  • Towards systems beyond 3G based on adaptive OFDMA transmission
  • 2007
  • In: Proceedings of the IEEE. - 0018-9219 .- 1558-2256. ; 95:12, s. 2432-2455
  • Research review (peer-reviewed)abstract
    • High data rates, high spectral efficiency, flexibility, and low delays over the air interface will be important features in next-generation wireless systems. The overall challenge will be packet scheduling and adaptive radio transmission for multiple users, via multiple antennas and over frequency-selective wideband channels. This problem needs to be structured to obtain feasible solutions. The basic simplifying assumptions used here are clustering of antennas into cells, orthogonal transmission by use of cyclic-prefix orthogonal frequency-division multiplexing (OFDM) and a time-scale separation view of the total link adaptation, scheduling and intercell coordination problem. Based on these assumptions, we survey techniques that adapt the transmission to the temporal, frequency, and spatial channel properties. We provide a systematic overview of the design problems, such as the dimensioning of the allocated time-frequency resources, the influence of duplexing schemes, adaptation control issues for downlinks and uplinks, timing issues, and their relation to the required performance of channel predictors. Specific design choices are illustrated by recent research within the Swedish Wireless IP program and the EU IST-WINNER project. The presented results indicate that high-performance adaptive OFDM transmission systems are indeed feasible, also for challenging scenarios that involve vehicular velocities, high carrier frequencies, and high bandwidths

    • © 2005 IEEE. Recent research has shown that a single-carrier system with frequency domain equalization can be a potential alternative to an OFDM-based system in a single user case. In this paper we propose and evaluate an adaptive single-carrier TDMA uplink with scheduling for broadband cellular packet data systems. Frequency division duplex (FDD) is assumed. Multiuser channel estimation and channel prediction are considered. The adaptive modulation scheme is optimized to maximize the throughput including also the ARQ part of the transmission. Frequency domain equalization is employed to combat the inter-symbol-interference (ISI). With the proposed design, the simulation results show that the multiuser diversity gain in spectral efficiency is limited in a wideband system, which indicates that an OFDM based system might be a more favorable candidate.

  • A study of value in agile software development organizations
  • 2017
  • In: Journal of Systems and Software. - : Elsevier BV. - 0164-1212 .- 1873-1228. ; 125, s. 271-288
  • Journal article (peer-reviewed)abstract
    • The Agile manifesto focuses on the delivery of valuable software. In Lean, the principles emphasise value, where every activity that does not add value is seen as waste. Despite the strong focus on value, and that the primary critical success factor for software intensive product development lies in the value domain, no empirical study has investigated specifically what value is. This paper presents an empirical study that investigates how value is interpreted and prioritised, and how value is assured and measured. Data was collected through semi-structured interviews with 23 participants from 14 agile software development organisations. The contribution of this study is fourfold. First, it examines how value is perceived amongst agile software development organisations. Second, it compares the perceptions and priorities of the perceived values by domains and roles. Third, it includes an examination of what practices are used to achieve value in industry, and what hinders the achievement of value. Fourth, it characterises what measurements are used to assure, and evaluate value-creation activities. (C) 2016 Elsevier Inc. All rights reserved.

  • An exploratory study of waste in software development organizations using agile or lean approaches : A multiple case study at 14 organizations
  • 2019
  • In: Information and Software Technology. - : Elsevier B.V.. - 0950-5849 .- 1873-6025. ; 107, s. 78-94
  • Journal article (peer-reviewed)abstract
    • Context: The principal focus of lean is the identification and elimination of waste from the process with respect to maximizing customer value. Similarly, the purpose of agile is to maximize customer value and minimize unnecessary work and time delays. In both cases the concept of waste is important. Through an empirical study, we explore how waste is approached in agile software development organizations. Objective: This paper explores the concept of waste in agile/lean software development organizations and how it is defined, used, prioritized, reduced, or eliminated in practice Method: The data were collected using semi-structured open-interviews. 23 practitioners from 14 embedded software development organizations were interviewed representing two core roles in each organization. Results: Various wastes, categorized in 10 different categories, were identified by the respondents. From the mentioned wastes, not all were necessarily waste per se but could be symptoms caused by wastes. From the seven wastes of lean, Task-switching was ranked as the most important, and Extra-features, as the least important wastes according to the respondents’ opinion. However, most companies do not have their own or use an established definition of waste, more importantly, very few actively identify or try to eliminate waste in their organizations beyond local initiatives on project level. Conclusion: In order to identify, recognize and eliminate waste, a common understanding, and a joint and holistic view of the concept is needed. It is also important to optimize the whole organization and the whole product, as waste on one level can be important on another, thus sub-optimization should be avoided. Furthermore, to achieve a sustainable and effective waste handling, both the short-term and the long-term perspectives need to be considered. © 2018 Elsevier B.V.

