    • Perturbation in the expression and signaling pathways of vascular endothelial growth factor (VEGF) has been linked to pathogenesis of hematologic malignancies. We investigated the expression and clinical importance of VEGF and two of its receptors, VEGFR-1 and VEGFR-2, in childhood precursor B-cell acute lymphoblastic leukemia (pre-B ALL) by using immunohistochemistry. These angiogenic proteins were expressed in the majority of leukemic bone marrow samples. Notably, pre-B ALL patients had significantly increased expression of VEGFR-1 compared with no expression in the nonmalignant group, indicating a link between VEGFR-1 protein expression and pre-B ALL. These novel findings suggest that VEGFR-1 may have clinical importance in childhood pre-B ALL.

    • This article analyses the factors influencing stakeholder participation in municipal energy and climate planning, together with stakeholder experiences of participation in such processes. The article presents findings from a survey of 60 Swedish municipalities and detailed case studies of energy and climate planning in five Swedish municipalities to explore which stakeholders are involved, how and why they are involved, and the impacts of participation on both stakeholder experiences. The results of this study provide insight into the role of stakeholders in municipal energy and climate planning.The study proceeds as follows: the Introduction is followed by a discussion of theoretical perspectives on stakeholder participation and energy and climate planning. The Methods used to conduct the study are presented, followed by Results and Analysis. In the subsequent Discussion, the authors propose a conceptual approach that may assist municipalities in development of energy and climate strategies. The Analysis and Discussion inform Conclusions in which the authors advocate early and active stakeholder engagement in energy and climate strategy planning and emphasise the possible utility of their conceptual approach in supporting stakeholder participation.

This paper describes the factors influencing development of municipal climate and energy plans in five Swedish municipalities and assesses the relevance and importance of these factors from theoretical and practical perspectives. The questions raised in the paper are: what are the characteristics of municipal climate and energy planning processes in the five Swedish municipalities, how do municipalities include stakeholders in the process, and in what ways do stakeholders influence outcomes? Results suggest that a number of key factors influence the development of municipal climate and energy strategies and their content. These include the importance of a clear, shared vision and engaged politicians; the size and organisational structure of the municipality and its willingness and capability to act; the organisation of the process and extent to which stakeholders have been involved and feel included; the need for clarity about financial aspects, such as planned financing of implementation; the need for greater clarity concerning selection of targets and their relevance to global climate and energy trends. By illustrating the characteristics of the municipal climate and energy planning processes in five Swedish municipalities, the study makes a contribution to the research on local governments and sustainable development in general, and specifically to the growing literature on municipal climate and energy planning. The study and its results may be used to inform policy-makers on the national and local levels about the factors influencing municipal energy planning and the importance of involving stakeholders and citizens in the strategic work to reduce climate impacts and energy consumption.

  • Sustainable Energy and Climate Strategies : lessons from planning processes in five Swedish Municipalities

    • This report forms part of the research project “Sustainable Energy and Climate Strategies – development and potential”, which is financed by the Swedish Energy Agency’s Sustainable Municipality programme. In this research project, case studies of the processes to develop energy strategies in five municipalities were prepared. The five municipalities were participants in the Sustainable Municipality programme’s second phase, which began in 2008, and represent different types of municipality, in terms of geography and population.This report presents analysis of the five case studies, using a policy theoretical perspective to focus on issues including how problems and solutions are identified and formulated, which solutions are proposed, which actors are included or excluded from the process, and which local resources are used or not used in the process. The report reflects on the implications of increasing stakeholder cooperation in energy planning processes and using different types of organisational approaches during the development of energy and climate strategies.Each case study began with an inventory of publically-available documents shaping the context for energy and climate strategies in each municipality. These documents were compiled in time lines showing the documents or decisions influencing energy planning in each municipality. Subsequently, group interviews were held with participants in planning processes in each of the five municipalities. In addition, individual interviews took place with stakeholders who had been active in the processes. Interviews were recorded and then transcribed. The results from the document study and interviews were then compiled in a summary of each municipality’s energy planning process, forming the basis for the analysis in this report.The case studies highlight both similarities and differences with regard to the organisation of work to develop and introduce energy strategies. All municipalities established, at an early stage, internal organisations for the process and throughout the processes, the organisational form, participants and their role, and division of tasks and responsibilities were fairly clear. For example, all five municipalities made use of Steering Groups and Working Groups, although the extent of the roles which these groups played – and the background of their members - varied. Four municipalities had a Reference Group in which external stakeholders were represented. A number of other constellations, including thematic working groups, were present in some but not all municipalities.The municipalities also took different steps in their energy planning processes and identified different drivers stimulating their activities. Participants from one municipality considered the Sustainable Municipality programme as a pivotal moment in their strategic energy work, whereas others felt the programme did not significantly influence their approach or outputs. The report identifies a number of factors influencing the development and implementation of municipal energy strategies, as well as a strategy’s scope and content.

    • PTEN and SHP1 are tumor suppressor genes involved in the regulation of cell cycle control and apoptosis. The authors investigated the protein expression of PTEN and SHP1, by immunohistochemistry in tissue microarrays from bone marrow samples in children, diagnosed with acute lymphoblastic leukaemia and nonmalignant controls. PTEN was overexpressed in diagnostic ALL samples, while SHP1 showed a low expression. Both proteins showed a significant difference in expression compared to nonmalignant controls. The roles of PTEN and SHP1 are not well investigated in pediatric leukemia and could in the future play a role as prognostic factors.

