  • Revealing the mechanisms of sexual isolation in a case of sympatric and parallel ecological divergence

    • Two ecotypes of a marine intertidal snail (Littorina saxatilis), living at different microhabitats and shore levels, have evolved in sympatry and in parallel across the Galician rocky shore. These ecotypes differ in many traits (including size) due to differential adaptation. They meet, mate assortatively, and partially hybridize at the mid shore where the two microhabitats overlap. The partial sexual isolation observed is claimed to be a side-effect of the size differences between ecotypes combined with a size assortative mating found in most populations of this species. We investigated this hypothesis using three complementary experimental approaches. First, we investigated which of the different shell variables contributed most to the variation in individual sexual isolation in the field by using two new statistics developed for that purpose: (1) pair sexual isolation and (2) r(i), which is based on the Pearson correlation coefficient. We found that size is the most important trait explaining the sexual isolation and, in particular, the males appear to be the key sex contributing to sexual isolation. Second, we compared the size assortative mating between regions: exposed rocky shore populations from north-westwern Spain (showing incomplete reproductive isolation due to size assortative mating) and protected Spanish and Swedish populations (showing size assortative mating but not reproductive isolation between ecomorphs). Most of the variation in size assortative mating between localities was significantly explained by the within-population level of variation on size. Third, we performed a laboratory male choice experiment, which further suggested that the choice is made predominantly on the basis of size. These results confirm the mechanism proposed to explain the sexual isolation in the Galician hybrid zone and thus support this case as a putative example of parallel incipient speciation.   • Sex and age differences in reflectance and biochemistry of carotenoid-based colour variation in the great tit Parus major

    • The plumage coloration in great tits (Parus major) is the subject of much behavioural and ecophysiological research, yet there is a lack of analyses of the natural colour variation and its mechanisms. We used reflectance spectrometry and high-performance liquid chromatography to explore individual, sexual and age-related variation in carotenoid coloration and pigmentation, paramount to the often presumed, but rarely substantiated, costs and 'honesty' of carotenoid displays. In adults, we found that sex was the strongest predictor of 'brightness' (higher in males) and of 'hue' (longer wavelength in females). There was no sex difference in 'carotenoid chroma' or carotenoid content of feathers which also was unrelated to adult age (1 or 2+ years) and condition. Similar patterns were revealed for nestlings. Regarding the biochemical 'signal content', 'carotenoid chroma', but not 'hue', was significantly related to the carotenoid content (lutein and zeaxanthin) of feathers. These results refute the previously assumed exaggeration of carotenoid pigmentation in male great tits, and question the condition-dependence of carotenoid coloration in this species. However, the sexual dimorphism in total reflectance or 'brightness', most likely due to melanins rather than carotenoids, may have implications for signalling or other adaptive explanations that need to be explored.

    • The relative importance of natural selection in the diversification of organisms. can be assessed indirectly using matrix correspondence. The present study determines the environmental and genetic correlates of microgeographical variation in the growth form, leaf form and flower morphology in populations of four Aeonium species from section Leuconium using partial regression methods. The phylogeny of the four species and the other 12 species in the section was deduced from amplified fragment length polymorphism (AFLP). Pubescence of floral organs and flower size correlate with the phylogeny while traits related to growth form, leaf form, flower construction and inflorescence size correlate with ecological factors. The variation in the latter four traits may therefore reflect selection by current ecological conditions while variation in pubescence and flower size may reflect historical events like neutral mutations, founder events and drift. Additionally, the morphological analyses revealed a large amount of variation in all traits within populations. This suggests a possible influence of microhabitat on the variation in morphology of Aeonium in the Canary Islands.   • Limited male dispersal in a social spider with extreme inbreeding

    • Cooperatively breeding animals commonly avoid incestuous mating through pre-mating dispersal. However, a few group-living organisms, including the social spiders, have low pre-mating dispersal, intra-colony mating, and inbreeding. This results in limited gene flow among colonies and sub-structured populations. The social spiders also exhibit female-biased sex ratios because survival benefits to large colonies favour high group productivity, which selects against 1 : 1 sex ratios. Although propagule dispersal of mated females may occasionally bring about limited gene flow, little is known about the role of male dispersal. We assessed the extent of male movement between colonies in natural populations both experimentally and by studying colony sex ratios over the mating season. We show that males frequently move to neighbouring colonies, whereas only 4% of incipient nests were visited by dispersing males. Neighbouring colonies are genetically similar and movement within colony clusters does not contribute to gene flow. Post-mating sex ratio bias was high early in the mating season due to protandry, and also in colonies at the end of the season, suggesting that males remain in the colony when mated females have dispersed. Thus, male dispersal is unlikely to facilitate gene flow between different matrilineages. This is consistent with models of non-Fisherian group-level selection for the maintenance of female biased sex ratios, which predict the elimination of male dispersal.

