    • Deciphering the assembly history of the Milky Way is a formidable task, which becomes possible only if one can produce high-resolution chrono-chemo-kinematical maps of the Galaxy. Data from large-scale astrometric and spectroscopic surveys will soon provide us with a well-defined view of the current chemo-kinematical structure of the Milky Way, but it will only enable a blurred view on the temporal sequence that led to the present-day Galaxy. As demonstrated by the (ongoing) exploitation of data from the pioneering photometric missions CoRoT, Kepler, and K2, asteroseismology provides the way forward: solar-like oscillating giants are excellent evolutionary clocks thanks to the availability of seismic constraints on their mass and to the tight age-initial mass relation they adhere to. In this paper we identify five key outstanding questions relating to the formation and evolution of the Milky Way that will need precise and accurate ages for large samples of stars to be addressed, and we identify the requirements in terms of number of targets and the precision on the stellar properties that are needed to tackle such questions. By quantifying the asteroseismic yields expected from PLATO for red giant stars, we demonstrate that these requirements are within the capabilities of the current instrument design, provided that observations are sufficiently long to identify the evolutionary state and allow robust and precise determination of acoustic-mode frequencies. This will allow us to harvest data of sufficient quality to reach a 10% precision in age. This is a fundamental prerequisite to then reach the more ambitious goal of a similar level of accuracy, which will be possible only if we have at hand a careful appraisal of systematic uncertainties on age deriving from our limited understanding of stellar physics, a goal that conveniently falls within the main aims of PLATO's core science. We therefore strongly endorse PLATO's current design and proposed observational strategy, and conclude that PLATO, as it is, will be a legacy mission for Galactic archaeology.

    • Analyses of data from spectroscopic and astrometric surveys have led to conflicting results concerning the vertical characteristics of the Milky Way. Ages are often used to provide clarity, but typical uncertainties of >40 per cent from photometry restrict the validity of the inferences made. Using the Kepler APOKASC sample for context, we explore the global population trends of two K2 campaign fields (3 and 6), which extend further vertically out of the Galactic plane than APOKASC. We analyse the properties of red giant stars utilizing three asteroseismic data analysis methods to cross-check and validate detections. The Bayesian inference tool PARAM is used to determine the stellar masses, radii, and ages. Evidence of a pronounced red giant branch bump and an [a/Fe] dependence on the position of the red clump is observed from the K2 fields radius distribution. Two peaks in the age distribution centred at similar to 5 and similar to 12 Gyr are found using a sample with sigma(age) < 35 per cent. In comparison with Kepler, we find the older peak to be more prominent for K2. This age bimodality is also observed based on a chemical selection of low-[alpha/Fe] (<= 0.1) and high-[alpha/Fe] (>0.1) stars. As a function of vertical distance from the Galactic mid-plane (|Z|), the age distribution shows a transition from a young to old stellar population with increasing |Z| for the K2 fields. Further coverage of campaign targets with high-resolution spectroscopy is required to increase the yield of precise ages achievable with asteroseismology.

  • Management of latent Mycobacterium tuberculosis infection: WHO guidelines for low tuberculosis burden countries

    • Latent tuberculosis infection (LTBI) is characterised by the presence of immune responses to previously acquired Mycobacterium tuberculosis infection without clinical evidence of active tuberculosis (TB). Here we report evidence-based guidelines from the World Health Organization for a public health approach to the management of LTBI in high risk individuals in countries with high or middle upper income and TB incidence of <100 per 100 000 per year. The guidelines strongly recommend systematic testing and treatment of LTBI in people living with HIV, adult and child contacts of pulmonary TB cases, patients initiating anti-tumour necrosis factor treatment, patients receiving dialysis, patients preparing for organ or haematological transplantation, and patients with silicosis. In prisoners, healthcare workers, immigrants from high TB burden countries, homeless persons and illicit drug users, systematic testing and treatment of LTBI is conditionally recommended, according to TB epidemiology and resource availability. Either commercial interferon-gamma release assays or Mantoux tuberculin skin testing could be used to test for LTBI. Chest radiography should be performed before LTBI treatment to rule out active TB disease. Recommended treatment regimens for LTBI include: 6 or 9 month isoniazid; 12 week rifapentine plus isoniazid; 3–4 month isoniazid plus rifampicin; or 3–4 month rifampicin alone.

