    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

    • Burst super-resolution has received increased attention in recent years due to its applications in mobile photography. By merging information from multiple shifted images of a scene, burst super-resolution aims to recover details which otherwise cannot be obtained using a simple input image. This paper reviews the NTIRE 2022 challenge on burst super-resolution. In the challenge, the participants were tasked with generating a clean RGB image with 4x higher resolution, given a RAW noisy burst as input. That is, the methods need to perform joint denoising, demosaicking, and super-resolution. The challenge consisted of 2 tracks. Track 1 employed synthetic data, where pixel-accurate high-resolution ground truths are available. Track 2 on the other hand used real-world bursts captured from a handheld camera, along with approximately aligned reference images captured using a DSLR. 14 teams participated in the final testing phase. The top performing methods establish a new state-of-the-art on the burst super-resolution task.

  • Genome-wide association analyses in Han Chinese identify two new susceptibility loci for amyotrophic lateral sclerosis

    • To identify susceptibility genes for amyotrophic lateral sclerosis (ALS), we conducted a genome-wide association study (GWAS) in 506 individuals with sporadic ALS and 1,859 controls of Han Chinese ancestry. Ninety top SNPs suggested by the current GWAS and 6 SNPs identified by previous GWAS were analyzed in an independent cohort of 706 individuals with ALS and 1,777 controls of Han Chinese ancestry. We discovered two new susceptibility loci for ALS at 1q32 (CAMK1G, rs6703183, P-combined = 2.92 x 10(-8), odds ratio (OR) = 1.31) and 22p11 (CABIN1 and SUSD2, rs8141797, P-combined = 2.35 x 10(-9), OR = 1.52). These two loci explain 12.48% of the overall variance in disease risk in the Han Chinese population. We found no association evidence for the previously reported loci in the Han Chinese population, suggesting genetic heterogeneity of disease susceptibility for ALS between ancestry groups. Our study identifies two new susceptibility loci and suggests new pathogenic mechanisms of ALS.

  • Deep Learning-Based Classification of Cancer Cell in Leptomeningeal Metastasis on Cytomorphologic Features of Cerebrospinal Fluid

    • Background: It is a critical challenge to diagnose leptomeningeal metastasis (LM), given its technical difficulty and the lack of typical symptoms. The existing gold standard of diagnosing LM is to use positive cerebrospinal fluid (CSF) cytology, which consumes significantly more time to classify cells under a microscope.Objective: This study aims to establish a deep learning model to classify cancer cells in CSF, thus facilitating doctors to achieve an accurate and fast diagnosis of LM in an early stage.Method: The cerebrospinal fluid laboratory of Xijing Hospital provides 53,255 cells from 90 LM patients in the research. We used two deep convolutional neural networks (CNN) models to classify cells in the CSF. A five-way cell classification model (CNN1) consists of lymphocytes, monocytes, neutrophils, erythrocytes, and cancer cells. A four-way cancer cell classification model (CNN2) consists of lung cancer cells, gastric cancer cells, breast cancer cells, and pancreatic cancer cells. Here, the CNN models were constructed by Resnet-inception-V2. We evaluated the performance of the proposed models on two external datasets and compared them with the results from 42 doctors of various levels of experience in the human-machine tests. Furthermore, we develop a computer-aided diagnosis (CAD) software to generate cytology diagnosis reports in the research rapidly.Results: With respect to the validation set, the mean average precision (mAP) of CNN1 is over 95% and that of CNN2 is close to 80%. Hence, the proposed deep learning model effectively classifies cells in CSF to facilitate the screening of cancer cells. In the human-machine tests, the accuracy of CNN1 is similar to the results from experts, with higher accuracy than doctors in other levels. Moreover, the overall accuracy of CNN2 is 10% higher than that of experts, with a time consumption of only one-third of that consumed by an expert. Using the CAD software saves 90% working time of cytologists.Conclusion: A deep learning method has been developed to assist the LM diagnosis with high accuracy and low time consumption effectively. Thanks to labeled data and step-by-step training, our proposed method can successfully classify cancer cells in the CSF to assist LM diagnosis early. In addition, this unique research can predict cancer’s primary source of LM, which relies on cytomorphologic features without immunohistochemistry. Our results show that deep learning can be widely used in medical images to classify cerebrospinal fluid cells. For complex cancer classification tasks, the accuracy of the proposed method is significantly higher than that of specialist doctors, and its performance is better than that of junior doctors and interns. The application of CNNs and CAD software may ultimately aid in expediting the diagnosis and overcoming the shortage of experienced cytologists, thereby facilitating earlier treatment and improving the prognosis of LM.

  • Small interfering RNA-directed targeting of toll-like receptor 4 inhibits human prostate cancer cell invasion, survival, and tumorigenicity

    • A major cause of tumor treatment failure is cancer cell metastasis. Toll-like receptor 4 (TLR4)-mediated signaling has been implicated in tumor cell invasion, survival, and metastasis in a variety of cancers. In this study, we investigated the biological roles of TLR4 in prostate metastatic cell invasion and survival, and the potential of gene silencing of TLR4 using small interfering RNA (siRNA) for treatment of cancer. In cultured human prostate cancer cell lines, TLR4 were higher PC3 and DU145 as compared with the poorly metastatic LNCaP indicating that up-regulation of TLR4 was positively correlated with metastasis of tumor cell. In the highly metastatic cancer cell PC3, gene silencing of TLR4 using siRNA significantly inhibited TLR4 mRNA expression and protein level. Knockdown of TLR4 in PC3 cells resulted in a dramatic reduction of tumor cell migration and invasion as indicated by a Matrigel invasion assay. Furthermore, TLR4 siRNA suppressed cell viability and ultimately caused the induction of apoptotic cell death. The effects were associated with abrogating TLR4-mediated signaling to downstream target molecules such as myeloid differentiation factor 88 (MyD88), adaptor-inducing IFN-beta (TRIF), and interferon regulatory factor-1 (IRF-1). In a mouse prostate cancer model, administration with the plasmid construct expressing siRNA for TLR4 obviously inhibited established tumor growth and survival. These studies revealed evidence of a multifaceted signaling network operating downstream of TLR4-mediated tumor cell invasion, proliferation, and survival. Thus, RNA interference-directed targeting of TLR4 may raise the potential of its application for cancer therapy.

