    • An integrated stratigraphic, paleoecological, and geochronological study of lake and fen sediments just beyond the terminus of Berendon Glacier provides insights into late Holocene climate, vegetation, and glacier change in the northern Coast Mountains of British Columbia. Cores collected from two small lakes in the foreland of Berendon Glacier and pits dug in a nearby fen record Little lee Age and older glacier advances. The first Little Ice Age advance in this area began more than 500 years ago and peaked in the early 17th century. An earlier Neoglacial advance began about 2800-3000 cal yr ago and may have lasted for hundreds of years. There is also evidence for an intervening advance of even smaller magnitude around 1200-1300 cal yr ago. The advances are broadly synchronous with those in other parts of western North America, indicating that they were caused by regional, possibly global, changes in climate. Plant communities within the study area did not change dramatically during the late Holocene. The ranges of some plants, however, likely retracted or extended near treeline in response to changes in mean temperatures of perhaps 1-2degreesC, as A well as changes in summer snow cover. The greatest changes in vegetation occurred within and just beyond the forefields of Berendon, Frank Mackie, and other nearby glaciers. The largest climate shifts of the last 3000 years took place during the late Little Ice Age and the last century. Climate warmed about 1-2degreesC during the 20th century, accompanied by a rise in treeline, an increase in coniferous tree cover in the subalpine zone, and an increase in the temperature and biological productivity of ponds.   • Effects of mechanical ventilation on platelet microparticles in bronchoalveolar lavage fluid

    • INTRODUCTION: Mechanical ventilation (MV) is considered to contribute to lung injury. Platelet membrane-derived microparticles (PMPs) are procoagulant and participate in the inflammatory process. The bronchoalveolar space could, besides plasma, be a site of origin of these microparticles. We evaluated the presence of these PMPs and two prostaglandin-derived metabolites in bronchoalveolar lavage fluid (BALF) regarding their possible relation to MV.MATERIALS AND METHODS: Before and after 1 h of MV, PMPs and prostaglandin metabolites were analyzed, in BALF from 14 anesthetized pigs, by flow cytometry and RIA, respectively. Tracheal mucus from five humans was analyzed for PMPs at extubation after surgery.RESULTS: Activated PMPs and prostaglandin metabolites were present in all BALF samples. The time needed to count 5000 cellular events was prolonged six-fold after 1 h of mechanical ventilation (p<0.001). The relative content of PMPs was constant in all samples. The PMPs were thrombogenic, i.e. they were fibrinogen, p-selectin and von Willebrand factor positive. Lavage did not per se affect the period necessary to count 5000 cellular events. PMPs in human tracheal mucus were in the same range as in the pig after 1 h of MV aiming at a PaCO(2) between 5.0 and 5.5 kPa.CONCLUSIONS: Activated PMPs are present in the pulmonary air-liquid interface. The prolongation of the time needed to count 5000 cellular events in BALF after MV indicates activation and adherence. Adherent microparticles bind neutrophils, which may aggravate pathological processes leading to pulmonary dysfunction. Evaluation of PMPs in BALF may be useful in evaluating strategies for lung-protective ventilator treatment.

  • Mutated V-H genes and preferential V(H)3-21 use define new subsets of mantle cell lymphoma

    • Mantle cell lymphoma (MCL) is believed to originate from a naive B cell. However, we recently demonstrated that a subset of MCL displayed mutated V-H genes. We also reported restricted use of certain V-H genes. To assess the prognostic impact of these new findings, we performed V-H gene analysis of 110 patients, revealing that 18 (16%) patients had mutated and 92 (84%) patients had unmutated V-H genes. Because the mutation rate was low in the mutated group (2.2%-6.7%), further investigation of the germline V-H gene in T cells from 5 patients with mutated V-H genes was carried out; results showed that the unrearranged V-H gene was identical to the published sequence. These data confirm that the base pair substitutions within the rearranged V-H genes represent hyper-mutations, and indicate germinal center exposure. However, V-H gene mutation status did not correlate with prognosis because there was no difference in clinical outcome between the unmutated and mutated groups. The most frequently used V-H genes were V(H)3-21 (21 patients) and V(H)4-34 (19 patients). A novel finding was that V(H)3-21(+) MCL almost exclusively ex-pressed X light chains and displayed highly restricted use of the V(lambda)3-19 gene. V(H)3-21(+) patients had longer median survival than the remaining patients (53 vs 34 months; P = .03), but they tended to be younger at diagnosis. The combined use Of V(H)3-21/V(lambda)3-19 suggests a possible role for antigen(s) in the pathogenesis of these tumors and indicates that V(H)3-21(+) patients constitute a new MCL entity.
