    • In the first production run of the WASA experiment at COSY, the eta decay into three neutral pions was measured in proton-proton interactions at a proton beam kinetic energy of 1.4 GeV. The Dalitz plot of the three pious was Studied using 1.2 x 10(5) fully reconstructed events. and the quadratic slope parameter alpha was determined to be -0.027 +/- 0.008(stat) +/- 0.005(syst).     • Exclusive and kinematically complete measurements of the double pionic fusion to He-3 have been performed in the energy region of the so-called ABC effect, which denotes a pronounced low-mass enhancement in the pi pi-invariant mass spectrum. The experiments were carried out with the WASA detector setup at COSY (the cooler synchrotron at Forschungszentrum Julich). Similar to the observations in the basic pn -> d pi(0)pi(0) reaction and in the dd -> He-4 pi(0)pi(0) reaction, the data reveal a correlation between the ABC effect and a resonance-like energy dependence in the total cross section. Differential cross sections are well described by the hypothesis of d* resonance formation during the reaction process in addition to the conventional t-channel Delta Delta mechanism. The deduced d* resonance width can be understood from collision broadening due to Fermi motion of the nucleons in initial and final nuclei.

  • Charge symmetry breaking in dd -> He-4 pi(0) with WASA-at-COSY

    • Charge symmetry breaking (CSB) observables are a suitable experimental tool to examine effects induced by quark masses on the nuclear level. Previous high precision data from TRIUMF and IUCF are currently used to develop a consistent description of CSB within the framework of chiral perturbation theory. In this work the experimental studies on the reaction dd -> He-4 pi(0) have been extended towards higher excess energies in order to provide information on the contribution of p-waves in the final state. For this, an exclusive measurement has been carried out at a beam momentum of p(d) = 1.2GeV/c using the WASA-at-COSY facility. The total cross section amounts to sigma(tot) =(118 +/- 18(stat) +/- 13(sys) +/- 8(ext)) p band first data on the differential cross section are consistent with s-wave pion production.

  • Measurement of the np -> np pi(0)pi(0) reaction in search for the recently observed d* (2380) resonance

    • Exclusive measurements of the quasi-free np --> np pi(0)pi(0) reaction have been performed by means of dp collisions at T-d = 2.27 GeV using the WASA detector setup at COSY. Total and differential cross sections have been obtained covering the energy region root s = (2.35-2.46) GeV, which includes the region of the ABC effect and its associated d* (2380) resonance. Adding the d* resonance amplitude to that for the conventional processes leads to a reasonable description of the data. The observed resonance effect in the total cross section is in agreement with the predictions of Faldt and Wilkin as well with those of Albadajedo and Oset. The ABC effect, i.e. the low-mass enhancement in the pi(0)pi(0)-invariant mass spectrum, is found to be very modest - if present at all, which might pose a problem to some of its interpretations.

    • We report on a high-statistics measurement of the most basic double-pionic fusion reaction over the energy region of the d (*)(2380) resonance by use of a polarized deuteron beam and observing the double fusion reaction in the quasifree scattering mode. The measurements were performed with the WASA detector setup at COSY. The data reveal substantial analyzing powers and confirm conclusions about the d(*) resonance obtained from unpolarized measurements. We also confirm the previous unpolarized data obtained under complementary kinematic conditions.

  • Measurements of branching ratios for eta decays into charged particles

    • The WASA-at-COSY experiment has collected 3 x 10(7) events with eta mesons produced via the reaction pd -> He-3 eta at T = 1.0 GeV. Using this data set, we evaluate the branching ratios of the decays eta -> pi(+)pi(-)gamma, eta -> e(+)e(-)gamma, eta -> pi(+)pi(-)e(+)e(-), and eta -> e(+)e(-)e(+)e(-). The branching ratios are normalized to the eta -> pi(+)pi(-) pi(0) decay. In addition an upper limit on a CP-violating asymmetry in eta -> pi(+)pi(-)e(+)e(-) is extracted.

