    • The collection of modern, spatially extensive pollen data is important for the interpretation of fossil pollen assemblages and the reconstruction of past vegetation communities in space and time. Modern datasets are readily available for percentage data but lacking for pollen accumulation rates (PARs). Filling this gap has been the motivation of the pollen monitoring network, whose contributors monitored pollen deposition in modified Tauber traps for several years or decades across Europe. Here we present this monitoring dataset consisting of 351 trap locations with a total of 2742 annual samples covering the period from 1981 to 2017. This dataset shows that total PAR is influenced by forest cover and climate parameters, which determine pollen productivity and correlate with latitude. Treeless vegetation produced PAR values of at least 140 grains cm(-2) yr(-1). Tree PAR increased by at least 400 grains cm(-2) yr(-1) with each 10% increase in forest cover. Pollen traps situated beyond 200 km of the distribution of a given tree species still collect occasional pollen grains of that species. The threshold of this long-distance transport differs for individual species and is generally below 60 grains cm(-2) yr(-1). Comparisons between modern and fossil PAR from the same regions show similar values. For temperate taxa, modern analogues for fossil PARs are generally found downslope or southward of the fossil sites. While we do not find modern situations comparable to fossil PAR values of some taxa (e.g. Corylus), CO2 fertilization and land use may cause high modern PARs that are not documented in the fossil record. The modern data are now publicly available in the Neotoma Paleoecology Database and aid interpretations of fossil PAR data.

  • Combined First-Principles and Experimental Study on the Microstructure and Mechanical Characteristics of the Multicomponent Additive-Manufactured Ti-35Nb-7Zr-5Ta Alloy

    • New & beta;-stabilizedTi-based alloys are highly promising forbone implants, thanks in part to their low elasticity. The natureof this elasticity, however, is as yet unknown. We here present combinedfirst-principles DFT calculations and experiments on the microstructure,structural stability, mechanical characteristics, and electronic structureto elucidate this origin. Our results suggest that the studied & beta;Ti-35Nb-7Zr-5Ta wt % (TNZT) alloy manufacturedby the electron-beam powder bed fusion (E-PBF) method has homogeneousmechanical properties (H = 2.01 & PLUSMN; 0.22 GPa and E = 69.48 & PLUSMN; 0.03 GPa) along the building direction,which is dictated by the crystallographic texture and microstructuremorphologies. The analysis of the structural and electronic properties,as the main factors dominating the chemical bonding mechanism, indicatesthat TNZT has a mixture of strong metallic and weak covalent bonding.Our calculations demonstrate that the softening in the Cauchy pressure(C & PRIME; = 98.00 GPa) and elastic constant C ̅ ( 44 ) = 23.84 GPa is the originof the low elasticity of TNZT. Moreover, the nature of this softeningphenomenon can be related to the weakness of the second and thirdneighbor bonds in comparison with the first neighbor bonds in theTNZT. Thus, the obtained results indicate that a carefully designedTNZT alloy can be an excellent candidate for the manufacturing oforthopedic internal fixation devices. In addition, the current findingscan be used as guidance not only for predicting the mechanical propertiesbut also the nature of elastic characteristics of the newly developedalloys with yet unknown properties.

  • New Ti-35Nb-7Zr-5Ta Alloy Manufacturing by Electron Beam Melting for Medical Application Followed by High Current Pulsed Electron Beam Treatment

    • High-current pulsed electron-beam (PEB) treatment was applied as a surface finishing procedure for Ti-35Nb-7Zr-5Ta (TNZT) alloy produced by electron beam melting (EBM). According to the XRD results the TNZT alloy samples before and after the PEB treatment have shown mainly the single body-centered cubic (bcc) beta-phase microstructures. The crystallite size, dislocation density, and microstrain remain unchanged after the PEB treatment. The investigation of the texture coefficient at the different grazing angle revealed the evolution of the crystallite orientations at the re-melted zone formed at the top of the bulk samples after the PEB treatment. The top-view SEM micrographs of the TNZT samples treated by PEB exhibited the bcc beta-phase grains with an average size of similar to 85 mu m. TEM analysis of as-manufactured TNZT alloy revealed the presence of the equiaxed beta-grains with the fine dispersion of nanocrystalline alpha and NbTi4 phases together with beta-Ti twins. Meanwhile, the beta phase regions free of alpha phase precipitation are observed in the microstructure after the PEB irradiation. Nanoindentation tests revealed that the surface mechanical properties of the melted zone were slightly improved. However, the elastic modulus and microhardness in the heat-affected zone and the deeper regions of the sample were not changed after the treatment. Moreover, the TNZT alloy in the bulk region manufactured by EBM displayed no significant change in the corrosion resistance after the PEB treatment. Hence, it can be concluded that the PEB irradiation is a viable approach to improve the surface topography of EBM-manufactured TNZT alloy, while the most important mechanical parameters remain unchanged.

