    • Background Autoantibodies are found in up to 80% of patients with idiopathic inflammatory myopathies (IIM) and are associated with distinct clinical phenotypes [1]. Autoantibodies targeting cytosolic 5´-nucleotidase 1A (anti-cN1A) are currently the only known serum biomarker for the subgroup inclusion body myositis (IBM) (2), although detected even in other autoimmune diseases.Objectives To identify new autoimmune targets in IIM by antigen bead array assay.Methods In a first cross-sectional exploratory study, 357 antigens representing 268 proteins were incubated with plasma samples from 219 IIM (108 Polymyositis (PM), 80 Dermatomyositis (DM) and 31 IBM) patients, 349 Systemic Lupus Erythematosus (SLE) patients and 306 population controls for screening of IgG reactivity by antigen bead array. All samples were identified in the local biobank of the Rheumatology clinic, Karolinska University Hospital. Interesting results obtained for the IBM subgroup were then validated in an independent larger cohort of 287 patients with IBM followed at nine European rheumatological or neurological centers. IBM serum samples were explored by antigen bead array and results validated by western blot. As controls, serum samples from 30 patients with PM and 30 with DM, HLA-matched with the IBM Swedish cohort, were included. Demographics, laboratory, clinical, and muscle biopsy data of the IBM cohort was retrieved.Results In the exploratory study IgG reactivity towards NADH dehydrogenase 1 α subcomplex 11 (NDUFA11), a subunit of the membrane-bound mitochondrial respiratory chain complex I, was discovered with higher frequency in the IBM (9,7%) than PM (2,8%) and DM samples (2,5%), although the difference was not statistically significant. Anti-NDUFA11 IgG was also found in 2,3% of SLE and 2,6% of population control samples. In the validation study anti-NDUFA11 autoantibodies were detected in 11/287 IBM patients (3,8%), 0/30 PM and 0/30 DM patients. Reactivity against NDUFA11 could be confirmed by western blot (Table 1, Figure 1). The eleven anti-NDUFA11 positive patients showed a trend of lower frequency of wheelchair/walker ever use and higher creatine kinase levels at time of IBM diagnosis compared to the anti-NDUFA11 negative group. Ragged red fibers were significantly more prevalent in anti-NDUFA11 positive than negative patients (p=0.04). Anti-cN1A autoantibodies were detected in 98/287 (34,1%) of IBM, 3/30 (10%) DM and 9/29 (31%) PM patients, p=0.03. Coexistence of anti NDUFA11 and anti-cN1A antibodies was observed in 3 IBM patients.Conclusion Our results reveal a new autoimmune target in the mitochondrial respiratory chain complex I that might be specifically associated with IBM.   • Comparing emission- and absorption-based gas-phase metallicities in GRB host galaxies at z =2-4 using JWST

    • Much of what is known of the chemical composition of the universe is based on emission line spectra from star-forming galaxies. Emission-based inferences are, nevertheless, model-dependent and they are dominated by light from luminous star-forming regions. An alternative and sensitive probe of the metallicity of galaxies is through absorption lines imprinted on the luminous afterglow spectra of long gamma ray bursts (GRBs) from neutral material within their host galaxy. We present results from a JWST/NIRSpec programme to investigate for the first time the relation between the metallicity of neutral gas probed in absorption by GRB afterglows and the metallicity of the star-forming regions for the same host galaxy sample. Using an initial sample of eight GRB host galaxies at z = 2.1–4.7, we find a tight relation between absorption and emission line metallicities when using the recently proposed ?^ metallicity diagnostic (±0.2 dex). This agreement implies a relatively chemically homogeneous multiphase interstellar medium and indicates that absorption and emission line probes can be directly compared. However, the relation is less clear when using other diagnostics, such as R23 and R3. We also find possible evidence of an elevated N/O ratio in the host galaxy of GRB 090323 at z = 4.7, consistent with what has been seen in other z > 4 galaxies. Ultimate confirmation of an enhanced N/O ratio and of the relation between absorption and emission line metallicities will require a more direct determination of the emission line metallicity via the detection of temperature-sensitive auroral lines in our GRB host galaxy sample.

