    • Using a sensitive and rapid method combining broad-range PCR amplification of bacterial 16S rDNA fragments and pyrosequencing for detection, identification and typing, we have found contaminating bacterial DNA in our reagents used for PCR. Identified bacteria are the water-borne bacterial genera Pseudomonas, Stenotrophomonas, Xanthomonas, Ralstonia and Bacillus. Our results are in concordance with recent reports of contaminated industrial water systems. In light of this conclusion, we believe that there is a need for increased awareness of possible contamination in uncertified widely used molecular biology reagents, including ultra-pure water. Since sequence-based 16S rDNA techniques are used in a variety of settings for bacterial typing and the characterization of microbial communities, we feel that future certification of molecular biology reagents, as free of nucleic acids, would be advantageous. ⌐ 2002 Federation of European Microbiological Societies.     • I den här artikeln diskuterar vi generella iakttagelser av Idrottslyftet utifrån Idrottsstödsutredningens slutsatser om Handslaget (1). Vi gör en kvalitativ bedömning utifrån erfarenheterna från våra fem förbundsvisa utvärderingar (Klätter-, Orientering-, Ridsport-, Sim och Taekwondoförbundet) men också utifrån kunskap om andra förbund, från andra utvärderare och från i sammanhanget relevanta nyckelpersoner inom idrottsrörelsen. RF är mer intresserade av kvalitativa data än kvantitativa, eftersom de kvantitativa uppgifterna ändå kommer in via projektrapporteringen. Här följer således ett försök att tolka och karakterisera ett antal iakttagelser som vi bedömer är generella för idrottsrörelsens idrottslyftsarbete.

    • Genom regeringens satsning, kallad” Idrottslyftet”, fick den svenska idrottsrörelsen under perioden 2007–2011 möjlighet att genomföra den hittills största satsningen någonsin för att utveckla barn-och ungdomsidrotten. Idrottsrörelsen har sammanlagt fått två miljarder kronor för att öppna dörrarna till idrotten för fler barn och ungdomar och utveckla verksamheten, så att de väljer att idrotta längre upp i åldrarna. Arbetet skulle utgå från ett jämställdhets-och jämlikhetsperspektiv och genomsyras av riktlinjerna i” Idrotten vill”.Denna rapport utvärderar detta arbete i fyra specialidrottsförbund: Svenska Ridsportförbundet, Svenska Klätterförbundet, Svenska Orienteringsförbundet, Svenska Simförbundet och Svenska Taekwondoförbundet.

    • OBJECTIVES: Severe chronic obstructive pulmonary disease affects and changes the lives of both affected persons and next of kin. There is a need for support and a sense of coherence to manage the life situation and minimize the symptom and caregiver burden. The aim of this study was to diverge or converge views of symptom burden, caregiver burden, the need for support, and sense of coherence in persons with chronic obstructive pulmonary disease and their next of kin to gain a deeper and broader knowledge and understanding.METHODS: A mixed methods study with data from interviews and four validated questionnaires from persons affected by chronic obstructive pulmonary disease in GOLD stages III and IV and their next of kin.RESULTS: Questionnaires from 112 persons affected by chronic obstructive pulmonary disease, and 71 next of kin, together with 25 and 21 interviews, show that; there is a difference between estimated symptoms and caregiver burden and experiences expressed in their own words. There is also a defect regarding meaningfulness, comprehensibility, and manageability affecting daily life. Symptoms and caregiver burden, together with the sense of coherence, strengthen the need for support.DISCUSSION: The complexity of the life situation leads to a need for supportive interventions to strengthen internal and external resources.

    • BACKGROUND: Chronic obstructive pulmonary disease (COPD) is an inflammatory and irreversible lung disease. For next of kin caregiver burden can be a consequence of the situation of being close to a person affected by a chronic disease and in need of help. When there is an imbalance between stressors and resources to cope with the situation, caregiver burden emerges. Knowledge is sparse about how the caregiver burden is experienced by the next of kin. Therefore, the aim of this study is to identify and describe the caregiver burden experienced by next of kin of persons with severe COPD.METHOD: Qualitative interviews with 21 next of kin were conducted. Thematic analysis was used in accordance with the six steps of Braun and Clarke.RESULTS: The next of kin experience caregiver burden as 1) changed roles in daily life 2) putting life on hold 3) to stand aside. The next of kin are in need of support to manage daily life.CONCLUSIONS: The caregiver burden affect the next of kin physically and emotionally. To prevent advance consequences, person-centered care can be used to support the next of kin in the situation.

    • The qualitative interview is a method to obtain knowledge about the persons own lived experience and the possibility to share the person’s own life experiences.A qualitative interview study with planed interviews  between 20 to 30 minutes related to the persons health. The interviews lasted for 40 to 70 minuets. At the beginning of each interview, most of the informants were affected by symptoms, which faded out during the interview. An interview is aiming for data collection but the conversation can be treating the affected person so the qualitative interview is more than just data collection-becoming an intervention.

    • Background: Chronic obstructive pulmonary disease (COPD) is a prevalent disease as with its symptom and treatment gives a burden for the affected person, family, health care and finances. Today the knowledge and understanding are sparse regarding COPD affected person´s own lived experiences about the symptom burden and its effect on their daily life.  Due to this knowledge gap the aim of this study was to identify and describe the symptom burden and its effect on daily life in people with COPD based on their own lived experiences.Subject and Method: Eleven males and fourteen females in GOLD stage III and IV within an age of 58-82 years were interviewed. An interview guide guided the face-to-face interviews. Data was analyzed with Thematic Analysis following the six steps according to Braun and Clarke.Results: The result show one theme; An altered everyday life. The altered everyday life lead to a need of support to handle the everyday life and  different strategies to live as desired. Persons with COPD need to take the day as it comes dependent on the status of the day. The life is not easy to plan, and the day must be taken as it comes. The life is handled with strategies in breathing techniques, take care of the home and garden as well as the emotions. Support from the next of kin, society and the health care is important.Conclusion: This study provides insight in that persons with COPD in stage III and IV have an altered life caused by the symptom burden. They must struggle with strategies to handle everyday life. There is a need of support from next of kin and society to facilitate life, but this support needs to be well-balanced.