    • Dayside magnetosphere interactions are essential for energy and momentum transport between the solar wind and the magnetosphere. In this study, we investigate a new phenomenon within this regime. Sudden enhancements of ion fluxes followed by repeating dropouts and recoveries were observed by Magnetospheric Multiscale on 5 November 2016, which is the very end of the recovery phase from a moderate geomagnetic storm. These repetitive flux variations display energy-dispersive characteristics with periods relevant to ion bounce motion, suggesting they are corresponding echoes. Alongside the flux variations, bipolar electric field impulses originating from external sources were detected. We traced the source region of the initial injection and found it is located near the spacecraft's position. To elucidate the underlying physics, a test-particle simulation is conducted. The results reveal that radial transport resulting from impulse-induced acceleration can give rise to these echoes. Observations demonstrate dayside magnetosphere interactions are more common than we previously considered, which warrants further research.

  • Physical frailty, adherence to ideal cardiovascular health and risk of cardiovascular disease : a prospective cohort study

    • Background: longitudinal evidence concerning frailty phenotype and the risk of cardiovascular disease (CVD) remained insufficient, and whether CVD preventive strategies exert low CVD risk on frail adults is unclear.Objectives: we aimed to prospectively evaluate the association of frailty phenotype, adherence to ideal cardiovascular health (CVH) and their joint associations with the risk of CVD.Methods: a total of 314,093 participants from the UK Biobank were included. Frailty phenotype was assessed according to the five criteria of Fried et al.: weight loss, exhaustion, low physical activity, slow gait speed and low grip strength. CVH included four core health behaviours (smoking, physical activity and diet) and three health factors (weight, cholesterol, blood pressure and glycaemic control). The outcome of interest was incident CVD, including coronary heart disease, heart failure and stroke.Results: compared with the non-frail people whose incident rate of overall CVD was 6.54 per 1,000 person-years, the absolute rate difference per 1,000 person-years was 1.67 (95% confidence interval, CI: 1.33, 2.02) for pre-frail and 5.00 (95% CI: 4.03, 5.97) for frail. The ideal CVH was significantly associated with a lower risk of all CVD outcomes. For the joint association of frailty and CVH level with incident CVD, the highest risk was observed among frailty accompanied by poor CVH with an HR of 2.92 (95% CI: 2.68, 3.18).Conclusions: our findings indicate that physical frailty is associated with CVD incidence. Improving CVH was significantly associated with a considerable decrease in CVD risk, and such cardiovascular benefits remain for the frailty population.

  • Arctic sea ice loss warmed the temperate East Asian winter in the mid-Holocene

    • The recent colder winters in midlatitude Eurasia have been proposed to result from Arctic sea-ice decline. However, large uncertainties remain regarding this link in the present variable climate. Here, we present ice-rafted debris records from the eastern Arctic and geochemical data from the temperate East China Sea to reconstruct Holocene changes in sea ice and the East Asian winter monsoon. Our reconstructions and climate numerical simulations revealed enhanced Arctic sea-ice decline but warmer winters in East Asia in the mid-Holocene than in the late Holocene. In the warmer mid-Holocene, enhanced Arctic sea-ice loss transferred more heat from intensive summer solar insolation to the winter atmosphere, suppressing meridional heat transport; thus, less high-latitude cold air moved to lower latitudes in Asia due to the weakened winter monsoon. Our findings imply that the colder winters in East Asia may not change the long-term trend toward winter warming in the context of Arctic sea-ice decline.

    • In this paper, we present a receiver front-end for 5 GHz wireless LAN in novel LCP (liquid crystal polymer) based system-on-package module. The module is based on embedded chip technologies for system-on-package, which eliminates the constraints of off-chip pad drive capability and hence improves electrical performance. Furthermore, the novel LCP material shows excellent RF and microwave performance. The quality factors of key passive components such as inductors integrated in LCP substrate with thin film technologies is as high as 60. The insertion loss of the bandpass filter is 3dB. The conversion gain of the receiver front-end is 20 dB and occupies 8.7mm by 3.6mm area.

    • BackgroundResolving the phylogeny of rapidly radiating lineages presents a challenge when building the Tree of Life. An Old World avian family Prunellidae (Accentors) comprises twelve species that rapidly diversified at the Pliocene-Pleistocene boundary.ResultsHere we investigate the phylogenetic relationships of all species of Prunellidae using a chromosome-level de novo assembly of Prunella strophiata and 36 high-coverage resequenced genomes. We use homologous alignments of thousands of exonic and intronic loci to build the coalescent and concatenated phylogenies and recover four different species trees. Topology tests show a large degree of gene tree-species tree discordance but only 40-54% of intronic gene trees and 36-75% of exonic genic trees can be explained by incomplete lineage sorting and gene tree estimation errors. Estimated branch lengths for three successive internal branches in the inferred species trees suggest the existence of an empirical anomaly zone. The most common topology recovered for species in this anomaly zone was not similar to any coalescent or concatenated inference phylogenies, suggesting presence of anomalous gene trees. However, this interpretation is complicated by the presence of gene flow because extensive introgression was detected among these species. When exploring tree topology distributions, introgression, and regional variation in recombination rate, we find that many autosomal regions contain signatures of introgression and thus may mislead phylogenetic inference. Conversely, the phylogenetic signal is concentrated to regions with low-recombination rate, such as the Z chromosome, which are also more resistant to interspecific introgression.ConclusionsCollectively, our results suggest that phylogenomic inference should consider the underlying genomic architecture to maximize the consistency of phylogenomic signal.