    • Melanophores are pigmented cells in lower vertebrates capable of quick color changes and thereby suitable as whole cell biosensors. In the frog dermis skin layer, the large and dark pigmented melanophore surrounds a core of other pigmented cells. Upon hormonal stimulation the black-brown pigment organelles will redistribute within the melanophore, and thereby cover or uncover the core, making complex color changes possible in the dermis. Previously, melanophores have only been cultured on flat surfaces. Here we mimic the three dimensional biological geometry in the frog dermis by culturing melanophores on fluorescent plastic microbeads. To demonstrate biosensing we use the hormones melatonin and α-melanocyte stimulating hormone (α-MSH) as lightening or darkening stimuli, respectively. Cellular responses were successfully demonstrated on single cell level by fluorescence microscopy, and in cell suspension by a fluorescence microplate reader and a previously demonstrated computer screen photo-assisted technique. The demonstrated principle is the first step towards "single well/multiple read-out" biosensor arrays based on suspensions of different selective-responding melanophores, each cultured on microbeads with distinctive spectral characteristics. By applying small amount of a clinical sample, or a candidate substance in early drug screening, to a single well containing combinations of melanophores on beads, multiple parameter read-outs will be possible. © 2004 Elsevier B.V. • Myosin V is the rate-determinative step in Xenopus melanophore aggregation
  • Other publication (other academic/artistic)abstract
    • In Xenopus melanophores, melatonin induce melanosome aggregation via activation of its receptor Mel1c, coupled to inhibitory G proteins, adenylate cyclase deactivation, cyclic adenosine 3':5'-monophosphate (cAMP) decrease, protein kinase A inhibition, protein phophatase 2A activation, and serine/threonine dephosphorylations. Myosin V is the motor protein responsible for transporting melanosomes along actin filaments. Myosin V has been demonstrated to be necessary for melanosome dispersion and to keep the dispersed state. We have previously shown that melatonin induce activation of phosphoinositide-3 kinase, mitogen-activated protein kinase and tyrosine phosphorylation of a 280-kDa protein. Here we characterize the kinetics of latrunculin A-induced aggregation, and show that latrunculin A aggregate melanophores with the same kinetics as melatonin. This indicates that the downstream mechanisms might be similar although their primary targets in the cells are totally different. We suggest that both drugs act by inhibiting myosin V, the rate-determinative step for melanosome aggregation. Our data suggest that dynein is not up regulated during aggregation, as previously suggested by others,

  • Phosphoinositide 3-kinase is involved in Xenopus and Labrus melanophore aggregation
  • 2003

  • Journal article (peer-reviewed)abstract
    • Melanophores are pigmented cells capable of quick colour changes through coordinated transport of their intracellular pigment granules. We demonstrate the involvement of phosphoinositide 3-kinase (PI3-K) in Xenopus and Labrus aggregation by the use of the PI3-K inhibitor, LY-294002. In Xenopus, wortmannin-insensitive PI3-K was found to be essential for the aggregation, mitogen-activated protein kinase (MAPK) activation and tyrosine phosphorylation of a 280-kDa protein, and for the maintenance of low cyclic adenosine 3':5'-monophosphate (cAMP) during the aggregated state. Pre-aggregated cells disperse completely to LY-294002 at 50-100 muM, involving a transient elevation in cAMP due to adenylate cyclase (AC) stimulation or to inhibition of cyclic nucleotide phosphodiesterase (PDE). The inactive analogue LY-303511 did not induce dispersion at the same concentrations. PDE4 and/or PDE2 was found to be involved in melanosome aggregation. The similar kinetics of LY-294002 and various PDE inhibitors indicates that the elevation of cAMP might be due to inhibition of PDE. In Labrus melanophores, LY-294002 had a less dramatic effect, probably due to less dependence on PDE in regulation of cAMP levels. In Xenopus aggregation, we suggest that melatonin stimulation of the Mel1c receptor via G(betagamma) activates PI3-K that, directly or indirectly via MAPK, activates PDE. (C) 2003 Elsevier Inc. • 2003
  • In: Pigment Cell Research. - : Wiley. - 0893-5785 .- 1600-0749. ; 16:3, s. 215-221
  • Journal article (peer-reviewed)abstract
    • Our objectives were to further characterize the signaling pathways in melatonin-induced aggregation in Xenopus melanophores, specifically to investigate a possible role of mitogen-activated protein kinase (MAPK). By Western blotting we found that melatonin activates MAPK, which precedes melanosome aggregation measured in a microplate reader. Activation of MAPK, tyrosine phosphorylation of a previously described 280-kDa protein, and melanosome aggregation are sensitive to PD98059, a selective inhibitor of MAPK kinase. The MAPK activation is also decreased by the adenylate cyclase stimulant forskolin. In summary, we found that MAPK is activated during melatonin-induced melanosome aggregation. Activation was decreased by an inhibitor of MAPK kinase, and by forskolin. In addition to inhibition of cyclic adenosine 3′,5′-monophosphate (cAMP), reduction in protein kinase A activity (PKA), and activation of protein phosphatase 2A, we suggest that melatonin receptors activate the MAPK cascade and tyrosine phosphorylation of the 280-kDa protein. Although the cAMP/PKA signaling pathway is the most prominent, our data suggest that simultaneous activation of the MAPK cascade is of importance to obtain a completely aggregated state. This new regulatory mechanism of organelle transport by the MAPK cascade might be important in other eukaryotic cells.

