Vi börjar med att beskriva IM och hur det kan användas i utbildningssammanhang. Därefter beskriver vi hur informell coachning går till, pedagogiska utmaningar, hur coachning kan organiseras och vilka verktyg som kan användas. Avslutningsvis reflekterar vi över hur coachning kan bidra till lärande.

    • Synthesis of the nonbilayer-prone α-monoglucosyldiacylglycerol (MGlcDAG) is crucial for bilayer packing properties and the                     lipid surface charge density in the membrane ofAcholeplasma laidlawii. The gene for the responsible, membrane-bound glucosyltransferase (alMGS) (EC 2.4.1.157) was sequenced and functionally cloned in Escherichia coli, yielding MGlcDAG in the recombinants. Similar amino acid sequences were encoded in the genomes of several Gram-positive                     bacteria (especially pathogens), thermophiles, archaea, and a few eukaryotes. All of these contained the typical EX7E catalytic motif of the CAZy family 4 of α-glycosyltransferases. The synthesis of MGlcDAG by a close sequence analog from                      Streptococcus pneumoniae (spMGS) was verified by polymerase chain reaction cloning, corroborating a connection between sequence and functional similarity                     for these proteins. However, alMGS and  spMGS varied in dependence on anionic phospholipid activators phosphatidylglycerol                     and cardiolipin, suggesting certain regulatory differences. Fold predictions strongly indicated a similarity for alMGS (and                     spMGS) with the two-domain structure of the E. coli MurG cell envelope glycosyltransferase and several amphipathic membrane-binding segments in various proteins. On the basis                     of this structure, the alMGS sequence charge distribution, and anionic phospholipid dependence, a model for the bilayer surface                     binding and activity is proposed for this regulatory enzyme.

    • This paper develops a typology of incumbent resistance to disruptive new logics. Although scholars of institutional change have studied public forms of resistance, a comprehensive understanding of how incumbents oppose disruptive new logics also necessitates attention to the quiet forms of resistance. Conceptualizing resistance as a form of institutional work, we draw on insights from the literatures on institutional change and social movements to develop a typology of public, hidden, and implicit resistance to disruptive logics. Broadening the understanding of resistance work to include its quiet forms enables institutional scholars to understand how field incumbents resist disruption and why such efforts may be successful. A broadened analysis of incumbent resistance is vital for theorizing the past and future resilience of some of the most central institutions of modern society, such as the carbon-based economy and democracy.

    • In membranes of Acholeplasma laidlawii two consecutively acting glucosyltransferases, the (i) alpha-monoglucosyl-diacylglycerol. (MGlcDAG) synthase (aIMGS) (EC 2.4.1.157) and the (ii) alpha-diglucosyl-DAG (DGlcDAG) synthase (alDGS) (EC 2.4.1.208), are involved in maintaining (i) a certain anionic lipid surface charge density and (ii) constant nonbilayer/bilayer conditions (curvature packing stress), respectively. Cloning of the aIDGS gene revealed related uncharacterized sequence analogs especially in several Gram-positive pathogens, thermophiles and archaea, where the encoded enzyme function of a potential Streptococcus pneumoniae DGS gene (cpoA) was verified. A strong stimulation of aIDGS by phosphatidylglycerol (PG), cardiolipin, or nonbilayer-prone 1,3-DAG was observed, while only PG stimulated CpoA. Several secondary structure prediction and fold recognition methods were used together with SWISS-MODEL to build three-dimensional model structures for three MGS and two DGS lipid glycosyltransferases. Two Escherichia coli proteins with known structures were identified as the best templates, the membrane surface-associated two-domain glycosyltransferase MurG and the soluble GlcNAc epimerase. Differences in electrostatic surface potential between the different models and their individual domains suggest that electrostatic interactions play a role for the association to membranes. Further support for this was obtained when hybrids of the N- and C-domain, and full size alMGS with green fluorescent protein were localized to different regions of the E. coli inner membrane and cytoplasm in vivo. In conclusion, it is proposed that the varying abilities to bind, and sense lipid charge and curvature stress, are governed by typical differences in charge (pI values), amphiphilicity, and hydrophobicity for the N- and (catalytic) C-domains of these structurally similar membrane-associated enzymes.