    • Reliable models of the thermally pulsing asymptotic giant branch (TP-AGB) phase are of critical importance across astrophysics, including our interpretation of the spectral energy distribution of galaxies, cosmic dust production, and enrichment of the interstellar medium. With the aim of improving sets of stellar isochrones that include a detailed description of the TP-AGB phase, we extend our recent calibration of the AGB population in the Small Magellanic Cloud (SMC) to the more metal-rich Large Magellanic Cloud (LMC). We model the LMC stellar populations with the TRILEGAL code, using the spatially resolved star formation history derived from the VISTA survey. We characterize the efficiency of the third dredge-up by matching the star counts and the K-s-band luminosity functions of the AGB stars identified in the LMC. In line with previous findings, we confirm that, compared to the SMC, the third dredge-up in AGB stars of the LMC is somewhat less efficient, as a consequence of the higher metallicity. The predicted range of initial mass of C-rich stars is between M-i approximate to 1.7 and 3 M-circle dot at Z(i) = 0.008. We show how the inclusion of new opacity data in the carbon star spectra will improve the performance of our models. We discuss the predicted lifetimes, integrated luminosities, and mass-loss rate distributions of the calibrated models. The results of our calibration are included in updated stellar isochrones publicly available.

    • The Vera C. Rubin Observatory will undertake the Legacy Survey of Space and Time, providing an unprecedented, volume-limited catalog of star clusters in the Southern Sky, including Galactic and extragalactic star clusters. The Star Clusters subgroup of the Stars, Milky Way and Local Volume Working Group has identified key areas where Rubin Observatory will enable significant progress in star cluster research. This roadmap represents our science cases and preparation for studies of all kinds of star clusters from the Milky Way out to distances of tens of megaparsecs.

    • The brain cytoplasmic (BC1) RNA is a non-coding RNA (ncRNA) involved in neuronal translational control. Absence of BC1 is associated with altered glutamatergic transmission and maladaptive behavior. Here, we show that pyramidal neurons in the barrel cortex of BC1 knock out (KO) mice display larger excitatory postsynaptic currents and increased spontaneous activity in vivo. Furthermore, BC1 KO mice have enlarged spine heads and postsynaptic densities and increased synaptic levels of glutamate receptors and PSD-95. Of note, BC1 KO mice show aberrant structural plasticity in response to whisker deprivation, impaired texture novel object recognition and altered social behavior. Thus, our study highlights a role for BC1 RNA in experience-dependent plasticity and learning in the mammalian adult neocortex, and provides insight into the function of brain ncRNAs regulating synaptic transmission, plasticity and behavior, with potential relevance in the context of intellectual disabilities and psychiatric disorders.

    • The PUMA project, a Specific Targeted Research Project (STREP) of the European Union EURATOM 6th Framework Program, is mainly aimed at providing additional key elements for the utilisation and transmutation of plutonium and minor actinides (neptunium and americium) in contemporary and future (high temperature) gas-cooled reactor design, which are promising tools for improving the sustainability of the nuclear fuel cycle. PUMA would also contribute to the reduction of Pu and MA stockpiles and to the development of safe and sustainable reactors for C02-free energy generation. The project runs from September 1, 2006 until August 31, 2009. PUMA also contributes to technological goals of the Generation IV International Forum. It contributes to developing and maintaining the competence in reactor technology in the EU and addresses European stakeholders on key issues for the future of nuclear energy in the EU. An overview is presented of the status of the project at mid-term.

    • To improve the public acceptance of the future nuclear power in Europe we have to demonstrate that the new reactors have significantly higher safety level compared to traditional reactors. The ESFR-SMART project (European Sodium Fast Reactor Safety Measures Assessment and Research Tools) aims at enhancing further the safety of Generation-IV SFRs and in particular of the commercial-size European Sodium Fast Reactor (ESFR) in accordance with the European Sustainable Nuclear Industrial Initiative (ESNII) roadmap and in close cooperation with the Advanced Sodium Technological Reactor for Industrial Demonstration (ASTRID) program. The project aims at 5 specific objectives: 1. Produce new experimental data in order to support calibration and validation of the computational tools for each defence-in-depth level. 2. Test and qualify new instrumentations in order to support their utilization in the reactor protection system. 3. Perform further calibration and validation of the computational tools for each defence-in-depth level in order to support safety assessments of Generation-IV SFRs, using the data produced in the project as well as selected legacy data. 4. Select, implement and assess new safety measures for the commercial-size ESFR, using the GIF methodologies, the FP7 CP-ESFR project legacy, the calibrated and validated codes and being in accordance with the update of the European and international safety frameworks taking into account the Fukushima accident. 5. Strengthen and link together new networks, in particular, the network of the European sodium facilities and the network of the European students working on the SFR technology. By addressing the industry, policy makers and general public, the project is expected to make a meaningful impact on economics, environment, EU policy and society. Selected results and milestones achieved during the first eighteen months of the project will be briefly presented, including − proposal of new safety measures for ESFR; − evaluation of ESFR core performance; − benchmarking of codes; − experimental programs; and − education and training.

