    • The Amazonian rainforest is arguably the most species-rich terrestrial ecosystem in the world, yet the timing of the origin and evolutionary causes of this diversity are a matter of debate. We review the geologic and phylogenetic evidence from Amazonia and compare it with uplift records from the Andes. This uplift and its effect on regional climate fundamentally changed the Amazonian landscape by reconfiguring drainage patterns and creating a vast influx of sediments into the basin. On this "Andean" substrate, a region-wide edaphic mosaic developed that became extremely rich in species, particularly in Western Amazonia. We show that Andean uplift was crucial for the evolution of Amazonian landscapes and ecosystems, and that current biodiversity patterns are rooted deep in the pre-Quaternary.

  • Key to Improved Ion Core Confinement in the JET Tokamak : Ion Stiffness Mitigation due to Combined Plasma Rotation and Low Magnetic Shear

    • New experimental evidence indicates that ion stiffness mitigation in the core of rotating plasmas, observed previously in JET, results from the combined effect of high rotational shear and low magnetic shear. Ionstiffness in the outer plasma region is found to remain very high irrespective of rotation. Dedicated experimentsin plasmas with different q profiles and rotation levels point to a larger effect of rotation in reducing stiffnesswhen the core q profile is made flatter. The results have implications for the understanding of improved ion coreconfinement in hybrid plasmas or Internal Transport Barriers, both characterized by high rotation and low magnetic shear. Experimental evidence in these scenarios is discussed. Simulations indicate that the physics behindthese results may lie in the ITG/TEM turbulence behavior at the transition between fully developed turbulenceand zonal flows quenching. These findings point to the need for future devices of achieving sufficient rotationalshear and capability of q profile manipulation to reach improved ion core confinement, which is an essentialfeature of Advanced Tokamak operation.

    • The potential effects of oral contraceptive (OC) and postmenopausal hormone (PMH) use are not well understood among BRCA1 or BRCA2 (BRCA1/2) deleterious mutation carriers with a history of breast cancer. We investigated the association between OC and PMH use and risk of contralateral breast cancer (CBC) in the WECARE (Women's Environment, Cancer, and Radiation Epidemiology) Study. The WECARE Study is a population-based case-control study of 705 women with asynchronous CBC and 1,398 women with unilateral breast cancer, including 181 BRCA1/2 mutation carriers. Risk-factor information was assessed by telephone interview. Mutation status was measured using denaturing high-performance liquid chromatography followed by direct sequencing in all participants. Outcomes, treatment, and tumor characteristics were abstracted from medical records. Ever use of OCs was not associated with risk among noncarriers (RR = 0.87; 95% CI = 0.66-1.15) or BRCA2 carriers (RR = 0.82; 95% CI = 0.21-3.13). BRCA1 carriers who used OCs had a nonsignificant greater risk than nonusers (RR = 2.38; 95% CI = 0.72-7.83). Total duration of OC use and at least 5 years of use before age 30 were associated with a nonsignificant increased risk among mutation carriers but not among noncarriers. Few women had ever used PMH and we found no significant associations between lifetime use and CBC risk among carriers and noncarriers. In conclusion, the association between OC/PMH use and risk of CBC does not differ significantly between carriers and noncarriers; however, because carriers have a higher baseline risk of second primaries, even a potential small increase in risk as a result of OC use may be clinically relevant.

    • The 'European Transport Simulator' (ETS) (Coster et al 2010 IEEE Trans. Plasma Sci. 38 2085-92, Kalupin et al 2011 Proc. 38th EPS Conf. on Plasma Physics (Strasbourg, France, 2011) vol 35G (ECA) P. 4.111) is the new modular package for 1D discharge evolution developed within the EFDA Integrated Tokamak Modelling (ITM) Task Force. It consists of precompiled physics modules combined into a workflow through standardized input/output data structures. Ultimately, the ETS will allow for an entire discharge simulation from the start up until the current termination phase, including controllers and sub-systems. The paper presents the current status of the ETS towards this ultimate goal. It discusses the design of the workflow, the validation and verification of its components on the example of impurity solver and demonstrates a proof-of-principles coupling of a local gyrofluid model for turbulent transport to the ETS. It also presents the first results on the application of the ETS to JET tokamak discharges with the ITER like wall. It studies the correlations of the radiation from impurity to the choice of the sources and transport coefficients.

    • Samaria-doped ceria-based composites with a 2:1 addition of Li and Na carbonates (or simple Na carbonate as admixture) were prepared mixing nanosized powders of the ceramic phase with the remaining constituents. Samples fired at relatively low temperatures (below 700 degrees C) were characterized by X-ray diffraction, scanning electron microscopy combined with energy dispersive spectroscopy and impedance spectroscopy in air. These composites showed a complex but homogeneous distribution of both phases, with one ceramic skeleton of bonded nanosized grains surrounded by the carbonate-based phase. Impedance spectroscopy data was used to confirm the impressive electrical conductivity of these materials, but also to put into evidence the complex nature of the charge transport process, clearly deviating from classical electrolytes.

    • Anaplastic lymphoma kinase (ALK) is a receptor tyrosine kinase (RTK), which is transiently expressed during development of the central and peripheral nervous system. ALK has been recently identified as a major neuroblastoma predisposition gene and activating mutations have also been identified in a subset of sporadic neuroblastoma tumors. Two hot spots of ALK mutations have been observed at positions F1174 and R1275. Here, we studied stably transfected cell lines expressing wild-type or F1174L- or R1275Q-mutated ALK in parallel with a neuroblastoma cell line (CLB-GE) in which the allele mutated at position F1174 is amplified. We observed that the mutated ALK variants were essentially intracellular and were largely retained in the reticulum/Golgi compartments. This localization was corroborated by a defect of N-linked glycosylation. Although the mutated receptors exhibited a constitutive activation, the minor pool of receptor addressed to the plasma membrane was much more tyrosine phosphorylated than the intracellular pool. The use of antagonist monoclonal antibodies suggested that the constitutive activity of the mutated receptors did not require the dimerization of the receptor, whereas adequate dimerization triggered by agonist monoclonal antibodies increased this activity. Finally, kinase inactivation of the mutated receptors restored maturation and cell-surface localization. Our results show that constitutive activation of ALK results in its impaired maturation and intracellular retention. Furthermore, they provide a rationale for the potential use of kinase inhibitors and antibodies in ALK-dependent tumors.Oncogene advance online publication, 17 January 2011; doi:10.1038/onc.2010.595.