    • This report is related to the BONUS project “Nutrient Cocktails in COAstal zones of the Baltic Sea” alias COCOA. The aim of BONUS COCOA is to investigate physical, biogeochemical and biological processes in a combined and coordinated fashion to improve the understanding of the interaction of these processes on the removal of nutrients along the land -sea interface. The report is especially related to BONUS COCOA WP 6 in which the main objective is extrapolation of results from the BONUS COCOA learning sites to coastal sites around the Baltic Sea in general. Specific objectives of this deliverable (D6.4) were to connect observed process rates with modelling data and ecosystem characteristics.In the report we made statistical analyses of observations from BONUS COCOA study sites together with results from the Swedish Coastal zone Model (SCM). Eight structural variables (water depth, temperature, salinity, bottom water concentrations of oxygen, ammonium, nitrate and phosphate, as well as nitrogen content in sediment) were found common to both the experimentally determined and the model data sets. The observed process rate evaluated in this report was denitrification. In addition regressions were tested between observed denitrification rates and several structural variables (lat itude, longitude, depth, light, temperature, salinity, grain class, porosity, loss of ignition, sediment organic carbon, total nitrogen content in the sediment, sediment carbon/nitrogen-ratio, sediment chlorphyll-a as well as bottom water concentrations of oxygen, ammonium, nitrate, and dissolved inorganic phosphorus and silicate) for pooled data from all learning sites.The statistical results showed that experimentally determined multivariate data set from the shallow, illuminated stations was mainly found to be similar to the multivariate data set produced by the SCM model. Generally, no strong correlations of simple relations between observed denitrification and available structural variables were found for data collected from all the learning sites. We found some non-significant correlation between denitrification rates and bottom water dissolved inorganic phosphorous and dissolved silica but the reason behind the correlations is not clear.We also developed and evaluated a theory to relate process rates to monitoring data and nutrient retention. The theoretical analysis included nutrient retention due to denitrification as well as burial of phosphorus and nitrogen. The theory of nutrient retention showed good correlations with model results. It was found that area-specific nitrogen and phosphorus retention capacity in a sub-basin depend much on mean water depth, water residence time, basin area and the mean nutrient concentrations in the active sediment layer and in the water column.

    • The aim of this study is to describe and explore how instant messaging (IM) can be used to support informal math coaching. We have studied two projects where university students use IM to coach K-12 students in mathematics. The coaches were interviewed with a focus on how informal coaching works by examining coaching challenges, how coaching can be organized, whether coaching should be anonymous or personal, which tools can be used and how informal math coaching supports learning. Research shows that generating and answering questions are important in the process of understanding and learning, which means that both the students and the coaches can learn math through this type of project. The coaches perceive informal math coaching as complementing online math forums. For students to learn effectively, the coaches need to be able to interpret the students' competence level in order to coach on a level that is within their development zone. It seems particularly challenging to coach at the right level when using IM and, therefore, it is important to establish a personal relationship with the students.

    • The glucocorticoid receptor (GR) is a nuclear hormone receptor that regulates key genes controlling development, metabolism, and the immune response. GR agonists are efficacious for treatment of inflammatory, allergic, and immunological disorders. Steroid hormone binding to the ligand-binding domain (LBD) of GR is known to change the structural and dynamical properties of the receptor, which in turn control its interactions with DNA and various co-regulators and drive the pharmacological response. Previous biophysical studies of the GR LBD have required the use of mutant forms to overcome issues with limited protein stability and high aggregation propensity. However, these mutant variants are known to also influence the functional response of the receptor. Here we report a successful protocol for protein expression, purification, and NMR characterization of the wildtype human GR LBD. We achieved chemical shift assignments for 90% of the LBD backbone resonances, with 216 out of 240 non-proline residues assigned in the 1H–15N TROSY spectrum. These advancements form the basis for future investigations of allosteric effects in GR signaling.

    • Increasingly, healthcare systems around the globe are looking to transition toward person-centered models of care. However, how to effectively support this complex transition is not clear. Here we forward service design as a key driver to aid in catalyzing this transformation. In this chapter, we integrate literature on service design (SD) and person-centered care (PCC) to better understand how a SD approach can aid in the transition toward PCC. Synthesized from existing literature, this chapter offers a framework for transitioning from the biomedical model toward PCC, highlighting key changes across four dimensions: contexts, roles, processes, and outcomes. We then show the alignment between PCC and SD across these dimensions and elucidate how SD can help to catalyze related changes with the support of specific methods. In doing so, this chapter offers a guide for healthcare practitioners looking to use SD to support the transformation toward PCC and builds a platform for future research at the intersection of SD and PCC.
