  • Ion counting efficiencies at the IGISOL facility

    • At the IGISOL-JYFLTRAP facility, fission mass yields can be studied at high precision. Fission fragments from a U target are passing through a Ni foil and entering a gas filled chamber. The collected fragments are guided through a mass separator to a Penning trap where their masses are identified. This simulation work focuses on how different fission fragment properties (mass, charge and energy) affect the stopping efficiency in the gas cell. In addition, different experimental parameters are varied (e. g. U and Ni thickness and He gas pressure) to study their impact on the stopping efficiency. The simulations were performed using the Geant4 package and the SRIM code. The main results suggest a small variation in the stopping efficiency as a function of mass, charge and kinetic energy. It is predicted that heavy fragments are stopped about 9% less efficiently than the light fragments. However it was found that the properties of the U, Ni and the He gas influences this behavior. Hence it could be possible to optimize the efficiency.

    • At the Jyväskylä Ion Guide Isotope Separator On-Line (IGISOL) facility, independent fission yields are measured employing the Penning-trap technique. Fission products are produced, e.g. by impinging protons on a uranium target, and are stopped in a gas-filled chamber. The products are collected by a flow of He gas and guided through a mass separator to a Penning trap, where their masses are identified. This work investigates how fission-product properties, such as mass and energy, affect the ion stopping efficiency in the gas cell. The study was performed using the Geant4 toolkit and the SRIM code. The main results show a nearly mass-independent ion stopping with regard to the wide spread of ion masses and energies, with a proper choice of uranium target thickness. Although small variations were observed, in the order of 5%, the results are within the systematic uncertainties of the simulations. To optimize the stopping efficiency while reducing the systematic errors, different experimental parameters were varied; for instance material thicknesses and He gas pressure. Different parameters influence the mass dependence and could alter the mass dependencies in the ion stopping efficiency.

  • Screening and prioritization of micropollutants in wastewaters from on-site sewage treatment facilities

    • A comprehensive screening of micropollutants was performed in wastewaters from on-site sewage treatment facilities (OSSFs) and urban wastewater treatment plants (WWTPs) in Sweden. A suspect screening approach, using high resolution mass spectrometry, was developed and used in combination with target analysis. With this strategy, a total number of 79 micropollutants were successfully identified, which belong to the groups of per- and polyfluoroalkyl substances (PFASs), pesticides, phosphorus-containing flame retardants (PFRs) and pharmaceuticals and personal care products (PPCPs). Results from this screening indicate that concentrations of micropollutants are similar in influents and effluents of OSSFs and WWTPs, respectively. Removal efficiencies of micropollutants were assessed in the OSSFs and compared with those observed in WWTPs. In general, removal of PFASs and PFRs was higher in package treatment OSSFs, which are based on biological treatments, while removal of PPCPs was more efficient in soil bed OSSFs. A novel comprehensive prioritization strategy was then developed to identify OSSF specific chemicals of environmental relevance. The strategy was based on the compound concentrations in the wastewater, removal efficiency, frequency of detection in OSSFs and on in silico based data for toxicity, persistency and bioaccumulation potential.

  • Long-term SARS-CoV-2-specific and cross-reactive cellular immune responses correlate with humoral responses, disease severity, and symptomatology

    • Background: Cellular immune memory responses post coronavirus disease 2019 (COVID-19) have been difficult to assess due to the risks of contaminating the immune response readout with memory responses stemming from previous exposure to endemic coronaviruses. The work herein presents a large-scale long-term follow-up study investigating the correlation between symptomology and cellular immune responses four to five months post seroconversion based on a unique severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)-specific peptide pool that contains no overlapping peptides with endemic human coronaviruses. Methods: Peptide stimulated memory T cell responses were assessed with dual interferon-gamma (IFN gamma) and interleukin (IL)-2 Fluorospot. Serological analyses were performed using a multiplex antigen bead array. Results: Our work demonstrates that long-term SARS-CoV-2-specific memory T cell responses feature dual IFN gamma and IL-2 responses, whereas cross-reactive memory T cell responses primarily generate IFN gamma in response to SARS-CoV-2 peptide stimulation. T cell responses correlated to long-term humoral immune responses. Disease severity as well as specific COVID-19 symptoms correlated with the magnitude of the SARS-CoV-2-specific memory T cell response four to five months post seroconversion. Conclusion: Using a large cohort and a SARS-CoV-2-specific peptide pool we were able to substantiate that initial disease severity and symptoms correlate with the magnitude of the SARS-CoV-2-specific memory T cell responses.

