  • Rotation and pulsation in Ap stars : first light results from TESS sectors 1 and 2

    • We present the first results from the Transiting Exoplanet Survey Satellite (TESS) on the rotational and pulsational variability of magnetic chemically peculiar A-type stars. We analyse TESS 2-min cadence data from sectors 1 and 2 on a sample of 83 stars. Five new rapidly oscillating Ap (roAp) stars are announced. One of these pulsates with periods around 4.7 min, making it the shortest period roAp star known to date. Four out of the five new roAp stars are multiperiodic. Three of these and the singly periodic one show the presence of rotational mode splitting. Individual frequencies are provided in all cases. In addition, seven previously known roAp stars are analysed. Additional modes of oscillation are found in some stars, while in others we are able to distinguish the true pulsations from possible aliases present in the ground-based data. We find that the pulsation amplitude in the TESS filter is typically a factor of 6 smaller than that in the B filter, which is usually used for ground-based observations. For four roAp stars we set constraints on the inclination angle and magnetic obliquity, through the application of the oblique pulsator model. We also confirm the absence of roAp-type pulsations down to amplitude limits of 6 and 13 mu mag, respectively, in two of the best characterized non-oscillating Ap (noAp) stars. We announce 27 new rotational variables along with their rotation periods, and provide different rotation periods for seven other stars. Finally, we discuss how these results challenge state-of-the-art pulsation models for roAp stars.

  • Randomized controlled trial of atorvastatin in mild to moderate Alzheimer disease : LEADe

    • BACKGROUND: There is some evidence that statins may have a protective and symptomatic benefit in Alzheimer disease (AD). The LEADe study is a randomized controlled trial (RCT) evaluating the efficacy and safety of atorvastatin in patients with mild to moderate AD.METHODS: This was an international, multicenter, double-blind, randomized, parallel-group study. Subjects had mild to moderate probable AD (Mini-Mental State Examination score 13-25), were aged 50-90 years, and were taking donepezil 10 mg daily for > or 3 months prior to screening. Entry low-density lipoprotein cholesterol levels (LDL-C) were > 95 and < 195 mg/dL. Patients were randomized to atorvastatin 80 mg/day or placebo for 72 weeks followed by a double-blind, 8-week atorvastatin withdrawal phase. Coprimary endpoints were changes in cognition (Alzheimer's Disease Assessment Scale-Cognitive Subscale [ADAS-Cog]) and global function (Alzheimer's Disease Cooperative Study Clinical Global Impression of Change [ADCS-CGIC]) at 72 weeks.RESULTS: A total of 640 patients were randomized in the study. There were no significant differences in the coprimary endpoints of ADAS-cog or ADCS-CGIC or the secondary endpoints. Atorvastatin was generally well-tolerated.CONCLUSIONS: In this large-scale randomized controlled trial evaluating statin therapy as a treatment for mild to moderate Alzheimer disease, atorvastatin was not associated with significant clinical benefit over 72 weeks. This treatment was generally well-tolerated without unexpected adverse events. Classification of evidence: This study provides Class II evidence that intensive lipid lowering with atorvastatin 80 mg/day in patients with mild to moderate probable Alzheimer disease (aged 50-90), taking donepezil, with low-density lipoprotein cholesterol levels between 95 and 195 mg/dL over 72 weeks does not benefit cognition (as measured by Alzheimer's Disease Assessment Scale-Cognitive Subscale) (p = 0.26) or global function (as measured by Alzheimer's Disease Cooperative Study Clinical Global Impression of Change) (p = 0.73) compared with placebo.

  • TESS Cycle 2 observations of roAp stars with 2-min cadence data

    • We present the results of a systematic search of the Transiting Exoplanet Survey Satellite (TESS) 2-min cadence data for new rapidly oscillating Ap (roAp) stars observed during the Cycle 2 phase of its mission. We find seven new roAp stars previously unreported as such and present the analysis of a further 25 roAp stars that are already known. Three of the new stars show multiperiodic pulsations, while all new members are rotationally variable stars, leading to almost 70 per cent (22) of the roAp stars presented being α2 CVn-type variable stars. We show that targeted observations of known chemically peculiar stars are likely to overlook many new roAp stars, and demonstrate that multiepoch observations are necessary to see pulsational behaviour changes. We find a lack of roAp stars close to the blue edge of the theoretical roAp instability strip, and reaffirm that mode instability is observed more frequently with precise, space-based observations. In addition to the Cycle 2 observations, we analyse TESS data for all-known roAp stars. This amounts to 18 further roAp stars observed by TESS. Finally, we list six known roAp stars that TESS is yet to observe. We deduce that the incidence of roAp stars amongst the Ap star population is just 5.5  per cent, raising fundamental questions about the conditions required to excite pulsations in Ap stars. This work, coupled with our previous work on roAp stars in Cycle 1 observations, presents the most comprehensive, homogeneous study of the roAp stars in the TESS nominal mission, with a collection of 112 confirmed roAp stars in total.