  • Three-nucleon dynamics in dp breakup collisions using the WASA detector at COSY-Julich

    • The differential cross section for the H-1(d, pp)n breakup reaction at deuteron beam energy of 340 MeV has been measured with the use of the WASA detector at COSY-Jillich. The set of proton-proton coincidences registered at the Forward Detector has been analyzed on a dense grid of kinematic variables, giving in total around 5600 data points. The cross-section data are compared to theoretical predictions based on the state-ofthe-art nucleon-nucleon potentials, combined with a three-nucleon force or the Coulomb interaction or carried out in a relativistic regime.

    • Various theoretical calculations based on QCD or hadronic interactions predict that in addition to the recently observed dibaryon resonance d*(2380) with I(J(P)) = 3(0(+)) there should also exist a dibaryon resonance with mirrored quantum numbers I(J(P)) = 3(0(+)). We report here on a search for such a NN-decoupled state in data on the pp -> pp pi(+) pi(+) pi(-) pi(-) reaction. Since no clear-cut evidence has been found, we give upper limits for the production cross section of such a resonance in the mass range 2280-2500 MeV.

    • We report on the search for the rare decay eta -> pi(0)e(+)e(-) which is of interest to study C violation in the electromagnetic interaction which would indicate contributions from physics beyond the Standard Model, since the allowed decay via a two-photon intermediate state is strongly suppressed. The experiment has been performed using the WASA-at-COSY installation, located at the COSY accelerator of the Forschungszentrum Julich, Germany. In total 3 x 10(7) events of the reaction pd -> He-3 eta have been recorded at an excess energy of Q = 59.8MeV. Based on this data set the C parity violating decay eta -> pi(0)gamma* -> pi(0)e(+)e(-) via a single-photon intermediate state has been searched for, resulting in new upper limits of Gamma(eta -> pi(0)e(+) e(-))/Gamma(eta -> pi(+) pi(-) pi(0)) < 3.28 x 10(-5) and Gamma(eta -> pi(0)e(+) e(-))/Gamma(eta -> all)< 7.5 x 10(-6) (CL=90%), respectively.

  • Backward single-pion production in the pd -> He-3 pi(0)reaction with WASA-at-COSY

    • New data on the production of single neutral pions in the pd -> He-3 pi(0) reaction are presented. For fifteen proton beam momenta between p(p) = 1.60GeV/c and p(p) = 1.74 GeV/c, differential cross sections are determined over a large fraction of the backward hemisphere. Since the only previous systematic measurements of single-pion production at these energies were made in collinear kinematics, the present work constitutes a significant extension of the current knowledge on this reaction. Even this far above the production threshold, significant changes are found in the behaviour of the angular distributions over small intervals in beam momentum.

  • Importance of d-wave contributions in the charge symmetry breaking reaction dd -> He-4 pi(0)

    • This letter reports a first quantitative analysis of the contribution of higher partial waves in the charge symmetry breaking reaction dd -> He-4 pi(0) using the WASA-at-COSY detector setup at an excess energy of Q = 60 MeV. The determined differential cross section can be parametrized as d sigma/d Omega = a + b cos(2) theta*, where theta* is the production angle of the pion in the center-of-mass coordinate system, and the results for the parameters are a = (1.55 +/- 0.46(stat)(-0.8)(+0.32)(syst)) pb/sr and b = (13.1 +/- 2.1(stat)(-2.7)(+1.0)(syst)) pb/sr. The data are compatible with vanishing p-waves and a sizable d-wave contribution. This finding should strongly constrain the contribution of the A isobar to the dd -> He-4 pi(0) reaction and is, therefore, crucial for a quantitative understanding of quark mass effects in nuclear production reactions.

