    • The International Cancer Genome Consortium (ICGC) was launched to coordinate large-scale cancer genome studies in tumours from 50 different cancer types and/or subtypes that are of clinical and societal importance across the globe. Systematic studies of more than 25,000 cancer genomes at the genomic, epigenomic and transcriptomic levels will reveal the repertoire of oncogenic mutations, uncover traces of the mutagenic influences, define clinically relevant subtypes for prognosis and therapeutic management, and enable the development of new cancer therapies.

  • The E-cadherin/AmotL2 complex organizes actin filaments required for epithelial hexagonal packing and blastocyst hatching

    • Epithelial cells connect via cell-cell junctions to form sheets of cells with separate cellular compartments. These cellular connections are essential for the generation of cellular forms and shapes consistent with organ function. Tissue modulation is dependent on the fine-tuning of mechanical forces that are transmitted in part through the actin connection to E-cadherin as well as other components in the adherens junctions. In this report we show that p100 amotL2 forms a complex with E-cadherin that associates with radial actin filaments connecting cells over multiple layers. Genetic inactivation or depletion of amotL2 in epithelial cells in vitro or zebrafish and mouse in vivo, resulted in the loss of contractile actin filaments and perturbed epithelial packing geometry. We further showed that AMOTL2 mRNA and protein was expressed in the trophectoderm of human and mouse blastocysts. Genetic inactivation of amotL2 did not affect cellular differentiation but blocked hatching of the blastocysts from the zona pellucida. These results were mimicked by treatment with the myosin II inhibitor blebbistatin. We propose that the tension generated by the E-cadherin/AmotL2/actin filaments plays a crucial role in developmental processes such as epithelial geometrical packing as well as generation of forces required for blastocyst hatching.

    • Membranes fabricated from bacteria-produced cellulose have many practical and technological advantages over other means of cellulose fiber production, however, their functionality for targeted applications is limited and requires complex, multi-stage processing, while the mechanisms underpinned the optimum improvements remain largely unknown. Focusing on one of the highest-demand applications in wastewater treatment, here we resolve the three persistent issues in bacterial cellulose membranes (BCMs), namely poor fibrillar network quality, insufficient functionality and unsatisfactory performance, and discover the counterintuitive, yet most effective mechanism of imparting the multifunctional properties. First, innovative application of the Hestrin-Shramm medium instead of the CM0986 medium stimulated Taonella mepensis to produce BCMs with higher yields, more uniform fibrils and developed fibrillar networks, higher crystallinity indexes, better mechanical and thermal properties. Second, a novel but facile electroadsorption method was developed to enhance BCM functionality via bonding a natural bactericidal peptide-Nisin to the deliberately activated terminals on the BCM surface, achieving great antibacterial activity, good durability, well-preserved nanoporous network, outstanding water retention and low toxicity. Third, due to the synergetic effects of surface adsorption, ionic bonding, physical retention and active microbial killing, the Nisin-decorated BCMs enabled outstanding organic dye removal and excellent disinfection performance, among the best in the available reports. Collectively, this study demonstrates that electroadsorption is a promising and potentially generic strategy to fabricate Nisin-decorated and functionalized BCMs for next-generation membrane filters toward water purification.

  • Roles of Acceptor Guests in Tuning the Organic Solar Cell Property Based on an Efficient Binary Material System with a Nearly Zero Hole-Transfer Driving Force

    • Sub-picosecond hole transfer has been recently observed in several narrow band gap nonfullerene small-molecule acceptor (NFA)-based binary blended organic solar cell (OSC) systems operating with negligible energetic driving forces. As the driving forces are near zero, how the added acceptor/donor guests tune the barrier-free hole-transfer dynamics of these systems remains very unclear. In this study, we report a new NFA (BTCT-2Cl) that conducts a sub-picosecond hole transfer (2 ps) for efficient photocurrent generation when pairing with PM6 though the energetic offset is only 0.02 eV. We observe that the added nonfullerene and PCBM components differently tune the charge generation and recombination when selectively exciting BTCT-2Cl. After adding PC71BM, the hole transfer from the host BTCT-2Cl to the host donor is greatly accelerated, with the rate significantly reduced to 0.29 ps and the charge generation becomes more efficient; on the contrary, recombination is prolonged and a larger fill factor is obtained after adding an NFA guest, here, IT-4F. The different tuning on the host binary hole-transfer dynamics is likely related with the phase crystallinity and the domain size changed after adding different acceptor guests. Over 16% efficiency is obtained on the PC71BM-based ternary device that outperforms the host binary and the IT-4F-based ternary solar cells (both showing over 15% efficiencies). The results clearly demonstrate that adding PCBM or NFA guests enables a very effective and different tuning on the hole-transfer rates and the recombination rates between the barrier-free host binary components, hence leading to efficient tuning on the short-circuit current density and fill factor, which outlines new strategies toward designing high-efficiency ternary blended OSC systems.