  • Comparative Study of Dayside Pulsating Auroras Induced by Ultralow-Frequency Waves

    • Pulsating auroras are usually observed with ultralow-frequency (ULF) waves in the Pc 3-5 band (period 10-600 s). These auroras are thought to result from interactions between energetic electrons and chorus waves, but their relationship with ULF waves remains an open question. In this study, we investigated this question by conducting a comparative study on two ULF wave events with pulsating auroras observed near the magnetic footprints. Conjugate observations from the Magnetospheric Multiscale mission and the Chinese Yellow River Station were used. In both events, lower-band chorus waves were observed, which were suggested to be connected with the auroral pulsations by wavelet analysis. The intensity of these waves oscillates at the period of the ULF waves, but the physics laid behind them differs by events. During the event of 22 January 2019, compressional ULF waves changed the threshold for the whistler anisotropy instability periodically, affecting the emission of chorus waves. In the event on 10 January 2016, poloidal ULF waves modulated the chorus wave generation by regulating electron temperature anisotropy through drift resonance. ULF waves in these events may originate from perturbations in the solar wind. We highlight the role of ULF waves in the solar wind-magnetosphere-ionosphere coupling, which requires further study.

    • Drift-bounce resonance between ultralow frequency (ULF) waves and ions is essential for ion energization in the magnetosphere. Here, we present the first comprehensive study of drift-bounce resonance in the dayside outer magnetosphere, where off-equatorial magnetic field minima would strongly distort ions' bounce and drift motion. A generalized theory is proposed, in which the effects of off-equatorial minima, time-evolving fields and ion bounce motion are taken into account. In consequence of these effects, ion pitch angle distributions undergo dramatic changes. In the presence of off-equatorial minima, the time-of-flight effect of ion bounce motion forms latitude-dependent dispersions besides "paw-track shaped" structures, while evolving wave fields cause time-dependent phase shifts in "paw-tracks." All the predicted signatures have been confirmed by 5 years of Magnetospheric Multiscale spacecraft data and numerical simulations. These results allow us to better understand the interactions between ULF waves and thermal ion species in global magnetospheric dynamics.

  • EDF-VD scheduling of mixed-criticality systems with degraded quality guarantees

    • This paper studies real-time scheduling of mixed-criticality systems where low-criticality tasks are still guaranteed some service in the high-criticality mode, with reduced execution budgets. First, we present a utilization-based schedulability test for such systems under EDF-VD scheduling. Second, we quantify the suboptimality of EDF-VD (with our test condition) in terms of speedup factors. In general, the speedup factor is a function with respect to the ratio between the amount of resource required by different types of tasks in different criticality modes, and reaches 4/3 in the worst case. Furthermore, we show that the proposed utilization-based schedulability test and speedup factor results apply to the elastic mixed-criticality model as well. Experiments show effectiveness of our proposed method and confirm the theoretical suboptimality results.

    • Isoflavonoids comprise a class of plant natural products with great nutraceutical, pharmaceutical and agricultural significance. Their low abundance in nature and structural complexity however hampers access to these phytochemicals through traditional crop-based manufacturing or chemical synthesis. Microbial bioproduction therefore represents an attractive alternative. Here, we engineer the metabolism of Saccharomyces cerevisiae to become a platform for efficient production of daidzein, a core chemical scaffold for isoflavonoid biosynthesis, and demonstrate its application towards producing bioactive glucosides from glucose, following the screening-reconstruction-application engineering framework. First, we rebuild daidzein biosynthesis in yeast and its production is then improved by 94-fold through screening biosynthetic enzymes, identifying rate-limiting steps, implementing dynamic control, engineering substrate trafficking and fine-tuning competing metabolic processes. The optimized strain produces up to 85.4 mg L−1 of daidzein and introducing plant glycosyltransferases in this strain results in production of bioactive puerarin (72.8 mg L−1) and daidzin (73.2 mg L−1). Our work provides a promising step towards developing synthetic yeast cell factories for de novo biosynthesis of value-added isoflavonoids and the multi-phased framework may be extended to engineer pathways of complex natural products in other microbial hosts.

    • A SUMMARY There is an urgent demand for all-polymer organic solar cells (AP-OSCs) to gain higher efficiency. Here, we successfully improve the performance to 16.09% by introducing a small amount of BN-T, a B <- N-type polymer acceptor, into the PM6:PY-IT blend. It has been found that BN-T makes the active layer, based on the PM6:PY-IT:BN-T ternary blend, more crystalline but meanwhile slightly reduces the phase separation, leading to enhancement of both exciton harvesting and charge transport. From a thermodynamic viewpoint, BN-T prefers to reside between PM6 and PY-IT, and the fraction of this fine-tunes the morphology. Besides, a significantly reduced nonradiative energy loss occurs in the ternary blend, along with the coexistence of energy and charge transfer between the two acceptors. The progressive performance facilitated by these improved properties demonstrates that AP-OSCs can possibly comparably efficient with those based on small molecule acceptors, further enhancing the competitiveness of this device type.

