    • We determine the strong coupling constant alpha(s) and its energy dependence from the p(T) dependence of the inclusive jet cross section in pp collisions at s=1.96 TeV. The strong coupling constant is determined over the transverse momentum range 50 < p(T)< 145 GeV. Using perturbative QCD calculations to order O(alpha(3)(s)) combined with O(alpha(4)(s)) contributions from threshold corrections, we obtain alpha(s)(M-Z)=0.1161(-0.0048)(+0.0041). This is the most precise result obtained at a hadron-hadron collider.

  • Localization and expression of spermadhesin PSP-I/PSP-II subunits in the reproductive organs of the boar

    • The epithelial localization and expression of the spermadhesin PSP-I and PSP-II subunits were determined in the testis, ductus epididymes (caput, corpus and cauda), seminal vesicles and bulbourethral glands of mature boars, using immunohistochemical, western blotting and RT-PCR methods. Immunohistochemistry showed positive labelling for PSP-I and PSP-II antibodies in the epithelium of seminal vesicles in all males tested. Positive immunolabelling, but with variable intensity, was also present in the epididymal epithelium (caput, corpus and cauda), although varying largely among segments and boars. Immunoreactivity was nearly or completely absent in the seminiferous epithelium and the bulbourethral gland, although SDS-PAGE and western blotting revealed the presence of PSP-I and PSP-II immunoreactive bands in all the tissue extracts, including the testis and the bulbourethral gland. mRNA amplification by RT-PCR using primers specific for PSP-I and PSP-II showed a trend similar to that observed for western blotting, i.e. intensity variation between tissues (even between segments of the same epididymis) and among boars. Our results indicate that the seminal vesicles are the main source of PSP-I and PSP-II spermadhesins, although epididymal segments, testis and the bulbourethral gland also participate in the expression of both proteins.

  • The effect of early menarche on later body composition and fat distribution in female adolescents: role of birth weight

    • <i>Background:</i> To examine the effect of early menarche (EM) on adolescent body composition and fat distribution, and to explore the possible contribution of birth weight (BW) to this relationship. <i>Methods:</i> From the cross-sectional AVENA study, 788 girls (aged 13–18.5 years) were selected. Post-menarcheal body composition was estimated by anthropometric methods. <i>Results:</i> BW Z-score was associated with age at menarche (β = 0.496, p < 0.001). EM (age of menarche <12 years) adolescents were shorter, had a higher body mass index (BMI), fat free mass index (FFMI), fat mass index (FMI) and waist circumference (all p < 0.01). BW was associated with height (p < 0.001), BMI Z-score (p < 0.01), FFMI (p < 0.01) and FMI (p < 0.05). These relationships did not change when the analysis was controlled for age of menarche. Height and FFM were lower in EM (p < 0.01), but these effects seem to be due to the BW (p < 0.01). <i>Conclusion:</i> EM was strongly associated with unhealthy body composition in female adolescents, but these relationships seem to be due to the programming effect of BW.

  • Accumulation of ubiquitin conjugates in a polyglutamine disease model occurs without global ubiquitin/proteasome system impairment

    • Aggregation-prone proteins have been suggested to overwhelm and impair the ubiquitin/proteasome system (UPS) in polyglutamine (polyQ) disorders, such as Huntington's disease (HD). Overexpression of an N-terminal fragment of mutant huntingtin (N-mutHtt), an aggregation-prone polyQ protein responsible for HD, obstructs the UPS in cellular models. Furthermore, based on the accumulation of polyubiquitin conjugates in brains of R6/2 mice, which express human N-mutHtt and are one of the most severe polyQ disorder models, it has been proposed that UPS dysfunction is a consistent feature of this pathology, occurring in both in vitro and in vivo models. Here, we have exploited transgenic mice that ubiquitously express a ubiquitin fusion degradation proteasome substrate to directly assess the functionality of the UPS in R6/2 mice or the slower onset R6/1 mice. Although expression of N-mutHtt caused a general inhibition of the UPS in PC12 cells, we did not observe an increase in the levels of proteasome reporter substrate in the brains of R6/2 and R6/1 mice. We show that the increase in ubiquitin conjugates in R6/2 mice can be primarily attributed to an accumulation of large ubiquitin conjugates that are different from the conjugates observed upon UPS inhibition. Together our data show that polyubiquitylated proteins accumulate in R6/2 brain despite a largely operative UPS, and suggest that neurons are able to avoid or compensate for the inhibitory effects of N-mutHtt.