  • Mechanistic study of transcription factor Sox18 during heart development

    • Heart development is a delicate and complex process regulated by coordination of various signaling pathways. In this study, we investigated the role of sox18 in heart development by modulating Wnt/β-Catenin signaling pathways. Our spatiotemporal expression analysis revealed that sox18 is mainly expressed in the heart, branchial arch, pharyngeal arch, spinal cord, and intersegmental vessels at the tailbud stage of Xenopus tropicalis embryo. Overexpression of sox18 in the X. tropicalis embryos causes heart edema, while loss-of-function of sox18 can change the signal of developmental heart marker gata4 at different stages, suggesting that sox18 plays an essential role in the development of the heart. Knockdown of SOX18 in human umbilical vein endothelial cells suggests a link between Sox18 and β-CATENIN, a key regulator of the Wnt signaling pathway. Sox18 negatively regulates islet1 and tbx3, the downstream factors of Wnt/β-Catenin signaling, during the linear heart tube formation and the heart looping stage. Taken together, our findings highlight the crucial role of Sox18 in the development of the heart via inhibiting Wnt/β-Catenin signaling.

    • Industrial composite fiber has been widely used in the high-end manufacturing because of its superior stiffness and strength. To ensure the quality of composite fiber placement in making high-end and lightweight aerospace products, it needs reliable composite fiber layup in-situ inspection. An accurate seam width measurement is an indispensable procedure during the layup inspection. However, the traditional seam measurements may fail in presence of surface curvature variance, curved edges between sheets, to name a few. This paper intentionally designs a novel 3D vision based Seam Width Measurement for Industrial Composite Fiber Layup In-situ Inspection (called 3D-SWiM). It aims to address three major challenges in measuring the seam width between the composite fiber sheets, namely, seam region over-segmentation, failures on region of interests (ROI) seam detection, seam width measurement inconsistency. Firstly, the region growing with semantic refinement is designed for 3D point cloud clustering, to overcome over-segmentation on seam regions. Afterwards, the gridding model with edge discrimination is applied to extract the 3D potential candidates in the edge areas. Eventually, we use the cubic B-spline fitting model to describe the seam region curves and the improved particle swarm optimization model to estimate the minimum and maximum distance between the seam edges. 3D-SWiM has been evaluated extensively on the developed composite fiber layup platform. The comparisons with the state-of-the-art methods (such as HT and GFM) on the region-of-interest segmentation, seam extraction and seam width measurement have been performed and the experimental results prove the competitive performance in composite fiber layup in-situ inspections.

    • An accurate and globally consistent navigation system is crucial for estimating the positions and attitudes of automated guided vehicles (AGVs) in digital workshops. A promising navigation technology for this purpose is tightly coupled visual-inertial fusion, which offers advantages such as quick response (QR), absolute scale, and accuracy. However, existing visual-inertial fusion systems have limitations, including long-term drift, tracking failures in textureless or poorly illuminated environments, and a lack of absolute references. To create a reliable and consistent AGV navigation framework and correct for long-term drift, we have designed a novel framework, globally consistent multiview visual-inertial fusion for AGV navigation (GCMVF-AGV). This framework uses a downward-looking QR vision sensor and a forward-looking visual-inertial sensor together to estimate AGV poses in real time. The downward camera provides absolute AGV positions and attitudes with reference to the global workshop frame. Furthermore, long-term visual-inertial drift, inertial biases, and velocities are periodically compensated between spatial intervals of QR codes by minimizing visual-inertial residuals with the rigid constraints of absolute poses estimated from the downward visual measurements. We have evaluated the proposed method on the developed AGV navigation platform, and experimental results demonstrate the position and attitude errors of less than 0.05 m and 2 degrees, respectively.