    • If cellulosic materials are to replace materials derived from non-renewable resources, it is necessary to overcome intrinsic limitations such as fragility, permeability to gases, susceptibility to water vapour and poor three-dimensional shaping. Novel properties or the enhancement of existing properties are required to expand the applications of cellulosic materials and will create new market opportunities. Here we have overcome the well-known restrictions that impede melt-processing of high cellulose content composites. Cellulose fibres, partially derivatised to dialcohol cellulose, have been used to manufacture three-dimensional high-density materials by conventional melt processing techniques, with or without the addition of a thermoplastic polymer. This work demonstrates the use of melt processable chemically modified cellulose fibres in the preparation of a new generation of highly sustainable materials with tuneable properties that can be tailored for specific applications requiring complex three-dimensional parts.

  • HbA1C and Cancer Risk in Patients with Type 2 Diabetes - A Nationwide Population-Based Prospective Cohort Study in Sweden

    • Background: Diabetes is associated with increased cancer risk. The underlying mechanisms remain unclear. Hyperglycemia might be one risk factor. HbA1c is an indicator of the blood glucose level over the latest 1 to 3 months. This study aimed to investigate association between HbA1c level and cancer risks in patients with type 2 diabetes based on real life situations. Methods: This is a cohort study on 25,476 patients with type 2 diabetes registered in the Swedish National Diabetes Register from 1997-1999 and followed until 2009. Follow-up for cancer was accomplished through register linkage. We calculated incidences of and hazard ratios (HR) for cancer in groups categorized by HbA1c <= 58 mmol/mol (7.5%) versus >58 mmol/mol, by quartiles of HbA1c, and by HbA1c continuously at Cox regression, with covariance adjustment for age, sex, diabetes duration, smoking and insulin treatment, or adjusting with a propensity score. Results: Comparing HbA1c >58 mmol/mol with <= 58 mmol/mol, adjusted HR for all cancer was 1.02 [95% CI 0.95-1.10] using baseline HbA1c, and 1.04 [95% CI 0.97-1.12] using updated mean HbA1c, and HRs were all non-significant for specific cancers of gastrointestinal, kidney and urinary organs, respiratory organs, female genital organs, breast or prostate. Similarly, no increased risks of all cancer or the specific types of cancer were found with higher quartiles of baseline or updated mean HbA1c, compared to the lowest quartile. HR for all cancer was 1.01 [0.98-1.04] per 1%-unit increase in HbA1c used as a continuous variable, with non-significant HRs also for the specific types of cancer per unit increase in HbA1c. Conclusions: In this study there were no associations between HbA1c and risks for all cancers or specific types of cancer in patients with type 2 diabetes.

    • The beta-actin gene encodes 1 of 6 different actin proteins. De novo heterozygous missense mutations in ACTB have been identified in patients with Baraitser-Winter syndrome (BRWS) and also in patients with developmental disorders other than BRWS, such as deafness, dystonia, and neutrophil dysfunction. We describe 2 different novel de novo missense ACTB mutations, c.208Camp;gt;G (p.Pro70Ala) and c.511Camp;gt;T (p.Leu171Phe), found by trio exome sequencing analysis of 2 unrelated patients: an 8-year-old boy with a suspected BRWS and a 4-year-old girl with unclear developmental disorder. The mutated residue in the first case is situated in the actin H-loop, which is involved in actin polymerization. The mutated residue in the second case (p.Leu171Phe) is found at the actin barbed end in the W-loop, important for binding to profilin and other actin-binding molecules. While the boy presented with a typical BRWS facial appearance, the girl showed facial features not recognizable as a BRWS gestalt as well as ventricular arrhythmia, cleft palate, thrombocytopenia, and gray matter heterotopia. We reviewed previously published ACTB missense mutations and ascertained that a number of them do not cause typical BRWS. By comparing clinical and molecular data, we speculate that the phenotypic differences found in ACTB missense mutation carriers might supposedly be dependent on the conformational change of ACTB.

  • Normal and Functional TP53 in Genetically Stable Myxoid/Round Cell Liposarcoma

    • Myxoid/round-cell liposarcoma (MLS/RCLS) is characterized by either the fusion gene FUS-DDIT3 or the less commonly occurring EWSR1-DDIT3 and most cases carry few or no additional cytogenetic changes. There are conflicting reports concerning the status and role of TP53 in MLS/RCLS. Here we analysed four MLS/RCLS derived cell lines for TP53 mutations, expression and function. Three SV40 transformed cell lines expressed normal TP53 proteins. Irradiation caused normal posttranslational modifications of TP53 and induced P21 expression in two of these cell lines. Transfection experiments showed that the FUS-DDIT3 fusion protein had no effects on irradiation induced TP53 responses. Ion Torrent AmpliSeq screening, using the Cancer Hotspot panel, showed no dysfunctional or disease associated alleles/mutations. In conclusion, our results suggest that most MLS/RCLS cases carry functional TP53 genes and this is consistent with the low numbers of secondary mutations observed in this tumor entity.