    • The thermally pulsing asymptotic giant branch (TP-AGB) experienced by low-and intermediate-mass stars is one of the most uncertain phases of stellar evolution and the models need to be calibrated with the aid of observations. To this purpose, we couple high-quality observations of resolved stars in the Small Magellanic Cloud (SMC) with detailed stellar population synthesis simulations computed with the TRILEGAL code. The strength of our approach relies on the detailed spatially resolved star formation history of the SMC, derived from the deep near-infrared photometry of the VISTA survey of the Magellanic Clouds, as well as on the capability to quickly and accurately explore a wide variety of parameters and effects with the COLIBRI code for the TP-AGB evolution. Adopting a well-characterized set of observations - star counts and luminosity functions - we set up a calibration cycle along which we iteratively change a few key parameters of the TP-AGB models until we eventually reach a good fit to the observations. Our work leads to identify two best-fitting models that mainly differ in the efficiencies of the third dredge-up and mass-loss in TP-AGB stars with initial masses larger than about 3 M-circle dot. On the basis of these calibrated models, we provide a full characterization of the TP-AGB stellar population in the SMC in terms of stellar parameters (initial masses, C/O ratios, carbon excess, mass-loss rates). Extensive tables of isochrones including these improved models are publicly available.

    • We estimate that there will be 13·7 million new cases of childhood cancer globally between 2020 and 2050. At current levels of health system performance (including access and referral), 6·1 million (44·9%) of these children will be undiagnosed. Between 2020 and 2050, 11·1 million children will die from cancer if no additional investments are made to improve access to health-care services or childhood cancer treatment. Of this total, 9·3 million children (84·1%) will be in low-income and lower-middle-income countries. This burden could be vastly reduced with new funding to scale up cost-effective interventions. Simultaneous comprehensive scale-up of interventions could avert 6·2 million deaths in children with cancer in this period, more than half (56·1%) of the total number of deaths otherwise projected. Taking excess mortality risk into consideration, this reduction in the number of deaths is projected to produce a gain of 318 million life-years. In addition, the global lifetime productivity gains of US$2580 billion in 2020–50 would be four times greater than the cumulative treatment costs of $594 billion, producing a net benefit of $1986 billion on the global investment: a net return of $3 for every $1 invested. In sum, the burden of childhood cancer, which has been grossly underestimated in the past, can be effectively diminished to realise massive health and economic benefits and to avert millions of needless deaths.

    • It is unclear whether coronary artery bypass grafting (CABG) improves survival compared with medical therapy (MT) in patients with stable coronary artery disease (CAD). The aim of this analysis was to perform an individual-patient data-pooled meta-analysis of contemporary randomized controlled trials that compared CABG and MT in patients with stable CAD.A systematic search was performed in January 2021 to identify randomized controlled trials enrolling adult patients with stable CAD, randomized to CABG or MT. Only trials using at least aspirin, beta-blockers, and statins in the MT arm were included. Individual patient data were obtained from all eligible studies and pooled. The primary outcome was all-cause mortality.Four trials involving 2523 patients (1261 CABG; 1262MT) were included with a median follow-up of 5.6 (4.0-9.2) years. CABG was associated with increased risk of all-cause mortality within 30days (hazard ratio [HR], 4.81; 95% confidence interval [CI], 1.95-11.83) but subsequent reduction in the long-term risk of death (HR, 0.79; 95% CI, 0.69-0.89). As such, the cumulative 10-year mortality rate was lower in patients treated with CABG compared with MT (45.1% vs 51.7%, respectively; odds ratio, 0.70; 95% CI, 0.58-0.85). Age and race were significant treatment effect modifier (interaction P=.003 for both).In patients with stable CAD, initial allocation to CABG was associated with greater periprocedural risk of death but improved long-term survival compared with MT. The survival advantage for CABG became significant after the fourth postoperative year and was particularly pronounced in younger and non-White patients.

    • We employed the VISTA near-infrared YJKssurvey of the Magellanic System (VMC) to analyse the Y, J, and Ks light curves of δCepheid stars (DCEPs) in the Large Magellanic Cloud (LMC). Our sample consists of 4408 objects accounting for 97 per cent of the combined list of OGLE IV and Gaia DR2 DCEPs. We determined a variety of period-luminosity (PL) and period-Wesenheit PW relationships for Fundamental (F) and First Overtone (1O) pulsators. We discovered for the first time a break in these relationships for 1O DCEPs at P= 0.58 d. We derived relative individual distances for DCEPs in the LMC with a precision of ∼1 kpc, calculating the position angle of the line of nodes and inclination of the galaxy: θ = 145.6 ± 1.0 deg and i = 25.7 ± 0.4 deg. The bar and the disc are seen under different viewing angles. We calculated the ages of the pulsators, finding two main episodes of DCEP formation lasting ∼40 Myr which happened 93 and 159 Myr ago. Likely as a result of its past interactions with the SMC, the LMC shows a non-planar distribution, with considerable structuring: the bar is divided into two distinct portions, the eastern and the western displaced by more than 1 kpc from each other. Similar behaviour is shown by the spiral arms. The LMC disc appears 'flared' and thick, with a disc scale height of h ∼0.97 kpc. This feature can be explained by strong tidal interactions with the Milky Way and/or the Small Magellanic Cloud or past merging events with now disrupted LMC satellites.