  • Pairing-quadrupole interplay in the neutron-deficient tin nuclei : First lifetime measurements of low-lying states in Sn-106,Sn-108

    • The lifetimes of the low-lying excited states 2(+) and 4(+) have been directly measured in the neutron-deficient Sn-106,Sn-108 isotopes. The nuclei were populated via a deep-inelastic reaction and the lifetime measurement was performed employing a differential plunger device. The emitted gamma rays were detected by the AGATA array, while the reaction products were uniquely identified by the VAMOS++ magnetic spectrometer. Large-Scale Shell-Model calculations with realistic forces indicate that, independently of the pairing content of the interaction, the quadrupole force is dominant in the B(E2; 2(1)(+) -> 0(g.s)(+)) values and it describes well the experimental pattern for Sn104-114 ; the B(E2;(+)(4) -> 2(1)(+)) values, measured here for the first time, depend critically on a delicate pairing-quadrupole balance, disclosed by the very precise results in Sn-108.

  • Evidence for octupole collectivity in 172Pt

    • Excited states in the extremely neutron-deficient nucleus 172Pt were populated via 96Ru(78Kr,2p) and 92Mo(83Kr,3n) reactions. The level scheme has been extended up to an excitation energy of  ~ 5 MeV and tentative spin-parity assignments up to Iπ = 18+. Linear polarization and angular distribution measurements were used to determine the electromagnetic E1 character of the dipole transitions connecting the positive-parity ground-state band with an excited side-band, firmly establishing it as a negative-parity band. The lowest member of this negative-parity structure was firmly assigned spin-parity 3-. In addition, we observed an E3 transition from this 3- state to the ground state, providing direct evidence for octupole collectivity in 172Pt. Large-scale shell model (LSSM) and total Routhian surface (TRS) calculations have been performed, supporting the interpretation of the 3- state as a collective octupole-vibrational state.

  • Lifetimes of core-excited states in semi-magic Rh-95

    • Lifetimes of negative-parity states have been determined in the neutron deficient semi-magic (N = 50) nucleus Rh-95. The fusion-evaporation reaction Ni-58(Ca-40, 3p) was used to populate high-spin states in Rh-95 at the Grand Accelerateur National d'Ions Lourds (GANIL) accelerator facility. The results were obtained using the Doppler Shift Attenuation Method (DSAM) based on the Doppler broadened line shapes produced during the slowing down process of the residual nuclei in a thick 6 mg/cm(2) metallic target. B(M1) and B(E2) reduced transition strengths are compared with predictions from large-scale shell-model calculations. state-of-the-art theory. Remarkably, the structural features up to moderate angular momentum of nuclei immediately below the N = Z = 50 shell closures can be described with high accuracy in a very simple way by shell-model calculations including only the g(9/2) and p(1/2) subshells. Of special interest is the neutron-proton pair coupling scheme which is expected to appear in the heaviest N=Z nuclei [1,2] and the seniority structure of the N = 50 isotones [3-7]. However, multiple core-excited states have been observed in the semi-magic nuclei of the Sn-100 region [8-10]. The theoretical study of those states is a challenging task, which requires a significantly larger model space for their interpretation. Transition probabilities between nuclear states provide important constraints for theoretical modelling of the structure of the nuclei of interest. Our previous lifetime study of the semimagic (N = 50) nucleus Ru-94 [ 11,12] provided information on the electromagnetic decay properties of neutron-core excited states. We now address lifetime measurements in its closest, more neutron deficient, isotone Rh-95 using the same DSAM technique. The experimental results have been interpreted within the framework of large-scale shell-model (LSSM) calculations.