    • Most of the Nordic region was ice-covered during the last (Weichselian) glaciation. During the postglacial period, plant and animal species recolonized the region from several directions and the geographic structuring of genetic variation within Nordic species may still contain a historic component that reflects patterns of postglacial immigration. The present investigation of 69 populations of Silene dioica represents the first large-scale allozyme study of a widespread herbaceous plant in the Nordic region. Although the frequencies of individual alleles showed a range of different geographic patterns, mapping of the axis scores from an ordination of variation at eight polymorphic loci revealed a division into two main geographic groups of populations. The broadly south-western and northeastern distributions of these two groups of populations suggest that immigration into the region may have involved both eastern and southern geographic sources. However, the geographic boundaries between the two groups of populations are diffuse, and the relatively low between-population component of genetic diversity (G(ST) = 16.4%) suggests a history of extensive gene dispersal by pollen.     • Genetic differentiation of Dactylorhiza majalis ssp. traunsteineri from the Alps, Scandinavia, and Britain was studied and compared with other allotetraploid members of the systematically challenging genus Dactylorhiza. One-hundred and eleven populations from altogether 18 taxa were analysed for eight polymorphic plastid markers and two size-variable fragments from the nuclear internal transcribed spacer (ITS) region. In total, 60 plastid haplotypes and six ITS alleles were found among the 737 individuals analysed. No clear differentiation between populations of ssp. traunsteineri from the three regions was revealed. However, ssp. traunsteineri was genetically differentiated from Dactylorhiza baumanniana, Dactylorhiza elata, and D. majalis ssp. sphagnicola, although the majority of allotetraploid taxa remained inseparable. Judging from the degree of concerted evolution in ITS, D. majalis ssp. alpestris may be regarded as a relatively old allotetraploid, whereas ssp. baltica and ssp. purpurella may be considerably younger. Based on plastid data, the Alp region had the highest genetic diversity followed by Scandinavia and Britain. The geographic distribution of haplotypes provided support for possible refugial areas around the Alps and for several independent immigration routes into Scandinavia after the last ice age.     • The present study aimed to investigate how the impact of several factors linked to geography would shape life-history traits in a gregarious species, using the pine processionary moth (PPM) Thaumetopoea pityocampa as a model system. PPM has a wide geographical distribution over the Mediterranean Basin, and it is a strictly gregarious species throughout larval development, where the total reproductive output of each female forms a colony. We reviewed both published and unpublished data on PPM from all over its distribution in the Mediterranean Basin and extracted data on fecundity, egg size, egg parasitoid mortality, flight period, and development time. These life-history traits were then related to location, expressed as latitude and altitude, local average temperatures, and host tree species. We found that PPM fecundity increaseed with latitude, concomitant with an increase in the length of development and an earlier onset of adult flight. These results are the opposite of that found in other Lepidoptera species with a wide geographical distribution, as well as in insects in general. We propose that a large colony size in PPM is important at higher latitudes because this confers an advantage for thermoregulation and tent building in areas where larvae have to face harsher conditions during the winter, thus shifting the optimal trade-off between the number and size of eggs with latitude. However, host tree species also affected the relationship between egg number and size and the optimal outcome of these traits is likely a compromise between different selection pressures.     • In this investigation, we have collected family-structured data from a partly self-compatible, outcrossing population of Brassica cretica to estimate and compare the effects of one-generation selfing on different types of characters. Inbreeding not only depressed characters that should be positively correlated with fitness irrespective of habitat, e.g. germinability, leaf number and inflorescence size, but also resulted in later flowering, smaller and more asymmetric flowers, and an increased production of basal branches. Population-level estimates of inbreeding depression were similar in magnitude to estimates reported in other wild plant species. There was a tendency for direct components of fitness to exhibit a stronger response to inbreeding than other characters, but only when the differences between selfed and outbred offspring were measured in standard deviation units. Family-level estimates of inbreeding depression were weakly correlated across characters. Given these and other observations, we hypothesize that the genetic basis of inbreeding depression varies across the life cycle and that changes in local inbreeding will lead to shifts in the mean phenotypes of B. cretica populations. However, judging from data on current levels of population divergence, quite large changes in inbreeding will be required to influence large-scale patterns of variation in this species.   • Species-specific song convergence in a moving hybrid zone between two passerines