  • The Beaker phenomenon and the genomic transformation of northwest Europe

    • From around 2750 to 2500 bc, Bell Beaker pottery became widespread across western and central Europe, before it disappeared between 2200 and 1800 bc. The forces that propelled its expansion are a matter of long-standing debate, and there is support for both cultural diffusion and migration having a role in this process. Here we present genome-wide data from 400 Neolithic, Copper Age and Bronze Age Europeans, including 226 individuals associated with Beaker-complex artefacts. We detected limited genetic affinity between Beaker-complex-associated individuals from Iberia and central Europe, and thus exclude migration as an important mechanism of spread between these two regions. However, migration had a key role in the further dissemination of the Beaker complex. We document this phenomenon most clearly in Britain, where the spread of the Beaker complex introduced high levels of steppe-related ancestry and was associated with the replacement of approximately 90% of Britain's gene pool within a few hundred years, continuing the east-to-west expansion that had brought steppe-related ancestry into central and northern Europe over the previous centuries.

    • The breakup of Laurasia to form the Northeast Atlantic Realm disintegrated an inhomogeneous collage of cratons sutured by cross-cutting orogens. Volcanic rifted margins formed that are underlain by magma-inflated, extended continental crust. North of the Greenland-Iceland-Faroe Ridge a new rift–the Aegir Ridge–propagated south along the Caledonian suture. South of the Greenland-Iceland-Faroe Ridge the proto-Reykjanes Ridge propagated north through the North Atlantic Craton along an axis displaced ~150 km to the west of the rift to the north. Both propagators stalled where the confluence of the Nagssugtoqidian and Caledonian orogens formed an ~300-km-wide transverse barrier. Thereafter, the ~150 × 300-km block of continental crust between the rift tips–the Iceland Microcontinent–extended in a distributed, unstable manner along multiple axes of extension. These axes repeatedly migrated or jumped laterally with shearing occurring between them in diffuse transfer zones. This style of deformation continues to the present day in Iceland. It is the surface expression of underlying magma-assisted stretching of ductile continental crust that has flowed from the Iceland Microplate and flanking continental areas to form the lower crust of the Greenland-Iceland-Faroe Ridge. Icelandic-type crust which underlies the Greenland-Iceland-Faroe Ridge is thus not anomalously thick oceanic crust as is often assumed. Upper Icelandic-type crust comprises magma flows and dykes. Lower Icelandic-type crust comprises magma-inflated continental mid- and lower crust. Contemporary magma production in Iceland, equivalent to oceanic layers 2–3, corresponds to Icelandic-type upper crust plus intrusions in the lower crust, and has a total thickness of only 10–15 km. This is much less than the total maximum thickness of 42 km for Icelandic-type crust measured seismically in Iceland. The feasibility of the structure we propose is confirmed by numerical modeling that shows extension of the continental crust can continue for many tens of millions of years by lower-crustal ductile flow. A composition of Icelandic-type lower crust that is largely continental can account for multiple seismic observations along with gravity, bathymetric, topographic, petrological and geochemical data that are inconsistent with a gabbroic composition for Icelandic-type lower crust. It also offers a solution to difficulties in numerical models for melt-production by downward-revising the amount of melt needed. Unstable tectonics on the Greenland-Iceland-Faroe Ridge can account for long-term tectonic disequilibrium on the adjacent rifted margins, the southerly migrating rift propagators that build diachronous chevron ridges of thick crust about the Reykjanes Ridge, and the tectonic decoupling of the oceans to the north and south. A model of complex, discontinuous continental breakup influenced by crustal inhomogeneity that distributes continental material in growing oceans fits other regions including the Davis Strait, the South Atlantic and the West Indian Ocean.

  • Oxidation of pharmaceuticals by chlorine dioxide in biologically treated wastewater

    • Biologically treated wastewater spiked with a mixture of 56 active pharmaceutical ingredients (APIs) was treated with 0-20 mg/L chlorine dioxide (ClO2) solution in laboratory-scale experiments. Wastewater effluents were collected from two wastewater treatment plants in Sweden, one with extended nitrogen removal (low COD) and one without (high COD). About one third of the tested APIs resisted degradation even at the highest ClO2 dose (20 mg/L), while others were reduced by more than 90% at the lowest ClO2 level (0.5 mg/L). In the low COD effluent, more than half of the APIs were oxidized at 5 mg/L ClO2, while in high COD effluent a significant increase in API oxidation was observed after treatment with 8 mg/L ClO2. This study illustrates the successful degradation of several APIs during treatment of wastewater effluents with chlorine dioxide.

