    • The cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental studies. Over the past decades, rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and habitats. Yet, in spite of the progress of molecular methods, knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains challenging. In order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database FungalTraits operating at genus and species hypothesis levels. Combining the information from previous efforts such as FUNGuild and Fun(Fun) together with involvement of expert knowledge, we reannotated 10,210 and 151 fungal and Stramenopila genera, respectively. This resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera, designed for rapid functional assignments of environmental studies. In order to assign the trait states to fungal species hypotheses, the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ITS sequences. On the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1% dissimilarity threshold.

    • Tumor cells use membrane type 1 matrix metalloproteinase (MT1-MMP) for invasion and metastasis. However, the signaling mechanisms that underlie MT1-MMP regulation in cancer have remained unclear. Using a systematic gain-of-function kinome screen for MT1-MMP activity, we have here identified kinases that significantly enhance MT1-MMP activity in tumor cells. In particular, we discovered an MT1-MMP/FGF receptor-4 (FGFR4) membrane complex that either stimulates or suppresses MT1-MMP and FGFR4 activities, depending on a tumor progression-associated polymorphism in FGFR4. The FGFR4-R388 allele, linked to poor cancer prognosis, increased collagen invasion by decreasing lysosomal MT1-MMP degradation. FGFR4-R388 induced MT1-MMP phosphorylation and endosomal stabilization, and surprisingly, the increased MT1-MMP in return enhanced FGFR4-R388 autophosphorylation. A phosphorylation-defective MT1-MMP was stabilized on the cell surface, where it induced simultaneous FGFR4-R388 internalization and dissociation of cell–cell junctions. In contrast, the alternative FGFR4-G388 variant down-regulated MT1-MMP, and the overexpression of MT1-MMP and particularly its phosphorylation-defective mutant vice versa induced FGFR4-G388 degradation. These results provide a mechanistic basis for FGFR4-R388 function in cancer invasion.

  • Clinical Characterization of Patients Diagnosed with Prostate Cancer and Undergoing Conservative Management : A PIONEER Analysis Based on Big Data

    • Background: Conservative management is an option for prostate cancer (PCa) patients either with the objective of delaying or even avoiding curative therapy, or to wait until palliative treatment is needed. PIONEER, funded by the European Commission Innovative Medicines Initiative, aims at improving PCa care across Europe through the application of big data analytics. Objective: To describe the clinical characteristics and long-term outcomes of PCa patients on conservative management by using an international large network of real-world data. Design, setting, and participants: From an initial cohort of >100 000 000 adult individuals included in eight databases evaluated during a virtual study-a-thon hosted by PIONEER, we identified newly diagnosed PCa cases (n = 527 311). Among those, we selected patients who did not receive curative or palliative treatment within 6 mo from diagnosis (n = 123 146). Outcome measurements and statistical analysis: Patient and disease characteristics were reported. The number of patients who experienced the main study outcomes was quantified for each stratum and the overall cohort. Kaplan-Meier analyses were used to estimate the distribution of time to event data. Results and limitations: The most common comorbidities were hypertension (35–73%), obesity (9.2–54%), and type 2 diabetes (11–28%). The rate of PCa-related symptomatic progression ranged between 2.6% and 6.2%. Hospitalization (12–25%) and emergency department visits (10–14%) were common events during the 1st year of follow-up. The probability of being free from both palliative and curative treatments decreased during follow-up. Limitations include a lack of information on patients and disease characteristics and on treatment intent. Conclusions: Our results allow us to better understand the current landscape of patients with PCa managed with conservative treatment. PIONEER offers a unique opportunity to characterize the baseline features and outcomes of PCa patients managed conservatively using real-world data. Patient summary: Up to 25% of men with prostate cancer (PCa) managed conservatively experienced hospitalization and emergency department visits within the 1st year after diagnosis; 6% experienced PCa-related symptoms. The probability of receiving therapies for PCa decreased according to time elapsed after the diagnosis.