  • 2022
  • Reports (other academic/artistic)abstract

    • The necessity of software as stand-alone products, and as central parts of non-traditional software products have changed how software products are developed. It started with the introduction of the agile manifesto and has resulted in a change of how software process improvements (SPI) are conducted. Although there are agile SPI methods and several agile practices for evaluating and improving current processes and ways-of-working, no method or practices for evaluating the backlog exists. To address this gap, the Backlog Assessment Method (BAM) was developed and applied in collaboration with Telenor Sweden. BAM enables agile organizations to assess backlogs, and assure that the backlog items are good-enough for their needs and well aligned with the decision process. The results from the validation show that BAM is feasible and relevant in an industrial environment, and it indicates that BAM is useful as a tool to perform analysis of items in a specific backlog. © The Author(s) 2019.

  • Prioritization of quality requirements : State of practice in eleven companies
  • 2011
  • In: 2011 IEEE 19th International Requirements Engineering Conference, RE 2011; Trento; 29 August 2011 through 2 September 2011. - Trento : IEEE. - 9781457709234 ; , s. 69-78, s. 69-78
  • Conference paper (peer-reviewed)abstract
    • Requirements prioritization is recognized as an important but challenging activity in software product development. For a product to be successful, it is crucial to find the right balance among competing quality requirements. Although literature offers many methods for requirements prioritization, the research on prioritization of quality requirements is limited. This study identifies how quality requirements are prioritized in practice at 11 successful companies developing software intensive systems. We found that ad-hoc prioritization and priority grouping of requirements are the dominant methods for prioritizing quality requirements. The results also show that it is common to use customer input as criteria for prioritization but absence of any criteria was also common. The results suggests that quality requirements by default have a lower priority than functional requirements, and that they only get attention in the prioritizing process if decision-makers are dedicated to invest specific time and resources on QR prioritization. The results of this study may help future research on quality requirements to focus investigations on industry-relevant issues.

    • In order to create a successful software product and assure its quality, it is not enough to fulfill the functional requirements, it is also crucial to find the right balance among competing quality requirements (QR). An extended, previosluy piloted, interview study was performed to identify specific challenges associated with the selection, trade-off, and management of QR in industrial practice. Data was collected through semi-structured interviews with eleven product managers and eleven project leaders from eleven software companies. The contribution of this study is fourfold: First, it compares how QR are handled in two cases, companies working in business-to-business markets, and companies that are working in business-to-consumer markets. These two are also compared in terms of impact on the handling of QRs. Second, it compares the perceptions and priorities of QR by product and project management respectively. Third, it includes an examination of the interdependencies among quality requirements perceived as most important by the practitioners. Fourth, it characterizes the selection and management of QR in down-stream development activities.