    • Svenska kommuner har en lång tradition av energiplanering. Dock har verkningsfullheten avkommunernas energistrategiska arbete varit föremål för diskussion. Syftet med det här projektet vardärför att analysera kommuners arbete med att ta fram och genomföra kommunala energistrategiersamt att reflektera över hur/om ett vidgat systemperspektiv genom ökad aktörssamverkan kan bidramed kunskaper till det lokala energistrategiska arbetet.Utifrån projektets resultat har vi sammanställt ett antal rekommendationer som stöd till kommuner ideras arbete med att införa, implementera och utveckla det energistrategiska arbetet. Dessarekommendationer sammanfattar också de huvudsakliga resultat som vi vill skicka med kommunernai deras arbete. Det energistrategiska arbetet bör hanteras som en del i en långsiktig verksamhet och intesom ett projekt Både kommunens politiska majoritet och opposition bör vara med i processen att utvecklaenergistrategin och de ska ha en aktiv roll under hela processen Informera och utbilda politiker om de lokala energisystemen för engagemang och delaktighet Vid målformulering bör de som berörs av målen involveras Koppla mål och åtgärder till den ekonomiska uppföljning som redan görs i kommunen. Energistrategen bör sitta i ledningsgruppen för att möjliggöra att det energistrategiskaarbetet blir en del av det övergripande strategiska arbetet För att säkra kontinuitet i arbetet bör energistrategen ha en tillsvidareanställning och inteprojektanställning Det behövs stöd för kommunernas utåtriktade energistrategiska arbete från regional ochnationell nivå. Formulera en öppen process med en bred regional systemsyn Att samverka kan bidra till mycket positivt samtidigt som det kan göra processen merkomplex.   • Management and stakeholder participation in local strategic energy planning : – examples from Sweden

    • Rapid environmental degradation and consumption of natural resources is a growing global concern, leading to the awareness that environmental sustainability is a critical new strategic objective. Municipalities are important actors when it comes to energy efficiency and improve environmental sustainability. One important tool to promote this work is implementing a local energy and/or climate strategy. How such local strategies are and could be designed in order to make a difference and promote sustainable energy solutions will be in focus for this paper. In order to promote strategic energy work at the local level, the, Swedish Energy Agency initiated a program called Sustainable Municipalities. As a part of this program participating municipalities were mandated to develop local energy and climate strategies.This paper aims at creating a general overview of approaches to, and uses of, local energy strategies among the participants in the Sustainable Municipalities program. This overview includes: analyzes of whether energy plans or strategies are present, which issues these plans address, what actors that are included, and whether follow-up is pursued.We will elucidate local energy strategies from a management perspective, and discuss if and how the composition of actors influence suggested goals and measures, if there were plans for implementation and follow-ups. Our theoretical ambitions are to integrate theories on how to manage an efficient energy strategic planning and on how to achieve successful stakeholder participation. This in order to discuss weaknesses and strengths in existing municipal energy planning practices and how to improve energy planning in relation to those theories.Document studies and structured telephone interviews with representatives from 60 municipalities were used to collect data. Around 75 percent of the municipalities in the study had adopted local energy strategies. This figure is surprisingly low; given that participation in the sustainable Municipalities program required energy strategies. In general, processes to develop a strategy included different parts of the municipality, and resulted in concrete measures. There were no correlations between included actors, and measures suggested and themes in the strategies. Most commonly occurring were strategies related to public buildings and other buildings owned by municipality owned companies. The most commonly suggested measures were related to transports. Almost all respondents claimed that they performed follow-ups and that this was planned for already in the set-up phase.Ten out of sixty respondents, who mainly were managers or strategists, meant that they were they alone were the main driving actors in strategic energy issues.The studied municipalities had not adopted energy plans or strategies to a higher degree compared to other Swedish municipalities. Cooperation is often broad within the municipalities but limited when it comes to external actors. The content of the energy plans do not always reflect the content of the measures and goals. The rather limited participation in the process could affect the impact and legitimacy of the strategies in the organizations.

These theories are used to discuss weaknesses and strengths in existing municipal energy planning practices and potential improvements.The results from this paper show that only 75% of the municipalities adopted local energy strategies. In addition, it was revealed that the processes to develop the strategies generally included several municipal actors although other stakeholders were rarely represented. The strategies resulted in concrete measures; however, there were no clear correlations between included actors, suggested measures, and identified themes in the strategies. The rather limited participation in the process could affect the impact and legitimacy of the strategies.

  • Översiktlig kartläggning av kommuners erfarenheter kring energi- och klimatstrategiarbete : en enkätstudie