    • In the past decades, the housing shortage in Sweden accumulated to a level that led to problems finding accommodation for many people, which created opportunities for the market producing multi-family houses. The market is dominated by concrete solutions whereas solutions using wood as a building material only comprise 9 % of the market. This market is highly competitive with many companies offering relatively similar products or services. One way to develop new business opportunities is to enhance other competence besides production technology that currently acts as a market development barrier. Hence, the material handling process at building sites is seen as a barrier that can contribute improving competitiveness.There are currently problems with material handling at many building sites of wood-building solutions in Sweden, materials arrive at the wrong time, waste of storage space, unnecessary tracking of materials or wrong quantities arriving at the sites. The purpose of the study is to investigate if Radio Frequency Identification (RFID) is a technology that can be used to achieve an efficient material handling process and if the wood building industry in Sweden is willing to implement the technology. Thus, investigate if material handling using RFID can improve time effectiveness, minimise waste and monitor moisture levels in the material efficiently, creating improved competitiveness. The study shows that the use of RFID improves time efficiency and control the material handling, indicating that RFID has potential to improve this process. Further, the study identifies possibilities by using RFID technology to minimize damages and control moisture levels.

  • Antibodies from serum and CSF of multiple sclerosis patients bind to oligodendroglial and neuronal cell-lines

    • Multiple sclerosis is a highly complex and heterogeneous disease. At the onset it often presents as a clinically isolated syndrome. Thereafter relapses are followed by periods of remissions, but eventually, most patients develop secondary progressive multiple sclerosis. It is widely accepted that autoantibodies are important to the pathogenesis of multiple sclerosis, but hitherto it has been difficult to identify the target of such autoantibodies. As an alternative strategy, cell-based methods of detecting autoantibodies have been developed. The objective of this study was to explore differences in the binding of antibodies from sera and CSF of multiple sclerosis patients and controls to oligodendroglial and neuronal cell-lines, related to antibody type, immunoglobulin (IgG/IgM), matrix (serum/CSF) and disease course. The oligodendroglial and neuronal cell-lines were expanded in tissue culture flasks and transferred to 96-well plates at a concentration of 50 000 cells/well followed by fixation and blocking with bovine serum albumin. Sera and CSF samples, from healthy controls and multiple sclerosis patients, were incubated with the fixed cells. Epitope binding of immunoglobulins (IgG and IgM) in sera and CSF was detected using biotinylated anti-human IgM and IgG followed by avidin conjugated to horseradish peroxidase. Horseradish peroxidase activity was detected with 3,3 ',5,5 '-tetramethylbenzidine substrate. Serum from 76 patients and 30 controls as well as CSF from 62 patients and 32 controls were investigated in the study. The binding was similar between clinically isolated syndrome patients and controls, whereas the largest differences were observed between secondary progressive multiple sclerosis patients and controls. Antibodies from multiple sclerosis patients (all disease course combined) bound more to all investigated cell-lines, irrespectively of matrix type, but binding of immunoglobulin G from CSF to human oligodendroglioma cell-line discriminated best between multiple sclerosis patients and controls with a sensitivity of 93% and a specificity of 96%. The cell-based enzyme linked immunosorbent assay (ELISA) was able to discriminate between multiple sclerosis patients and controls with a high degree of accuracy. The disease course was the major determinant for the antibody binding. Nazir et al. measured antibody binding to neuronal and oligodendroglial cell-lines with a cell-based ELISA. They report that this method could discriminate between multiple sclerosis patients and healthy controls with a high degree of accuracy. The disease course was the major determinant for the antibody binding.

  • Cerebrospinal Fluid Metals and the Association with Cerebral Small Vessel Disease.

    • Brain metal homeostasis is essential for brain health, and deregulation can result in oxidative stress on the brain parenchyma.Our objective in this study was to focus on two hemorrhagic MRI manifestations of small vessel disease [cerebral microbleeds (CMBs) and cortical superficial siderosis (cSS)] and associations with cerebrospinal fluid (CSF) iron levels. In addition, we aimed to analyze CSF biomarkers for dementia and associations with CSF metal levels.This is a cross-sectional study of 196 patients who underwent memory clinic investigation, including brain MRI. CSF was collected and analyzed for metals, amyloid-β (Aβ) 42, total tau (T-tau), and phosphorylated tau (P-tau), and CSF/serum albumin ratios. Statistical analyses were performed using generalized linear models.No significant difference was found between CSF metal levels across diagnostic groups. Higher iron and copper levels were associated with higher CSF levels of Aβ42, T-tau, P-tau, and CSF/serum albumin ratios (p<0.05). Zinc was associated with higher CSF/serum albumin ratios. There was no significant association between CMBs or cSS and CSF iron levels. An increase in CSF iron with the number of CMBs was seen in APOEɛ4 carriers.CSF iron levels are elevated with cerebral microbleeds in APOEɛ4 carriers, with no other association seen with hemorrhagic markers of small vessel disease. The association of elevated CSF iron and copper with tau could represent findings of increased neurodegeneration in these patients.
