  • Alterations of the WNT7A Gene in Clear Cell Renal Cell Carcinomas

    • WNT7A (wingless-type MMTV integration site family, member 7A) is a known tumor suppressor gene of non-small cell lung carcinomas (NSCLC) and is frequently inactivated due to CpG-island hypermethylation in human cancers. The members of WNT family are involved in cell signaling and play crucial roles in cancer development. In the present work hypermethylation of the WNT7A gene was detected in 66% (29/44) of analyzed clear cell renal cell carcinomas (RCCs) using methyl-specific PCR (MSP). Moreover, bisulfite sequencing confirmed intensive hypermethylation of the 5-CpG island of the WNT7A gene. Methylation analysis revealed positive correlations between tumor stage, Fuhrman nuclear grade and WNT7A hypermethylation. Additionally, restoration of WNT7A gene expression in the A498 cell line by 5-aza-2-deoxycytidine treatment confirmed a direct contribution of hypermethylation in silencing of the WNT7A gene. High frequency of loss of heterozygosity (LOH) was demonstrated on chromosome 3p25 in regions surrounding the WNT7A gene. The frequent down-regulation of WNT7A gene expression was detected in 88% (15/17) of clear cell RCCs. We have also shown that the WNT7A gene possesses tumor suppression function by colony-formation and cell proliferation assays in RCC cell lines. In summary, the WNT7A gene is inactivated by genetic/epigenetic alterations in clear cell RCC and demonstrates tumor suppressor properties.

    • A new quasi-one-dimensional compound Ni8Bi8SI has been synthesized and its crystal structure determined from single-crystal X-ray diffraction data. The structure of Ni8Bi8SI consists of [(infinityNi8Bi8S)-Ni-1] columns separated by iodine atoms. Conductivity and magnetic susceptibility measurements (down to 4.2 K) show that Ni8Bi8SI is a one-dimensional metal and exhibits Pauli paramagnetic properties. These observations are in good agreement with the results from electronic structure calculations. An analysis of the chemical bonding employing difference electron charge density maps reveals strong multicenter Ni-Bi bonds and pair Ni-S interactions within the [(infinityNi8Bi8S)-Ni-1] columns. Only electrostatic interactions are inferred between the columns and iodine atoms.

    • The new compound Ni(8)Bi(8)Sl(2) has been synthesized and its crystal structure determined by X-ray crystallography. The structure contains one-dimensional (1D) cations (1)(infinity)[Ni8Bi8S](2+) separated by iodine anions. The geometry of the columns is similar to that of the recently reported (1)(infinity)[Ni8Bi8S](+), and the main difference between them is only their formal charge. Electronic structure calculations and physical properties measurements were performed to analyze the influence of the number of valence electrons on the bonding and properties of compounds containing these 1D cations. It was shown that the removal of one electron (i.e., (1)(infinity)[Ni8Bi8S](+) --> (1)(infinity)[Ni8Bi8S](2+)) mainly affects the Ni-S bonding within the cation and essentially has no influence on the intermetallic Ni-Bi bonding. It was found that Ni(8)Bi(8)Sl(2) containing double-charged columns has conductivity properties more similar to a pure 1D metal than the congener Ni(8)Bi(8)Sl containing mono-charged columns.

    • A group of five Gram-negative, aerobic, halotolerant, non-pigmented bacteria isolated from shallow sediment samples and invertebrate specimens collected from the Gulf of Alaska and the Sea of Japan was subjected to taxonomic study. On the basis of 16S rRNA gene sequence analysis, the novel isolates were affiliated to the genus Cobetia, sharing the highest sequence similarity of 99.3-99.9% with Cobetia marina DSM 4741(T). DNA-DNA hybridization experiments between and among the novel strains and C. marina DSM 4741(T) and Cobetia crustatorum JCM 15644(T) revealed that the five strains represent three separate genospecies, which could be differentiated in their morphological, physiological and biochemical characteristics. Halomonas halodurans NBRC 15607(T) was included in this study as it has recently been reported to exhibit high 16S rRNA gene sequence similarity to C. marina DSM 4741(T), and it showed a high DNA relatedness value of 96% with C. marina DSM 4741(T), indicating that they belong to the same species. On the basis of phylogenetic analysis, DNA-DNA hybridization and phenotypic characterization, three novel species are proposed, named Cobetia amphilecti sp. nov. (type strain KMM 1561(T) =NRIC 0815(T) =CCUG 49560(T)), Cobetia litoralis sp. nov. (type strain KMM 3880(T) =NRIC 0814(T) =CCUG 49563(T)) and Cobetia pacifica sp. nov. (type strain KMM 3879(T) =NRIC 0813(T) =CCUG 49562(T)). It is also proposed that Halomonas halodurans is a later heterotypic synonym of Cobetia marina, and emended descriptions of the genus Cobetia and the species Cobetia marina are provided.

    • This work demonstrates the use of imidazolium-based poly(ionic liquid)s (PILs) as efficient dispersants of multiwalled carbon nanotubes (MWCNTs). With these polymeric dispersants, highly stable fine dispersions of MWCNTs (inks) can be easily prepared in aqueous media and applied for rather simple but efficient surface modification of screen-printed electrodes (SPEs). Such a modification of SPEs remarkably increases the electroactive surface area and accelerates the electron transfer rate due to synergistic combination of specific features of MWCNTs such as strong adsorptive property and high specific surface with the advantages of PILs like ion conductivity and dispersability. We further show that the PIL/MWCNT-modified SPEs can be beneficially utilized for direct electrochemical analysis of double stranded DNA (dsDNA). Specifically, it is exemplified by the direct electrooxidation of guanine and adenine bases in salmon testes dsDNA chosen as a model system. The linear ranges for the determination of dsDNA correspond to 5-500 mu g/mL for the oxidative peak of guanine and 0.5-50 mu g/mL for the oxidative peak of adenine. This makes direct electrochemical dsDNA detection with the use of the easy-preparable PIL/MWCNT-modified SPEs strongly competing to currently applied spectral and fluorescent techniques. Furthermore, we show that the developed constructs are capable of sensing a single point mutation in the 12-bases single-stranded DNA fragments. Such detection is of high clinical significance in choosing an adequate anticancer treatment, where the electrochemical identification of the point mutation could offer time and cost benefits.