    • Kommuner har en viktig roll i att bidra till positiv utveckling inom miljö- och hållbarhetsområdet genom myndighetsutövande, som fastighetsägare, informatör och ägare av energibolag et cetera. Alla kommuner måste också, enligt svensk lagstiftning upprätta en energiplan, men flera studier visar att efterlevnaden av denna lag är låg och att dess verkningsfullhet är ifrågasatt. Därför är det intressant att studera och analysera om och hur det energistrategiska arbetet ska utformas för att göra skillnad i lokala energiplaneringsprocesser. Den här rapporten presenterar resultaten från en enkätundersökning som genomfördes vintern 2010/2011 som en deletapp av forskningsprojektet ”Hållbara energi- och klimatstrategier – lärdomar och utvecklingspotential”. Syftet med undersökningen var att ge en övergripande ögonblicksbild av Uthållig kommun-kommunernas arbetet med energistrategier. Enkätundersökningen riktades till de som står som kontaktpersoner för respektive kommun i Uthållig kommun och behandlade frågor såsom: huruvida det finns en energistrategi, vilka områden som omfattas av strategin, vilka aktörer som varit med och utarbetat den, vilka aktörer som driver energi- och klimatarbetet i kommunen, samt om energistrategiarbetet utvärderas eller följs upp. Av de 66 kommuner som kontaktades valde 60 att delta i undersökningen, vilket resulterar i en svarsfrekvens på 92 procent. Resultaten från undersökningen visar att deltagarna i uthållig kommun inte har antagit energistrategier i större omfattning än övriga svenska kommuner och att strategierna främst tagits fram i samverkan inom kommunen och med de kommunala bolagen. Mindre än hälften av kommunerna har samverkat med externa aktörer som industri, medborgare eller handel i framtagandet av strategierna. Förankringen av det energi- och klimatstrategiska arbetet i de kommunala organisationerna tycks vara god och både politiker och tjänstemän är i stor utsträckning drivande. De vanligaste åtgärdsområdena i strategierna är energieffektivisering i kommunala fastigheter och transporter. Majoriteten av kommunerna har genomfört konkreta åtgärder för att implementera strategin och alla säger sig arbeta med uppföljning. Ett stort antal olika benämningar på energistrategier identifierades bland enkätsvaren och variationerna på vad som uppges ingå i energi- och klimatstrategierna är också stor. Att variationerna finns kan förklaras med att det finns fördelar för förankringen av arbetet om arbetsmetoder och semantik anpassas för de lokala förutsättningarna och rådande kultur. Detta kan också leda till osäkerhet när det gäller jämförbarhet med andra kommuner och för samverkan med andra kommuner. Detta kommer att studeras vidare i projektets kommande etapp.

  • Size distribution of colloidal trace metals and organic carbon during a coastal bloom in the Baltic Sea

    • The physico-chemical speciation of organic carbon and selected metals was measured during a coastal bloom in Ekhagen Bay, Baltic Sea, using ultrafiltration.One important objective with the study was to see if any depletion of trace metals could be measured in the directly bioavailable fraction (<1000 Da, the soluble low molecular weight fraction, LMW) during a plankton bloom. Filters with five different cut-offs were used (1 kD (1000 Da), 5 kD, 10 kD, 100 kD and 0.22 μm) in order to delineate the size distribution of colloidal organic carbon (COC) and trace metals. During the bloom in May, LMW Al, Co, Cu, Mn and Ni concentrations decreased although the colloidal and particulate concentrations were relatively high. Data show that desorption of colloidal and particulate bound trace metals to the LMW fraction was slower than the process depleting the LMW fraction. Estimates of the maximum active uptake of Cu, Ni and Mn by the phytoplankton, and the loss of non-bioactive Al from the LMW fraction, indicate that processes other than active uptake by phytoplankton must contribute to the observed depletion of trace metals in the LMW fraction. Hence, in order to estimate the bioavailable pool of trace metals for plankton during bloom conditions, these other processes must be understood and quantified. Transparent Exopolymeric Particles (TEP, reflecting sugar-rich phytoplankton exudates) increased around eight times during the plankton bloom. We hypothesize that the formation of TEP is a process that might be important for the transfer of trace metals from the LMW to the particulate fraction during the phytoplankton bloom, but the significance of TEP for this depletion in Baltic Sea surface water remains to be shown.

    • We have reported that transmembrane mucin MUC17 binds PDZ protein PDZK1, which retains MUC17 apically in enterocytes. MUC17 and transmembrane mucins MUC3 and MUC12 are suggested to build the enterocyte apical glycocalyx. Carbachol (CCh) stimulation of the small intestine results in gel-forming mucin secretion from goblet cells, something that requires adjacent enterocytes to secrete chloride and bicarbonate for proper mucin formation. Surface labeling and confocal imaging demonstrated that apically expressed MUC17 in Caco-2 cells and Muc3(17) in murine enterocytes were endocytosed upon stimulation with CCh. Relocation of MUC17 in response to CCh was specific as MUC3 and MUC12 did not relocate following CCh stimulation. MUC17 colocalized with PDZK1 under basal conditions, while MUC17 relocated to the terminal web and into early endosomes after CCh stimulation. CCh stimulation concomitantly internalized the Na(+/)H(+) exchanger 3 (NHE3) and recruited cystic fibrosis transmembrane conductance regulator (CFTR) to the apical membranes, a process that was important for CFTR-mediated bicarbonate secretion necessary for proper gel-forming mucin unfolding. The reason for the specific internalization of MUC17 is not understood, but it could limit the diffusion barrier for ion secretion caused by the apical enterocyte glycocalyx or alternatively act to sample luminal bacteria. Our results reveal well-orchestrated mucus secretion and trafficking of ion channels and the MUC17 mucin.