  • Software architecture as a means of communication in a globally distributed software development context
  • 2012
  • Conference paper (peer-reviewed)abstract
    • The management and coordination of globally distributed development poses many new challenges, including compensating for informal implicit communication, which is aggravated by heterogeneous social and engineering traditions between development sites. Although much research has gone into identifying challenges and working with practical solutions, such as tools for communication, little research has focused on comparing communication mechanisms in terms of their ability to provide large volumes of rich information in a timely manner. Data was collected through in-depth interviews with eleven practitioners and twenty-eight responses through a web-based questionnaire from three product lines at an international software development organization. This paper assesses the relative importance of ten commonly used communication mechanisms and practices across local and global development sites. The results clearly indicate that some communication mechanisms are more important than others in providing large volumes of rich information in a timely manner. The prevalence of architecture in providing rich information in large volumes for both local and global communication can be clearly observed.

  • Panax ginseng induces anterograde transport of pigment organelles in Xenopus melanophores
  • 2008
  • In: Journal of Ethnopharmacology. - : Elsevier. - 0378-8741 .- 1872-7573. ; 119:1, s. 17-23
  • Journal article (peer-reviewed)abstract
    • Melanophores from Xenopus laevis are pigmented cells, capable of quick colour changes through cyclic adenosine 3':5'-monophosphate (cAMP) coordinated transport of their intracellular pigment granules, melanosomes. In this study we use the melanophore cell line to evaluate the effects of Panax ginseng extract G115 on organelle transport. Absorbance readings of melanophore-coated microplates, Correlate-EIA direct cAMP enzyme immunoassay kit, and western blot were used to measure the melanosome movement and changes in intracellular signalling. We show that Panax ginseng induces a fast concentration-dependent anterograde transport of the melanosomes. No significant increase in the cAMP level was seen and pre-incubation of melanophores with the protein kinase C (PKC) inhibitor EGF-R Fragment 651-658 (M-EGF) only partly decreased the ginseng-induced dispersion. We also demonstrate that Panax ginseng, endothelin-3 (ET-3) and alpha-melanocyte stimulating hormone (MSH) stimulate an activation of mitogen activated protein kinase (MAPK). Pre-incubation with M-EGF decreased the MAPK activity induced by ET-3 and MSH, but again only marginally affected the response of Panax ginseng. Thus, in melanophores we suggest that Panax ginseng stimulates an anterograde transport of pigment organelles via a non-cAMP and mainly PKC-independent pathway.

  • Genome-wide meta-analysis identifies 56 bone mineral density loci and reveals 14 loci associated with risk of fracture.

  • In: Nature genetics. - : Springer Science and Business Media LLC. - 1546-1718 .- 1061-4036. ; 44:5, s. 491-501
  • Journal article (peer-reviewed)abstract
    • Bone mineral density (BMD) is the most widely used predictor of fracture risk. We performed the largest meta-analysis to date on lumbar spine and femoral neck BMD, including 17 genome-wide association studies and 32,961 individuals of European and east Asian ancestry. We tested the top BMD-associated markers for replication in 50,933 independent subjects and for association with risk of low-trauma fracture in 31,016 individuals with a history of fracture (cases) and 102,444 controls. We identified 56 loci (32 new) associated with BMD at genome-wide significance (P < 5 × 10(-8)). Several of these factors cluster within the RANK-RANKL-OPG, mesenchymal stem cell differentiation, endochondral ossification and Wnt signaling pathways. However, we also discovered loci that were localized to genes not known to have a role in bone biology. Fourteen BMD-associated loci were also associated with fracture risk (P < 5 × 10(-4), Bonferroni corrected), of which six reached P < 5 × 10(-8), including at 18p11.21 (FAM210A), 7q21.3 (SLC25A13), 11q13.2 (LRP5), 4q22.1 (MEPE), 2p16.2 (SPTBN1) and 10q21.1 (DKK1). These findings shed light on the genetic architecture and pathophysiological mechanisms underlying BMD variation and fracture susceptibility.

  • 2003
  • In: Biosensors & bioelectronics. - 0956-5663 .- 1873-4235. ; 19:1, s. 35-41
  • Journal article (peer-reviewed)abstract
    • Melanophores, dark pigment cells from the frog Xenopus laevis, have the ability to change light absorbance upon stimulation by different biological agents. Hormone exposure (e.g. melatonin or α-melanocyte stimulating hormone) has been used here as a reversible stimulus to test a new compact microplate reading platform. As an application, the detection of the asthma drug formoterol in blood plasma samples is demonstrated. The present system utilizes a computer screen as a (programmable) large area light source, and a standard web camera as recording media enabling even kinetic microplate reading with a versatile and broadly available platform, which suffices to evaluate numerous bioassays. Especially in the context of point of care testing or self testing applications these possibilities become advantageous compared with highly dedicated comparatively expensive commercial systems.