  • Evaluating synergies and trade-offs among Sustainable Development Goals (SDGs) : Explorative analyses of development paths in South Asia and Sub-Saharan Africa

    • Understanding the linkages between multiple targets of Sustainable Development Goals (SDGs) may help to integrate different sectoral programmes and develop coherent cross-sectoral policy to explore synergies. Synergy is interaction among two or more actions, which will lead to an impact greater or less than the sum of individual effects. Therefore, synergy can be positive or negative (trade-off). This paper aims at developing an analytical framework to evaluate sectoral linkages and examine potential synergies and trade-offs among various SDGs' goals and targets. Synergies and trade-offs related to energy access (SDG7), clean water and sanitation access (SDG6), food security and sustainable agriculture (SDG2) and poverty alleviation (SDG1) have been evaluated from the perspective of developing countries using examples from South Asia (Bangladesh, Nepal, and Sri Lanka) and Sub-Saharan Africa (Ghana, Ethiopia and Rwanda), and historical data for the period between 1990 and 2012. The analytical framework includes both qualitative and quantitative methods. Network analysis technique has been used for exploring the conceptual linkage among different indicators, and capturing the targets associated with SDGs. Advanced Sustainability Analysis (ASA) developed under the European framework programme has been used for quantifying the synergies and trade-offs among sustainability indicators. The analysis showed strong synergy among various SDG targets. Interestingly, the potential synergy differs from country to country and over time. Ghana and Sri Lanka had relatively higher potential synergy, whereas Rwanda and Nepal had relatively lower potential synergy among the various targets. Higher synergy values were evidenced in those cases where the policy have recognized and emphasized on linkages among cross-sectoral targets.

  • Role of socio-cultural factors on changes in fitness and adiposity in youth : A 6-year follow-up study

    • BACKGROUND AND AIMS: Optimal cardio-respiratory fitness and adiposity levels are tightly related to health in youth. We analysed changes in fitness and adiposity in young individuals from two countries, and examined the role of maternal education in these changes.METHODS AND RESULTS: A 6-year follow-up study was conducted on 483 Estonian children (9 years) and 466 Swedish children (9-10 years) and adolescents (15 years). Fitness was assessed by a maximal bike test, and total and central adiposity were indirectly estimated by skinfolds (Slaughter's equation for fat mass) and waist circumference. At follow-up, fitness and adiposity had increased in the children cohort (P ≤ 0.001), while small or no change occurred in the adolescent cohort. In the children cohort, Estonian participants had a lower increase in fitness and a higher increase in adiposity (total and central) than Swedish participants. Higher maternal education increased the odds of remaining fit (top quartile) by half and reduced the risk of remaining fat (top quartile) by half; odds ratios = 1.56 (1.00-2.43), 0.50 (0.32-0.77) and 0.61 (0.39-0.94) for fitness, total and central adiposity, respectively.CONCLUSIONS: Our data suggest that the socioeconomic situation of a country might influence key cardiovascular risk factors (fitness and adiposity), being at higher risk for a low-middle income country (Estonia) than a higher income country (Sweden). The findings stress the role of socioeconomic status, particularly maternal education, in the maintenance of healthy fitness and adiposity levels from childhood into later life. Preventive efforts have to be taken from early age.

    • This study identified LTBP-2 as a pleiotropic tumor suppressor in nasopharyngeal carcinoma, which safeguards against critical malignant behaviors of tumor cells. LTBP-2 expression was significantly decreased or lost in up to 100% of NPC cell lines (7/7) and 80% of biopsies (24/30). Promoter hypermethylation was found to be involved in LTBP-2 silencing. Using a tetracycline-regulated inducible expression system, we unveiled functional roles of LTBP-2 in suppressing colony formation, anchorage-independent growth, cell migration, angiogenesis, VEGF secretion, and tumorigenicity. Three-dimensional culture studies suggested the involvement of LTBP-2 in maintenance of tumor cell dormancy in a growth factor favorable microenvironment.

  • Active commuting to school in children and adolescents: an opportunity to increase physical activity and fitness

    • Aims: The purpose was to describe the patterns of commuting to school in young people and to examine its associations with physical activity (PA) and cardiorespiratory fitness. Methods: The sample comprised 2271 Estonian and Swedish children and adolescents (1218 females) aged 9—10 years and 15—16 years. Data were collected in 1998/99. Mode of commuting to and from school was assessed by questionnaire. Time spent (min/day) in PA and average PA (counts/min) was measured by accelerometry. Cardiorespiratory fitness was assessed by means of a maximal cycle ergometer test. Results: Sixty-one percent of the participants reported active commuting to school (ACS). Estonian youth showed lower levels of ACS than Swedish (odds ratio, 0.64; 95% confidence interval, 0.53—0.76) and girls reported lower levels than boys (0.74; 0.62—0.88). ACS boys showed higher PA levels than non-ACS boys for moderate, vigorous, MVPA, and average PA levels (all p ≤ 0.01). Participants who cycled to school had higher cardiorespiratory fitness than walkers or passive travellers (p < 0.001). Conclusions: Nearly two-thirds of the participants actively commuted to school. ACS may provide an opportunity to increase levels of daily PA, especially in boys, and cardiorespiratory fitness, especially if cycling. Public health strategies should develop and test ACS patterns to get more evidence and promote bike-friendly environments.