    • MUC2, the major colonic mucin, forms large polymers by N-terminal trimerization and C-terminal dimerization. Although the assembly process for MUC2 is established, it is not known how MUC2 is packed in the regulated secretory granulae of the goblet cell. When the N-terminal VWD1-D2-D'D3 domains (MUC2-N) were expressed in a goblet-like cell line, the protein was stored together with full-length MUC2. By mimicking the pH and calcium conditions of the secretory pathway we analyzed purified MUC2-N by gel filtration, density gradient centrifugation, and transmission electron microscopy. At pH 7.4 the MUC2-N trimer eluted as a single peak by gel filtration. At pH 6.2 with Ca(2+) it formed large aggregates that did not enter the gel filtration column but were made visible after density gradient centrifugation. Electron microscopy studies revealed that the aggregates were composed of rings also observed in secretory granulae of colon tissue sections. The MUC2-N aggregates were dissolved by removing Ca(2+) and raising pH. After release from goblet cells, the unfolded full-length MUC2 formed stratified layers. These findings suggest a model for mucin packing in the granulae and the mechanism for mucin release, unfolding, and expansion.

    • Our model of the MUC2 mucin shows a well-organized netlike gel that is cross-linked by six different covalent and noncovalent bonds. When the MUC2 mucin is packed in the mucin granule it is organized by an amino-terminal concatenated ring platform formed at high calcium and low pH. This packing allows an ordered release and a normal mucin expansion when calcium is removed and pH increased by bicarbonate. This process is defective in the absence of cystic fibrosis transmembrane conductance regulator (CFTR)-dependent bicarbonate transport. The expanded secreted mucin is suggested to be self-organizing by properties inherited in the MUC2 mucin and by proteolytic processes.

    • Between 1967 and 1972, Swedish futures studies emerged as a distinct political and intellectual endeavor. In this development, the humanities and social sciences played a special role, especially through the idea that human knowledge and knowledge about a “human system” could be brought to bear on societal problems and used to forge a new approach to the future. We focus here on two scholars, historian Birgitta Odén and geographer Torsten Hägerstrand, proposing that they were important f igures in developing a new engagement with problems of social and human time in Swedish social science and humanities research.

    • Ongoing urban exploitation is increasing pressure to transform urban green spaces, while there is increasing awareness that greenery provides a range of important benefits to city residents. In efforts to help resolve associated problems we have developed a framework for integrated assessments of ecosystem service (ES) benefits and values provided by urban greenery, based on the ecosystem service cascade model. The aim is to provide a method for assessing the contribution to, and valuing, multiple ES provided by urban greenery that can be readily applied in routine planning processes. The framework is unique as it recognizes that an urban greenery comprises several components and functions that can contribute to multiple ecosystem services in one or more ways via different functional traits (e.g. foliage characteristics) for which readily measured indicators have been identified. The framework consists of five steps including compilation of an inventory of indicator; application of effectivity factors to rate indicators' effectiveness; estimation of effects; estimation of benefits for each ES; estimation of the total ES value of the ecosystem. The framework was applied to assess ecosystem services provided by trees, shrubs, herbs, birds, and bees, in green areas spanning an urban gradient in Gothenburg, Sweden. Estimates of perceived values of ecosystem services were obtained from interviews with the public and workshop activities with civil servants. The framework is systematic and transparent at all stages and appears to have potential utility in the existing spatial planning processes.

    • This study deals with the meanings and conditions of motherhood for some Muslim women in Sweden, the majority of which live in Stockholm. The analysis is based mainly on 16 in-depth interviews with women who self-identify as Muslims. A majority of them were born in Sweden.In this study, motherhood and mothering are defined as intentional care work situated within, and shaped by, specific social, cultural and historical contexts. I examine which gendered, religious and spatial meanings are associated with mothering and Muslim identity in a Swedish transnational context. The analysis shows that religion (for most of the women) constitutes an interpretational frame for motherhood and for how children should be mothered into good Muslims and citizens. The women represent Islam as a facilitating religion by making a distinction between religion and culture. The study also shows how the women approach the problem of maintaining the children’s Muslim identities and their self-esteem in a secularized and islamophobic Swedish context, and how they stress the importance of the child developing a strong inner self. Based on the women’s own experiences of having been singled out as different, they respond to a racist logic associated with certain norms and conceptions about what counts as freedom or oppression. The interviews also reveal a transnational aspect of their mothering in which they consider what other places can offer their children. The thesis shows on the one hand how an authentic Muslim identity is related to ideas about Muslim places and origins; on the other hand it demonstrates how the women’s ambivalent affinities with Sweden, and (what they consider to be) Swedish and Muslim values, destabilize such an unambiguous connection. These ambivalent identifications show how the women’s conditional affinities become relevant for how they speak about motherhood and mothering and for how they relate to questions concerning “the good of the child”.

  • Psychological risk factors for exercise dependence

    • The main aim of this study was to investigate if exercisers' personality characteristics were associated with exercise dependence. Specifically, the purpose was to examine if anxiety, obsessive passion, and physical appearance orientation were associated to an increased risk for exercise dependence. Participants were 330 exercisers from exercise groups, sport clubs and university sport science classes in the southwest of Sweden. Data were analysed using CHAID (Chi-squared Automatic Interaction Detection) analysis. The CHAID analysis indicated that anxiety was the main predictor of exercise dependence. More specifically, 12.7% more exercisers who experienced high levels of anxiety symptoms (i.e. scores above 6), were, in comparison to the exercises experiencing low levels of anxiety, classified as ?at risk for exercise dependence?. For exercisers that reported low levels of anxiety symptoms (i.e. scores below 7), obsessive passion for exercise was a positive statistically significant predictor (absolute risk difference?=?8.6%). Overall, the results highlight anxiety as a main risk factor behind exercise dependence. Also, the risk of exercise dependence may increase either from obsessive passion or as a coping strategy for anxiety. Furthermore, results may illustrate two types of exercise dependence; ?primary? exercise dependence driven mainly by an obsessive passion for exercise and ?secondary? exercise dependence where exercise function as a strategy to cope with anxiety.

