    • Objective: To describe the experience of physical activity in patients with heart failure who were randomized to a control group in which they received exercise advice and motivational support.Design: A qualitative descriptive interview study, using qualitative content analysis.Patients: Fifteen patients with heart failure.Methods: Interviews with members of the control group in a physical activity trial (Heart Failure-Wii study), who received exercise advice and telephone follow-ups.Results: The experience of physical activity was described by 4 categories: (1) affected by study participation; (ii) impact of having heart failure; (iii) mixed feelings when physically active; and (iv) influence of the social and physical environment. Study participation made patients feel encouraged or obliged, or they gained a sense of security in connection with becoming more physically active. Symptoms, side-effects and barriers could hamper their physical activity. They changed their type and level of physical activity, or used resources or equipment to become physically active. Patients experienced physical and emotional improvements and developed routines and structures, and their environment and caregivers were able to influence their physical activity.Conclusion: The role of study participation, having heart failure, and psychological and environmental factors describe the experiences of physical activity in patients with heart failure who were randomized to a control group, and are important in evaluating and motivating patients' physical activity.

  • Multiple sulfur-isotopic evidence for a shallowly stratified ocean following the Triassic-Jurassic boundary mass extinction

    • The cause of the Triassic-Jurassic (Tr-J) boundary biotic crisis, one of the ‘Big Five’ mass extinctions of the Phanerozoic, remains controversial. In this study, we analyzed multiple sulfur-isotope compositions (δ33S, δ34S and δ36S) of pyrite and Spy/TOC ratios in two Tr-J successions (Mariental, Mingolsheim) from the European Epicontinental Seaway (EES) in order to better document ocean-redox variations during the Tr-J transition. Our results show that upper Rhaetian strata are characterized by 34S-enriched pyrite, low Spy/TOC ratios, and values of Δ33Spy (i.e., the deviation from the mass-dependent array) lower than that estimated for contemporaneous seawater sulfate, suggesting an oxic-suboxic depositional environment punctuated by brief anoxic events. The overlying Hettangian strata exhibit relatively 34S-depleted pyrite, high Δ33Spy, and Spy/TOC values, and the presence of green sulfur bacterial biomarkers indicate a shift toward to euxinic conditions. The local development of intense marine anoxia thus postdated the Tr-J mass extinction, which does not provide support for the hypothesis that euxinia was the main killing agent at the Tr-J transition. Sulfur and organic carbon isotopic records that reveal a water-depth gradient (i.e., more 34S-, 13C-depleted with depth) in combination with Spy/TOC data suggest that the earliest Jurassic EES was strongly stratified, with a chemocline located at shallow depths just below storm wave base. Shallow oceanic stratification may have been a factor for widespread deposition of black shales, a large positive shift in carbonate δ13C values, and a delay in the recovery of marine ecosystems following the Tr-J boundary crisis.

    • Mucosal administration of Ag conjugated to cholera toxin B subunit (CTB) can efficiently induce peripheral immunologic tolerance, so-called oral tolerance, associated with development of Foxp3(+)CD25(+)CD4(+) regulatory T (Treg) cells. Using an established sublingual tolerization regimen with Ag(OVA)/CTB conjugate, wherein CTB mediates Ag uptake and presentation by most B lymphocytes irrespective of their Ag specificity, we have assessed the importance of B cells for induction of Ag-specific Treg cells and oral tolerance. We found that Treg cells are reduced in microMT(-/-) B cell-deficient mice compared with wild-type (WT) mice. After sublingual Ag/CTB treatment, Treg cells increased much more in WT than in microMT(-/-) mice; however, adoptive transfer of B cells before treatment normalized Treg cell development and functional oral tolerance. B cells from OVA/CTB-treated mice expressed more IL-10 and less CD86 than control B cells. Adoptive transfer of these cells before parenteral immunization with OVA led to efficient suppression of proliferation and to induction of apoptotic depletion of Ag-specific CD25(-)CD4(+) effector T cells associated with the expansion of Treg cells. However, also OVA/CTB-treated microMT(-/-) mice could suppress the immune response to parenteral immunization with OVA, which was associated with a strong increase in Foxp3(-)CD4(+) T cells expressing LAP/TGF-beta. Our results indicate that mucosal tolerance comprises at least two separate pathways: one being B cell dependent and associated with expansion of Treg cells and Treg-mediated suppression and depletion of effector T cells, and one being B cell independent and associated with development of Foxp3(-)LAP(+)TGF-beta(+) regulatory T cells.

    • Coronavirus disease 2019 (COVID-19) is a recently-emerged infectious disease that has caused millions of deaths, where comprehensive understanding of disease mechanisms is still unestablished. In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection.

    • Accurately estimating the state of health (SoH) of batteries is indispensable for the safety, reliability, and optimal energy and power management of electric vehicles. However, from a data-driven perspective, complications, such as dynamic vehicle operating conditions, stochastic user behaviors, and cell-to-cell variations, make the estimation task challenging. This work develops a data-driven multi-model fusion method for SoH estimation under arbitrary usage profiles. All possible operating conditions are categorized into six scenarios. For each scenario, an appropriate feature set is extracted to indicate the SoH. Based on the obtained features, four machine learning algorithms are applied individually to train SoH estimation models using time-series data. In addition to the estimates at the current time step, a histogram data-based and online adaptive model is taken from previous work for predicting the next-step SoH. Then, a Kalman filter is applied to systematically fuse the results of all the estimation and prediction models. Experimental data collected from different types of batteries operated under diverse profiles verify the effectiveness and practicability of the developed method, as well as its superiority over individual models.