  • Isoscalar single-pion production in the region of Roper and d*(2380) resonances

    • Exclusive measurements of the quasi-free pn -> pp pi(-) and pp -> pp pi(0) reactions have been performed by means of pd collisions at T-p= 1.2 GeV using the WASA detector setup at COSY. Total and differential cross sections have been obtained covering the energy region T-p= 0.95-1.3 GeV (root s = 2.3-2.46 GeV), which includes the regions of triangle(1232), N*(1440) and d*(2380) resonance excitations. From these measurements the isoscalar single-pion production has been extracted, for which data existed so far only below T-p = 1 GeV. We observe a substantial increase of this cross section around 1 GeV, which can be related to the Roper resonance N*(1440), the strength of which shows up isolated from the triangle resonance in the isoscalar (N pi)(I=0) invariant-mass spectrum. No evidence for a decay of the dibaryon resonance d*(2380) into the isoscalar (NN pi)(I=0) channel is found. An upper limit of 180 mu b (90% CL.) corresponding to a branching ratio of 9% has been deduced.

    • Using the production reactions pd -> He-3 omega and pp -> pp omega, the Dalitz plot distribution for the omega -> pi(+)pi(-)pi(0)decay is studied with the WASA detector at COSY, based on a combined data sample of (4.408 +/- 0.042) x 10(4) events. The Dalitz plot density is parametrised by a product of the P-wave phase space and a polynomial expansion in the normalised polar Dalitz plot variables Z and phi. For the first time, a deviation from pure P-wave phase space is observed with a significance of 4.1 sigma. The deviation is parametrised by a linear term 1+2 alpha Z, with alpha determined to be +0.147 +/- 0.036, consistent with the expectations of rho-meson-type final-state interactions of the P-wave pion pairs.   • Spin Dependence of eta Meson Production in Proton-Proton Collisions Close to Threshold

    • Taking advantage of the high acceptance and axial symmetry of the WASA-at-COSY detector, and the high polarization degree of the proton beam of COSY, the reaction →pp→ppη has been measured close to threshold to explore the analyzing power Ay. The angular distribution of Ay is determined with the precision improved by more than 1 order of magnitude with respect to previous results, allowing a first accurate comparison with theoretical predictions. The determined analyzing power is consistent with zero for an excess energy of Q=15  MeV, signaling s-wave production with no evidence for higher partial waves. At Q=72  MeV the data reveal strong interference of Ps and Pp partial waves and cancellation of (Pp)2 and Ss∗Sd contributions. These results rule out the presently available theoretical predictions for the production mechanism of the η meson.

    • New data on both total and differential cross sections of the production of eta mesons in proton-deuteron fusion to He-3 eta in the excess energy region 13.6 MeV <= Q(eta) <= 80.9 MeV are presented. These data have been obtained with the WASA-at-COSY detector setup located at the Forschungszentrum Julich, using a proton beam at 15 different beam momenta between p(p) = 1.60 GeV/c and p(p) = 1.74 GeV/c. While significant structure of the total cross section is observed in the energy region 20 MeV less than or similar to Q(eta) less than or similar to 60 MeV, a previously reported sharp variation around Q(eta) approximate to 50 MeV cannot be confirmed. Angular distributions show the typical forward- peaking that was noted earlier. For the first time, it is possible to study the development of these angular distributions with rising excess energy over a wide interval.