  • Psychosocial Predictors of Drop-Out from Organised Sport : A Prospective Study in Adolescent Soccer

    • In recent years an increased drop-out rate in adolescents' soccer participation has been observed. Given the potentially adverse consequences of drop-out from soccer, more information about risk factors for drop-out is warranted. In the current study, Classification and Regression Tree (CRT) analysis was used to investigate demographic and motivational factors associated with an increased risk of drop-out from adolescent soccer. The results of this study indicate that older age, experiencing less autonomy support from the coach, less intrinsic motivation, being female, and lower socioeconomic status are factors associated with an increased risk of drop-out. An interpretation of the results of this study is that coaches play a central part in creating a sports context that facilitates motivation and continued soccer participation. Based on the findings of the current study we propose that soccer clubs implement theoretically informed coach education programs to help coaches adopt autonomy-supportive coaching strategies.

  • Study of excited Ξ baryons with the P¯ ANDA detector

    • The study of baryon excitation spectra provides insight into the inner structure of baryons. So far, most of the world-wide efforts have been directed towards N∗ and Δ spectroscopy. Nevertheless, the study of the double and triple strange baryon spectrum provides independent information to the N∗ and Δ spectra. The future antiproton experiment P¯ANDA will provide direct access to final states containing a Ξ¯ Ξ pair, for which production cross sections up to μb are expected in p¯p reactions. With a luminosity of L= 10 31 cm- 2 s- 1 in the first phase of the experiment, the expected cross sections correspond to a production rate of ∼106events/day. With a nearly 4 π detector acceptance, P¯ANDA will thus be a hyperon factory. In this study, reactions of the type p¯p → Ξ¯ +Ξ∗ - as well as p¯p → Ξ¯ ∗ +Ξ- with various decay modes are investigated. For the exclusive reconstruction of the signal events a full decay tree fit is used, resulting in reconstruction efficiencies between 3 and 5%. This allows high statistics data to be collected within a few weeks of data taking.

    • The antiproton experiment PANDA at FAIR is designed to bring hadron physics to a new level in terms of scope, precision and accuracy. In this work, its unique capability for studies of hyperons is outlined. We discuss ground-state hyperons as diagnostic tools to study non-perturbative aspects of the strong interaction, and fundamental symmetries. New simulation studies have been carried out for two benchmark hyperon-antihyperon production channels: p¯ p→ Λ¯ Λ and p¯ p→ Ξ¯ +Ξ-. The results, presented in detail in this paper, show that hyperon-antihyperon pairs from these reactions can be exclusively reconstructed with high efficiency and very low background contamination. In addition, the polarisation and spin correlations have been studied, exploiting the weak, self-analysing decay of hyperons and antihyperons. Two independent approaches to the finite efficiency have been applied and evaluated: one standard multidimensional efficiency correction approach, and one efficiency independent approach. The applicability of the latter was thoroughly evaluated for all channels, beam momenta and observables. The standard method yields good results in all cases, and shows that spin observables can be studied with high precision and accuracy already in the first phase of data taking with PANDA.

    • Goblet cells and their main secretory product, mucus, have long been poorly appreciated; however, recent discoveries have changed this and placed these cells at the center stage of our understanding of mucosal biology and the immunology of the intestinal tract. The mucus system differs substantially between the small and large intestine, although it is built around MUC2 mucin polymers in both cases. Furthermore, that goblet cells and the regulation of their secretion also differ between these two parts of the intestine is of fundamental importance for a better understanding of mucosal immunology. There are several types of goblet cell that can be delineated based on their location and function. The surface colonic goblet cells secrete continuously to maintain the inner mucus layer, whereas goblet cells of the colonic and small intestinal crypts secrete upon stimulation, for example, after endocytosis or in response to acetyl choline. However, despite much progress in recent years, our understanding of goblet cell function and regulation is still in its infancy.

    • Platelet-derived growth factor-B (PDGFB) is necessary for normal cardiovascular development, but the relative importance of different cellular sources of PDGFB has not been established. Using Cre-lox techniques, we show here that genetic ablation of Pdgfb in endothelial cells leads to impaired recruitment of pericytes to blood vessels. The endothelium-restricted Pdgfb knockout mutants also developed organ defects including cardiac, placental and renal abnormalities. These defects were similar to those observed in Pdgfb null mice. However, in marked contrast to the embryonic lethality of Pdgfb null mutants, the endothelium-specific mutants survived into adulthood with persistent pathological changes, including brain microhemorrhages, focal astrogliosis, and kidney glomerulus abnormalities. This spectrum of pathological changes is reminiscent of diabetic microangiopathy, suggesting that the endothelium-restricted Pdgfb knockouts may serve as models for some of the pathogenic events of vascular complications to diabetes.     • Childhood CNS tumor survivors risk health and functional impairments that threaten normal psychological development and self-perception. This study investigated the extent to which health and functional ability predict adult survivors' body image (BI) and self-confidence regarding sports and physical activity. The study cohort covered 708 eligible ≥18year old CNS tumor survivors, and data from 528 (75%) were analyzed. Disability was estimated using the Health Utilities Index™ Mark2/3, a multidimensional self-report instrument. Physical self-confidence in terms of BI and sports/physical activity-related self-confidence (SPAS) were assessed using the BI and the Sports/Athletics modules of a standardized self-report assessment scale. In adjusted regression models, global health and functional status (GHFS) predicted BI (B=0.94, 95% CI 0.69-1.19) and SPAS (B=0.79, 95% CI 0.55-1.04). Emotion and pain, and to a lesser degree cognition, speech and vision disability, were associated with poorer BI and SPAS. Gender, sub-diagnosis, and time since diagnosis influenced the relationship between health status and physical self-confidence outcomes. Females had poorer GHFS, BI and SPAS than males. Decreased health and functional ability following childhood CNS cancer intrudes on physical self-confidence, with females being at heightened risk for both disability and negative self-confidence. Identified disability and gender-related risk calls for a follow-up plan that integrates treatment of psychological sequelae in lifetime monitoring of childhood CNS tumor survivors to restore and protect self-image and self-confidence, essential mental health correlates. An expanded plan should recognize the need for such services, optimizing life-long quality of survival for CNS tumor survivors.