    • Background: Tendons are a crucial component of the musculoskeletal system and responsible for transmission forces derived from muscle to bone. Patients with tendon injuries are often observed with decreased collagen production and matrix degeneration, and healing of tendon injuries remains a challenge as a result of limited understanding of tendon biology. Recent studies highlight the contribution of miR-378a on the regulation gene expression during tendon differentiation. Methods: We examined the tendon microstructure and tendon repair with using miR-378a knock-in transgenic mice, and the tendon-derived stem cells were also isolated from transgenic mice to study their tenogenic differentiation ability. Meanwhile, the expression levels of tenogenic markers were also examined in mouse tendon-derived stem cells transfected with miR-378a mimics during tenogenic differentiation. With using online prediction software and luciferase reporter assay, the binding target of miR-378a was also studied. Results: Our results indicated miR-378a impairs tenogenic differentiation and tendon repair by inhibition collagen and extracellular matrix production both in vitro and in vivo. We also demonstrated that miR-378a exert its inhibitory role during tenogenic differentiation through binding at TGFβ2 by luciferase reporter assay and western blot. Conclusions: Our investigation suggests that miR-378a could be considered as a new potential biomarker for tendon injury diagnosis or drug target for a possible therapeutic approach in future clinical practice.

    • The safety of lithium-ion batteries (LIBs) is paramount for all users. One effective way to improve safety is incorporating heat-resistant polyimide (PI) separators, which can increase the thermal stability of batteries and minimize the risk of thermal runaway. However, preparing PI separators with both an ideal pore structure and adequate mechanical properties remains as a challenge. Here, we introduced decabromodiphenyl ethane (DBDPE) and cellulose nanofibers (CNFs) into PI and produced a hybrid separator with an outstanding pore structure and excellent mechanical properties. Aided with DBDPE, the separators attain a well-defined and uniform pore size (20 nm), while demonstrating high porosities (78%) through phase inversion processes. Owing to the addition of CNFs, the mechanical properties of the separators were significantly improved, with a tensile strength of 25.4 MPa and an elastic modulus of 550.1 MPa. Moreover, the separators demonstrate high ion conductivity (0.45 mS cm-1), excellent thermal-dimensional stability (up to 200 degrees C), remarkable flame retardancy, and outstanding electrolyte wettability. At room temperature, the batteries with the separators demonstrate comparable performance with those of polypropylene (PP) separators. However, when subjected to thermal shock treatments, the batteries with the separators outperform those with PP, showcasing their superior performance. The work introduces a novel strategy for designing high-performance separators, thereby paving the way for advancements in the fabrication of LIBs with enhanced safety features. A porous, robust, and thermally stable hybrid separator was developed to solve the dilemma between desired pore structures and mechanical properties in polyimide separators by introducing decabromodiphenyl ethane and cellulose nanofibers.

    • Spinterface engineering has shown quite important roles in organic spintronics as it can improve spin injection or extraction. In this study, 11,11,12,12-tetracyanonaptho-2,6-quinodimethane (TNAP) is introduced as an interfacial layer for a prototype interface of Fe/TNAP. An element-specific investigation of the electronic and magnetic structures of Fe/TNAP system by use of near edge X-Ray absorption fine structure (NEXAFS) and X-ray magnetic circular dichroism (XMCD) is reported. Strong hybridization between TNAP and Fe and induced magnetization of N atoms in TNAP molecule are observed. XMCD sum rule analysis demonstrates that the adsorption of TNAP reduces the spin moment of Fe by 12%. In addition, induced magnetization in N K-edge of TNAP is also found with other commonly used ferromagnets in organic spintronics, such as La0.7Sr0.3MnO3 and permalloy, which makes TNAP a very promising molecule for spinterface engineering in organic spintronics.

  • Reducing supply risk of critical materials for clean energy via foreign direct investment

    • Existing research on the security of the supply of critical materials for clean energy generally aggregates information at the country level, a practice that obscures the extensive role of foreign direct investment (FDI) in the production of critical materials. FDI refers to an ownership stake in a company or project by an overseas investor. Here we establish a database for global mining of lithium, cobalt, nickel and platinum at company level, covering 240 countries and regions. We show that 47% of lithium, 71% of cobalt, 41% of nickel and 34% of platinum mined in 2019 were under FDI. We then explore how FDI may affect supply risks by proposing a supply risk index that allocates production of the critical materials to the country of origin of investors instead of the country where production is located. We present upper and lower bounds of the supply risk index that reflect scenarios where either all investors or only state investors prioritize the home-country demand, respectively. This study presents an approach for assessing the national supply risks of critical materials, considering the geographical allocation of FDI.

    • The high-mountain Third Pole (TP) in Asia is undergoing rapid warming, profoundly impacting river discharge. Changes in precipitation and the degradation of glaciers and permafrost exert a substantial impact on TP rivers, affecting millions downstream. Nevertheless, conventional estimation methods that rely on in -situ observations and models face considerable challenges due to inconsistent data quality and availability, complex terrain, and difficulty in representing hydrological processes. Here, we harnessed the power of high-resolution (3 m) Dove/ PlanetScope satellite images to meticulously measure river width and propose a novel formula for estimating discharge. This approach facilitates a coherent assessment of discharge at various river section shapes. Its efficacy is demonstrated through applying to thirteen high-mountain TP rivers, revealing an estimated total mountain discharge of 3786 billion m 3 during 2017 - 2021, equaling water from approximately 167 Great Lakes. These findings have immediate implications for integrated water resources management at the poorly-gauged transboundary basins of Asia.