    • The search for He-4-eta bound states was performed with the WASA-at-COSY facility via the measurement of the excitation function for the dd -> (3)Hen pi(0) and dd -> (3)Hep pi(-) processes. The deuteron beam momentum was varied continuously between 2.127 GeV/c and 2.422 GeV/c, corresponding to the excess energy for the dd -> He-4 eta reaction ranging from Q = 70 MeV to Q = 30 MeV. The luminosity was determined based on the dd -> (3)Hen reaction and the quasi-free proton proton scattering via dd -> ppn(spectator)n(spectator) reactions. The excitation functions, determined independently for the measured reactions, do not reveal a structure which could be interpreted as a narrow mesic nucleus. Therefore, the upper limits of the total cross sections for the bound state production and decay in dd -> (4He-eta)(bound) -> (3)Hen pi(0) and dd -> (He-4-eta)(bound) -> (3)Hep pi(-) processes were determined taking into account the isospin relation between the both of the considered channels. The results of the analysis depend on the assumptions of the N* (1535) momentum distribution in the anticipated mesic-He-4. Assuming, as in the previous works, that this is identical with the distribution of nucleons bound with 20 MeV in He-4, we determined that (for the mesic bound state width in the range from 5 MeV to 50 MeV) the upper limits at 90% confidence level are about 3 nb and about 6 nb for n pi(0) and p pi(-) channels, respectively. However, based on the recent theoretical findings of the N*(1535) momentum distribution in the N*-He-3 nucleus bound by 3.6 MeV, we find that the WASA-at-COSY detector acceptance decreases and hence the corresponding upper limits are 5 nb and 10 nb for n pi(0) and p pi(-) channels respectively.     • A detector setup for the cooler storage ring CELSIUS is described. The setup detects particles produced in interactions between the internal beam and a cluster-jet target. Particles emitted in the forward direction are measured by means of arrays of plastic scintillators and proportional counters. Particles, particularly photons, emitted more isotropically are measured by means of two calorimeters containing CsI(Na) crystals. The performance of the setup is given for neutral meson production in proton-proton and proton-deuteron interactions in the energy range 290-1360 MeV.

    • Context. EECep is an unusual long-period (5.6 yr) eclipsing binary discovered during the mid-twentieth century. It undergoes almost-grey eclipses that vary in terms of both depth and duration at different epochs. The system consists of a Be type star and a dark dusty disk around an invisible companion. EECep together with the widely studied epsilon Aur are the only two known cases of long-period eclipsing binaries with a dark, dusty disk component responsible for periodic obscurations.Aims. Two observational campaigns were carried out during the eclipses of EECep in 2003 and 2008/9 to verify whether the eclipsing body in the system is indeed a dark disk and to understand the observed changes in the depths and durations of the eclipses.Methods. Multicolour photometric data and spectroscopic observations performed at both low and high resolutions were collected with several dozen instruments located in Europe and North America. We numerically modelled the variations in brightness and colour during the eclipses. We tested models with different disk structure, taking into consideration the inhomogeneous surface brightness of the Be star. We considered the possibility of disk precession.Results. The complete set of observational data collected during the last three eclipses are made available to the astronomical community. The 2003 and 2008/9 eclipses of EECep were very shallow. The latter is the shallowest among all observed. The very high quality photometric data illustrate in detail the colour evolution during the eclipses for the first time. Two blue maxima in the colour indices were detected during these two eclipses, one before and one after the photometric minimum. The first (stronger) blue maximum is simultaneous with a "bump" that is very clear in all the UBV(RI)(C) light curves. A temporary increase in the I-band brightness at the orbital phase similar to 0.2 was observed after each of the last three eclipses. Variations in the spectral line profiles seem to be recurrent during each cycle. The Na I lines always show at least three absorption components during the eclipse minimum and strong absorption is superimposed on the H alpha emission.Conclusions. These observations confirm that the eclipsing object in EECep system is indeed a dark, dusty disk around a low luminosity object. The primary appears to be a rapidly rotating Be star that is strongly darkened at the equator and brightened at the poles. Some of the conclusions of this work require verification in future studies: (i) a complex, possibly multi-ring structure of the disk in EECep; (ii) our explanation of the "bump" observed during the last two eclipses in terms of the different times of obscuration of the hot polar regions of the Be star by the disk; and (iii) our suggested period of the disk precession (similar to 11-12 P-orb) and predicted depth of about 2(m) for the forthcoming eclipse in 2014.

    • The quasifree pp -> pp pi(+)pi(-) reaction has been measured by means of pd collisions at T-p = 1.2 GeV using the WASA detector setup at COSY enabling exclusive and kinematically complete measurements. Total and differential cross sections have been extracted for the energy region T-p = 1.08-1.36 GeV (root s = 2.35-2.46 GeV) covering thus the regions of N* (1440 ) and Delta(1232)Delta(1232) resonance excitations. Calculations describing these excitations by t-channel meson exchange as well as isospin relations based on the pp -> pp pi(0)pi(0) data underpredict substantially the measured total cross section. The calculations are also at variance with specific experimental differential cross sections. An isotensor Delta N dibaryon resonance with I(J(P)) = 2(1(+) ) produced associatedly with a pion is able to overcome these deficiencies. Such a dibaryon was predicted by Dyson and Xuong [Phys. Rev. Lett. 13, 815 (1964)] and more recently calculated by A. Gal and H. Garcilazo [Nucl. Phys. A 928, 73 (2014)].