    • Moving hybrid zones are receiving increasing attention. However, so far little is known about the proximate mechanisms underlying these movements. Signalling behaviour, by individuals engaged in interspecific sexual and aggressive interactions, may play a crucial role. In this study, we investigated song variation within a moving hybrid zone between two warblers, Hippolais polyglotta and H. icterina. In these species, song is involved in interspecific territoriality and, probably, in mixed pairings. We showed that allopatric populations of the two species are clearly acoustically differentiated. However, interspecific differences faded out in sympatry as a result of an overall pattern of convergence. Unexpectedly, the two species converged for different song parameters, namely temporal parameters for H. icterina and syntax for H. polyglotta. Hybridization and interspecific competition could explain convergence in H. icterina. Instead, in H. polyglotta we suggest that local adaptation to habitat and interspecific learning might contribute to convergence. We particularly stress that cross-species learning, by maintaining high levels of interspecific interactions, may influence the movement of the zone. (C) 2003 The Linnean Society of London.

    • We investigated allozyme variation in 34 populations of the perennial herb Silene nutans from Sweden and northern Finland, areas that were ice-covered during the last (Weichselian) glaciation. The present geographical structure of genetic variation in S. nutans in Sweden and northern Finland appears to have been mainly shaped by ancient historical processes. Patterns of variation in allele frequencies suggest two major postglacial immigration routes into Sweden, with populations entering the area from both the south and the east and forming a contact zone with admixed populations in central Sweden. While estimates of within-population genetic diversity and allelic richness are significantly correlated with present population size and geographical position (latitude), population size is not correlated with latitude. Low genetic diversity in the northern populations is more likely to have resulted from ancient stochastic events during the process of immigration than from recent population fragmentation. F-IS values are high and increase with latitude. Evidence of recent bottlenecks was detected in several southern Swedish populations: these can be interpreted in terms of population fragmentation as a result of anthropogenic disturbance.     • Sexual selection against immigrants is a mechanism that can regulate premating isolation between populations but, so far, few field studies have examined whether males can discriminate between immigrant and resident females. Males of the damselfly Calopteryx splendens show mate preferences and are able to force pre-copulatory tandems. We related male mate responses to the ecological characteristics of female origin, geographic distances between populations, and morphological traits of females to identify factors influencing male mate discrimination. Significant heterogeneity between populations in male mate responses towards females was found. In some populations, males discriminated strongly against immigrant females, whereas the pattern was reversed or nonsignificant in other populations. Immigrant females were particularly attractive to males when they came from populations with similar predation pressures and densities of conspecifics. By contrast, immigrant females from populations with strongly dissimilar predation pressures and conspecific densities were not attractive to males. Differences in the abiotic environment appeared to affect mating success to a lesser degree. This suggests that male mate discrimination is context-dependent and influenced by ecological differences between populations, a key prediction of ecological speciation theory. The results obtained in the present study suggest that gene-flow is facilitated between ecologically similar populations.     • Resource polymorphism has been suggested to be a platform for speciation. In some cases resource polymorphism depends on phenotypic plasticity but in other cases on genetic differences between morphotypes, which in turn has been suggested to be the ongoing development of a species pair. Here we study environmentally induced morphological differences in two age classes of Arctic char (Salvelinus alpinus) influencing char performance and diet in relation to resource availability. We found that structurally complex habitats with relatively lower zooplankton densities gave rise to individuals with a deeper body, and a downward positioned tip of the snout compared with individuals from structurally simple habitats with relatively higher zooplankton densities for both age classes. Environment also had an effect on foraging efficiency on zooplankton, with fish from structurally simple habitats had a higher foraging rate than fish from structurally complex habitats. Diet analyses showed that resource use in char mainly depends on the relative abundance of different resources. Therefore, to gain further understanding of resource polymorphism we suggest that future studies must include population dynamic feedbacks by the resources on the consumers.