    • Food losses and food waste (FLW) have become a global concern in recent years and emerge as a priority in the global and national political agenda (e.g., with Target 12.3 in the new United Nations Sustainable Development Goals). A good understanding of the availability and quality of global FLW data is a prerequisite for tracking progress on reduction targets, analyzing environmental impacts, and exploring mitigation strategies for FLW. There has been a growing body of literature on FLW quantification in the past years; however, significant challenges remain, such as data inconsistency and a narrow temporal, geographical, and food supply chain coverage. In this paper, we examined 202 publications which reported FLW data for 84 countries and 52 individual years from 1933 to 2014. We found that most existing publications are conducted for a few industrialized countries (e.g., the United Kingdom and the United States), and over half of them are based only on secondary data, which signals high uncertainties in the existing global FLW database. Despite these uncertainties, existing data indicate that per-capita food waste in the household increases with an increase of per-capita GDP. We believe that more consistent, in-depth, and primary-data-based studies, especially for emerging economies, are badly needed to better inform relevant policy on FLW reduction and environmental impacts mitigation.

    • Conjugated polymers are regarded as promising candidates for dopant-free hole-transport materials (HTMs) in efficient and stable perovskite solar cells (PSCs). Thus far, the vast majority of polymeric HTMs feature structurally complicated benzo[1,2-b:4,5-b']dithiophene (BDT) analogs and electron-withdrawing heterocycles, forming a strong donor-acceptor (D-A) structure. Herein, a new class of phenanthrocarbazole (PC)-based polymeric HTMs (PC1, PC2, and PC3) has been synthesized by inserting a PC unit into a polymeric thiophene or selenophene chain with the aim of enhancing the pi-pi stacking of adjacent polymer chains and also to efficiently interact with the perovskite surface through the broad and planar conjugated backbone of the PC. Suitable energy levels, excellent thermostability, and humidity resistivity together with remarkable photoelectric properties are obtained via meticulously tuning the conformation and elemental composition of the polymers. As a result, PSCs containing PC3 as dopant-free HTM show a stabilized power conversion efficiency (PCE) of 20.8% and significantly enhanced longevity, rendering one of the best types of PSCs based on dopant-free HTMs. Subsequent experimental and theoretical studies reveal that the planar conformation of the polymers contributes to an ordered and face-on stacking of the polymer chains. Furthermore, introduction of the "Lewis soft" selenium atom can passivate surface trap sites of perovskite films by Pb-Se interaction and facilitate the interfacial charge separation significantly. This work reveals the guiding principles for rational design of dopant-free polymeric HTMs and also inspires rational exploration of small molecular HTMs.

    • In the present study, porous silicon carbide ceramics were prepared via spark plasma sintering at relatively low temperatures using Al2O3 and CeO2 as sintering additives. Sacrificial template was selected as the pore forming mechanism, and gelcasting was used to fix the slurry in a short time. The evolution process of the microstructures during different steps was observed by SEM. The influence of the sintering temperature and sintering additives on the shrinkage and porosity of the samples was studied. The microstructures of different samples were characterized, and the mechanical properties were also evaluated.

    • Porous silicon carbide ceramics with high open porosity and large pore size are usually applied as filters for the cleaning of hot dust gases. However, the large porosity and large pore size will decrease the mechanical properties. In the present study, porous SiC ceramics were prepared using bentonite as bonding phase. The effects of sintering temperature on the microstructure and the compressive strength were studied. Bentonite could melt and spread on the surface of silicon carbide particles at all the sintering temperatures. However, the bonding effects were very different at different temperatures.

    • The properties of proton (0 eV < E < 40 keV) in the plasma sheet are examined by means of a superposed epoch analysis, using 115 magnetotail plasma injection events which are identified from Cluster magnetotail orbit time in between 2001 and 2004. All events distribute in magnetic local time from 20 p.m. to 04 a. in. Five classes of magnetotail injection events are found to be similar with the geosynchronous observation: (1) pure ion injections; (2) ion injections followed a few minutes later by an electron injection; (3) simultaneous ion and electron injections; (4) electron injections followed a few minutes later by an ion injection; (5) pure electron injections. Proton shows a significant increase in temperature and density at the onset, and injects earthward with an increasing velocity more than the pre-injection average one. Super-posed epoch analysis on the simultaneous observation data of dusk-dawn electric field from the EFW (Electric Field and Waves) instrument, we found two different electric field configurations: ( I) electric field increases suddenly at the onset and the value is positive; (2) electric field changes the direction at the onset, and turns into a negative value. The simulation results of velocity vector after injection in equatorial plain, calculated in static magnetic (T89c) and electric (Volland-Stern) field models, agree with the statistical results mostly, and that suggests the electric drift caused by dawn-dusk convection electric field is one of the mechanisms of the particles injected earthward in magnetotail(-18 R-E < R < -10R(E)).

  • YOLOv8-QR : An improved YOLOv8 model via attention mechanism for object detection of QR code defects

    • Defect detection in Quick Response (QR) codes has important implications for downstream tasks. However, QR code defects include small objects and complex backgrounds, which makes their recognition effect poor. To address the above problems, we proposed a model based on YOLOv8, called YOLOv8-QR, to detect QR code defects. Specifically, first, the non-local attention (Non-local) module is introduced into the backbone of the YOLOv8 to enhance the interactive ability of the feature. The Non-local captures the correlation information of long-distance dependencies between features by calculating the attention weight between any positions. In addition, the contextual information required for representation learning of different defective objects is different. To extract multi-scale features, a Large Selective Kernel Network (LSKNet) was introduced. LSKNet dynamically adjusts the convolution receptive field of the neck fusion network and effectively uses the receptive field to capture the background information of different objects, thereby improving the representation ability of the model. To improve the defect detection accuracy of small objects in QR codes, the Normalized Gaussian Wasserstein Distance (NGWD) is introduced to replace the Intersection over Union (IoU) optimization function that is sensitive to the position deviation of objects and is not conducive to the regression of multi-scale objects. To verify the effectiveness of the model, the QR dataset was constructed and a series of experiments were conducted based on this dataset. The results show that the mAP50 and mAP50:95 of the YOLOv8-QR reach 95.5% and 65%, which are 3.8% and 2.3% higher than YOLOv8 respectively. The proposed YOLOv8-QR can better adapt to the needs of QR code defect detection in actual industrial environments. Our code is available at https://github.com/Code-of-Liujie/YOLOv8-QR.git. © 2024