  • Validated Reference Panel from Renewable Source of Genomic DNA Available for Standardization of Blood Group Genotyping

    • Extended blood group genotyping is an invaluable tool used for prevention of alloimmunization. Genotyping is particularly suitable when antigens are weak, specific antisera are unavailable, or accurate phenotyping is problematic because of a disease state or recent transfusions. In addition, genotyping facilitates establishment of mass-scale patient-matched donor databases. However, standardization of genotyping technologies has been hindered by the lack of reference panels. A well-characterized renewable reference panel for standardization of blood group genotyping was developed. The panel consists of genomic DNA lyophilized and stored in glass vials. Genomic DNA was extracted in bulk from immortalized lymphoblastoid cell lines, generated by Epstein-Barr virus transformation of peripheral blood lymphocytes harvested from volunteer blood donors. The panel was validated by an international collaborative study involving 28 laboratories that tested each DNA panel member for 41 polymorphisms associated with 17 blood group systems. Overall, analysis of genotyping results showed >98% agreement with the expected outcomes, demonstrating suitability of the material for use as reference. Highest levels of discordance were observed for the genes CR1, CD55, BSG, and RHD. Although limited, observed inconsistencies and procedural limitations reinforce the importance of reference reagents to standardize and harmonize results. Results of stability and accelerated degradation studies support the suitability of this panel for use as reference reagent for blood group genotyping assay development and standardization.

  • Optimization of N-Piperidinyl-Benzimidazolone Derivatives as Potent and Selective Inhibitors of 8-Oxo-Guanine DNA Glycosylase 1

    • 8-oxo Guanine DNA Glycosylase 1 is the initiating enzyme within base excision repair and removes oxidized guanines from damaged DNA. Since unrepaired 8-oxoG could lead to G : C→T : A transversion, base removal is of utmost importance for cells to ensure genomic integrity. For cells with elevated levels of reactive oxygen species this dependency is further increased. In the past we and others have validated OGG1 as a target for inhibitors to treat cancer and inflammation. Here, we present the optimization campaign that led to the broadly used tool compound TH5487. Based on results from a small molecule screening campaign, we performed hit to lead expansion and arrived at potent and selective substituted N-piperidinyl-benzimidazolones. Using X-ray crystallography data, we describe the surprising binding mode of the most potent member of the class, TH8535. Here, the N-Piperidinyl-linker adopts a chair instead of a boat conformation which was found for weaker analogues. We further demonstrate cellular target engagement and efficacy of TH8535 against a number of cancer cell lines.

  • Capacity building on decision support for air pollution policies – results from Nordic-Russian co-operation

    • In 2010 the Nordic Council of Ministers initiated a research project with the aims to enable capacity building on the EMEP/MSC-W model, expand data inventories in the Russian Federation, and to develop a Russian version of the GAINS model. The project activities were: Identification of regions to be modelled; EMEP/MSC-W model calculations and capacity building; GAINS model development and adaptation; Emissions & Data inventories and consistency checks; GAINS model scenario analysis. The project resulted in an updated GAINS Russia model, regionalised and sector-specific emission inventories, region-specific source-receptor calculations, and EMEP/MSC-W model training activities. Finally, regionalised emission abatement scenarios were analysed and showed large differences between regions in terms of potential emission reductions and emission abatement costs. The Russian Federation has now established analytical capacity related to the GAINS and EMEP/MSC-W models, the GAINS Russia model has been updated and improved, and a process for preparing input data inventories has been initiated. The Russian Federation can now launch independent research on cost effective emission reductions in the Russian Federation and analyse consequences on human health and the environment.
