  • Not All Lectins Are Equally Suitable for Labeling Rodent Vasculature

    • The vascular system is vital for all tissues and the interest in its visualization spans many fields. A number of different plant-derived lectins are used for detection of vasculature; however, studies performing direct comparison of the labeling efficacy of different lectins and techniques are lacking. In this study, we compared the labeling efficacy of three lectins: Griffonia simplicifolia isolectin B4 (IB4); wheat germ agglutinin (WGA), and Lycopersicon esculentum agglutinin (LEA). The LEA lectin was identified as being far superior to the IB4 and WGA lectins in histological labeling of blood vessels in brain sections. A similar signal-to-noise ratio was achieved with high concentrations of the WGA lectin injected during intracardial perfusion. Lectins were also suitable for labeling vasculature in other tissues, including spinal cord, dura mater, heart, skeletal muscle, kidney, and liver tissues. In uninjured tissues, the LEA lectin was as accurate as the Tie2-eGFP reporter mice and GLUT-1 immunohistochemistry for labeling the cerebral vasculature, validating its specificity and sensitivity. However, in pathological situations, e.g., in stroke, the sensitivity of the LEA lectin decreases dramatically, limiting its applicability in such studies. This work can be used for selecting the type of lectin and labeling method for various tissues.

  • Atrial fibrillation as a clinical characteristic of arrhythmogenic right ventricular cardiomyopathy : Experience from the Nordic ARVC Registry

    • Background: Recent studies in arrhythmogenic right ventricular cardiomyopathy (ARVC) patients have drawn attention to atrial fibrillation (AF) as an arrhythmic manifestation of ARVC and as an indicator of atrial involvement in the disease progression. We aimed to assess the prevalence of AF in the Scandinavian cohort of ARVC patients and to evaluate its association with disease clinical manifestations. Methods: Study sample comprised of 293 definite ARVC patients by 2010 Task Force criteria (TFC2010) and 141 genotype-positive family members (total n = 434, 43% females, median age at ARVC diagnosis 41 years [interquartile range (IQR) 28–52 years]). ARVC diagnostic score was calculated as the sum of major (2 points) and minor (1 point) criteria in all categories of the TFC2010. Results: AF was diagnosed in 42 patients (10%): in 41 patients with definite ARVC diagnosis (14%) vs in one genotype-positive family member (1%), p < 0.001. The median age at AF onset was 51 (IQR 38–58) years. The prevalence of AF was related to the ARVC diagnostic score: it significantly increased starting with the diagnostic score 4 (2% in those with score 3 vs 13% in those with score 4, p = 0.023) and increased further with increased diagnostic score (Somer's d value is 0.074, p < 0.001). Conclusion: AF is seen in 14% of definite ARVC patients and is related to the severity of disease phenotype thus suggesting AF being an arrhythmic manifestation of this cardiomyopathy indicating atrial myocardial involvement in the disease progression.

  • Evolution of P-wave indices during long-term follow-up as markers of atrial substrate progression in arrhythmogenic right ventricular cardiomyopathy

    • AIMS: Patients with arrhythmogenic right ventricular cardiomyopathy (ARVC) have increased prevalence of atrial arrhythmias indicating atrial involvement in the disease. We aimed to assess the long-term evolution of P-wave indices as electrocardiographic (ECG) markers of atrial substrate during ARVC progression.METHODS AND RESULTS: We included 100 patients with a definite ARVC diagnosis according to 2010 Task Force criteria [34% females, median age 41 (inter-quartile range 30-55) years]. All available sinus rhythm ECGs (n = 1504) were extracted from the regional electronic ECG databases and automatically processed using Glasgow algorithm. P-wave duration, P-wave area, P-wave frontal axis, and prevalence of abnormal P terminal force in lead V1 (aPTF-V1) were assessed and compared at ARVC diagnosis, 10 years before and up to 15 years after diagnosis.Prior to ARVC diagnosis, none of the P-wave indices differed significantly from the data at ARVC diagnosis. After ascertainment of ARVC diagnosis, P-wave area in lead V1 decreased from -1 to -30 µV ms at 5 years (P = 0.002). P-wave area in lead V2 decreased from 82 µV ms at ARVC diagnosis to 42 µV ms 10 years after ARVC diagnosis (P = 0.006). The prevalence of aPTF-V1 increased from 5% at ARVC diagnosis to 18% by the 15th year of follow-up (P = 0.004). P-wave duration and frontal axis did not change during disease progression.CONCLUSION: Initial ARVC progression was associated with P-wave flattening in right precordial leads and in later disease stages an increased prevalence of aPTF-V1 was seen.