  • Seasonal migration patterns of Siberian Rubythroat (Calliope calliope) facing the Qinghai-Tibet Plateau

    • Background Small songbirds respond and adapt to various geographical barriers during their annual migration. Global flyways reveal the diverse migration strategies in response to different geographical barriers, among which are high-elevation plateaus. However, few studies have been focused on the largest and highest plateau in the world, the Qinghai-Tibet Plateau (QTP) which poses a significant barrier to migratory passerines. The present study explored the annual migration routes and strategies of a population of Siberian Rubythroats (Calliope calliope) that breed on the north-eastern edge of the QTP. Methods Over the period from 2021 to 2023, we applied light-level geolocators (13 deployed, seven recollected), archival GPS tags (45 deployed, 17 recollected), and CAnMove multi-sensor loggers (with barometer, accelerometer, thermometer, and light sensor, 20 deployed, six recollected) to adult males from the breeding population of Siberian Rubythroat on the QTP. Here we describe the migratory routes and phenology extracted or inferred from the GPS and multi-sensor logger data, and used a combination of accelerometric and barometric data to describe the elevational migration pattern, flight altitude, and flight duration. All light-level geolocators failed to collect suitable data. Results Both GPS locations and positions derived from pressure-based inference revealed that during autumn, the migration route detoured from the bee-line between breeding and wintering grounds, leading to a gradual elevational decrease. The spring route was more direct, with more flights over mountainous areas in western China. This different migration route during spring probably reflects a strategy for faster migration, which corresponds with more frequent long nocturnal migration flights and shorter stopovers during spring migration than in autumn. The average flight altitude (1856 +/- 781 m above sea level) was correlated with ground elevation but did not differ between the seasons. Conclusions Our finding indicates strong, season-dependent impact of the Qinghai-Tibet Plateau on shaping passerine migration strategies. We hereby call for more attention to the unexplored central-China flyway to extend our knowledge on the environment-migration interaction among small passerines.

    • ALICE (A Large Ion Collider Experiment) is the LHC (Large Hadron Collider) experiment devoted to investigating the strongly interacting matter created in nucleus-nucleus collisions at the LHC energies. The ALICE ITS, Inner Tracking System, consists of six cylindrical layers of silicon detectors with three different technologies; in the outward direction: two layers of pixel detectors, two layers each of drift, and strip detectors. The number of parameters to be determined in the spatial alignment of the 2198 sensor modules of the ITS is about 13,000. The target alignment precision is well below 10 mu m in some cases (pixels). The sources of alignment information include survey measurements, and the reconstructed tracks from cosmic rays and from proton-proton collisions. The main track-based alignment method uses the Millepede global approach. An iterative local method was developed and used as well. We present the results obtained for the ITS alignment using about 10(5) charged tracks from cosmic rays that have been collected during summer 2008, with the ALICE solenoidal magnet switched off.

    • Background: Severe neutrophilic asthma is resistant to treatment with glucocorticoids. The immunomodulatory protein macrophage migration inhibitory factor (MIF) promotes neutrophil recruitment to the lung and antagonises responses to glucocorticoids. We hypothesised that MIF promotes glucocorticoid resistance of neutrophilic inflammation in severe asthma.Methods: We examined whether sputum MIF protein correlated with clinical and molecular characteristics of severe neutrophilic asthma in the Unbiased Biomarkers for the Prediction of Respiratory Disease Outcomes (U-BIOPRED) cohort. We also investigated whether MIF regulates neutrophilic inflammation and glucocorticoid responsiveness in a murine model of severe asthma in vivo.Results: MIF protein levels positively correlated with the number of exacerbations in the previous year, sputum neutrophils and oral corticosteroid use across all U-BIOPRED subjects. Further analysis of MIF protein expression according to U-BIOPRED-defined transcriptomic-associated clusters (TACs) revealed increased MIF protein and a corresponding decrease in annexin-A1 protein in TAC2, which is most closely associated with airway neutrophilia and NLRP3 inflammasome activation. In a murine model of severe asthma, treatment with the MIF antagonist ISO-1 significantly inhibited neutrophilic inflammation and increased glucocorticoid responsiveness. Coimmunoprecipitation studies using lung tissue lysates demonstrated that MIF directly interacts with and cleaves annexin-A1, potentially reducing its biological activity.Conclusion: Our data suggest that MIF promotes glucocorticoid-resistance of neutrophilic inflammation by reducing the biological activity of annexin-A1, a potent glucocorticoid-regulated protein that inhibits neutrophil accumulation at sites of inflammation. This represents a previously unrecognised role for MIF in the regulation of inflammation and points to MIF as a potential therapeutic target for the management of severe neutrophilic asthma.

    • Asthma is a chronic inflammatory disease primarily characterized by inflammation and reversible bronchoconstriction. It is currently one of the leading causes of morbidity and mortality in the world. Oxidative stress further complicates the pathology of the disease. The current treatment strategies for asthma mainly involve the use of anti-inflammatory agents and bronchodilators. However, long-term usage of such medications is associated with severe adverse effects and complications. Hence, there is an urgent need to develop newer, novel, and safe treatment modalities for the management of asthma. This has therefore prompted further investigations and detailed research to identify and develop novel therapeutic interventions from potent untapped resources. This review focuses on the significance of oxidative stressors that are primarily derived from both mitochondrial and non-mitochondrial sources in initiating the clinical features of asthma. The review also discusses the biological scavenging system of the body and factors that may lead to its malfunction which could result in altered states. Furthermore, the review provides a detailed insight into the therapeutic role of nutraceuticals as an effective strategy to attenuate the deleterious effects of oxidative stress and may be used in the mitigation of the cardinal features of bronchial asthma.