    • A method is presented for reconstructing the geometry, the pith, the knots and the local fibre orientations in timber boards, based on X-ray computed tomography scans. The local fibre deviations around knots were found by a new algorithm, based on image analysis. The experimental data comprised tomography scans, eigenfrequency measurements and four-point bending tests of 20 Norway spruce boards. 3D and 1D finite element models of the pure bending zone of the bending tests were created, accounting for the exact board geometry and the reconstructed fibre deviations. A purely density based, a purely eigenfrequency based, and a mixed constitutive law were compared. Model estimations showed a high coefficient of determination (R2) for global modulus of elasticity (MoE) (R2⩽0.93), local MoE (R2⩽0.87), bending strength (R2⩽0.83), and the location of initial failure. Constitutive laws accounting for eigenfrequency showed the most accurate results. In the future, adapting the method for logs could enable analyses of boards before sawing.

    • Non-destructive testing of wood for prediction of strength is significantly influenced by wood density and moisture content. A sensor capable of measuring both density and moisture content would be a good tool to aid in predicting the strength of sawn timber. This study was carried out to investigate the possibility of calibrating a prediction model for the moisture content and density of Scots pine (Pinus sylvestris) using microwave sensors. The material was initially at green moisture content, and thereafter dried in several steps to zero moisture content. At each step all the samples were weighted, scanned with a microwave camera (Satimo 9.4 GHz) and CT scanned with a medical CT scanner (Siemens Somatom AR.T.). The output variables from the microwave camera were used as predictors, and CT images correlated with known moisture content were used as response variables. Multivariate models to predict moisture content and density were calibrated using partial least squares (PLS) regression. The result shows that it is possible to predict both moisture content and density with very high accuracy using microwave sensors

    • The TGF beta family member growth differentiation factor-9 (GDF-9) is an oocyte-derived factor that is essential for mammalian ovarian folliculogenesis. GDF-9 mRNAs have been shown to be expressed in the human ovarian follicle from the primary follicle stage onward, and recombinant GDF-9 has been shown to promote human ovarian follicle growth in vitro. In this study with primary cultures of human granulosa-luteal (hGL) cells, we investigated whether recombinant GDF-9 activates components of the Smad signaling pathways known to be differentially activated by TGF beta and the bone morphogenetic proteins (BMPs). As with TGF beta, GDF-9 treatment caused the phosphorylation of endogenous 53-kDa proteins detected in Western blots with antiphospho-Smad2 antibodies (alpha PS2). However, unlike BMP-2, GDF-9 did not activate the phosphorylation of antiphospho-Smad1 antibody (alphaPS1)-immunoreactive proteins in hGL cells. Infection of hGL cells with an adenovirus expressing Smad2 (Ad-Smad2) confirmed that GDF-9 activates specifically phosphorylation of the Smad2 protein. Infection of hGL cells with Ad-Smad7, which expresses the inhibitory Smad7 protein, suppressed the levels of both GDF-9-induced endogenous and adenoviral alpha PS2-reactive proteins. Furthermore, GDF-9 increased the steady state levels of inhibin beta(B)-subunit mRNAs in hGL cells and strongly stimulated the secretion of dimeric inhibin B. Again, Ad-Smad7 blocked GDF-9-stimulated inhibin B production in a concentration-dependent manner. We identify here for the first time distinct molecular components of the GDF-9 signaling pathway in the human ovary. Our data suggest that GDF-9 mediates its effect through the pathway commonly activated by TGF beta and activin, but not that activated by many BMPs. The results are also consistent with the suggestion that in addition to endocrine control of inhibin production by gonadotropins, a local paracrine control of inhibin production is likely to occur via oocyte-derived factors in the human ovary.