    • Objective The aim of this study was to quantify the physiological workload of manual laborers in industrial sugarcane and assess the effect of receiving a rest, shade, and hydration intervention to reduce heat stress exposure risk.Methods In an observational study, physiological workload was evaluated for burned cane cutters (BCC), seed cutters (SC) and drip irrigation repair workers (DIRW) using heart rate (HR) recorded continuously (Polar (R)) across a work shift. Workers' percentage of maximal HR (%HRmax), time spent in different HR zones, and estimated core temperature (ECTemp) were calculated. The effect of increasing rest across two harvests was evaluated for BCC and SC.Results A total of 162 workers participated in this study [52 BCC (all male), 71 SC (13 female) and 39 DIRW (16 female)]. Average %HRmax across a work shift was similar between BCC and SC (BCC: 58%, SC: 59%), but lower in DIRW (51%). BCC and SC spent similar proportions of work shifts at hard/very hard intensities (BCC: 13%, SC: 15%), versus DIRW who worked mostly at light (46%) or light-moderate (39%) intensities. SC maximum ECTemp reached 38.2 degrees C, BCC 38.1 degrees C; while DIRW only reached 37.7 degrees C. Females performed at a higher %HRmax than males across work shifts (SC 64% versus 58%; DIRW 55% versus 49%). An additional rest period was associated with a lower average %HRmax across a work shift in BCC.Conclusion In this setting, BCC and SC both undertake very physiologically demanding work. Females maintained a higher workload than male co-workers. Regulated rest periods each hour, with water and shade access, appears to reduce physiological workload/strain.

  • Human genetic structure in Northwest France provides new insights into West European historical demography

    • The demographical history of France remains largely understudied despite its central role toward understanding modern population structure across Western Europe. Here, by exploring publicly available Europe-wide genotype datasets together with the genomes of 3234 present-day and six newly sequenced medieval individuals from Northern France, we found extensive fine-scale population structure across Brittany and the downstream Loire basin and increased population differentiation between the northern and southern sides of the river Loire, associated with higher proportions of steppe vs. Neolithic-related ancestry. We also found increased allele sharing between individuals from Western Brittany and those associated with the Bell Beaker complex. Our results emphasise the need for investigating local populations to better understand the distribution of rare (putatively deleterious) variants across space and the importance of common genetic legacy in understanding the sharing of disease-related alleles between Brittany and people from western Britain and Ireland.

  • In-situ temperatures and thermal properties of the East Siberian Arctic shelf sediments : Key input for understanding the dynamics of subsea permafrost

    • Significant reserves of methane (CH4) are held in the Arctic shelf, but the release of CH4 to the overlying ocean and, subsequently, to the atmosphere has been believed to be restricted by impermeable subsea permafrost, which has sealed the upper sediment layers for thousands of years. Our studies demonstrate progressive degradation of subsea permafrost which controls the scales of CH4 release from the sediment into the water-atmospheric system. Thus, new knowledge about the thermal state of subsea permafrost is crucial for better understanding of the permafrost -hydrate system and associated CH4 release from the East Siberian Arctic Shelf (ESAS) – the broadest and shallowest shelf in the World Ocean, which contains about 80% of subsea permafrost and giant pools of hydrates. Meanwhile, the ESAS, still presents large knowledge gaps in many aspects, especially with respect to subsea permafrost distribution and physical properties of bottom sediments. New field data show that the ESAS has an unfrozen (ice-free) upper sediment layer, which in-situ temperature is −1.0 to −1.8 °C and 0.6оС above the freezing point. On one hand, these cold temperature patterns may be related to the presence of subsea permafrost, which currently primarily occurs in the part of the ESAS that is shallower than 100 m, while ice-bearing sediments may also exist locally under deeper water in the Laptev Sea. On the other hand, the negative bottom sediment temperatures of −1.8 °C measured on the Laptev Sea continental slope sediments underlying water columns as deep as down to 330 m may result from dissociation of gas hydrates or possibly from dense water cascading down from the shelf. In contrast, data collected on recent expeditions in the northern Laptev shelf, zones of warmer bottom temperatures are coinciding with methane seeps, likely induced by seismic and tectonic activity in the area. These warm temperatures are not seen in the East Siberian Sea area, not even in areas of methane seeps, yet with little seismic activity.The thermal conductivity and heat capacity of bottom sediments recorded in the database of thermal parameters for the ESAS areas mainly depend on their lithification degree (density or porosity), moisture content, and particle size distribution. The thermal conductivity and heat capacity average about 1.0 W/(m·K) and 2900 kJ/(m3·K), with ±20% and ±10% variance, respectively, in all sampled Arctic sediments to a sub-bottom interval of 0–0.5 m.