    • A recent trend of sea level shows constant rising. Sea level rise has caused asignificant risk to seaside areas. This study examines the potential effect ofclimate change and rising sea levels on coastal regions and evaluates the susceptibilityof coastal areas in Inverloch, Melbourne Australia. A model ofHypothetically Flooded Zones, based on LiDAR data was built, processed andmanipulated in ArcGIS. Through applying this model, the effect of rising sealevel on the infrastructures such as buildings, dwellings, roads, land use andthe population was assessed. Elevation data sets of varying resolution and accuracyhave been processed to show the improved quality of LiDAR datacontributes to a more precise delineation of flood-prone coastal lands. Afterthe susceptible areas to sea level rise were delineated, the worst-case scenariowas calculated (based on the increase in sea level projected for 2100) and itwould impacts about 0.86% of roads, 221 of different building infrastructures.This method can be used in other areas to protect the coasts due to rapidchanges caused by climate change.

    • Climate change can impact coastal areas in different ways, including flooding, storm surges, and beach erosion. Of these, flooding has a major impact on the operation of coastal drainage systems. This paper develops a new flood screening model using a LiDAR based digital elevation model (DEM) to improve the implementation of Victorian’s coastal flooding risk assessment and management. Hydrological elevation models are directed towards protection from cloudbursts and applied to rising sea level. The aim is to simulate water flow on the ground and in streams, and the resulting accumulation of water in depressions of the blue spot using DEM. Due to the presence of pipes, watercourses, bridges and channels it was required that the DEM data to be lowered. The reservoirs of rain will prevent seawater from flowing across the stream channel into land. The rain drain will be open during normal sea levels to allow rain water in the river to move and flow in to the sea. Traditionally, geographic information system (GIS) assists with spatial data management, but lacks modelling capability for complex hydrology problems and cannot be relied upon by decision-makers in this sector. Functionality improvements are therefore required to improve the processing or analytical capabilities of GIS in hydrology. This research shows how the spatial data can be primarily processed by GIS adopting the spatial analysis routines associated with hydrology. The objective of this paper is to outline the importance of GIS technology for coastal flood management. Following a definition of the coastal flood, and, short description of its peculiarities and the urgency of its management, this paper describes the use of GIS technology in coastal flood management, its advantages and the consideration for accuracy. This is followed by the information and LiDAR data required for coastal flood management and the application area in coastal flood management. This paper method is presented to conduct a first high-resolution DEM screening to detect the degree and capacities of the sinks in the coastal landscape. When their capacities are established, the rain volumes received during a rainstorm from their coastal catchments are saved as attributes to the pour points. The conclusion emphases the importance of a geographical information system in coastal flood management for efficient data handling and analysis of geographically related data. Local governments at risk of coastal flooding that use the flood screening model can use to determine appropriate land use controls to manage long-term flood risk to human settlements.

    • Soft materials are usually defined as materials made of mesoscopic entities, often self-organised, sensitive to thermal fluctuations and to weak perturbations. Archetypal examples are colloids, polymers, amphiphiles, liquid crystals, foams. The importance of soft materials in everyday commodity products, as well as in technological applications, is enormous, and controlling or improving their properties is the focus of many efforts. From a fundamental perspective, the possibility of manipulating soft material properties, by tuning interactions between constituents and by applying external perturbations, gives rise to an almost unlimited variety in physical properties. Together with the relative ease to observe and characterise them, this renders soft matter systems powerful model systems to investigate statistical physics phenomena, many of them relevant as well to hard condensed matter systems. Understanding the emerging properties from mesoscale constituents still poses enormous challenges, which have stimulated a wealth of new experimental approaches, including the synthesis of new systems with, e.g. tailored self-assembling properties, or novel experimental techniques in imaging, scattering or rheology. Theoretical and numerical methods, and coarse-grained models, have become central to predict physical properties of soft materials, while computational approaches that also use machine learning tools are playing a progressively major role in many investigations. This Roadmap intends to give a broad overview of recent and possible future activities in the field of soft materials, with experts covering various developments and challenges in material synthesis and characterisation, instrumental, simulation and theoretical methods as well as general concepts.

    • With an ever-increasing deployment density of probe and fixed sensors, massive vehicular trajectory data is available and show a promising foundation to improve the observability of dynamic traffic demand pattern. However, due to technical and privacy issues, the raw trajectories are not always complete and the paths and destinations between discontinuous trajectory nodes are usually missing. This paper proposes a probabilistic method to jointly reconstruct the missing driving path and destination location of vehicles with incomplete trajectory data. One problem-specific HMM-structured model incorporating spatial and temporal analysis (ST-HMM) is constructed to define the matching probability between observed data and possible movement. Two algorithms, namely candidate set generation and best-match search algorithms, are developed to seek the most possible one as matching result. It can implement end-to-end processing from incomplete trajectory data to complete and connective paths and destinations for the target vehicle. The proposed method is tested based on field-test data and city-wide road network. Compared with two benchmark methods, the proposed method improved the matching accuracy in terms of both path identification and destination inference. Additionally, sensitivity analyses on the size of training dataset and candidate set were performed. We believe that experiment results of these sensitivity analyses can help to provide guidance on data sensing and candidate generation.

