  • Genetic variants associated with platelet count are predictive of human disease and physiological markers

    • Platelets play an important role in hemostasis and other aspects of vascular biology. We conducted a meta-analysis of platelet count GWAS using data on 536,974 Europeans and identified 577 independent associations. To search for mechanisms through which these variants affect platelets, we applied cis-expression quantitative trait locus, DEPICT and IPA analyses and assessed genetic sharing between platelet count and various traits using polygenic risk scoring. We found genetic sharing between platelet count and counts of other blood cells (except red blood cells), in addition to several other quantitative traits, including markers of cardiovascular, liver and kidney functions, height, and weight. Platelet count polygenic risk score was predictive of myeloproliferative neoplasms, rheumatoid arthritis, ankylosing spondylitis, hypertension, and benign prostate hyperplasia. Taken together, these results advance understanding of diverse aspects of platelet biology and how they affect biological processes in health and disease.

    • This paper describes the design of a compact polarizer that rotates the linear polarization of a fully metallic geodesic Luneburg lens antenna for applications requiring polarization diversity. The polarization rotation is achieved by loading the radiating aperture of the antenna with two metallic screens. The integrated antenna operates in the Ka-band, from 25 to 31 GHz, showing around 20% bandwidth with S11 below -10 dB, and a scanning range of 100°.

    • In this letter, a parallel-plate waveguide half-Luneburg geodesic lens multiple-beam antenna optimized for additive manufacturing is designed and validated experimentally in the K$_{\mathrm{a}}$-band. Two prototypes were manufactured in AlSi$_{10}$Mg through laser powder-bed fusion and measured in an anechoic chamber. The compact and lightweight prototype optimized for additive manufacturing demonstrates excellent RF performance while significantly reducing mass and mechanical complexity. Specifically, misalignment errors present in previous studies are solved, improving sidelobe level by up to 10 dB. A maximum realized gain of 22.1 dBi is measured at 28 GHz. This single-piece, compact, and lightweight design is particularly attractive for applications having mass restrictions and limited space like millimeter-wave systems on board small satellites.

    • While macro- and microscopic kidney development appear to proceed normally in mice that lack Foxi1, electron microscopy reveals an altered ultrastructure of cells lining the distal nephron. Northern blot analyses, cRNA in situ hybridizations, and immunohistochemistry demonstrate a complete loss of expression of several anion transporters, proton pumps, and anion exchange proteins expressed by intercalated cells of the collecting ducts, many of which have been implicated in hereditary forms of distal renal tubular acidosis (dRTA). In Foxi1-null mutants the normal epithelium with its two major cell types - principal and intercalated cells - has been replaced by a single cell type positive for both principal and intercalated cell markers. To test the functional consequences of these alterations, Foxi1(-/-) mice were compared with WT littermates in their response to an acidic load. This revealed an inability to acidify the urine as well as a lowered systemic buffer capacity and overt acidosis in null mutants. Thus, Foxi1(-/-) mice seem to develop dRTA due to altered cellular composition of the distal nephron epithelium, thereby denying this epithelium the proper gene expression pattern needed for maintaining adequate acid-base homeostasis.