    • We reconstructed Holocene paleoceanography of the Sherard Osborn Fjord (SOF), N Greenland, and Lincoln Sea in the eastern Arctic Ocean using sediment properties and micropaleontology from cores obtained during the Ryder 2019 Expedition. Our aims were to better understand faunal indicators of water mass influence on Ryder Glacier and the Lincoln Sea at water depths >500 m. Benthic microfaunal reflect glacio-marine interval during late deglaciation ~10.5 to 8.5 ka (kiloannum) during the Holocene Thermal Maximum (HTM) with dominant benthic foraminiferal species Cassidulina neoteretis, Cassidulina reniforme, and the ostracode Rabilimis mirabilis. Casssidulina neoteretis is considered an indicator of Atlantic Water (AW) throughout the Arctic Ocean and Nordic Seas; C. reniforme reflects glacio-marine conditions from the retreating Ryder Glacier. Deglaciation was followed by a period of elevated productivity and diverse ostracode faunal assemblages that suggest AW influence from 8.5 to 6 ka in the Lincoln Sea and inside SOF. The Holocene occurrence of the ostracode species Acetabulastoma arcticum, that appears in low numbers in the Lincoln Sea and briefly (~ 4–3 ka) in SOF, reflects the presence of variable sea ice in this region. Based on the similarities of the Lincoln Sea and fjord ostracodes to modern and glacial-deglacial faunas from the central Arctic Ocean, the AW influence likely originates from recirculation of AW water from the central Arctic Basin. In general, our results suggest a strong but temporally varying influence of AW during the entire 10.5 kyr record of the Lincoln Sea and SOF.

  • Evolution of cytokine production capacity in ancient and modern European populations

    • As our ancestors migrated throughout different continents, natural selection increased the presence of alleles advantageous in the new environments. Heritable variations that alter the susceptibility to diseases vary with the historical period, the virulence of the infections, and their geographical spread. In this study we built polygenic scores for heritable traits that influence the genetic adaptation in the production of cytokines and immune-mediated disorders, including infectious, inflammatory, and autoimmune diseases, and applied them to the genomes of several ancient European populations. We observed that the advent of the Neolithic was a turning point for immune-mediated traits in Europeans, favoring those alleles linked with the development of tolerance against intracellular pathogens and promoting inflammatory responses against extracellular microbes. These evolutionary patterns are also associated with an increased presence of traits related to inflammatory and auto-immune diseases.

    • We have developed an efficient approach to generate functional induced dopaminergic (DA) neurons from adult human dermal fibroblasts. When performing DA neuronal conversion of patient fibroblasts with idiopathic Parkinson's disease (PD), we could specifically detect disease-relevant pathology in these cells. We show that the patient-derived neurons maintain age-related properties of the donor and exhibit lower basal chaperone-mediated autophagy compared with healthy donors. Furthermore, stress-induced autophagy resulted in an age-dependent accumulation of macroautophagic structures. Finally, we show that these impairments in patient-derived DA neurons leads to an accumulation of phosphorylated alpha-synuclein, the classical hallmark of PD pathology. This pathological phenotype is absent in neurons generated from induced pluripotent stem cells from the same patients. Taken together, our results show that direct neural reprogramming can be used for obtaining patient-derived DA neurons, which uniquely function as a cellular model to study age-related pathology relevant to idiopathic PD.

    • Permanent ponds are valuable freshwater systems and biodiversity hotspots. They provide diverse ecosystem services (ESs), including water quality improvement and supply, food provisioning, and biodiversity support, despite significant pressure from multiple anthropogenic stressors and the impacts of ongoing global change. However, ponds are largely overlooked in management plans and legislation, and ecological research has focused on large freshwater ecosystems, such as rivers or lakes. Protection of ponds is often insufficient or indirectly provided via associated habitats such as wetlands. This situation is likely exacerbated by the lack of a full-scale understanding of the importance of ponds. In this review, we provide a detailed overview of permanent ponds across Europe, including their usages and the biodiversity they support. By discussing the concepts of pondscape and metacommunity theory, we highlight the importance of connectivity among and between ponds and identified fluxes of emerging insects as another ES of ponds. Those insects are rich in essential nutrients such as polyunsaturated fatty acids (PUFAs), delivered through them to the terrestrial environment; however, the extent and impact of this ES remains largely unexplored. Several potential stressors, especially related to ongoing global change, that influence pond diversity and integrity are discussed. We provide our insights on future pond management. Adaptive measures, taking into account the pond system per se within the pondscape, are the most promising to mitigate the loss of natural ponds and restore and conserve natural small waterbodies as refuges and diversity hotspots in increasingly urbanized landscapes.

