  • Large-scale proteomics analysis of human ovarian cancer for biomarkers

    • Ovarian cancer is usually found at a late stage when the prognosis is often bad. Relative survival rates decrease with tumor stage or grade, and the 5-year survival rate for women with carcinoma is only 38%. Thus, there is a great need to find biomarkers that can be used to carry out routine screening, especially in high-risk patient groups. Here, we present a large-scale study of 64 tissue samples taken from patients at all stages and show that we can identify statistically valid markers using nonsupervised methods that distinguish between normal, benign, borderline, and malignant tissue. We have identified 217 of the significantly changing protein spots. We are expressing and raising antibodies to 35 of these. Currently, we have validated 5 of these antibodies for use in immunohistochemical analysis using tissue microarrays of healthy and diseased ovarian, as well as other, human tissues.

  • Diagnosis of patients with raised serum calcium level in primary care, Sweden.

    • OBJECTIVE: To study the diagnosis of hypercalcaemic patients and to evaluate whether frequent analyses of serum calcium can detect more patients with hypercalcaemia. DESIGN: Retrospective study of serum calcium analyses performed during the time period 1992-2000 and of the medical records of patients with elevated serum calcium levels between 1995 and 2000. SETTING: Primary care in Tibro, Sweden. SUBJECTS: Patients from the local community attending the primary healthcare centre. MAIN OUTCOME MEASURES: Frequency of serum calcium analyses, hypercalcaemic patients, and their diagnosis. RESULTS: Doubling the number of serum calcium analyses did not increase the detected number of raised calcium levels. On the other hand, more frequent parathyroid hormone (PTH) analyses resulted in a corresponding increase in detected high PTH levels. In Tibro, 15% (n = 22) of the patients with hypercalcaemia were diagnosed with primary hyperparathyroidism, giving a rate of 0.22%. This is comparable to the prevalence in other population studies. Over 40% (n = 9) of patients with primary hyperparathyroidism in the study had only slightly raised serum calcium levels (2.55-2.60 mmol/l). In 70% (n = 99) of the cases, the cause of hypercalcaemia was unknown. The second most common diagnosis was skeletal disorders followed by kidney disease. CONCLUSION: An increase in the number of serum calcium analyses did not result in increased detection of raised calcium levels. In contrast, an increase in the number of PTH analyses resulted in increased detection of primary hyperparathyroidism. Therefore, PTH analyses should be used more frequently.

  • Nuclear expression of mu-opioid receptors in a human mesothelial cell line

    • 1 Possibly acting via mu-opioid receptors (MORs), morphine inhibits the formation ofexperimentally induced postoperative abdominal adhesions in rats. Mesothelial cells mayparticipate in adhesion formation by secreting mediators that interfere negatively withfibrinolysis. Morphine may prevent adhesions by inhibiting the release of pro-adhesionmediators from mesothelial cells. This study aimed to investigate whether human mesothelialcells express MOR-1; if so, such could constitute a site of action for morphine in adhesionprevention.2 Cells from Met-5A, a human mesothelial cell line were seeded and prepared forimmunocytochemistry and Western blotting.3 Immunocytochemistry showed MOR-1 expression in mesothelial cells, predominantly in thenuclei. Western blotting showed two bands (c. 35 and 50 kDa) which correspond to thoseobtained with a control lysate from cells known to express MORs. In addition, we foundMOR-1 expression with nuclear and cytoplasmatic localization in biopsies from humanabdominal adhesions.4 The current findings may suggest that morphine could interact directly with mesothelial cellsvia MOR-1 receptors, and thereby modulate adhesion formation, possibly by interfering withthe release of pro-adhesion factors from these cells

  • Membrane-integration characteristics of two ABC transporters, CFTR and P-glycoprotein

    • To what extent do corresponding transmembrane helices in related integral membrane proteins have different membrane-insertion characteristics? Here, we compare, side-by-side, the membrane insertion characteristics of the 12 transmembrane helices in the adenosine triphosphate-binding cassette (ABC) transporters, P-glycoprotein (P-gp) and the cystic fibrosis transmembrane conductance regulator (CFTR). Our results show that 10 of the 12 CFTR transmembrane segments can insert independently into the ER membrane. In contrast, only three of the P-gp transmembrane segments are independently stable in the membrane, while the majority depend on the presence of neighboring loops and/or transmembrane segments for efficient insertion. Membrane-insertion characteristics can thus vary widely between related proteins.