    • The level of gene flow is an important factor influencing genetic differentiation between populations. Typically, geographic distance is considered to be the major factor limiting dispersal and should thus only influence the degree of genetic divergence at larger spatial scales. However, recent studies have revealed the possibility for small-scale genetic differentiation, suggesting that the spatial scale considered is pivotal for finding patterns of isolation by distance. To address this question, genetic and morphological differentiation were studied at two spatial scales (range 2–13 km and range 300 m to 2 km) in the perch (Perca fluviatilis L.) from the east coast archipelago of Sweden, using seven microsatellite loci and geometric morphometrics. We found highly significant genetic differentiation between sampled locations at both scales. At the larger spatial scale, the distance not affecting the level of divergence. At the small scale, however, we found subtle patterns of isolation by distance. In addition, we also found morphological divergence between locations, congruent with a spatial separation at a microgeographic scale, most likely due to phenotypic plasticity. The present study highlights the importance of geographical scale and indicates that there might be a disparity between the dispersal capacity of a species and the actual movement of genes. Thus, how we view the environment and possible barriers to dispersal might have great implications for our ability to fully understand the evolution of genetic differentiation, local adaptation, and, in the end, speciation.

    • Crypsis and aposematism are often regarded as two opposite protective strategies. However, there is large variation in prey appearance within both strategies. In this article, we investigated the conspicuousness of the aposematic red-and-black firebug, Pyrrhocoris apterus, by presenting images of natural and digitally manipulated phenotypes in their natural habitat on a computer screen to human predators, and comparing the detection times. We asked whether the natural colour pattern can be made more or less conspicuous by rearranging the spatial distribution of colour elements. Hence, we created a phenotype in which the black colour elements were moved to the body outline to test for a possible disruptive effect. In the black and red manipulations, we removed one of the two colours, creating two uniform colour variants. We found that some of our manipulations increased, but none reduced, the detection time significantly; this indicates that the naturally coloured firebug is highly conspicuous. The detection time varied among backgrounds and there was a significant relationship between detection time and chromatic similarity between the bug and the background for the natural and black phenotypes. Although background colour composition has an important effect on the signal, we argue that the coloration of P. apterus has evolved for high conspicuousness.

    • Rapid evolutionary change is increasingly being recognized as commonplace, but the evolutionary consequences for species and ecosystems under human-induced selection regimes have not been explored in detail, although many species occur in such environments. In a common garden experiment and with amplified fragment length polymorphism markers, we examined whether genetic differentiation has taken place between spatially intermixed populations of creeping thistles Cirsium arvense (Asteraceae) collected from a natural habitat (maritime shores), a semi-natural habitat (road verges) and arable fields under two management regimes: conventional and organic farming. Populations of C. arvense have altered genetically and locally adapted their growth patterns with changed land use. Although plants from different habitats showed similar total biomass production, shoot and root production was higher for maritime populations, suggesting selection for increased competitive ability. Competitive ability then declined in the order semi-natural, conventional farms and organic farms. Thistles in arable fields may be more selected for tolerance against disturbances from herbicides and mechanical weed control. In addition, early shoot sprouting and genetic analysis showed differentiation between plants originating from conventional farms and farms that were converted to organic 9–30 years ago, suggesting some adaptation to altered crop cultivation practices

  • Population history and genetic structure of a circumpolar species : the arctic fox

    • The circumpolar arctic fox Alopex lagopus thrives in cold climates and has a high migration rate involving long-distance movements. Thus, it differs from many temperate taxa that were subjected to cyclical restriction in glacial refugia during the Ice Ages. We investigated population history and genetic structure through mitochondrial control region variation in 191 arctic foxes from throughout the arctic. Several haplotypes had a Holarctic distribution and no phylogeographical structure was found. Furthermore, there was no difference in haplotype diversity between populations inhabiting previously glaciated and unglaciated regions. This suggests current gene flow among the studied populations, with the exception of those in Iceland, which is surrounded by year-round open water. Arctic foxes have often been separated into two ecotypes: ‘lemming’ and ‘coastal’. An analysis of molecular variance suggested particularly high gene flow among populations of the ‘lemming’ ecotype. This could be explained by their higher migration rate and reduced fitness in migrants between ecotypes. A mismatch analysis indicated a sudden expansion in population size around 118 000 BP, which coincides with the last interglacial. We propose that glacial cycles affected the arctic fox in a way opposite to their effect on temperate species, with interglacials leading to short-term isolation in northern refugia.

