    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • A rare class of supernovae (SNe) is characterized by strong interaction between the ejecta and several solar masses of circumstellar matter (CSM) as evidenced by strong Balmer-line emission. Within the first few weeks after the explosion, they may display spectral features similar to overluminous Type Ia SNe, while at later phase their observation properties exhibit remarkable similarities with some extreme case of Type IIn SNe that show strong Balmer lines years after the explosion. We present polarimetric observations of SN 2018evt obtained by the ESO Very Large Telescope from 172 to 219 d after the estimated time of peak luminosity to study the geometry of the CSM. The non-zero continuum polarization decreases over time, suggesting that the mass-loss of the progenitor star is aspherical. The prominent H α emission can be decomposed into a broad, time-evolving component and an intermediate-width, static component. The former shows polarized signals, and it is likely to arise from a cold dense shell (CDS) within the region between the forward and reverse shocks. The latter is significantly unpolarized, and it is likely to arise from shocked, fragmented gas clouds in the H-rich CSM. We infer that SN 2018evt exploded inside a massive and aspherical circumstellar cloud. The symmetry axes of the CSM and the SN appear to be similar. SN 2018evt shows observational properties common to events that display strong interaction between the ejecta and CSM, implying that they share similar circumstellar configurations. Our preliminary estimate also suggests that the circumstellar environment of SN 2018evt has been significantly enriched at a rate of ∼0.1 M⊙ yr−1 over a period of >100 yr.

    • BACKGROUND: Gelatin is traditionally produced from mammals and widely applied in the food industry. The production is tedious, time-consuming and environment-unfriendly, while the application is restricted because of zoonosis risk and religious sentiment.RESULTS: Gelatin was extracted by hot water from sturgeon swim bladder after defatting with alcohol and hexane. The yield reached to 94.15% under the optimized conditions of 50 degrees C, 30 min and 10 mL g(-1). Its amino acid and subunit profiles were similar to type I collagen. Compared to commercial porcine, bovine and piscine gelatins, it exhibited higher whiteness (3.38), emulsion activity (171.76 m(2) g(-1)), gel strength (853.23 g), water-holding capacity (92.37%) and viscoelasticity (0.03). But the transmittance (40.56% at 450 nm and 59.07% at 620 nm), emulsion stability (30.09 min), foam expansion (203.00) and stability (26.92), gelling (16.88 degrees C) and melting temperature (21.80 degrees C) were lower. While the pH (6.87) and viscosity (28.60 mPa s) were moderate. Moreover, it made better hydrogels and nanofibers.CONCLUSION: Gelatin was extracted from sturgeon swim bladder using a clean and efficient approach, and exhibited unique properties and great potential for the food industry.

    • Bone resorption occurs after bone grafting, however, contemporaneous reconstruction of the innervation of the bone graft is a potential treatment to maintain the bone mass of the graft. The innervation of bone is an emerging research topic. To understand the potential molecular mechanisms of bone innervation after bone grafting, we collected normal iliac bone tissue as well as bone grafts with or without innervation from nine patients 1 year after surgery and performed RNA sequencing. We identified differentially expressed genes) from these samples and used the gene ontology and Kyoto Encyclopedia of Genes and Genomes databases for functional enrichment and signaling pathway analysis. In parallel, we established protein-protein interaction networks to screen functional modules. Based on bioinformatic results, we validated in vitro the osteogenic differentiation potential of rat bone marrow mesenchymal stem cells (BMMSCs) after calcitonin gene-related peptide (CGRP) stimulation and the expression of p38 MAPK and Wnt6/β-catenin pathways during osteogenesis. Our transcriptome analysis of bone grafts reveals functional modules and signaling pathways of innervation which play a vital role in the structural and functional integration of the bone graft. Simultaneously, we demonstrate that CGRP regulates the differentiation of BMMSCs through p38 MAPK and Wnt6/β-catenin.

    • Immunoglobulin family and carbohydrate vascular addressins encoded by Madcam1 and St6gal1 control lymphocyte homing into intestinal tissues, regulating immunity and inflammation. The addressins are developmentally programmed to decorate endothelial cells lining gut post-capillary and high endothelial venules (HEV), providing a prototypical example of organ- and segment-specific endothelial specialization. We identify conserved NKX-COUP-TFII composite elements (NCCE) in regulatory regions of Madcam1 and St6gal1 that bind intestinal homeodomain protein NKX2-3 cooperatively with venous nuclear receptor COUP-TFII to activate transcription. The Madcam1 element also integrates repressive signals from arterial/capillary Notch effectors. Pan-endothelial COUP-TFII overexpression induces ectopic addressin expression in NKX2-3+ capillaries, while NKX2-3 deficiency abrogates expression by HEV. Phylogenetically conserved NCCE are enriched in genes involved in neuron migration and morphogenesis of the heart, kidney, pancreas and other organs. Our results define an NKX-COUP-TFII morphogenetic code that targets expression of mucosal vascular addressins.