    • The excitation function for the pd -> dp pi(0) reaction has been measured byWASA-at-COSY experiment with the aim of searching for He-3-h mesic nuclei. The measurement in the vicinity of h meson production was performed using a ramped proton beam. The data analysis and interpretation was carried out with the assumption that the h mesic helium decays via the formation of an intermediate N*(1535) resonance. No direct signal of the. mesic nucleus is observed in the excitation function. We determine a new improved upper limit for the total cross section for the bound state production and decay in the pd -> (He-3-eta)(bound) -> dp pi(0) process. It varies between 13 nb to 24 nb for the bound state with width in the range Gamma is an element of ( 5, 50) MeV.

    • We report on the experimental search for the bound state of an eta meson and He-3 nucleus performed using the WASA-at-COSY detector setup. In order to search for the eta-mesic nucleus decay, the pd -> (3)He2(gamma) and pd -> (3)He6 gamma channels have been analysed. These reactions manifest the direct decay of the ti meson bound in a He-3 nucleus. This non-mesonic decay channel has been considered for the first time. When taking into account only statistical errors, the obtained excitation functions reveal a slight indication for a possible bound state signal corresponding to a He-3-eta nucleus width Gamma above 20 MeV and binding energy B-s between 0 and 15 MeV. However, the determined cross sections are consistent with zero in the range of the systematic uncertainty. Therefore, as final result we estimate only the upper limit for the cross section of the eta-mesic He-3 nucleus formation followed by the eta meson decay which varies between 2 nb and 15 nb depending on possible bound state parameters.

    • The reaction pp → ppπ0 has been measured using electron-cooled protons incident on an internal gas-jet target at seven different incident beam energies, from 280.7 MeV (1 MeV above the reaction threshold) up to 310.2 MeV. The pions were measured by their decay photons. In the overlapping energy region, the measured total cross sections agree with those measured in a recent Indiana experiment. The angular distributions are consistent with a 3P0 → 1S0S0 transition in the full energy range studied. The kinematical distributions are well described when taking into account the final state and the Coulomb interactions.

    • The pp → ppη reaction has been measured at six energies close to threshold, from 1258 MeV to 1352 MeV, using an internal cluster gas jet target in the CELSIUS storage ring. The η is detected through its decay photons, in an array of CsI detectors, and the forward-going protons are detected in a plastic scintillator spectrometer. A complete event reconstruction is obtained at the higher energies in the measured interval. The new data, together with earlier data, give an accurate determination of the energy dependence close to threshold. The influence of the η-proton FSI is seen in the total cross section data as well as in a Dalitz plot of the η-p invariant mass distributions.

    • A search for rare lepton decays of the eta meson was performed using the WASA detector at CELSIUS. Two candidates for double Dalitz decay eta -> e(+)e(-)e(+)e(-) events are reported with a background of 1.3 +/- 0.2 events. This allows to set an upper limit to the branching ratio of 9.7x10(-5) (90% CL). The branching ratio for the decay eta -> e(+)e(-)gamma is determined to (7.8 +/- 0.5(stat)+/- 0.8(syst))x10(-3) in agreement with world average value. An upper limit (90% CL) for the branching ratio for the eta -> e(+)e(-) decay is 2.7x10(-5) and a limit for the sum of the eta ->mu(+)mu(-)mu(+)mu(-) and eta ->pi(+)pi(-)mu(+)mu(-) decays is 3.6x10(-4).

