  • Effects of Hydroquinone on Cytoskeletal Organization and Intracellular Transport in Cultured Xenopus laevis Melanophores and Fibroblasts

    • Hydroquinone is used as a skin-lightening agent, it is also present in different chemical products and cigarette smoke. It is believed to inhibit melanin production in melanocytes by inhibiting the key enzyme tyrosinase. In the present study, we show that hydroquinone had severe effects on microtubules and actin filaments in cultured Xenopus laevis melanophores as studied by immunohistochemistry. It affected the intracellular transport of melanosomes, induced bundling of microtubules and disassembly of actin filaments at 10 and 50μM, and at 100μM proper adhesion to the substrate was lost. Effects occurred at lower concentrations than what previously has been stated to be cytotoxic, and the results show that tyrosinase is not the only cellular target. The cytoskeleton is of utmost importance for the function of all cells and across species. Our data has therefore to be considered in the discussions about the use of hydroquinone for bleaching of skin.

  • Classification of Raman Spectra to Detect Hidden Explosives

    • Raman spectroscopy is a laser-based vibrational technique that can provide spectral signatures unique to a multitude of compounds. The technique is gaining widespread interest as a method for detecting hidden explosives due to its sensitivity and ease of use. In this letter, we present a computationally efficient classification scheme for accurate standoff identification of several common explosives using visible-range Raman spectroscopy. Using real measurements, we evaluate and modify a recent correlation-based approach to classify Raman spectra from various harmful and commonplace substances. The results show that the proposed approach can, at a distance of 30 m, or more, successfully classify measured Raman spectra from several explosive substances, including nitromethane, trinitrotoluene, dinitrotoluene, hydrogen peroxide, triacetone triperoxide, and ammonium nitrate.

  • Internal and External Knowledge – Innovation of Export Varieties

    • Firms in local industries maintain their capability to generate innovations by simultaneously exploiting internal and external knowledge resources. The paper introduces the notion variety triplet to distinguish individual export varieties, where a triplet is a unique combination of a firm, a product code and a destination country. For each date the set of variety triplets in each local industry records all remaining past product innovations. In view of this the paper examines how internal and external knowledge of local industries influence the industry’s scope and value of export varieties. The paper contributes to existing knowledge firstly by introducing variables that measure a local industry’s access to external supply of knowledge, divided into local and extra-local supply. Secondly, the paper sheds light on how internal and external knowledge influence the scope of product innovations in local industries, with firm-level data from Sweden. Thirdly, the paper compares the influence of knowledge on the entire set of variety triplets and on a separate set of recently introduced varieties.

    • Alzheimer's disease (AD) is the most common neurodegenerative disease, and is clinically characterized by cognitive disturbances and the accumulation of the amyloid beta (A beta) peptides in plaques in the brain. Recent studies have shown the links between AD and the immediate-early gene Arc (activity-regulated cytoskeleton-associated protein), involved in synaptic plasticity and memory consolidation. For example, AD mouse models show a decreased expression of Arc mRNA in the brain. In additional, acute A beta application to brain slices leads to a widespread ARC protein diffusion, unlike the normal defined localization to synapses. In this study, we investigated genetic variation in human ARC and the risk of developing AD. To this end, we genotyped 713 subjects diagnosed with AD and 841 controls without dementia. ARC was sequenced in a group of healthy individuals, and seven previously known SNPs and three novel SNPs were identified. Two of the newly found SNPs were intronic and one, +2852(G/A), was located in the 3'UTR. Three tag SNPs were selected, including the novel SNP +2852(G/A), to relate to risk of AD, Mini Mental State Examination (MMSE) scores and cerebrospinal fluid (CSF) biomarker levels of total tau (T-tau), hyperphosphorylated tau181 (P-tau(181)) and A beta(1-42). The AA genotype of the newly found 3'-UTR SNP +2852(A/G), was associated with a decreased risk of AD (p (c) = 0.005; OR = 0.74; 95 % CI: 0.61-0.89). No associations of single SNPs or haplotypes with MMSE score or CSF biomarkers were found. Here we report a novel ARC SNP associated with a reduced risk of developing AD. To our knowledge, this is the first study associating a gene variant of ARC with any disease. The location of the SNP within the 3'UTR indicates that dendritic targeting of ARC mRNA could be involved in the molecular mechanisms underlying this protective function. However, further investigation of the importance of this SNP for ARC function, ARC processing and the pathology of AD is needed.

  • No Association of VEGF Polymorphims with Alzheimer's Disease

    • The vascular hypothesis of Alzheimer's disease (AD) has brought the vascular endothelial growth factor (VEGF) into focus. The genomic region including the VEGF gene has been linked to AD and single nucleotide polymorphisms (SNPs) of the VEGF have in previous studies been associated with AD risk. To further evaluate these findings, we genotyped two SNPs in the VEGF gene (rs699947 [-2578]) and rs1570360 [-1154]) by TaqMan Allelic Discrimination in a study sample including AD patients (n = 801) and controls (n = 286). In a subgroup of the population these SNPs were analyzed in relation to APOE epsilon 4 genotype, to cerebrospinal fluid biomarkers (T-tau, P-tau, and beta(42)-Amyloid) as well as to neuropathological markers for AD (neurofibrillary tangles and senile plaques). No significant associations with risk for AD or any of the studied biomarkers could be found in this study, thus not supporting VEGF as being a major risk gene for AD.