  • Public Health Measures to Address the Impact of Climate Change on Population Health-Proceedings from a Stakeholder Workshop

    • Background: The World Health Organization identified climate change as the 21st century's biggest health threat. This study aimed to identify the current knowledge base, evidence gaps, and implications for climate action and health policymaking to address the health impact of climate change, including in the most underserved groups. Methods: The Horizon-funded project ENBEL ('Enhancing Belmont Research Action to support EU policy making on climate change and health') organised a workshop at the 2021-European Public Health conference. Following presentations of mitigation and adaptation strategies, seven international researchers and public health experts participated in a panel discussion linking climate change and health. Two researchers transcribed and thematically analysed the panel discussion recording. Results: Four themes were identified: (1) 'Evidence is key' in leading the climate debate, (2) the need for 'messaging about health for policymaking and behaviour change' including health co-benefits of climate action, (3) existing 'inequalities between and within countries', and (4) 'insufficient resources and funding' to implement national health adaptation plans and facilitate evidence generation and climate action, particularly in vulnerable populations. Conclusion: More capacity is needed to monitor health effects and inequities, evaluate adaptation and mitigation interventions, address current under-representations of low- or middle-income countries, and translate research into effective policymaking.

  • Darwinian Selection Discriminates Young Athletes: the Relative Age Effect in Relation to Sporting Performance

    • Background: The relative age effect (RAE) is a worldwide phenomenon, allowing sport participation and elite selection to be based on birthdate distribution. Negative consequences include both a narrow, non-optimal elite selection and negative health effects on entire populations. This study investigated the RAE and athletic performance in multiple individual sports in Sweden. Methods: Birthdates of athletes born between the years 1922 and 2015 were collected across 4-month periods (tertiles: T1, T2, T3) from cross-country skiing (N = 136,387), orienteering (N = 41,164), athletics (N = 14,503), alpine skiing (N = 508), E-sports (N = 47,030), and chess (N = 4889). In total, data from 244,560 athletes (women: N = 79,807, men: N = 164,753) was compared to the complete parent population of 5,390,954 births in Sweden during the same years. Chi-squared statistics compared parent and cohort distributions stratified by sport, sex, and age. Results: A significantly skewed distribution of birthdates was present in all sports, both sexes, and most age groups. The largest RAEs are seen in children where T1 often constitutes 40–50% and T3, 20–25% of the population. In E-sports, an inversed RAE was seen in adults. In most investigated sports, birthdate distribution was correlated to performance in children but not in adults. Conclusions: Skewed birthdate distributions were consistently prevalent in all investigated individual sports in Sweden, both physically demanding and cognitive/skill-based. As sport participation is related to total level of physical activity, both present and future, failing to address the RAE issue at an early age will result not only in a narrow and arbitrary selection for adult elite athletes but also in a negative impact on public health.

    • Resistance exercise has been shown to induce an acute hormonal response. The purpose of this study was to examine upper-body adaptations and the endocrine response to strength training in men and women when subjected to two different types of lower-body resistance training protocols. Nine males and eight females were assigned to either a Heavy Load (HL) (N = 10) or Mixed Load (ML) (N = 7) training routine three times per week for ten weeks. The HL-group executed low-volume, high-load resistance exercise for both lower and upper-body (4-6 reps at 80-90% of one repetition maximum (1-RM), three-minute inter-set rest). The ML-group performed the HL-protocol for the upper-body, but a high-volume, moderate-load protocol for the lower body (10-15 reps at 60-70% of 1-RM, one-minute inter-set rest). Volume load, 1-RM strength and hormonal measurements were analyzed by repeated-measures linear mixed models. Both groups increased their 1-RM in all assessments (p < 0.01) with no significant difference between groups at any time. Growth hormone (GH), testosterone and bioavailable testosterone (T/SHBG) increased in both groups during a standardized exercise session (p < 0.01) with ML having a greater increase in GH. The notion that acute elevations in anabolic hormones is important for muscle strength adaptation cannot be supported by the present study.