  • Methods for Compression of Feedback in Adaptive Multi-carrier 4G Schemes
  • 2008
  • In: Wireless Personal Communications. - : Springer Science and Business Media LLC. - 1572-834X .- 0929-6212. ; 47:1, s. 101-112
  • Journal article (peer-reviewed)abstract
    • In this paper, several algorithms for compressing the feedback of channel quality information are presented and analyzed. These algorithms are developed for a proposed adaptive modulation scheme for future multi-carrier 4G mobile systems. These strategies compress the feedback data and, used together with opportunistic scheduling, drastically reduce the feedback data rate. Thus the adaptive modulation schemes become more suitable and efficient to be implemented in future mobile systems, increasing data throughput and overall system performance.

  • Is effect of (S;S)-formoterol due to contamination of (R;R)-formoterol?
  • Other publication (other academic/artistic)abstract
    • Formoterol is a long acting selective ß2-adrenoceptor (ß2-AR) agonist of the so-called third generation of ß-adrenoceptor agonists. lt also has an onset action comparable to most short acting ß2-AR agonists. Formoterol has two chiral centres making four enantiomers possible. In this study we have examined (R;R)- and (S;S)-formoterol relaxing effect on guinea pig tracheal ring preparations, affinity to human ß2-AR in transfected COS-7 cells and the ability to influence pigment movement in frog melanophores with stable expression of human ß2AR. We also compared single concentration curves versus cumulative concentration curves on guinea pig tracheal preparations. In all three systems the (R;R)-formoterol is the most potent ß2AR agonist compered to (S;S)-formoterol with eudismic ratios ranging from 11 to 75. We also measure and theoretically calculated the effect of (S;S)-formoterol. VVhen the contamination of (R;R)-formoterol was subtracted the (S;S)-formoterol had effect, although approximately 72 times less then (R;R)-formoterol. We conclude that (R;R)-formoterol is the most potent ß2-AR agonist in three different systems and that (S;S)-formoterol posses an ß2-AR effect. We also show that cumulative concentration curves have higher EC50 values compered to single concentration curves and that this might be a consequence of recaptor desensitisation.

  • Built Environments to Support Rehabilitation for People With Stroke From the Hospital to the Home (B-Sure) : Protocol for a Mixed Method Participatory Co-Design Study
  • 2023
  • In: JMIR Research Protocols. - 1929-0748. ; 12
  • Journal article (peer-reviewed)abstract
    • BACKGROUND: A global trend is to move rehabilitation closer to people's neighborhoods and homes. Still, little attention has been given to how the built environment outside the hospital setting might impact rehabilitation and recovery for stroke survivors.OBJECTIVE: The overarching objective of this project is to develop conceptual models of built environments that support stroke rehabilitation and recovery outside the hospital setting. Specifically, the project will explore factors and characteristics of the built environment that support people with stroke and their families and identify innovative built environments that can be designed for local health care. The project will examine facilitators and obstacles for implementing built environmental solutions and evaluate the potential benefits, feasibility, and acceptability.METHODS: The project uses a mixed methods design approach with 3 phases. In phase 1, factors and characteristics of the built environment for rehabilitation will be identified. Based on the results from phase 1, phase 2 will involve co-designing prototypes of environments to support the rehabilitation process for people with stroke. Finally, the prototypes will be evaluated in phase 3. Qualitative and quantitative methods will include a literature review, a concept mapping (CM) study, stakeholder interviews, prototype development, and testing. The project will use multidimensional scaling, hierarchical cluster analysis, descriptive statistics for quantitative data, and content analysis for qualitative data. Location analysis will rely on the location-allocation model for network problems, and the rule-based analysis will be based on geographic information systems data.RESULTS: As of the submission of this protocol, ethical approval for the CM study and the interview study has been obtained. Data collection is planned to start in September 2023 and the workshops later in the same year. The scoping review is ongoing from January 2023. The CM study is ongoing and will be finalized in the spring of 2024. We expect to finish the data analysis in the second half of 2024. The project is a 3-year project and will continue until December 2025.CONCLUSIONS: We aim to determine how new environments could better support a person's control over their day, environment, goals, and ultimately control over their recovery and rehabilitation activities. This "taking charge" approach would have the greatest chance of transferring the care closer to the patient's home. By co-designing with multiple stakeholders, we aim to create solutions with the potential for rapid implementation. The project's outcomes may target other people with frail health after a hospital stay or older persons in Sweden and anywhere else. The impact and social benefits include collaboration between important stakeholders to explore how new environments can support the transition to local health care, co-design, and test of new conceptual models of environments that can promote health and well-being for people post stroke.INTERNATIONAL REGISTERED REPORT IDENTIFIER (IRRID): DERR1-10.2196/52489.