    • We studied gene flow and bottleneck events in the population history of locally isolated citril finches endemic to European mountains. For the present study, we used two genetic markers with different rates of evolution: a fast evolving mitochondrial marker (ATPase6/8) and a more slowly evolving nuclear marker (02401). Populations north of the Pyrenees showed in general fewer haplotypes and a considerable lower nucleotide and gene diversity than the Iberian populations. Unexpectedly, we found very little genetic variability in the fast evolving mitochondrial marker, arguing for a strong and relatively recent bottleneck event in the species population history. This pattern potentially reflects a sudden decrease of crucial resources during Mid-Holocene (mountain pine, Scots pine, and black pine) and a subsequent breakdown of the population. The bottleneck could also have been caused or coincide with a selective sweep in the mitochondrion. By contrast, the slowly evolving nuclear marker showed a much higher variability. This marker probably reflects major gene flow along a potential expansion pathway from the Eastern Pyrenees, northwards to the populations of Central Europe, and southwards to the more fragmented populations of central and southern Spain. The population of the Western Pyrenees (Navarra) appears to be cut-off from this major gene flow and our data indicate a certain degree of partial isolation, probably reflecting more ancient events (e.g. the separation in distinct refuge sites during the last glacial maximum).

    • Understanding the ultimate causes for the presence of polymorphisms within populations requires knowledge of how the expression of discrete morphs is regulated. In the present study, we explored the determination mechanism of a colour dimorphism in larvae of the butterfly Pararge xiphia (Satyrinae: Nymphalidae) with the ultimate aim of understanding its potential adaptive value. Last-instar larvae of P. xiphia develop into either a green or a brown morph, although all individuals are invariably green during the preceding three instars. A series of laboratory experiments reveal that morph development is strongly environmentally dependent and not the result of alternative alleles at one locus. Photoperiod, temperature, and in particular larval density, all influenced morph determination. The strong effect of a high larval density in inducing the brown morph parallels other known cases of density-dependent melanization in Lepidopteran larvae. Because melanization is often correlated with increased immune function, this type of determination mechanism is expected to be adaptive. However, the ecology and behaviour of P. xiphia larvae suggests that increased camouflage under high-density conditions may be an additional adaptive explanation. We conclude that the colour dimorphism of P. xiphia larvae is determined by a developmental threshold that is influenced both by heredity and by environmental conditions, and that selection for increased immune function and camouflage under high-density conditions may be responsible for maintaining the dimorphism. © 2009 The Linnean Society of London, Biological Journal of the Linnean Society, 2009, 98, 256–266.

    • The aim of the present paper is to explore the role of the character in phylogenetic systematics. I argue that too much emphasis is put on particular characters rather than congruence both in the choice of phylogenetic hypotheses and in taxonomic decisions. This means that the logical priority of the tree over the characters is neglected. To a large extent, this is a result of not paying enough attention to the individuality thesis which states that clades are historical individuals and hence contingent in nature.

    • In many birds, carotenoids have dual functions as irreversible plumage pigments and as physiologically essential vitamins and antioxidants. They must be obtained through the diet and may therefore be a limiting resource, a constraint that is likely to vary with factors such as sex, habitat, and time of year. In the present study, we investigated signs of carotenoid limitation in great tits, Parus major, in relation to sex, season, year, and within an urban versus a rural habitat. The two main carotenoids, lutein and zeaxanthin, were analysed by high-performance liquid chromatography in the plasma and in the yellow carotenoid-based breast feathers. We found that plasma carotenoid concentrations were significantly influenced by sex, season, and year, but not by urban versus rural habitat. At moult, plasma concentration was positively correlated with feather pigmentation, independent of body condition and sex. During the breeding season, however, circulating carotenoid concentrations were negatively related to the feather pigmentation (i.e. from previous autumn moult). We suggest that great tits are carotenoid deprived before leaf emergence, and that carotenoid utilization and limitations are sex-specific, but that there are neither any obvious honesty-maintaining costs of pigmentation, nor any fitness consequences of the colour variation.