    • The time dependence of ice-nucleating particle (INP) activity is known to exist, yet for simplicity it is often omitted in atmospheric models as an approximation. Hitherto, only limited experimental work has been done to quantify this time dependency, for which published data are especially scarce regarding ambient aerosol samples and longer timescales. In this study, the time dependence of INP activity is quantified experimentally for six ambient environmental samples. The experimental approach includes a series of hybrid experiments with alternating constant cooling and isothermal experiments using a recently developed cold-stage setup called the Lund University Cold-Stage (LUCS). This approach of observing ambient aerosol samples provides the optimum realism for representing their time dependence in any model. Six ambient aerosol samples were collected at a station in rural Sweden representing aerosol conditions likely influenced by various types of INPs: marine, mineral dust, continental pristine, continental-polluted, combustion-related and rural continental aerosol. Active INP concentrations were seen to be augmented by about 40% to 100% (or 70% to 200%), depending on the sample, over 2h (or 10h). Mineral dust and rural continental samples displayed the most time dependence. This degree of time dependence observed was comparable to, but weaker than, that seen in previous published works. A general tendency was observed for the natural timescale of the freezing to dilate increasingly with time. The fractional freezing rate was observed to decline steadily with the time since the start of isothermal conditions following a power law. A representation of time dependence for incorporation into schemes of heterogeneous ice nucleation that currently omit it is proposed. Our measurements are inconsistent with the simplest purely stochastic model of INP activity, which assumes that the fractional freezing rate of all unfrozen drops is somehow constant and would eventually overpredict active INPs. In reality, the variability of efficiencies among INPs must be treated with any stochastic theory.

    • The coronavirus disease 2019 (COVID-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has unmasked mankind's vulnerability to biological threats. Although higher age is a major risk factor for disease severity in COVID-19, several predisposing risk factors for mortality are related to low cardiorespiratory and metabolic fitness, including obesity, cardiovascular disease, diabetes, and hypertension. Reaching physical activity (PA) guideline goals contribute to protect against numerous immune and inflammatory disorders, in addition to multi-morbidities and mortality. Elevated levels of cardiorespiratory fitness, being non-obese, and regular PA improves immunological function, mitigating sustained low-grade systemic inflammation and age-related deterioration of the immune system, or immunosenescence. Regular PA and being non-obese also improve the antibody response to vaccination. In this review, we highlight potential physiological, cellular, and molecular mechanisms that are affected by regular PA, increase the host antiviral defense, and may determine the course and outcome of COVID-19. Not only are the immune system and regular PA in relation to COVID-19 discussed, but also the cardiovascular, respiratory, renal, and hormonal systems, as well as skeletal muscle, epigenetics, and mitochondrial function.