    • Hur blir marknadsorientering till? Såväl i marknadsorienteringslitteraturen som i företag framhålls vikten av att sätta ”kunden i fokus”. Genom att svara mot kundernas behov och önskemål för-väntas företagen vinna konkurrensfördelar och därmed bättre lönsamhet. Men ”kunden i fokus” är abstrakt och talar egentligen inte om för företagen hur en orientering mot kunden går till. Denna studie belyser hur ett försäkringsbolag på olika sätt försöker orientera sig mot kunden och kundens kontext, det vill säga den marknad kunden befinner sig på. Litteraturgenomgången visar att en stor del av tidigare forskning inom marknadsorienteringsfältet utgår från att marknadsori-entering blir till genom behandling av marknadsinformation. Detta är dock ett begränsat sätt att se på marknadsorientering och hur företag lär om kundens behov och önskemål. Det utgår från att information alltid är tillgänglig och att människor vet vilken information som är intressant. Ett grundantagande om att verkligheten ser likadan ut, oavsett vem som tolkar den, framträder och detta ifrågasätts i den här studien. Ett alternativt perspektiv på marknadsorientering, som tar sin utgångspunkt i att organisationer består av olikheter och mångfald snarare än homogenitet, pre-senteras därför. Informationsbehandling är inte tillräckligt för att lära om kundens behov och önskemål, människor lär i interaktioner, det är när olikheter och variation får diskuteras som kun-skap växer. I analysen utvecklas förståelse för vad i ett tjänsteföretag som försvårar respektive underlättar för sociala interaktioner i ett marknadsorienteringsperspektiv. När människor förstår varandra tack vare gemensam praktik, delade fysiska och mentala rum och kan tala med varandra med ett delat organisatoriskt språk bidrar detta till att marknadsorientering blir till. Föreställningar om kunden och kundens kontext konstrueras på många skilda platser i företaget, marknadsorienter-ing blir mångfacetterad. En slutsats som dras är att mångfalden i sig inte utgör ett problem, utan det är vad mångfalden gör för marknadsorienterandet. De skilda föreställningarna leder till en mängd olika ageranden, men även till att människor väljer att inte engagera sig alls eftersom de inte ser någon mening med olika marknadsorienterade aktiviteter. Marknadsorienteringens kontext, i form av den horisontella och vertikala specialisering som försäkringsbolaget är indelat i, influerar också interaktioner. Interaktioner mellan avdelningar och enheter, mellan det operativa och det strategiska arbetet influeras av föreställningar om tid, intern konkurrens, engagemang och otydlig kommunikation och en uppdelning av människor i ”vi” och ”dom”. I företaget finns ett styrsystem som bidrar till att människor prioriterar den individuella tiden, den som ger resultat i form positiva resultat i de månatliga mätningarna, före den kollektiva tiden i vilken människor kan dela erfarenheter och utveckla kunskaper om kunden. Styrsystemet medverkar också till att interaktioner mellan enheter med samma specialisering försvåras, ut-formningen av systemet bidrar till konkurrens snarare än samverkan. Interaktioner mellan männi-skor med olika specialiseringar influeras av engagemang, människor finner marknadsorientering betydelsefullt vilket innebär att aktiviteter som överbrygger specialiserade gränser skapas. Interak-tionerna influeras också av otydlig kommunikation. Slutsatsen av detta är att gränser och normer får stor betydelse för om marknadsorientering blir till eller inte. Marknadsorienteringens presta-tionsparadox handlar om att klara av att parallellt hantera intern effektivitet och extern fokusering på kunden och kundens kontext.

    • Two-dimensional SIDS-PAGE gel electrophoresis using post-run staining is widely used to measure the abundances of thousands of protein spots simultaneously. Usually, the protein abundances of two or more biological groups are compared using biological and technical replicates. After gel separation and staining, the spots are detected, spot volumes are quantified, and spots are matched across gels. There are almost always many missing values in the resulting data set. The missing values arise either because the corresponding proteins have very low abundances (or are absent) or because of experimental errors such as incomplete/over focusing in the first dimension or varying run times in the second dimension as well as faulty spot detection and matching. In this study, we show that the probability for a spot to be missing can be modeled by a logistic regression function of the logarithm of the volume. Furthermore, we present an algorithm that takes a set of gels with technical and biological replicates as input and estimates the average protein abundances in the biological groups from the number of missing spots and measured volumes of the present spots using a maximum likelihood approach. Confidence intervals for abundances and p-values for differential expression between two groups are calculated using bootstrap sampling. The algorithm is compared to two standard approaches, one that discards missing values and one that sets all missing values to zero. We have evaluated this approach in two different gel data sets of different biological origin.   • Somatostatin effects on the proteome of the LNCaP cell-line

    • Some clinical results indicate that somatostatin (sms) might be useful in the treatment of advanced prostate cancer (HRPC). Because of its transient in vivo half-life only more stable derivatives of sms are of interest in this context. Recent studies have shown that natural sms can be conjugated to a carbohydrate (smsdx) with preservation of sms-like effects on the prostatic tumor cell proteome. The present study identifies some of the affected proteins in an effort to elucidate pathways and proteins that might be of importance for the potential usefulness of sms treatment in HRPC. After incubating the LNCaP cell-line with sms14/smsdx, comparative proteomics was used for analysing and identifying affected proteins. Protein expression patterns were analysed with two-dimensional polyacrylamide gel electrophoresis and mass spectrometry. Catalytic mitochondrial and mitochondrial-associated proteins were significantly affected (fold change approximately 2 or higher) and they were in general up-regulated. Apoptosis-related proteins were both up-regulated (VDAC1, VDAC2) and down-regulated (PRDX2, TCTP). The fold change was >2 for PRDX2 and <2 for the others. There was a strong agreement between sms and smsdx on the up- and down-regulation of proteins. Sms/smsdx triggered up-regulation of catalytic mitochondrial proteins and seemed to affect apoptosis-related proteins. This could indicate important pathways on which smsdx might be able to curb the progression of HRPC. The results encourage a pending clinical phase II study.

    • The purpose was to develop an extension finger-force measurement device, and investigate the intra-individual repeatability. The design of the measuring device allows single finger force and whole hand measurements, and the repeatability error on extension finger forces was measured, both on the whole hand, as well as on individual fingers. The tests showed that a repeatability error of less then 15 % can be achieved for single finger measurements and less then 21 % for whole hand measurements.