  • Evidence for Late-stage Eruptive Mass Loss in the Progenitor to SN2018gep, a Broad-lined Ic Supernova : Pre-explosion Emission and a Rapidly Rising Luminous Transient

    • We present detailed observations of ZTF18abukavn (SN2018gep), discovered in high-cadence data from the Zwicky Transient Facility as a rapidly rising (1.4 +/- 0.1 mag hr(-1)) and luminous (M-g,M- peak = -20 mag) transient. It is spectroscopically classified as a broad-lined stripped-envelope supernova (Ic-BL SN). The high peak luminosity (L-bol greater than or similar to 3 x 10(44) erg s(-1)), the short rise time (t(rise) = 3 days in g band), and the blue colors at peak (g-r similar to -0.4) all resemble the high-redshift Ic-BL iPTF16asu, as well as several other unclassified fast transients. The early discovery of SN2018gep (within an hour of shock breakout) enabled an intensive spectroscopic campaign, including the highest-temperature (T-eff greater than or similar to 40,000 K) spectra of a stripped-envelope SN. A retrospective search revealed luminous (M-g similar to M-r approximate to -14 mag) emission in the days to weeks before explosion, the first definitive detection of precursor emission for a Ic-BL. We find a limit on the isotropic gamma-ray energy release E-gamma,E- iso < 4.9 x 10(48) erg, a limit on X-ray emission L-X < 10(40) erg s(-1), and a limit on radio emission nu L-v less than or similar to 10(37) erg s(-1). Taken together, we find that the early (< 10 days) data are best explained by shock breakout in a massive shell of dense circumstellar material (0.02 M-circle dot) at large radii (3 x 10(14) cm) that was ejected in eruptive pre-explosion mass-loss episodes. The late-time (> 10 days) light curve requires an additional energy source, which could be the radioactive decay of Ni-56.

    • Lanthanide upconversion nanoparticles (UCNPs) have beenextensivelyexplored as biomarkers, energy transducers, and information carriersin wide-ranging applications in areas from healthcare and energy toinformation technology. In promoting the brightness and enrichingthe functionalities of UCNPs, core-shell structural engineeringhas been well-established as an important approach. Despite its importance,a strong limiting issue has been identified, namely, cation intermixingin the interfacial region of the synthesized core-shell nanoparticles.Currently, there still exists confusion regarding this destructivephenomenon and there is a lack of facile means to reach a delicatecontrol of it. By means of a new set of experiments, we identify andprovide in this work a comprehensive picture for the major physicalmechanism of cation intermixing occurring in synthesis of core-shellUCNPs, i.e., partial or substantial core nanoparticle dissolutionfollowed by epitaxial growth of the outer layer and ripening of theentire particle. Based on this picture, we provide an easy but effectiveapproach to tackle this issue that enables us to produce UCNPs withhighly boosted optical properties.

  • Optimal charging plan for electric bus considering time-of-day electricity tariff

    • Purpose: The purpose of this study is to develop an optimization method for charging plans with the implementation of time-of-day (TOD) electricity tariff, to reduce electricity bill. Design/methodology/approach: Two optimization models for charging plans respectively with fixed and stochastic trip travel times are developed, to minimize the electricity costs of daily operation of an electric bus. The charging time is taken as the optimization variable. The TOD electricity tariff is considered, and the energy consumption model is developed based on real operation data. An optimal charging plan provides charging times at bus idle times in operation hours during the whole day (charging time is 0 if the bus is not get charged at idle time) which ensure the regular operation of every trip served by this bus. Findings: The electricity costs of the bus route can be reduced by applying the optimal charging plans. Originality/value: This paper produces a viable option for transit agencies to reduce their operation costs.

    • Advances in cosmic microwave background (CMB) science depend on increasing the number of sensitive detectors observing the sky. New instruments deploy large arrays of superconducting transition-edge sensor (TES) bolometers tiled densely into ever larger focal planes. High multiplexing factors reduce the thermal loading on the cryogenic receivers and simplify their design. We present the design of focal-plane modules with an order of magnitude higher multiplexing factor than has previously been achieved with TES bolometers. We focus on the novel cold readout component, which employs microwave SQUID multiplexing (μmux). Simons Observatory will use 49 modules containing 70,000 bolometers to make exquisitely sensitive measurements of the CMB. We validate the focal-plane module design, presenting measurements of the readout component with and without a prototype detector array of 1728 polarization-sensitive bolometers coupled to feedhorns. The readout component achieves a 95% yield and a 910 multiplexing factor. The median white noise of each readout channel is 65 pA √Hz . This impacts the projected SO mapping speed by <8%, which is less than is assumed in the sensitivity projections. The results validate the full functionality of the module. We discuss the measured performance in the context of SO science requirements, which are exceeded.