    • Background: P-wave indices reflect atrial abnormalities contributing to atrial fibrillation (AF). We aimed to assess a comprehensive set of P-wave characteristics for prediction of incident AF in a population-based setting. Methods: Malmö Preventative Project (MPP) participants were reexamined in 2002–2006 with electrocardiographic (ECG) and echocardiographic examinations and followed for 5 years. AF-free subjects (n = 983, age 70 ± 5 years, 38% females) with sinus rhythm ECGs were included in the study. ECGs were digitally processed using the Glasgow algorithm. P-wave duration, axis, dispersion, P-terminal force in lead V1 and interatrial block (IAB) were evaluated. ECG risk score combining the morphology, voltage and length of P-wave (MVP score) was calculated. New-onset diagnoses of AF were obtained from nation-wide registers. Results: During follow up, 66 patients (7%) developed AF. After adjustment for age and gender, the independent predictors of AF were abnormal P-wave axis > 75° (HR 1.63 CI95% 1.95–11.03) and MVP score 4 (HR 6.17 CI 95% 1.76–21.64), both correlated with LA area: Person r − 0.146, p < 0.001 and 0.192, p < 0.001 respectively. Advanced IAB (aIAB) with biphasic P-wave morphology in leads III and aVF was the most prevalent variant of aIAB and predicted AF in a univariate model (HR 2.59 CI 95% 1.02–6.58). Conclusion: P-wave frontal axis and MVP score are ECG-based AF predictors in the population-based cohort. Our study provides estimates for prevalence and prognostic importance of different variants of aIAB, providing a support to use biphasic P-wave morphology in lead aVF as the basis for aIAB definition.

  • A framework for exploring blockchain technology in supply chain management

    • The purpose of the study is to identify and explore different applications of blockchain technology in supply chain management and suggest a framework for their analysis. The study addresses researchers' conflicting opinions regarding the hype of blockchain and clarifies which minor applications are hyped and which are currently more applicable. That helps supply chain practitioners appraise the different applications of blockchains before it is too late to revise a decision regarding their use. The framework was developed based on a systematic literature review of blockchain-related articles, and it was examined with semi-structured interviews with four companies that applied the technology. Findings from the study suggested that traceability and supply chain finance seemed to be the most applicable applications of blockchain technology in supply chain management. But, light was also shed on other applications such as: compliance with standards, supply chain integration and digitalization of the transactions. This study was restricted by the availability of applied cases, as blockchain remains an emerging technology. Therefore, this study cannot be considered a case research, but a literature review followed by a limited number of interviews. However, the study resulted in a conceptual framework that forms the basis for further exploratory studies with more empirical data.

    • Applying blockchain technology for information sharing in supply chain is driven by many factors, but developing trust is one of the most proposed. However, trust is a multidimensional, intangible concept without an agreed-upon definition. Whereas some argue that trust is the main driver of blockchain technology, others have found a negative relationship. This study focuses on how applying blockchain in supply chain management can influence trust and proposes a corresponding research agenda. Trust and blockchain technology discussions are scattered throughout the literature. Thus, a systematic literature review was performed based on a conceptual trust framework. This study discovered a gap in linking trust theories to blockchain technology applications especially in supply chain management, and provided insights into trust’s reciprocal nature. Current literature strongly expects trust as a consequence for blockchain adoption if considered in the technology. Simultaneously, trust in supply chain partners is strongly expected as an antecedent to blockchain because it requires openness in information sharing. Thus, propositions and agenda for future research are suggested. The research is limited to literature findings due to the immaturity and low scalability of blockchain technology adoption; however, the most reviewed articles in less than two years old, increase the results’ accuracy.