  • Peripheral inflammatory disease associated with centrally activated IL-1 system in humans and mice.
  • 2012
  • In: Proceedings of the National Academy of Sciences of the United States of America. - : Proceedings of the National Academy of Sciences. - 0027-8424 .- 1091-6490. ; 109:31, s. 12728-33
  • Journal article (peer-reviewed)abstract
    • During peripheral immune activation caused by an infection or an inflammatory condition, the innate immune response signals to the brain and causes an up-regulation of central nervous system (CNS) cytokine production. Central actions of proinflammatory cytokines, in particular IL-1β, are pivotal for the induction of fever and fatigue. In the present study, the influence of peripheral chronic joint inflammatory disease in rheumatoid arthritis (RA) on CNS inflammation was investigated. Intrathecal interleukin (IL)-1β concentrations were markedly elevated in RA patients compared with controls or with patients with multiple sclerosis. Conversely, the anti-inflammatory IL-1 receptor antagonist and IL-4 were decreased in RA cerebrospinal fluid (CSF). Tumor necrosis factor and IL-6 levels in the CSF did not differ between patients and controls. Concerning IL-1β, CSF concentrations in RA patients were higher than in serum, indicating local production in the CNS, and there was a positive correlation between CSF IL-1β and fatigue assessments. Next, spinal inflammation in experimental arthritis was investigated. A marked increase of IL-1β, IL-18, and tumor necrosis factor, but not IL-6 mRNA production, in the spinal cord was observed, coinciding with increased arthritis scores in the KBxN serum transfer model. These data provide evidence that peripheral inflammation such as arthritis is associated with an immunological activation in the CNS in both humans and mice, suggesting a possible therapeutic target for centrally affecting conditions as fatigue in chronic inflammatory diseases, for which to date there are no specific treatments.

  • T4 - Telematics for Totally Transparent Transports
  • 2005
  • In: 2005 IEEE Intelligent Transportation Systems Conference (ITSC). - New York : IEEE Press. - 0780392159 ; , s. 467-472
  • Conference paper (peer-reviewed)abstract
    • Transports can be made safer, more secure and efficient by help of telemetry and tracking on-line in real time. T4 is a system architecture aimed to support the development of telematic services for transparent tracking and surveillance monitoring of goods transported by different means on a global scale. The main idea is to focus on the transported pallets or parcels instead of the vehicles moving them. To enable rapid response to new customer requirements and to support remote management of field equipment, software implemented services are designed, packaged, deployed and mediated using XML, Java and the OSGi software technology standards.

  • CD163+ macrophages monitor enhanced permeability at the blood-dorsal root ganglion barrier
  • 2024
  • In: Journal of Experimental Medicine. - : Rockefeller University Press. - 0022-1007 .- 1540-9538. ; 221:2
  • Journal article (peer-reviewed)abstract
    • In dorsal root ganglia (DRG), macrophages reside close to sensory neurons and have largely been explored in the context of pain, nerve injury, and repair. However, we discovered that most DRG macrophages interact with and monitor the vasculature by sampling macromolecules from the blood. Characterization of the DRG vasculature revealed a specialized endothelial bed that transformed in molecular, structural, and permeability properties along the arteriovenous axis and was covered by macrophage-interacting pericytes and fibroblasts. Macrophage phagocytosis spatially aligned with peak endothelial permeability, a process regulated by enhanced caveolar transcytosis in endothelial cells. Profiling the DRG immune landscape revealed two subsets of perivascular macrophages with distinct transcriptome, turnover, and function. CD163(+) macrophages self-maintained locally, specifically participated in vasculature monitoring, displayed distinct responses during peripheral inflammation, and were conserved in mouse and man. Our work provides a molecular explanation for the permeability of the blood-DRG barrier and identifies an unappreciated role of macrophages as integral components of the DRG-neurovascular unit.

  • 2011
  • Reports (other academic/artistic)abstract