    • Starting from two simple units of anthracene and 2,2'-dipyridylamine, a series of new luminescent compounds 1-8 were designed and synthesized by a combined strategy of changing the connection mode between the two units, extending the conjugation size, and introducing an additional electron donor. Photophysical properties of 1-8 were investigated and discussed on the basis of solvatochromic behaviour, theoretical calculations, crystal structure, and optimized structures. Interestingly, the emission wavelengths of these compounds could be successfully tuned from violet to red both in solutions and the solid-state, and prominent positive solvatochromism was observed for the compounds with a D-p-A framework. Consider compound 7 as an example, it shows peaks at 526 nm and 627 nm in cyclohexane and DMSO, respectively. Meanwhile, the quantum yield was decreased from 0.80 in cyclohexane to 0.12 in DMSO. The introduction of bulky groups was demonstrated to be effective for suppressing the aggregation effect and thus improving the solid state emission quantum yield. These results indicate that the combined structure modulation strategy offers a powerful tool for tuning the emission behaviour. To demonstrate the possibility of practical applications, 2 was employed as the emitting material for the fabrication of deep-blue organic light-emitting diodes (OLEDs), which showed a maximum external quantum efficiency of 2.2%. The CIE coordinates of (0.15, 0.08) are indicative of excellent blue color purity.

    • In this note, we investigate relations between tree-level off-shell currents in nonlinear sigma model. Under Cayley parametrization, all odd-point currents vanish. We propose and prove a generalized U(1) identity for even-point currents. The off-shell U(1) identity given in [1] is a special case of the generalized identity studied in this note. The on-shell limit of this identity is equivalent with the on-shell KK relation. Thus this relation provides the full off-shell correspondence of tree-level KK relation in nonlinear sigma model.

    • Mixed-criticality models are an emerging paradigm for the design of real-time systems because of their significantly improved resource efficiency. However, formal mixed-criticality models have traditionally been characterized by two impractical assumptions: once any high-criticality task overruns, all low-criticality tasks are suspended and all other high-criticality tasks are assumed to exhibit high-criticality behaviors at the same time. In this paper, we propose a more realistic mixed-criticality model, called the flexible mixed-criticality (FMC) model, in which these two issues are addressed in a combined manner. In this new model, only the overrun task itself is assumed to exhibit high-criticality behavior, while other high-criticality tasks remain in the same mode as before. The guaranteed service levels of low-criticality tasks are gracefully degraded with the overruns of high-criticality tasks. We derive a utilization-based technique to analyze the schedulability of this new mixed-criticality model under EDF-VD scheduling. During run time, the proposed test condition serves an important criterion for dynamic service level tuning, by means of which the maximum available execution budget for low-criticality tasks can be directly determined with minimal overhead while guaranteeing mixed-criticality schedulability. Experiments demonstrate the effectiveness of the FMC scheme compared with state-of-the-art techniques.

    • 30-m Global Land Cover (GLC) data products permit the detection of land cover changes at the scale of most human land activities, and are therefore used as fundamental information for sustainable development, environmental change studies, and many other societal benefit areas. In the past few years, increasing efforts have been devoted to the accuracy assessment of GlobeLand30 and other finer-resolution GLC data products. However, most of them were conducted either within a limited percentage of map sheets selected from a global scale or in some individual countries (areas), and there are still many areas where the uncertainty of 30-m resolution GLC data products remains to be validated and documented. In order to promote a comprehensive and collaborative validation of 30-m GLC data products, the GEO Global Land Cover Community Activity had organized a project from 2015 to 2017, to examine and explore its major problems, including the lack of international agreed validation guidelines and on-line tools for facilitating collaborative validation activities. With the joint effort of experts and users from 30 GEO member countries or participating organizations, a technical specification for 30-m GLC validation was developed based on the findings and experiences. An on-line validation tool, GLCVal, was developed by integrating land cover validation procedures with the service computing technologies. About 20 countries (regions) have completed the accuracy assessment of GlobeLand30 for their territories with the guidance of the technical specification and the support of GLCVal.

    • This paper investigates the power allocation problems for non-orthogonal multiple access with coordinated direct and relay transmission (CDRT-NOMA), where a base station (BS) communicates with its nearby user directly, while communicating with its far user only through a dedicated relay node (RN). The RN is assumed to operate in either half-duplex relaying (HDR) mode or full-duplex relaying (FDR) mode. Based on instantaneous channel state information (CSI), the dynamic power allocation problems under HDR and FDR schemes are formulated respectively, with the objective of maximizing the minimum user achievable rate. After demonstrating the quasi-concavity of the considered problems, we derive the optimal closed-form power allocation policies under the HDR scheme and the FDR scheme. Then, a hybrid relaying scheme dynamically switching between HDR and FDR schemes is further designed. Moreover, we also study the fixed power allocation problems for the considered CDRT-NOMA systems based on statistical CSI so as to optimize the long-term system performance. The simulations show that our proposed power allocation policies can significantly enhance the performance of CDRT-NOMA systems.

    • NOMA is a promising radio access technology for 5G wireless systems. The core of NOMA is to support multiple users in the same resource block via power or code domain multiplexing, which provides great enhancement in spectrum efficiency and connectivity. Meanwhile, with the recent advance in self-interference (SI) cancelation techniques, FD wireless communication has become a feasible technology enabling radios to receive and transmit simultaneously. This article aims to investigate the combination of these two emerging technologies. At first, several typical scenarios and protocols are presented to illustrate the application of the FD technique in NOMA systems. Then, a novel NOMA system with FD base stations (BSs) based on C-RAN is proposed. Furthermore, power allocation policies are discussed for the proposed scheme, and simulation results are provided to demonstrate its superiority. Finally, challenges and research opportunities of FD NOMA systems are also identified to stimulate future research.