  • Comparison of Magnetic Resonance Angiography and Digital Subtraction Angiography for the Assessment of Infrapopliteal Arterial Occlusive Lesions, Based on the TASC II Classification Criteria

    • This paper aimed to study the agreement and repeatability, both intra- and interobserver, of infrapopliteal lesion assessment with magnetic resonance angiography (MRA), using the TransAtlantic Inter-Society Consensus (TASC) II criteria, with perioperative digital subtraction angiography (DSA) as a reference. Sixty-eight patients with an MRA preceding an endovascular infrapopliteal revascularization were included. Preoperative MRAs and perioperative DSAs were evaluated in random order by three independent observers using the TASC II classification. The results were analyzed using visual grading characteristics (VGC) analysis and Krippendorffs alpha. No systematic difference was found between modalities: area under the VGC curve (AUC(VGC)) = 0.48 (p = 0.58) or intraobserver; AUC(VGC) for Observer 1 and 2 respectively, 0.49 (p = 0.85) and 0.53 (p = 0.52) for MRA compared with 0.54 (p = 0.30) and 0.49 (p = 0.81) for DSA. Interobserver differences were seen: AUC(VGC) of 0.63 (p < 0.01) for DSA and 0.80 (p < 0.01) for MRA. These results were confirmed using Krippendorffs alpha for the three observers showing 0.13 (95% confidence interval (CI) -0.07-0.31) for MRA and 0.39 (95% CI 0.23-0.53) for DSA. Poor interobserver agreement was also found in the choice of a target vessel on preoperative MRA: Krippendorffs alpha = 0.19 (95% CI 0.01-0.36). In conclusion, infrapopliteal lesions can be reliably determined on preoperative MRA, but interobserver variability regarding the choice of a target vessel is a major concern that appears to affect the overall TASC II grade.

  • Sponge Takeover from End-Permian Mass Extinction to Early Induan Time : Records in Central Iran Microbial Buildups

    • The end-Permian mass extinction was the most severe biotic crisis in Earth’s history. In its direct aftermath, microbial communities were abundant on shallow-marine shelves around the Tethys. They colonized the space left vacant after the dramatic decline of skeletal metazoans. The presence of sponges and sponge microbial bioherms has largely gone unnoticed due to the sponges’ size and the cryptic method of preservation. In addition to sponge dominated facies recently described in South Armenia and Northwestern Iran, we describe here sponge-microbial bioherms cropping out in two well-known Permian-Triassic boundary localities: the Kuh-e Hambast section, south-east of Abadeh city and the more distal Shahreza section, near Isfahan. In both sections, the extinction horizon is located at the top of an upper Changhsingian ammonoid-rich nodular limestone, called Paratirolites limestone. At Kuh-e Hambast, the overlying decimetric thick shale deposit called “boundary clay,” the latest Permian in age, is conformably overlain by well-dated transgressive basal Triassic platy limestone containing four successive levels of decimeter to meter scale, elongated to form cup-shaped mounds made of branching columnar stromatolites. Sponge fibers from possibly keratose demosponge, are widely present in the lime mudstone matrix. At the Shahreza section, above the extinction level, the boundary clay is much thicker (3 m), with thin platy limestone intervals, and contains two main levels of decimeter to meter scale mounds of digitate microbialite crossing the Permian-Triassic boundary with similar sponge fibers. Three levels rich in thrombolite domes can be seen in the overlying 20 m platy limestone of earliest Triassic age. Sponge fibers and rare spicules are present in their micritic matrix. These sponge fibers and spicules which are abundant in the latest Permian post-extinction boundary clay, followed microbial buildups during the Griesbachian time.

  • The Brittle-Ductile Transition in Porous Limestone : Failure Mode, Constitutive Modeling of Inelastic Deformation and Strain Localization

    • Understanding of the mechanics of the brittle-ductile transition (BDT) in porous limestone is significantly more challenging than for sandstone because of the lack of consistent acoustic emission activity in limestone, meaning that one must rely on alternative techniques. In this paper, we investigate systematically the failure modes in Indiana limestone using X-ray microComputed Tomography imaging (μCT) and Digital Volume Correlation (DVC). Our new mechanical data show that the envelope for the onset of shear-enhanced compaction can be well approximated by an elliptical cap. The DVC analysis revealed the development of shear bands through the BDT, but no evidence of compaction bands. The shear band angles were between 29° and 46° with respect to the maximum principal stress. Compiling these new results with published data on Purbeck and Leitha limestones, we showed that inelastic compaction in each of these dual porosity allochemical limestones was in a good agreement with the normality condition, as defined in plasticity theory. Comparison of the observed failure modes with predictions based on bifurcation analysis showed that the shear band angles are consistently smaller than the theoretical predictions.