    • Although, in some insect taxa, wing shape is remarkably invariant, the wings of Anisopteran dragonflies show considerable variation among genera. Because wing shape largely determines the high energetic costs of flight, it may be expected that interspecific differences are partly due to selection. In the present study, we examined the roles of long-distance migration and high-manoeuvrability mate guarding in shaping dragonfly wings, using a phylogeny-based comparative method, and geometric morphometrics to quantify wing shape. The results obtained show that migration affects the shape of both front and hind wings, and suggest that mate guarding behaviour may also have an effect, especially on the front wing. These effects on front wing shape are at least partly independent. Our findings are interesting when compared with the geographically widespread and ecologically diverse dipterans Acalyptratae (including the genus Drosophila). The wings in that group are similar in function and structure, but show strikingly low levels of interspecific variation.

    • Faroe house mice are a 'classic' system of rapid and dramatic morphological divergence highlighted by J. S. Huxley during the development of the Modern Synthesis. In the present study, we characterize these charismatic mice using modern molecular techniques, examining specimens from all Faroe islands occupied by mice. The aims were to classify the mice within the modern house mouse taxonomy (i.e. as either Mus musculus domesticus or Mus musculus musculus) using four molecular markers and a morphological feature, and to examine the genetic diversity and possible routes of colonization using mitochondrial (mt) control region DNA sequences and microsatellite data (15 loci). Mice on the most remote islands were characterized as M. m. domesticus and exhibited exceptionally low genetic diversity, whereas those on better connected islands were more genetically diverse and had both M. m. musculus and M. m. domesticus genetic elements, including one population which was morphologically M. m. musculus-like. The mtDNA data indicate that the majority of the mice had their origins in south-western Norway (or possibly southern Denmark/northern Germany), and probably arrived with the Vikings, earlier than suggested by Huxley. The M. m. musculus genetic component appears to derive from recent mouse immigration from Denmark.

    • New Caledonian crows (Corvus moneduloides) are prolific tool users in captivity and in the wild, and have an inherited predisposition to express tool-oriented behaviours. To further understand the evolution and development of tool use, we compared the development of object manipulation in New Caledonian crows and common ravens (Corvus corax), which do not routinely use tools. We found striking qualitative similarities in the ontogeny of tool-oriented behaviour in New Caledonian crows and food-caching behaviour in ravens. Given that the common ancestor of New Caledonian crows and ravens was almost certainly a caching species, we therefore propose that the basic action patterns for tool use in New Caledonian crows may have their evolutionary origins in caching behaviour. Noncombinatorial object manipulations had similar frequencies in the two species. However, frequencies of object combinations that are precursors to functional behaviour increased in New Caledonian crows and decreased in ravens throughout the study period, ending 6 weeks post-fledging. These quantitative observations are consistent with the hypothesis that New Caledonian crows develop tool-oriented behaviour because of an increased motivation to perform object combinations that facilitate the necessary learning.

    • An explanation for female multiple mating when males offer no material benefits but sperm remains elusive, largely because of a lack of empirical support for the genetic benefits hypothesis. We used 21 microsatellite markers to test for multiple paternities among 88 litters of roe deer, Capreolus capreolus, and to investigate the inbreeding avoidance hypothesis as a potential mechanism for the evolution of female multiple mating. From paternity analyses, we found that 13.5% of polytocous litters were sired by more than one male. We also found that a half-sib relationship was more likely than a full-sib relationship for 20.5% of all litters. This is the first report of multiple paternities in a territorial ungulate species. In support of the inbreeding avoidance hypothesis, we found that parents who were strongly related produced offspring with lower individual heterozygosity that survived less well during their first summer than fawns with unrelated parents. In addition, fawns from multiple paternity litters survived their first summer better than fawns from single paternity litters. However, it remains unclear whether all female multiple paternity events in this species are provoked by an initial consanguineous mating.
