  • STAT4 Associates with SLE through two independent effects that correlate with gene expression and act additively with IRF5 to increase risk

    • OBJECTIVES: To confirm and define the genetic association of STAT4 and systemic lupus erythematosus, investigate the possibility of correlations with differential splicing and/or expression levels, and genetic interaction with IRF5. METHODS: 30 tag SNPs were genotyped in an independent set of Spanish cases and controls. SNPs surviving correction for multiple tests were genotyped in 5 new sets of cases and controls for replication. STAT4 cDNA was analyzed by 5'-RACE PCR and sequencing. Expression levels were measured by quantitative PCR. RESULTS: In the fine-mapping, four SNPs were significant after correction for multiple testing, with rs3821236 and rs3024866 as the strongest signals, followed by the previously associated rs7574865, and by rs1467199. Association was replicated in all cohorts. After conditional regression analyses, two major independent signals represented by SNPs rs3821236 and rs7574865, remained significant across the sets. These SNPs belong to separate haplotype blocks. High levels of STAT4 expression correlated with SNPs rs3821236, rs3024866 (both in the same haplotype block) and rs7574865 but not with other SNPs. We also detected transcription of alternative tissue-specific exons 1, indicating presence of tissue-specific promoters of potential importance in the expression of STAT4. No interaction with associated SNPs of IRF5 was observed using regression analysis. CONCLUSIONS: These data confirm STAT4 as a susceptibility gene for SLE and suggest the presence of at least two functional variants affecting levels of STAT4. Our results also indicate that both genes STAT4 and IRF5 act additively to increase risk for SLE.

    • Reaction of sodium azide with manganese(II) and 4-amino-1,2,4-triazol or 3-chloropyridine leads to the 1D systems [Mn(N-3)(2) (4-amtr)(2)](,) (1) or {[Mn(N-3)(2)(3-Clpy)(2)]center dot 1/2(3-Clpy)}(n) (2), respectively. Compound 1 crystallises in the C2/m (monoclinic) group and consists of chains of Mn(II) cations bridged by double mu(1,3)-azido bridges in trans arrangement. Compound 2 crystallises in the I2/a (monoclinic) group and in this case the 1D system shows alternating double mu(1,1) and mu(1,3) bridges.   • Fiber composition of packaging grade papers as determined by the Graff “C” staining test

    • A qualitative and quantitative analysis of the fiber components of 15 representative papers that are used for the production of corrugated board was carried out by the Graff "C" staining test. The method of processing of softwood, hardwood, and nonwood fibers was determined under a light microscope by their color reactions with the stain. All papers, due to the use of recycled pulp raw materials in their manufacturing, were found to incorporate in their furnish fibers that had been produced with a variety of pulping processes: chemical, mechanical, and semi-mechanical. The recycled-based papers (recycled-liner and recycled-medium) were proved to be the most variable comprising 12-15 different fiber components, while in some of the semi-chemicals only up to 7 components were identified. The weight percentages of the fiber components calculated by the application of weight factors showed that in almost all papers the most important fiber component from a quantitative standpoint was hardwood unbleached kraft followed by softwood unbleached kraft. Besides hardwood unbleached semi-chemical pulp and mechanical softwood pulp that were also plentiful in the papers, there was a smaller number of other components which sum, however, accounted for a significant fraction in the total furnish weight. The results taken on the total softwood, hardwood, and nonwood fibers content of the papers demonstrate that Graff "C" staining test is adequate to analyze both the structure and quality of packaging grade papers in practical industrial testing.

    • Understanding the influence that heterogeneous recycled raw materials have on packaging-grade paper performance offers great potential value to the corrugated board and packaging industry. Fifty-seven linerboards and corrugating medium were selected to represent all the variety of paper grades available on the Spanish market at the moment for the production of corrugated board. The origin of softwood, hardwood, and nonwood fibers and their percentages by weight were determined with light microscopy and standard fiber analysis techniques.

    • Within EUROMET a regional key comparison (EUROMET.M.P-K1.a) was performed in order to compare national vacuum standards in the pressure range from 0.1 Pa to 1000 Pa. The participants were BNM-LNE (France), CEM (Spain), OMH (Hungary), IMGC-CNR (Italy), NPL (United Kingdom), MIKES (Finland), PTB (Germany), NMi (The Netherlands), SP (Sweden) and UME (Turkey). IMGC-CNR acted as pilot laboratory. The measurements were carried out from November 1998 to April 2002. The chosen pressure values (from 0.1 Pa to 1000 Pa) cover the most commonly required range for calibration in low-pressure applications. The transfer standards were commercially available capacitance diaphragm gauges (CDGs): one of them was prepared for the comparison by BNM-LNE (Fr) and two were prepared by IMGC-CNR (It). Two sensors had 133 Pa full scale (one absolute and one relative used as absolute) and one 1333 Pa full scale (absolute). For the two 133 Pa full scale sensors seven pressure steps were generated between 0.1 Pa and 100 Pa; for the 1333 Pa full scale sensor nine pressure steps were generated generally between 0.1 Pa and 1000 Pa. The uncertainty of the generated pressure was reported by each participant in the tables of the results that consisted in the generated pressure value, the uncertainty of the generated pressure, the reading of the gauge and the temperature of the standard at each target pressure. The pilot laboratory has analysed the results, after application of the correction for thermal transpiration, in terms of gauge factors for each gauge, and the combined uncertainty was evaluated by considering, besides the component due to the standards, taking into account the components due to the transfer standards to guarantee a uniform uncertainty analysis for all the participants. At each target pressure a EUROMET reference pressure was calculated; finally the difference between the pressures generated by each laboratory from the reference values was calculated and compared with its expanded uncertainty. The results of most of the laboratories showed a good agreement with the reference values. Only a few values of two laboratories were significantly off the reference values. From the available data a linkage to the CCM.P-K4 key comparison results from 1 Pa to 1000 Pa was possible by means of the results of three laboratories in the (1-30) Pa range and two for the (100-1000) Pa range who took part in both the comparisons.     • Java is a successful programming environment and its use has grown from little embedded applications until enterprise network servers based on J2EE. This intensive use of Java demands the validation of their fault tolerance mechanisms to avoid unexpected behavior of the applications at runtime. This paper describes the design and implementation of a fault injector for the "Exhaustif®" SWIFI tool. Aspecific fault model for java applications that include class corruption/substitution at loading time, method call interception and unexpected exception thrown is proposed. The injector uses the JVMTI (JavaVirtual Machine Tool Interface) to perform bytecode instrumentation at runtime to carry out the fault model previously defined. Finally a XML formalization of the specific Java fault model is proposed. This approach, JVMTI + XML fault model description, provides complete independency between the system under test and the fault injection tool, as well the interoperability with another SWIFI tools.
