    • The Atlantic cod (Gadus morhua) is economically one of the world's most important marine species-a species presently suffering from heavy overexploitation throughout its range of distribution. Although not fully understood, the Atlantic cod is believed to be structured into populations in a rather complex manner, whereby both highly migratory and more confined ocean-spawning stocks coexist with stationary coastal populations. Owing to the complex population structure, little is presently known about how overexploitation of offshore stocks may affect other segments of the species. Here, we use microsatellite DNA analyses of coastal and offshore cod in combination with oceanographic modelling to investigate the population structure of Atlantic cod in the North Sea-Skagerrak area and evaluate the potential for larval transport into coastal populations. Our results suggest an extensive but temporally variable drift of offshore cod larvae into coastal populations. In a year (2001) with high inflow of North Sea waters into the Skagerrak we find that juvenile cod caught along the Skagerrak coast are predominantly of North Sea origin, whereas in a year (2000) with low inflow juveniles appear to be of local origin. These findings indicate that offshore cod may influence coastal cod populations over large distances.

  • Autochthonous versus allochthonous carbon sources of bacteria: Results from whole-lake C-13 addition experiments

    • Organic substrates for pelagic bacteria are derived from dissolved organic carbon (DOC) in the water column. DOC is a heterogeneous mixture of molecules, some of which are imported from the watershed (allochthonous DOC) and others that are produced by autotrophs within the system (autochthonous DOC). We examined the importance of autochthonous versus allochthonous DOC in supporting the growth of pelagic bacteria by manipulating the C-13 content of autochthonous sources in a whole-lake experiment. (NaHCO3)-C-13 was added daily to two small forested lakes for a period of 42 d, thereby strongly labeling autochthonous primary production. To obtain bacterial carbon isotopes, bacteria were regrown in vitro in particle-free lake water and in situ in dialysis tubes; little difference was found between the two methods. The contribution of autochthonous versus allochthonous carbon to the bacterial biomass was estimated by applying a two-member mixing model using a C-13 of -28parts per thousand as the allochthonous end member. The autochthonous end member, which varied over time, was estimated indirectly by several approaches. The bacterial biomass consisted of 35-70% allochthonous carbon. This result confirms the often-stated hypothesis that autochthonous carbon alone does not support bacterial production. On the other hand, autochthonous DOC was preferentially utilized relative to terrestrial DOC. On the basis of C-13 measurements, only 13% of the DOC standing stock was of recent autochthonous origin, but it supported 30-65% of bacterial production.

    • DNA microarrays can be used to identify gene expression changes characteristic of human disease. This is challenging, however, when relevant differences are subtle at the level of individual genes. We introduce an analytical strategy, Gene Set Enrichment Analysis, designed to detect modest but coordinate changes in the expression of groups of functionally related genes. Using this approach, we identify a set of genes involved in oxidative phosphorylation whose expression is coordinately decreased in human diabetic muscle. Expression of these genes is high at sites of insulin-mediated glucose disposal, activated by PGC-1alpha and correlated with total-body aerobic capacity. Our results associate this gene set with clinically important variation in human metabolism and illustrate the value of pathway relationships in the analysis of genomic profiling experiments.

    • Fibril formation of islet amyloid polypeptide (IAPP) is associated with cell death of the insulin-producing pancreatic beta-cells in patients with Type 2 Diabetes Mellitus. A likely cause for the cytotoxicity of human TAPP is that it destroys the barrier properties of the cell membrane. Here, we show by fluorescence confocal microscopy on lipid vesicles that the process of hIAPP amyloid formation is accompanied by a loss of barrier function, whereby lipids are extracted from the membrane and taken up in the forming amyloid deposits. No membrane interaction was observed when preformed fibrils were used. It is proposed that lipid uptake from the cell membrane is responsible for amyloid-induced membrane damage and that this represents a general mechanism underlying the cytotoxicity of amyloid forming proteins.     • PURPOSE: Previous studies have indicated that carcinoembryonic antigen (CEA) might be a suitable immunotherapeutic target in colorectal carcinoma (CRC). The aim of the present study was to analyze the immunological and clinical effects of vaccination with CEA together with the adjuvant granulocyte/macrophage colony-stimulating factor (GM-CSF).EXPERIMENTAL DESIGN: Twenty-four resected CRC patients without macroscopic disease were immunized seven times with recombinant CEA at four different dose levels over a 12-month period. Half of the patients received GM-CSF (80 microg/day for 4 consecutive days) at each immunization. Patients were monitored immunologically for 36 months and clinically for 76 months. T-cell response was evaluated by a [(3)H]thymidine incorporation assay, and IgG response was determined by ELISA.RESULTS: Minor local side effects were common. All 12 patients (100%) in the GM-CSF group developed a CEA-specific T-cell as well as an IgG response. The corresponding figures in the CEA alone group were 9 of 12 (75%) and 8 of 12 (66%), respectively. GM-CSF significantly augmented the amplitude of the T-cell response and the IgG titers. No dose-response relationship was noted. The immune responses at 12 months persisted 24 months after the last vaccination. Anti-CEA IgG titers were associated with increased survival (P < 0.05), whereas standard prognostic factors had no relationship, with the exception of serum CEA value.CONCLUSIONS: Vaccination with recombinant CEA and GM-CSF appears to be a nontoxic regimen inducing potent and durable antigen-specific IgG and T-cell response. The results of this study justify more extensive trials with recombinant CEA protein for immunotherapy of CRC.

    • Intraspecific brood parasites lay their eggs in the nests of conspecifics. There are a number of methods for detecting intraspecific brood parasitism (IBP) in birds based on egg morphology. Here we test Eadie's (1989) method, which calculates the Euclidean distances between eggs in a given clutch in a three-dimensional space (weight, length and width). A parasitised clutch is predicted to contain an egg (or eggs) that is significantly different from the clutch's other eggs. Data from three species were analysed. Our captive zebra finch Taeniopygia guttata clutches did not include any instances of IBP, the wild jackdaw Corvus monedula data were unlikely to contain any, and for the goldeneye Bucephala clangula data set we had an observational estimate of IBP. We simulated IBP in the zebra finch, jackdaw and goldeneye data to test whether the method reliably detects an experimentally 'parasitised' clutch. We show that the distributions of the test statistics greatly overlap in 'parasitised' and unmodified clutches, and are dependent on the clutch size. We therefore conclude that the method can only be used with caution, after calibrating it for a given population.