  • Comorbidity Burden in Severe and Nonsevere Asthma: A Nationwide Observational Study (FINASTHMA)

    • Background: Asthma, affecting more than 330 million people worldwide, is associated with a high level of morbidity, mortality, and socioeconomic costs. Objective: In this cross-sectional study, we analyzed the comorbidity burden in patients with severe asthma compared with nonsevere asthma and investigated the role of corticosteroid use on the risk of comorbidities. Methods: All adults (≥18 y) with a diagnosis of asthma (International Classification of Diseases–10th revision code J45.x) between 2014 and 2017 were identified and data were collected until 2018 from Finnish nationwide registers. Asthma was defined as continuously or transiently severe or nonsevere based on annual dispensed inhaled corticosteroids (ICS), oral corticosteroids (OCS), and hospitalizations. Results: Of 193,730 adult identified patients diagnosed with asthma, 86.3% had nonsevere, 8.1% transiently severe, and 5.6% continuously severe asthma. Excess prevalence of pneumonia was observed in continuously (22%) and transiently severe (14%) compared with nonsevere patients after adjusting for age and sex. Cataract, osteoporosis, obesity, heart failure, and atrial fibrillation were also more frequent in severe asthma patients. The ICS and/or OCS use contributed to the risk of several comorbidities in a dose-dependent manner, particularly pneumonia, osteoporosis, obesity, heart failure, and atrial fibrillation. High OCS use and the presence of comorbidities were associated with increased health care resource use. Conclusions: Patients with severe asthma have a high burden of comorbidities, especially pneumonia. Many of the comorbidities have a strong dose-dependent association with ICS and OCS treatment, suggesting that corticosteroid doses should be carefully evaluated in clinical practice.

    • Sustainable Development Goals (SDGs) comprises of 17 different goals with 169 targets. Understanding how these multiple targets and goals cut across different sectors and are linked may help to design and appraise common strategies and cross-sectoral policies towards integrated development programmes. This paper aims at developing a method for evaluating the synergies and trade-offs among various goals and targets within SDGs using both qualitative and quantitative approaches. We have analyzed synergy across four SDGs in developing countries, namely energy access (SDG 7), clean water and sanitation access (SDG 6), food security and sustainable agriculture (SDG 2) and poverty alleviation (SDG 1). These specific SDGs have been identified as priority areas in the Rio+20. Various targets related to the four SDGs have been analyzed in this paper with the use of indicators. Developing countries viz. Nepal, Bangladesh, Sri Lanka from South Asia and Ghana, Ethiopia and Rwanda from Africa have been chosen for the analysis. We analyze interactions among goals, and examine how the accomplishment of one target under a specific goal might help to attend multiple goals (mutually reinforcing or synergies), or how the accomplishment of targets under one specific goal might impact targets under another goal (imposing conditions or trade-offs). Network analysis technique has been used for exploring the conceptual linkage among these different indicators capturing the various targets associated with SDGs. Advanced Sustainability Analysis (ASA) approach developed under the European framework programme has been used for quantifying the synergy and trade-off among various SDGs. Based on the analysis, some coherent policy measures across energy, water/sanitation, food and poverty alleviation nexus are suggested.

    • Combination therapies in metastatic hormone-sensitive prostate cancer (mHSPC), which include the addition of an androgen receptor signaling inhibitor and/or docetaxel to androgen deprivation therapy, have been a game changer in the management of this disease stage. However, these therapies come with their fair share of toxicities and side effects. The goal of this observational study is to report drug-related adverse events (AEs), which are correlated with systemic combination therapies for mHSPC. Determining the optimal treatment option requires large cohorts to estimate the tolerability and AEs of these combination therapies in “real-life” patients with mHSPC, as provided in this study. We use a network of databases that includes population-based registries, electronic health records, and insurance claims, containing the overall target population and subgroups of patients defined by unique certain characteristics, demographics, and comorbidities, to compute the incidence of common AEs associated with systemic therapies in the setting of mHSPC. These data sources are standardised using the Observational Medical Outcomes Partnership Common Data Model. We perform the descriptive statistics as well as calculate the AE incidence rate separately for each treatment group, stratified by age groups and index year. The time until the first event is estimated using the Kaplan-Meier method within each age group. In the case of episodic events, the anticipated mean cumulative counts of events are calculated. Our study will allow clinicians to tailor optimal therapies for mHSPC patients, and they will serve as a basis for comparative method studies.