    • Tumor cells often carry mutations in genes that control cell survival, and become resistant to signals that trigger cell death. Yet, some cell death pathways remain intact in tumor cells. If identified, these pathways might be exploited to selectively remove tumor cells. HAMLET (human α-lactalbumin made lethal to tumor cells) is a protein-lipid complex derived from human milk that activates cell death programs in tumor cells but not in healthy differentiated cells. We use HAMLET as a tool to identify apoptosis and apoptosis-like cell death mechanisms in tumor cells and to understand if these mechanisms differ between tumor and healthy cells. HAMLET interacts with the cell surface, translocates into the cytoplasm and accumulates in cell nuclei, where it disrupts the chromatin. Recent in vivo studies have shown that HAMLET maintains the tumoricidal activity in glioblastoma, papilloma and bladder cancer models, with no significant side effects. The results suggest that HAMLET should be explored as a new therapeutic agent with selectivity for the tumor and with little toxicity for adjacent healthy tissue. Such therapies are a much-needed complement to conventional treatments, to reduce the side effects and improve the selectivity.

    • AIMS AND OBJECTIVES:To explore and describe how parents of preterm and/or sick infants in neonatal care perceive their sleep.BACKGROUND:Parents experience many stressful situations when their newborn infant is preterm and/or sick. This affects bonding. By developing more family-centred care units with single-family rooms, parents are given the opportunity to stay and care for their newborn infant(s) 24 hours a day. Lack of sleep may affect new parents' ability to cope with the many challenges they face on a daily basis.DESIGN:A phenomenographic study with an inductive and exploratory design.METHODS:Semi-structured interviews were conducted with twelve parents of infants in neonatal care between January-March 2012. To describe variations in perception of the phenomenon, data were analysed using phenomenography.FINDINGS:Four descriptive categories were identified within the phenomenon sleep in parents of preterm and/or sick infants in neonatal care: impact of stress on sleep; how the environment affects sleep; keeping the family together improves sleep; and, how parents manage and prevent tiredness.CONCLUSION:Anxiety, uncertainty and powerlessness have a negative influence on sleep. This can be decreased by continuous information, guidance and practical support. Skin-to-skin care was perceived as a stress-reducing factor that improved relaxation and sleep and should be encouraged by the nurse. The parents also mentioned the importance of being together. Having a private place where they could relax and take care of themselves and their newborn infant improved sleep. It was also desirable to involve older siblings in order to decrease feelings of loneliness, sadness and isolation.RELEVANCE FOR CLINICAL PRACTICE:Improved parental sleep in neonatal care may help the families cope with the situation and facilitate problem-solving, emotional regulation and the transition to parenthood.

    • IntroductionSleep is important for mental and emotional health. For parents staying in the hospital with their preterm and/or sick infant, lack of sleep may affect their ability to handle the situation, supporting their infant, and participate in decision-making. Moreover, when a child is born preterm, parents may experience stress that potentially affects their ability to interact and bond with the infant. PurposeTo describe parents’ perceptions of what it is like to sleep in a neonatal intensive care unit (NICU) in a room nearby or in the same room as their infant. MaterialTwelve parents (eight mothers and four fathers) of infants born between week 29 and 36, in three different hospitals, were included. Eight of the parents slept in the same room as their infants and four parents slept in parents’ rooms in the NICU.   MethodsParents were interviewed with open-ended questions. Data was analysed with a phenomenographic method according to Marton and Both. ResultsFive descriptive categories in the phenomenon of parents’ perception of how it is to sleep  in a NICU  were  identified; Transition to parenthood, How parents perceive and manage their tiredness, A feeling of being out of control, Different forms of support and Environment. ConclusionsParents in the NICU are vulnerable, in a stressful situation, with an infant in need of neonatal intensive care. At the same time they are going through a complicated transition to parenthood. Hence sleep is important for the parents in several aspects, and NICU staff needs to acknowledge and promote parents’ sleep.

