    • Concerns have been raised about the abuse potential of pregabalin. Therefore, the aim of our study was to characterize patients dispensed pregabalin at higher than the maximum allowed dose in a cohort study based on data extracted from Swedish national registers. All patients dispensed at least three prescriptions of pregabalin between July 2006 and December 2009 were included (n = 48,550). The daily dose was defined as the amount of pregabalin dispensed divided by the number of days between the second and third dispensings. Associations between sociodemographic and clinical variables and dispensing pregabalin at a dose exceeding the maximum daily allowed dose (600 mg) were investigated in multivariate regression models. Of the patients dispensed pregabalin during the study period, 8.5 % were dispensed a dose that exceeded the maximum daily allowed dose. A previous addictive disorder drug treatment or diagnosis was present in 20 and 31 % of patients dispensed pregabalin within and exceeding the recommended dose range, respectively. Our analysis revealed that those patients at increased risk of being dispensed pregabalin at higher than the maximum allowed dose were male [adjusted odds ratio (aOR) 1.40, 95 % confidence interval (CI) 1.31-1.49], were between 18 and 29 years of age compared with those aged a parts per thousand yen65 years (aOR 1.62, 95 % CI 1.45-1.82), had a low income (aOR 1.24, 95 % CI 1.10-1.40), had epilepsy compared with no diagnosis (aOR 1.41, 95 % CI 1.10-1.81), had a previous substance use disorder treatment or diagnosis (aOR 1.41, 95 % CI 1.31-1.52) or had previously been dispensed high doses of drugs with abuse potential (aOR 1.77, 95 % CI 1.62-1.94). Based on our results we conclude that patients at a high risk of addiction and patients with epilepsy are more likely to be dispensed pregabalin at higher than the maximum approved daily dose.

    • Hydrogen sensing based on Pd nanoalloys has shown great promise in the past decades and could potentially be part of a solution that enables a safe future hydrogen economy. There are, however, remaining challenges related to, e.g., long-term stability and a need for further optimization of these systems. To efficiently span the possible combinations of alloyants, composition, and nanostructure geometry, computational methods are invaluable. This thesis focuses on two aspects of sensor optimization: surface segregation and hydrogen sensitivity, using multi-scale modeling approaches. Alloying with metals such as Au and Cu is necessary to overcome issues related to hysteresis and CO poisoning. At the same time, it introduces additional difficulties related to the chemical order such as surface segregation, which is directly related to long-term stability. In this thesis, the surface composition of Pd alloyed with Au or Cu is studied as a function of H2 pressure using Monte Carlo simulations based on cluster expansions parametrized against ab-initio calculations. For Pd-Au, an increased H2 concentration abruptly switches the surface from Au to Pd dominant. For Pd-Cu, the change with H2 concentration is much more gradual with non-monotonic tendencies, with an overall surplus of Pd in most conditions. The sensing principle is based on the shift in optical response upon H absorption. The magnitude of the sensor readout at a certain H2 pressure depends on nanoparticle geometry and alloy composition. In this thesis, extinction spectra are calculated for Pd-Au-H nanodisks using electrodynamic simulations and the corresponding H sensitivity is analyzed. It is found that the H sensitivity depends on the nanodisk diameter, mainly due to the interplay between a localized surface plasmon and an interband transition which becomes more apparent for smaller nanodisks.

    • In recent years, the concept of peer-to-peer computing has gained renewed interest for sharing resources within and between organizations or individuals. This article describes a decentralized resource management system (DRMS) that uses a network of workstations for the execution and storage of high-level architecture (HLA) federations/federates in a peer-to-peer environment. The implementation of DRMS is based on the open-source project JXTA, which represents an attempt to standardize the peer-to-peer domain. DRMS is part of a Web-based simulation environment supporting collaborative design, development, and execution of HLA federations. This study evaluates the possibilities of using peer-to-peer technology for increasing the reuse and availability of simulation components within the defense modelling and simulation community. More specifically, it addresses the necessary adjustments of simulation components to conform to the requirements of the DRMS and shows that JXTA could provide the foundation for a distributed system that increases the possibilities for reusing simulation components.

    • Aim: To investigate the effect of carbon monoxide (CO) in the inspired air as anticipated during peak hours of traffic in polluted megalopolises on cerebral, respiratory and leg muscle oxygenation during a constant-power test (CPT). In addition, since O2 breathing is used to hasten elimination of CO from the blood, we examined the effect of breathing O2 following exposure to CO on cerebral and muscle oxygenation during a subsequent exercise test under CO conditions. Methods: Nine men participated in three trials: (i) 3-h air exposure followed by a control CPT, (ii) 1-h air and 2-h CO (18.9 ppm) exposure succeeded by a CPT under CO conditions (CPTCO A), and (iii) 2-h CO and 1-h 100% normobaric O2 exposure followed by a CPT under CO conditions (CPTCO B). All exercise tests were performed at 85% of peak power output to exhaustion. Oxygenated (D[O2Hb]), deoxygenated (D[HHb]) and total (D[tHb]) haemoglobin in cerebral, intercostal and vastus lateralis muscles were monitored with near-infrared spectroscopy throughout the CPTs. Results: Performance time did not vary between trials. However, the vastus lateralis and intercostal D[O2Hb] and D[tHb] were lower in CPTCO A than in CPT. During the CPTCO B, the intercostal D[O2Hb] and D[tHb] were higher than in the CPTCO A. There were no differences in cerebral oxygenation between the trials. Conclusion: Inspiration of 18.9 ppm CO decreases oxygenation in the vastus lateralis and serratus anterior muscles, but does not affect performance. Breathing normobaric O2 moderates the CO-induced reductions in muscle oxygenation, mainly in the intercostals, but does not affect endurance.
