    • Two nanocomposites containing CdS nanoparticles in polymeric matrices were studied using the photoluminescence (PL) and optically detected magnetic resonance (ODMR) methods. Due to equal sizes of NPs in the composites (~5 nm) but different matrices – the oxygen-containing polymer PVA (polyvinyl alcohol) and oxygen-free polymer PEI (polyethyleneimine) – differences of nanocomposites properties are predominantly caused by different interfacial conditions. ODMR spectra have revealed five types of centers related to the PL emission – four centers involved in radiative recombination and one center related to non-radiative recombination processes. The oxygen-related interfacial center in CdS/PVA (LK1-center) and sulfur vacancy center in CdS/PEI (Vs-center) were identified.

    • Silver birch (Betula pendula) is a pioneer boreal tree that can be induced to flower within 1 year. Its rapid life cycle, small (440-Mb) genome, and advanced germplasm resources make birch an attractive model for forest biotechnology. We assembled and chromosomally anchored the nuclear genome of an inbred B. pendula individual. Gene duplicates from the paleohexaploid event were enriched for transcriptional regulation, whereas tandem duplicates were overrepresented by environmental responses. Population resequencing of 80 individuals showed effective population size crashes at major points of climatic upheaval. Selective sweeps were enriched among polyploid duplicates encoding key developmental and physiological triggering functions, suggesting that local adaptation has tuned the timing of and cross-talk between fundamental plant processes. Variation around the tightly-linked light response genes PHYC and FRS10 correlated with latitude and longitude and temperature, and with precipitation for PHYC. Similar associations characterized the growth-promoting cytokinin response regulator ARR1, and the wood development genes KAK and MED5A.

    • BackgroundSalivary cell secretion (SCS) plays a critical role in blood feeding by medicinal leeches, making them of use for certain medical purposes even today.ResultsWe annotated the Hirudo medicinalis genome and performed RNA-seq on salivary cells isolated from three closely related leech species, H. medicinalis, Hirudo orientalis, and Hirudo verbana. Differential expression analysis verified by proteomics identified salivary cell-specific gene expression, many of which encode previously unknown salivary components. However, the genes encoding known anticoagulants have been found to be expressed not only in salivary cells. The function-related analysis of the unique salivary cell genes enabled an update of the concept of interactions between salivary proteins and components of haemostasis.ConclusionsHere we report a genome draft of Hirudo medicinalis and describe identification of novel salivary proteins and new homologs of genes encoding known anticoagulants in transcriptomes of three medicinal leech species. Our data provide new insights in genetics of blood-feeding lifestyle in leeches.

  • A cerato-platanin-like protein HaCPL2 from Heterobasidion annosum sensu stricto induces cell death in Nicotiana tabacum and Pinus sylvestris

    • The cerato-platanin family is a group of small secreted cysteine-rich proteins exclusive for filamentous fungi. They have been shown to be involved in the interactions between fungi and plants. Functional characterization of members from this family has been performed mainly in Ascomycota, except Moniliophthora perniciosa. Our previous phylogenetic analysis revealed that recent gene duplication of cerato-platanins has occurred in Basidiomycota but not in Ascomycota, suggesting higher functional diversification of this protein family in Basidiomycota than in Ascomycota. In this study, we identified three cerato-platanin homologues from the basidiomycete conifer pathogen Heterobasidion annosum sensu stricto. Expression of the homologues under various conditions as well as their roles in the H. annosum s.s.-Pinus sylvestris (Scots pine) pathosystem was investigated. Results showed that HaCPL2 (ceratoplatanin-like protein 2) had the highest sequence similarity to cerato-platanin from Ceratocystis platani and hacpl2 was significantly induced during nutrient starvation and necrotrophic growth. The treatment with recombinant HaCPL2 induced cell death, phytoalexin production and defense gene expression in Nicotiana tabacum. Eliciting and cell death-inducing ability accompanied by retardation of apical root growth was also demonstrated in Scots pine seedlings. Our results suggest that HaCPL2 might contribute to the virulence of H. annosum s.s. by promoting plant cell death.

    • We report on the observation of the long-lasting low-temperature photoluminescence decay in the hybrid system nano-CdS/polyvinyl alcohol with a characteristic time of about 1.7 s. The origin of the phosphorescence is ascribed to the accumulation of photo-excited excitons in the traps within the polymeric matrix with subsequent transfer of the excitation to the embedded CdS nanoparticles.     • CdS nanoparticles have been synthesized in aqueous solution using polyvinyl alcohol (PVA) as a capping reagent. The effects of exposure by ultraviolet (UV) light on optical properties of nanocomposites consisting of colloidal CdS nanoparticles and a polymer PVA matrix were studied by employing photoluminescence (PL) spectroscopy. It is shown that UV-induced changes of the photoluminescence intensity in PVA are caused by creation and healing of non-radiative recombination centers. It is also concluded that in the nanocomposites, the UV-induced changes of the PL intensity are predominantly governed by processes at the NP/PVA interface.

  • Interfacial bonding in a CdS/PVA nanocomposite: A Raman scattering study

    • Raman spectroscopy is employed to characterize the bonding between CdS nanoparticles (NPs) and a polyvinyl alcohol (PVA) as well as structural changes in the polymeric matrix caused by incorporation of NPs. It is shown that after the formation of CdS NPs the vibrations of carbonyl groups in acetate residuals of PVA and of CO groups at the macromolecules ends disappear. Formation of NPs also leads to an increased degree of hydrogen bonding and crystallinity of the hybrid material as compared with the unloaded polymer. The observed changes are ascribed to the formation of coordinative bonds and hydrogen between the CdS nanoparticles and polymeric macromolecules. The scheme of this interfacial bonding is also proposed.

    • Participation of a polymeric media in light-emitting processes of composite nano-CdS/polyvinyl alcohol is studied by probing different absorption-emission routes via adjustment of excitation wavelengths. It is shown that the polymeric constituent of the composite contributes chiefly to the photoluminescence excitation processes via absorption and excitation transfer to the embedded CdS nanoparticles while the composite emission occurs mostly within the nanoparticles.
