of fibrin glue among the surgical community.

    • Cavity formation is an important obstacle impeding regeneration after spinal cord injury and bridging strategies are essential to provide physical substrate allowing axons to grow across the lesion site. In this study we evaluated effects of biodegradable tubular conduit made from poly-beta-hydroxybutyrate (PHB) scaffold with predominantly unidirectional fiber orientation and supplemented with cultured adult Schwann cells on axonal regeneration after cervical spinal cord injury in adult rats. After transplantation into the injured spinal cord, plain PHB conduit was well-integrated into posttraumatic cavity and induced modest astroglial reaction. Regenerating axons were found mainly outside the PHB with only single fibers crossing the host-graft interface. No host Schwann cells migrated into the graft. In contrast, when suspension of adult Schwann cells was added to the PHB during transplantation, neurofilament-positive axons filled the conduit and became associated with the implanted cells. Although rubrospinal fibers did not enter the PHB, numerous raphaespinal and CGRP-positive axons were found within the conduit. Modification of PHB surface with fibronectin, laminin or collagen significantly increased Schwann cell attachment and proliferation in vitro. However, transplantation of PHB conduit pre-coated with fibronectin and seeded with Schwann cells did not alter axonal growth response. The results demonstrate that a PHB scaffold promotes attachment, proliferation and survival of adult Schwann cells and supports marked axonal regeneration within the graft.

    • Developing autonomous vehicles is technically complex and up to now research has focused on technical improvement and operative safety. As the level of automation increases the role of the driver will change; from controlling every movement of the vehicle into becoming an operator/passenger. Little is known about how this new context will affect the social experiences with and within the vehicle. This workshop focuses on three different kinds of social experience and socializing, namely; between other road users and the autonomous car, the social activities taking place within the autonomous car, and lastly the relationship between the car and the operator. The workshop aims at exploring possible practices, research and design directions of autonomous vehicles in relation to these social experiences. A human-centered design approach is the core of the workshop, with playful field excursions and ideation sessions.

    • Peripheral nerve injuries are often associated with loss of nerve tissue and require autologous nerve grafts to provide a physical substrate for axonal growth. Biosynthetic neural conduits could be an alternative treatment strategy in such injuries. The present study investigates the long-term effects of a tubular fibrin conduit on neuronal regeneration, axonal sprouting and recovery of muscle weight following peripheral nerve injury and repair in adult rats. Sciatic axotomy was performed proximally in the thigh to create a 10-mm gap between the nerve stumps. The injury gap was bridged by using a 14-mm-long fibrin glue conduit, entubulating 2mm of the nerve stump at each end. A reversed autologous nerve graft was used as a control. The regenerative response from sensory and motor neurones was evaluated following retrograde labelling with Fast Blue fluorescent tracer. In control experiments, at 16 weeks following peripheral nerve grafting, 5184 (+/-574 standard error of mean (SEM)) sensory dorsal root ganglion neurones and 1001 (+/-37 SEM) spinal motor neurones regenerated across the distal nerve-graft interface. The fibrin conduit promoted regeneration of 60% of sensory neurones and 52% of motor neurones when compared to the control group. The total number of myelinated axons in the distal nerve stump in the fibrin-conduit group reached 86% of the control and the weight of gastrocnemius and soleus muscles recovered to 82% and 89% of the controls, respectively. The present results suggest that a tubular fibrin conduit can be used to promote neuronal regeneration following peripheral nerve injury.