    • Removal of six active pharmaceutical ingredients in wastewater was investigated using chlorine dioxide (ClO2) or peracetic acid (PAA) as chemical oxidants. Four non-steroidal anti-inflammatory drugs (ibuprofen, naproxen, diclofenac and mefenamic acid) and two lipid-regulating agents (gemfibrozil and clofibric acid, a metabolite of clofibrate) were used as target substances at 40 mu g/L initial concentration. Three different wastewaters types originating from two wastewater treatment plants (WWTPs) were used. One wastewater was collected after extended nitrogen removal in activated sludge, one after treatment with high-loaded activated sludge without nitrification, and one from the final effluent from the same plant where nitrogen removal was made in trickling filters for nitrification and moving-bed biofilm reactors for denitrification following the high-loaded plant. Of the six investigated compounds, only clofibric acid and ibuprofen were not removed when treated with ClO2 up to 20 mg/L. With increasing PAA dose up to 50 mg/L, significant removal of most of the pharmaceuticals was observed except for the wastewater with the highest chemical oxygen demand (COD). This indicates that chemical oxidation with ClO2 could be used for tertiary treatment at WWTPs for active pharmaceutical ingredients, whereas PAA was not sufficiently efficient.

    • Ozonation to achieve removal of pharmaceuticals from wastewater effluents, with pH values in the upper and lower regions of the typical range for Swedish wastewater, was investigated. The main aim was to study the effects of varying pH values (6.0 and 8.0), and if small additions of H2O2 prior to ozone treatment could improve the removal and lower the reaction time. The effluents studied differed in their chemical characteristics, particularly in terms of alkalinity (65.3-427 mg center dot l(-1) HCO3-), COD (18.2-41.8 mg center dot l(-1)), DOC (6.9-12.5 mg center dot l(-1)), ammonium content (0.02-3.6 mg center dot l(-1)) and specific UV absorbance (1.78-2.76 l center dot mg(-1)center dot m(-1)). As expected, lower ozone decomposition rates were observed in the effluents at pH 6.0 compared to pH 8.0. When pH 8.0 effluents were ozonated, a higher degree of pharmaceutical removal occurred in the effluent with low specific UV absorbance. For pH 6.0 effluents, the removal of pharmaceuticals was most efficient in the effluent with the lowest organic content. The addition of H2O2 had no significant effect on the quantitative removal of pharmaceuticals but enhanced the ozone decomposition rate. Thus, H2O2 addition increased the reaction rate. In practice, this will mean that the reactor volume needed for the ozonation of wastewater effluents can be reduced.

    • AIM/HYPOTHESIS: Studies have suggested a link between insulin resistance and mitochondrial dysfunction in skeletal muscles. Our primary aim was to investigate the effect of aerobic training on mitochondrial respiration and mitochondrial reactive oxygen species (ROS) release in skeletal muscle of obese participants with and without type 2 diabetes. METHODS: Type 2 diabetic men (n = 13) and control (n = 14) participants matched for age, BMI and physical activity completed 10 weeks of aerobic training. Pre- and post-training muscle biopsies were obtained before a euglycaemic-hyperinsulinaemic clamp and used for measurement of respiratory function and ROS release in isolated mitochondria. RESULTS: Training significantly increased insulin sensitivity, maximal oxygen consumption and muscle mitochondrial respiration with no difference between groups. When expressed in relation to a marker of mitochondrial density (intrinsic mitochondrial respiration), training resulted in increased mitochondrial ADP-stimulated respiration (with NADH-generating substrates) and decreased respiration without ADP. Intrinsic mitochondrial respiration was not different between groups despite lower insulin sensitivity in type 2 diabetic participants. Mitochondrial ROS release tended to be higher in participants with type 2 diabetes. CONCLUSIONS/INTERPRETATION: Aerobic training improves muscle respiration and intrinsic mitochondrial respiration in untrained obese participants with and without type 2 diabetes. These adaptations demonstrate an increased metabolic fitness, but do not seem to be directly related to training-induced changes in insulin sensitivity.

    • Mitochondrial energy production involves the movement of protons down a large electrochemical gradient via ATP synthase located on the folded inner membrane, known as cristae. In mammalian skeletal muscle, the density of cristae in mitochondria is assumed to be constant. However, recent experimental studies have shown that respiration per mitochondria varies. Modelling studies have hypothesized that this variation in respiration per mitochondria depends on plasticity in cristae density, although current evidence for such a mechanism is lacking. In the present study, we confirm this hypothesis by showing that, in human skeletal muscle, and in contrast to the current view, the mitochondrial cristae density is not constant but, instead, exhibits plasticity with long-term endurance training. Furthermore, we show that frequently recruited mitochondria-enriched fibres have significantly increased cristae density and that, at the whole-body level, muscle mitochondrial cristae density is a better predictor of maximal oxygen uptake rate than muscle mitochondrial volume. Our findings establish an elevating mitochondrial cristae density as a regulatory mechanism for increasing metabolic power in human skeletal muscle. We propose that this mechanism allows evasion of the trade-off between cell occupancy by mitochondria and other cellular constituents, as well as improved metabolic capacity and fuel catabolism during prolonged elevated energy requirements.
