    • Poly(ADP-ribose) polymerase-1 (PARP-1) plays the active role of "nick sensor" during DNA repair and apoptosis, when it synthesizes ADP-ribose from NAD(+) in the presence of DNA strand breaks. Moreover, PARP-1 becomes a target of apoptotic caspases, which originate two proteolytic fragments of 89 and 24 kDa. The precise relationship between PARP-1 activation and degradation during apoptosis is still a matter of debate. In human Hep-2 cells driven to apoptosis by actinomycin D, we have monitored PARP-1 activity by the mAb 10H, which is specific for the ADP-ribose polymers, and we have observed that poly(ADP-ribose) synthesis is a very early response to the apoptotic stimulus. The analysis of the presence and fate of the p89 proteolytic fragment revealed that PARP-1 proteolysis by caspases is concomitant with poly(ADP-ribose) synthesis and that p89 migrates from the nucleus into the cytoplasm in late apoptotic cells with advanced nuclear fragmentation.

  • "Is 'junk food' an 'healthy' concept?" the challenges of the current debate From policy making back to science

    • Purpose - The term "junk food" is for the most part currently used in the widest political and media debate, without reference to something tangible. The purpose of this paper is to pinpoint the delicate issues involved in moving towards a trans-national, unified, consensual definition of the term "junk food", including social, economic, cultural, nutritional and methodological problems. Design/methodology/approach - Departing from the work done by international bodies (WHO, FAO-Codex Alimentarius, OECD, EFSA and European Commission) authors descriptively investigate possible background elements able to frame the surrounding debate about "junk food"("nutrient profiles", labelling provisions, institutional aspects, etc.). Findings - Presently there is a lack of a global consensus and of scientific basis to define clearly what constitutes "junk food" either on regional areas or globally. Despite of good metrics able to classify foods according to their nutritional quality, policy making relies yet on the concept that only diets or single nutrients can be focused as "good or bad". Practical implications - A "junk food" taxonomy could be helpful to harmonize trade policies in internal markets (i.e. the EU) and at global level; but also to gain a wider social support for "hard" policy measures intending to counteract non-communicable-diseases (NCDs), and more generally, obesity and overweight. Social implications - A clear basis for "junk food" definition could be the first step to introduce otherwise controversial and easily opposable public health policies and campaigns, due to private interests of the different stakeholders. Even consumers may perceive food policies (in the sake of "food taxes" or "traffic light labelling") as unfair, whereas not robust scientific ground has been previously given at the highest possible level. Originality/value - The value of this descriptive paper consists in addressing the shortcomings of global and regional nutritional policies framework in front of the emerging trend of "globesity". Conclusions stress the need to find support for broader food policies (labelling, taxes, education, bans, etc.) which currently are on the rise but lack fundamental aspects of scientific and hence social support.

In response to this need, we calculated the Mediterranean diet score (MDS) in 2-9-year-old children from the Identification and prevention of dietary- and lifestyle-induced health effects in children and infants (IDEFICS) eight-country study.

  • Development of the Choices 5-Level Criteria to Support Multiple Food System Actions

    • In 2008, the Choices International Foundation developed its logo criteria, identifying best-in-class food products. More advanced, global and graded nutrient profiling systems (NPSs) are needed to substantiate different national nutrition policies. The objective of this work was to extend Choices NPS to identify five levels of the healthiness of food products, so that the Choices NPS can also be used to support other nutrition policies, next to front-of-pack labelling. Based on the same principles as the previous logo criteria, four sets of threshold criteria were determined using a combination of compliance levels, calculated from a large international food group-specific database, the Choices logo criteria, and WHO-NPSs developed to restrict marketing to children. Validation consisted of a comparison with indicator foods from food-based dietary guidelines from various countries. Some thresholds were adjusted after the validation, e.g., because intermediate thresholds were too lenient. This resulted in a new international NPS that can be applied to different contexts and to support a variety of health policies, to prevent both undernutrition and obesity. It can efficiently evaluate mixed food products and represents a flexible tool, applicable in various settings and populations.

  • Bayesian multi-source regression and monocyte-associated gene expression predict BCL-2 inhibitor resistance in acute myeloid leukemia

    • The FDA recently approved eight targeted therapies for acute myeloid leukemia (AML), including the BCL-2 inhibitor venetoclax. Maximizing efficacy of these treatments requires refining patient selection. To this end, we analyzed two recent AML studies profiling the gene expression and ex vivo drug response of primary patient samples. We find that ex vivo samples often exhibit a general sensitivity to (any) drug exposure, independent of drug target. We observe that this “general response across drugs” (GRD) is associated with FLT3-ITD mutations, clinical response to standard induction chemotherapy, and overall survival. Further, incorporating GRD into expression-based regression models trained on one of the studies improved their performance in predicting ex vivo response in the second study, thus signifying its relevance to precision oncology efforts. We find that venetoclax response is independent of GRD but instead show that it is linked to expression of monocyte-associated genes by developing and applying a multi-source Bayesian regression approach. The method shares information across studies to robustly identify biomarkers of drug response and is broadly applicable in integrative analyses.