    • Background: The underlying cause of a urethral stricture can sometimes be obscure. It is possible that an injury to the urethra induces an immunological cascade that generates scar tissue and fibrosis, eventually resulting in a stricture. If such immuno-logical reactions could be better elucidated, immunological therapies could possibly emerge. Objective: To evaluate if ectopic germinal centres exist in urethral stricture disease. Design, setting, and participants: Resected stricture specimens from 45 patients undergoing open bulbar urethroplasty with excision and anastomosis were assessed. Histopathological characteristics, such as fibrosis (grade I & ndash;III), inflammation, and scle-rosis, were evaluated using immunostaining for CD3 (T cells), CD20 (B cells), and CD21 (follicular dendritic cells). Outcome measurements and statistical analysis: The primary outcome measure was the presence or absence of a germinal centre. The secondary outcome was evaluation of any correlation between the degree of fibrosis and germinal centres. Fisher & rsquo;s exact test was used for univariate analysis. Results and limitations: In six patients, ectopic germinal centres were found. In ten patients, there was no inflammation at all. There was no correlation found between the degree of fibrosis and the abundance of immunohistochemically detected immune cells. Conclusions: Ectopic germinal centres, with B and T cells as well as follicular dendritic cell networks, do exist in urethral stricture disease. This finding may open up for novel research avenues on the possibility of adopting immunological treatments for urethral stricture disease. Patient summary: In patients with a narrowing of the urethra due to any kind of trauma, we looked for the presence of centres of immunological reaction in urethral tissue. We identified these immunological centres (also called germinal centres) in some patients. This intriguing finding suggests that immunological treatments may have potential for men with scar tissue in a narrowed urethra. (c) 2021 The Authors.     • The UN emphasises the importance of collaboration and integrated approaches to effectively implement the SDGs. Much of the action will have to take place locally where municipalities will play an important role in coordinating the efforts towards SDG fulfilment. They are constant local actors close to citizens and they can also influence other actors through their strategies. This paper reflects on how the SDGs can be integrated into existing strategies in order to avoid parallel non-effective processes and to avoid the risk of the SDGs to become marginalised. Furthermore, the paper discusses roles and preconditions of municipalities in the SDG implementation process. This study focuses the implementation of the SDGs into a regional municipality’s strategic planning and management, Region Östergötland, Sweden. The challenges and opportunities connected to implementing the SDGs will be problematized, and the paper gives recommendations on how this type of organisations can implement the SDGs taking advantage from qualities in already existing management and working procedures.

    • A new social movement was formed in Sweden in 1961: Kampanjen mot atomvapen (KMA, “The Campaign for Nuclear Disarmament”). Opposing the military’s plans to acquire Swedish nuclear bombs and protesting against the escalating tensions of the Cold War, KMA in the years 1961‒1963 rallied young people and students and introduced several new forms of collective political actions in the Swedish repertoire of contention. One example was peace or disarmament marches between Swedish cities while another consisted of illegal sit-downs. This article analyzes these new forms of collective action, focusing on the question of how they spread to Sweden. It is here argued that KMA must be seen as a Swedish part of a broad, transnational social movement against nuclear weapons in the years around 1960. The most important national branch of this movement was the British Campaign for Nuclear Disarmament (CND) formed in the late 1950s. Swedish activists travelled to London and participated in CND activities, learning how to organize “easter marches” for disarmament as well as sit-downs. Back in Sweden, they adopted these new forms of collective action (and were, in turn, visited by British as well as Danish activists). For example, they arranged illegal sit-downs outside the Soviet Embassy in Stockholm to protest atmospheric nuclear bomb tests in the fall of 1961 and organized peace marches to military facilities in the vicinity of the capital as well as from the city of Södertälje to Stockholm. KMA also used the same symbols as CND, such as the by now well-known circular peace symbol designed by British artist Gerald Holtom. This means that not only new forms of collective action were introduced but also new political identities and ways of organizing new forms of social movements. In turn, this turns KMA into an important precursor of the broader (and more studied) radicalization of Swedish youth in the late 1960s.

    • Objective: In Sweden, cyclists, pedestrians, and moped riders share the space on combined pedestrian and cycle paths, and their speeds may differ greatly. As a starting point, this article studies the speed component and how cyclists' speed varies at pedestrian and cycle paths depending on the day, week, and year; road user composition; and road design.Methods: Three data sources were used: 1) Existing measurements of cycle speed and flow at 17 sites in 3 different Swedish municipalities; 2) complementary measurements of cycle speed and flow at 5 sites; and 3) roadside observations of bicycle types at these 5 new sites.Results: The average speed of cyclists on the paths varied between 12.5 and 26.5 km/h. As expected, the lower average speeds were found in uphill directions, near intersections, and on paths with high pedestrian flows. The higher speeds were found in downhill directions and on commuter routes. In all, 70%-95% of road users observed on pedestrian and cycle paths were cyclists, and 5%-30% were pedestrians. The 2 sites with the highest proportion of electric-assisted bicycles and racer bicycles also had the highest average speeds.Conclusions: The relationship between bicycle type and measured speed was not entirely clear, but the results suggest that paths with higher proportions of electric and racer bicycles have higher average speeds. There also appears to be a connection between average speed and the width of the distribution; that is, the higher the average speed, the wider the speed distribution. More research is needed on how speed levels and speed variance affect accident risk.