    • A high performance apparatus has been designed and built by the H8-RD22 collaboration for the study of channeling and volume reflection phenomena in the interaction of 400 GeVc protons with bent silicon crystals, during the 2006 data taking in the external beamline H8 of the CERN SPS. High-quality silicon short crystals were bent by either anticlastic or quasimosaic effects. Alignment with the highly parallel (8 μrad divergence) proton beam was guaranteed through a submicroradian goniometric system equipped with both rotational and translational stages. Particle tracking was possible by a series of silicon microstrip detectors with high-resolution and a parallel plate gas chamber, triggered by various scintillating detectors located along the beamline. Experimental observation of volume reflection with 400 GeVc protons proved true with a deflection angle of (10.4±0.5) μrad with respect to the unperturbed beam, with a silicon crystal whose (111) planes were parallel to the beam. © 2008 American Institute of Physics.

    • Extracellular vesicles derived from mesenchymal stem cells (MSCs) represent a novel approach for regenerative and immunosuppressive therapy. Recently, cytochalasin B-induced microvesicles (CIMVs) were shown to be effective drug delivery mediators. However, little is known about their immunological properties. We propose that the immunophenotype and molecular composition of these vesicles could contribute to the therapeutic efficacy of CIMVs. To address this issue, CIMVs were generated from murine MSC (CIMVs-MSCs) and their cytokine content and surface marker expression determined. For the first time, we show that CIMVs-MSCs retain parental MSCs phenotype (Sca-1(+), CD49e(+), CD44(+), CD45(-)). Also, CIMVs-MSCs contained a cytokine repertoire reflective of the parental MSCs, including IL-1 beta, IL-2, IL-3, IL-4, IL-5, IL-6, IL-9, IL-10, IL-12(p40), IL-13, IL-17, CCL2, CCL3, CCL4, CCL5, CCL11, G-CSF, GM-CSF and TNF-alpha. Next, we evaluated the immune-modulating properties of CIMVs-MSCs in vivo using standard preclinical tests. MSCs and CIMVs-MSCs reduced serum levels of anti-sheep red blood cell antibody and have limited effects on neutrophil and peritoneal macrophage activity. We compared the immunomodulatory effect of MSCs, CIMVs and EVs. We observed no immunosuppression in mice pretreated with natural EVs, whereas MSCs and CIMVs-MSCs suppressed antibody production in vivo. Additionally, we have investigated the biodistribution of CIMVs-MSCs in vivo and demonstrated that CIMVs-MSCs localized in liver, lung, brain, heart, spleen and kidneys 48 h after intravenous injection and can be detected 14 days after subcutaneous and intramuscular injection. Collectively our data demonstrates immunomodulatory efficacy of CIMVs and supports their further preclinical testing as an effective therapeutic delivery modality.

We used time-resolved serial femtosecond crystallography at an x-ray free electron laser to visualize conformational changes in bR from nanoseconds to milliseconds following photoactivation. An initially twisted retinal chromophore displaces a conserved tryptophan residue of transmembrane helix F on the cytoplasmic side of the protein while dislodging a key water molecule on the extracellular side. The resulting cascade of structural changes throughout the protein shows how motions are choreographed as bR transports protons uphill against a transmembrane concentration gradient.

    • Aquaporin-4 (AQP4) plays an important role in the basolateral movement of water in the collecting duct. Here we show that this water channel can be dynamically regulated. Water permeability (P-f) was measured in individual LLC-PK1 cells that were transiently transfected with AQP4. To identify which cells were transfected, AQP4 was tagged at the NH2 terminus with green fluorescent protein. Transfected cells showed a strong fluorescent signal in basolateral membrane and a low-to-negligible signal in the cytosol and apical membrane. Activation of protein kinase C (PKC) with phorbol 12,13-dibutyrate (PDBu) significantly decreased P-f of cells expressing AQP4 but had no effect on neighboring untransfected cells. No redistribution of AQP4 in response to PDBu was detected. Dopamine also decreased the P-f in transfected cells. The effect was abolished by the PKC inhibitor Ro 31-8220. Reduction of AQP4 water permeability by PDBu and dopamine was abolished by point mutation of Ser(180), a consensus site for PKC phosphorylation. We conclude that PKC and dopamine decrease AQP4 water permeability via phosphorylation at Ser(180) and that the effect is likely mediated by gating of the channel.

