  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Novel similarities in the free-surface profiles and velocities of solitary waves traveling over a very steep beach

    • This study investigates experimentally similarity and Froude number similitude (FNS) in the dimensionless flow features of three solitary waves traveling on a 1:3 sloping beach. These three waves, designated as cases A, B, and C, respectively, have different heights H-0 (=5.8 cm, 2.9 cm, and 1.815 cm) and still water depths h(0) (=16.0 cm, 8.0 cm, and 5.0 cm), but identical ratios H-0/h(0) (=0.363). A high-speed particle image velocimetry system is employed to obtain the free surface profiles (FSPs) and velocity fields/profiles. These features include the free surface elevation (FSE)/FSP time series; velocity fields and profiles, positions, and propagation speeds of flow demarcation curves; times and maximum onshore distances of the maximum run-up heights (MRHs); and times and onshore distances of hydraulic jumps for cases A and B. When the swash tip of a solitary wave reaches the MRH, the contact point becomes almost immobile for a short time interval, with the contact angle changing from obtuse, via right, to acute angle. For cases A and B, the similarities in the dimensionless MRHs and times, at which the run-down motions of the wave tips start, are affirmed. These facts highlight that the swash tips and contact points are subject to complex interactions among gravity force, viscous friction, and surface tension of fluid. Case C, having the smallest length scale, is only focused on the arrival or starting time of the MRH or run-down motion and the MRH and used as a counterexample to demonstrate the absence of similarity or FNS due to the relatively prominent effects of viscous friction and surface tension.

    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

    • With the implementation of the 11th edition of the International Classification of Diseases (ICD-11) and the publication of the metabolic dysfunction-associated fatty liver disease (MAFLD) nomenclature in 2020, it is important to establish consensus for the coding of MAFLD in ICD-11. This will inform subsequent revisions of ICD-11.Using the Qualtrics XM and WJX platforms, questionnaires were sent online to MAFLD-ICD-11 coding collaborators, authors of papers, and relevant association members.A total of 890 international experts in various fields from 61 countries responded to the survey. We also achieved full coverage of provincial-level administrative regions in China. 77.1% of respondents agreed that MAFLD should be represented in ICD-11 by updating NAFLD, with no significant regional differences (77.3% in Asia and 76.6% in non-Asia, p=0.819). Over 80% of respondents agreed or somewhat agreed with the need to assign specific codes for progressive stages of MAFLD (i.e. steatohepatitis) (92.2%), MAFLD combined with comorbidities (84.1%), or MAFLD subtypes (i.e., lean, overweight/obese, and diabetic) (86.1%).This global survey by a collaborative panel of clinical, coding, health management and policy experts, indicates agreement that MAFLD should be coded in ICD-11. The data serves as a foundation for corresponding adjustments in the ICD-11 revision.

  • Social and moral psychology of COVID-19 across 69 countries

    • The COVID-19 pandemic has affected all domains of human life, including the economic and social fabric of societies. One of the central strategies for managing public health throughout the pandemic has been through persuasive messaging and collective behaviour change. To help scholars better understand the social and moral psychology behind public health behaviour, we present a dataset comprising of 51,404 individuals from 69 countries. This dataset was collected for the International Collaboration on Social & Moral Psychology of COVID-19 project (ICSMP COVID-19). This social science survey invited participants around the world to complete a series of moral and psychological measures and public health attitudes about COVID-19 during an early phase of the COVID-19 pandemic (between April and June 2020). The survey included seven broad categories of questions: COVID-19 beliefs and compliance behaviours; identity and social attitudes; ideology; health and well-being; moral beliefs and motivation; personality traits; and demographic variables. We report both raw and cleaned data, along with all survey materials, data visualisations, and psychometric evaluations of key variables.

  • Global Fully Distributed Parameter Regionalization Based on Observed Streamflow From 4,229 Headwater Catchments

    • All hydrological models need to be calibrated to obtain satisfactory streamflow simulations. Here we present a novel parameter regionalization approach that involves the optimization of transfer equations linking model parameters to climate and landscape characteristics. The optimization was performed in a fully spatially distributed fashion at high resolution (0.05 degrees), instead of at lumped catchment scale, using an unprecedented database of daily observed streamflow from 4,229 headwater catchments (<5,000 km(2)) worldwide. The optimized equations were subsequently applied globally to produce parameter maps for the entire land surface including ungauged regions. The approach was evaluated using the Kling-Gupta efficiency (KGE) and a gridded version of the hydrological model HBV. Tenfold cross validation was used to evaluate the generalizability of the approach and to obtain an ensemble of parameter maps. For the 4,229 independent validation catchments, the regionalized parameters yielded a median KGE of 0.46. The median KGE improvement (relative to uncalibrated parameters) was 0.29, and improvements were obtained for 88% of the independent validation catchments. These scores compare favorably to those from previous large catchment sample studies. The degree of performance improvement due to the regionalized parameters did not depend on climate or topography. Substantial improvements were obtained even for independent validation catchments located far from the catchments used for optimization, underscoring the value of the derived parameters for poorly gauged regions. The regionalized parameters-available via -should be useful for hydrological applications requiring accurate streamflow simulations.