    • The purpose of this paper is to enhance the understanding of cyclists’ speed on pedestrian and cycle paths and to understand how cyclists adapt their speed to other road users and the surrounding environment. The paper is based on a study where three different data sources were used: existing measurements of cycle speed and flow in three different Swedish municipalities, Eskilstuna, Linköping and Stockholm (19 sites); new measurements of cycle speed and flow in Linköping (4 sites) and Stockholm (1 site); and road side observations of bicycle types at these five sites.The average speed of cyclists on the paths varied between 15 and 25 kilometres per hour. As expected, the lower average speeds were found in uphill directions, near intersections and on paths with high pedestrian flows. The higher speeds were found in downhill directions and on commuter routes. In all, 70–95 percent of the road users observed on pedestrian and cycle paths were cyclists while 5–30 percent were pedestrians. The most common type of bicycle used was a comfort bike, followed by a trekking bike. Electric assisted bicycles and racer bikes occurred at all sites, with a proportion of 1–10 percent and 1–15 percent, respectively. The relationship be-tween bicycle type and measured speed was not entirely clear, but the paths with higher pro-portions of electric and racer bikes generally had higher speeds. There also appears to be a con-nection between average speed and the width of the distribution – the higher the average speed, the wider the speed distribution. More research is needed on how this affects the accident risk.

    • På gång- och cykelvägar blandas många olika trafikanter med skilda hastighetsanspråk. Skillnader i hastighet skulle kunna vara en orsak till brister i samspel trafikanterna emellan och till att incidenter och olyckor uppstår. Syftet med detta projekt är att få ökad kunskap om cyklisters hastigheter på gång- och cykelvägar och förståelse för hur cyklisterna anpassar hastigheten till omgivande trafikanter och trafikmiljö. Tre olika typer av data har använts: Hastighet och cykelflöde från tidigare mätningar i kommunerna Eskilstuna, Linköping och Stockholm på totalt 18 platser; hastighet och cykelflöde i Linköping (4 platser) och Stockholm (1 plats) från nya mätningar; observationer avseende typ av cyklar vid dessa fem platser. Utöver detta har vi också sammanställt några utvalda referenser.Data visar att cyklisternas medelhastighet på utvald sträcka varierar mellan 15 och 25 kilometer i timmen. Som väntat förekommer de lägre medelhastigheterna i uppförsbacke, vid närhet till korsning och vid stort fotgängarflöde medan de högre framför allt märks i nedförsbacke och på pendlingsstråk. Vi kan inte påvisa någon generell ökning av cyklisternas hastighet i ett längre tidsperspektiv, vare sig i medelhastighet eller andel cyklister som håller högre fart än 30 kilometer i timmen. Däremot har cykelflödet ökat på många platser. Det innebär att antalet cyklister som håller en hög hastighet, exempelvis över 30 kilometer i timmen, ökar, även om inte andelen ökar. Detta kan felaktigt uppfattas som ökade medelhastigheter. Ungefär 70–95 procent av de trafikanter som observerades på gång- och cykelvägarna var cyklister och cirka 5–30 procent var fotgängare. En ytterst liten andel var mopedister, endast 0,2 procent. Komfortcykeln var den vanligast förekommande typen av cykel, följt av trekkingcykel. Elcykel förekom på alla platser, men i varierande omfattning från 1 till 10 procent. Racercykel förekom också på alla platser, mellan 1–15 procent. Sambandet mellan typen av cykel och hastighetsanspråk är inte helt entydigt, men resultaten stöder påståendet att cyklister på elcykel och racercykel har högre hastighetsanspråk.

  • Hyper-osmolarity and calcium chelation: Effects on cystic fibrosis mucus

    • A non-functional Cystic Fibrosis Transmembrane conductance Regulator (CFTR) leads to the disease cystic fibrosis (CF). Although the CFTR is expressed in multiple organs, pulmonary disease is the major cause of illness and death in patients with CF. Stagnant mucus, causing airway obstruction, bacterial overgrowth, persistent inflammation and tissue destruction characterizes the disease, but how the defect in CFTR function is coupled to the mucus phenotype is still controversial. We have recently shown that bicarbonate ions passing through CFTR are necessary for proper unfolding of the MUC2 mucin, thus highlighting the importance of bicarbonate ion transport via the CFTR and the ability of these ions to raise the pH and chelate calcium bound to the mucin as the important steps in forming normal mucus. In order to find potential CF treatments and expand our knowledge about the usefulness of bicarbonate as an active ingredient in formulations to alleviate mucus plugging, we used an Ussing-type chamber and explants from the F508del-CFTR mutant mouse ileum to test the effect of calcium chelators on mucus attachment, either in isolation or in combination with osmolytes such as mannitol or hypertonic saline. We found that increasing the concentration of bicarbonate, both alone or in combination with increased osmolarity of the solution, detached the otherwise attached CF mucus. (C) 2015 Elsevier B.V.     • Peyer's patches (PPs) are collections of lymphoid follicles in the small intestine, responsible for scanning the intestinal content for foreign antigens such as soluble molecules, particulate matter as well as intact bacteria and viruses. The immune cells of the patch are separated from the intestinal lumen by a single layer of epithelial cells, the follicle-associated epithelium (FAE). This epithelium covers the dome of the follicle and contains enterocyte-like cells and M cells, which are particularly specialized in taking up antigens from the gut. However, the presence and number of goblet cells as well as the presence of mucus on top of the FAE is controversial. When mouse ileal PPs were mounted in a horizontal Ussing-type chamber, we could observe a continuous mucus layer at mounting and new, easily removable mucus was released from the villi on the patch upon stimulation. Confocal imaging using fluorescent beads revealed a penetrable mucus layer covering the domes. Furthermore, immunostaining of FAE from mice, rats and humans with a specific antibody against the main component of intestinal mucus, the MUC2 mucin, clearly identify mucin-containing goblet cells. Transmission electron micrographs further support the identification of mucus releasing goblet cells on the domes of PPs in these species.