    • The water channel aquaporin 4 (AQP4) is abundantly expressed in astrocytes and provides a mechanism by which water permeability of the plasma membrane can be regulated. Astrocytes play a key role in the clearance of both potassium (K+) and glutamate released during neuronal activity. Emerging evidence suggests that AQP4 facilitates K+ clearance by astrocytes and contributes to recovery of neuronal excitability. Here we report that AQP4 can assemble with its regulator metabotropic glutamate receptor 5 (mGluR5) and with Na,K-ATPase; the enzyme responsible for active K+ transport and for establishing the electrochemical gradient across the cell plasma membrane. We have, by use of pull down assays in rat brain tissue, identified the segment in the AQP4 NH2-terminus containing the amino acid residues 23-32 as the site for interaction with Na,K-ATPase catalytic subunit and with mGluR5. Mutagenesis studies revealed that the AQP4 amino acids K27 and W30 are of key importance for interaction with both Na,K-ATPase and mGluR5. To confirm that interaction also occurs within intact cells, we have performed fluorescence resonance energy transfer (FRET) studies in primary astrocytes derived from rat striatum. The results indicate close proximity of wild type AQP4 and Na,K-ATPase in the plasma membrane of rat astrocytes. FRET efficiencies observed with the mutants AQP4 K27A and AQP4 W30A were significantly lower, highlighting the importance of these residues for the interaction between AQP4 and Na,K-ATPase. We conclude that AQP4/Na,K-ATPase/mGluR5 can form a macromolecular complex/transporting microdomain in astrocytes. This complex may be of functional importance for the regulation of water and K+ homeostasis in the brain, as well as for neuron-astrocyte metabolic crosstalk. (C) 2010 IBRO.     • Background: Plant mitochondrial genomes (mitogenomes) can be structurally complex while their size can vary from similar to 222 Kbp inBrassica napusto 11.3 Mbp inSilene conica. To date, in comparison with the number of plant species, only a few plant mitogenomes have been sequenced and released, particularly for conifers (the Pinaceae family). Conifers cover an ancient group of land plants that includes about 600 species, and which are of great ecological and economical value. Among them, Siberian larch (Larix sibiricaLedeb.) represents one of the keystone species in Siberian boreal forests. Yet, despite its importance for evolutionary and population studies, the mitogenome of Siberian larch has not yet been assembled and studied. Results: Two sources of DNA sequences were used to search for mitochondrial DNA (mtDNA) sequences: mtDNA enriched samples and nucleotide reads generated in the de novo whole genome sequencing project, respectively. The assembly of the Siberian larch mitogenome contained nine contigs, with the shortest and the largest contigs being 24,767 bp and 4,008,762 bp, respectively. The total size of the genome was estimated at 11.7 Mbp. In total, 40 protein-coding, 34 tRNA, and 3 rRNA genes and numerous repetitive elements (REs) were annotated in this mitogenome. In total, 864 C-to-U RNA editing sites were found for 38 out of 40 protein-coding genes. The immense size of this genome, currently the largest reported, can be partly explained by variable numbers of mobile genetic elements, and introns, but unlikely by plasmid-related sequences. We found few plasmid-like insertions representing only 0.11% of the entire Siberian larch mitogenome. Conclusions: Our study showed that the size of the Siberian larch mitogenome is much larger than in other so far studied Gymnosperms, and in the same range as for the annual flowering plantSilene conica(11.3 Mbp). Similar to other species, the Siberian larch mitogenome contains relatively few genes, and despite its huge size, the repeated and low complexity regions cover only 14.46% of the mitogenome sequence.

Understanding these mechanisms of action requires knowledge of how the proteins couple to their fluid, hydrated lipid membrane environment. We present here current studies in computational and experimental membrane protein biophysics, and show how they address outstanding challenges in understanding the complex environmental effects on the structure, function, and dynamics of membrane proteins.