  • RADSex : A computational workflow to study sex determination using restriction site-associated DNA sequencing data

    • The study of sex determination and sex chromosome organization in nonmodel species has long been technically challenging, but new sequencing methodologies now enable precise and high-throughput identification of sex-specific genomic sequences. In particular, restriction site-associated DNA sequencing (RAD-Seq) is being extensively applied to explore sex determination systems in many plant and animal species. However, software specifically designed to search for and visualize sex-biased markers using RAD-Seq data is lacking. Here, we present RADSex, a computational analysis workflow designed to study the genetic basis of sex determination using RAD-Seq data. RADSex is simple to use, requires few computational resources, makes no prior assumptions about the type of sex-determination system or structure of the sex locus, and offers convenient visualization through a dedicated R package. To demonstrate the functionality of RADSex, we re-analysed a published data set of Japanese medaka, Oryzias latipes, where we uncovered a previously unknown Y chromosome polymorphism. We then used RADSex to analyse new RAD-Seq data sets from 15 fish species spanning multiple taxonomic orders. We identified the sex determination system and sex-specific markers in six of these species, five of which had no known sex-markers prior to this study. We show that RADSex greatly facilitates the study of sex determination systems in nonmodel species thanks to its speed of analyses, low resource usage, ease of application and visualization options. Furthermore, our analysis of new data sets from 15 species provides new insights on sex determination in fish.

  • Substantial increase in future fluvial flood risk projected in China’s major urban agglomerations

    • Urban land will face high fluvial flood risk against the background of climate change and urban expansion. The effect of urban spatial expansion, instead of densification of assets within existing urban cells, on flood risk has rarely been reported. Here, we project the future flood risk of seven urban agglomerations in China, home to over 750 million people. The inundated urban land areas in the future are projected to be 4 to 19 times that at present. Without considering the urban spatial expansion, the inundated urban land areas will be underestimated by 10-50%. Urban land is more likely to be inundated than non-urban land, and the newly-developed urban land will be inundated more easily than the historical urban land. The results demonstrate the urgency of integrating climate change mitigation, reasonable urban land expansion, and increased flood protection levels to minimize the flood risk in urban land.

  • Roles of Acceptor Guests in Tuning the Organic Solar Cell Property Based on an Efficient Binary Material System with a Nearly Zero Hole-Transfer Driving Force

    • Sub-picosecond hole transfer has been recently observed in several narrow band gap nonfullerene small-molecule acceptor (NFA)-based binary blended organic solar cell (OSC) systems operating with negligible energetic driving forces. As the driving forces are near zero, how the added acceptor/donor guests tune the barrier-free hole-transfer dynamics of these systems remains very unclear. In this study, we report a new NFA (BTCT-2Cl) that conducts a sub-picosecond hole transfer (2 ps) for efficient photocurrent generation when pairing with PM6 though the energetic offset is only 0.02 eV. We observe that the added nonfullerene and PCBM components differently tune the charge generation and recombination when selectively exciting BTCT-2Cl. After adding PC71BM, the hole transfer from the host BTCT-2Cl to the host donor is greatly accelerated, with the rate significantly reduced to 0.29 ps and the charge generation becomes more efficient; on the contrary, recombination is prolonged and a larger fill factor is obtained after adding an NFA guest, here, IT-4F. The different tuning on the host binary hole-transfer dynamics is likely related with the phase crystallinity and the domain size changed after adding different acceptor guests. Over 16% efficiency is obtained on the PC71BM-based ternary device that outperforms the host binary and the IT-4F-based ternary solar cells (both showing over 15% efficiencies). The results clearly demonstrate that adding PCBM or NFA guests enables a very effective and different tuning on the hole-transfer rates and the recombination rates between the barrier-free host binary components, hence leading to efficient tuning on the short-circuit current density and fill factor, which outlines new strategies toward designing high-efficiency ternary blended OSC systems.