    • Bathymetry (seafloor depth), is a critical parameter providing the geospatial context for a multitude of marine scientific studies. Since 1997, the International Bathymetric Chart of the Arctic Ocean (IBCAO) has been the authoritative source of bathymetry for the Arctic Ocean. IBCAO has merged its efforts with the Nippon Foundation-GEBCO-Seabed 2030 Project, with the goal of mapping all of the oceans by 2030. Here we present the latest version (IBCAO Ver. 4.0), with more than twice the resolution (200 x 200m versus 500 x 500m) and with individual depth soundings constraining three times more area of the Arctic Ocean (similar to 19.8% versus 6.7%), than the previous IBCAO Ver. 3.0 released in 2012. Modern multibeam bathymetry comprises similar to 14.3% in Ver. 4.0 compared to similar to 5.4% in Ver. 3.0. Thus, the new IBCAO Ver. 4.0 has substantially more seafloor morphological information that offers new insights into a range of submarine features and processes; for example, the improved portrayal of Greenland fjords better serves predictive modelling of the fate of the Greenland Ice Sheet.     • PURPOSE: To evaluate vertical acceleration, vector magnitude, non-wear time, valid day classifications and valid period classifications, in the data processing phase when using the ActiGraph GT3X accelerometer in non-ambulant children and adolescents with cerebral palsy (CP).MATERIAL AND METHODS: Accelerometer data retrieved from 33 non-ambulant children and adolescents (4-17 years) with CP, were analysed. Comparisons of i) vertical acceleration versus vector magnitude, ii) two different non-wear times, iii) three different settings to classify a day as valid and iv) two different settings to classify a period as valid were made.RESULTS AND CONCLUSIONS: Vector magnitude and a non-wear time of at least 90 consecutive minutes statistically significantly increased minutes recorded per day, especially for sedentary time. There was a statistically significant difference in numbers of valid days depending on time criteria set to determine a valid day, whereas there was no statistically significant difference in valid periods using 3 compared to 4 days. This study suggests using the pre-settings in ActiLife; vector magnitude, non-wear time of 90 consecutive minutes, 500 minutes recorded per day with periods of at least 3 valid days when assessing physical activity objectively by the ActiGraph GT3X accelerometer in non-ambulant children and adolescents with CP.     • In order to allow for a computationally efficient estimation of radar reflections, one commonly assumes the reflecting target to be in the far-field of the sensing array, such that the impinging wavefront is modeled as a linear phase-shift along the array. This works well in most radar scenarios, but causes significant performance degradation for close-range radar systems, wherein the curvature of the impinging wavefront may not be neglected. In this work, we examine how the used far-field assumption limits the resulting performance, illustrating how the (misspecified) Cramér-Rao lower bound (CRLB), taking the model mismatch into account, significantly devi-ates from the true CRLB for close-range targets. We further introduce a robust estimator that allows for the waveform cur-vature, showing that this computationally efficient estimator allows for superior performance for close-range reflectors.

    • Mass spectrometry (MS) and proteomics offer comprehensive characterization and identification of microorganisms and discovery of protein biomarkers that are applicable for diagnostics of infectious diseases. The use of biomarkers for diagnostics is widely applied in the clinic and the use of peptide biomarkers is increasingly being investigated for applications in the clinical laboratory. Respiratory-tract infections are a predominant cause for medical treatment, although, clinical assessments and standard clinical laboratory protocols are time-consuming and often inadequate for reliable diagnoses. Novel methods, preferably applied directly to clinical samples, excluding cultivation steps, are needed to improve diagnostics of infectious diseases, provide adequate treatment and reduce the use of antibiotics and associated development of antibiotic resistance. This study applied nano-liquid chromatography (LC) coupled with tandem MS, with a bioinformatics pipeline and an in-house database of curated high-quality reference genome sequences to identify species-unique peptides as potential biomarkers for four bacterial pathogens commonly found in respiratory tract infections (RTIs): Staphylococcus aureus; Moraxella catarrhalis; Haemophilus influenzae and Streptococcus pneumoniae. The species-unique peptides were initially identified in pure cultures of bacterial reference strains, reflecting the genomic variation in the four species and, furthermore, in clinical respiratory tract samples, without prior cultivation, elucidating proteins expressed in clinical conditions of infection. For each of the four bacterial pathogens, the peptide biomarker candidates most predominantly found in clinical samples, are presented. Data are available via ProteomeXchange with identifier PXD014522. As proof-of-principle, the most promising species-unique peptides were applied in targeted tandem MS-analyses of clinical samples and their relevance for identifications of the pathogens, i.e. proteotyping, was validated, thus demonstrating their potential as peptide biomarker candidates for diagnostics of infectious diseases.

    • The majority of long-duration (>2 s) gamma-ray bursts (GRBs) arise from the collapse of massive stars, with a small proportion created from the merger of compact objects. Most of these systems form via standard stellar evolution pathways. However, a fraction of GRBs may result from dynamical interactions in dense environments. These channels could also contribute substantially to the samples of compact object mergers detected as gravitational wave sources. Here we report the case of GRB 191019A, a long GRB (a duration of T90 = 64.4 ± 4.5 s), which we pinpoint close (⪅100 pc projected) to the nucleus of an ancient (>1 Gyr old) host galaxy at z = 0.248. The lack of evidence for star formation and deep limits on any supernova emission disfavour a massive star origin. The most likely route for progenitor formation is via dynamical interactions in the dense nucleus of the host. The progenitor, in this case, could be a compact object merger. These may form in dense nuclear clusters or originate in a gaseous disc around the supermassive black hole. Identifying, to the best of our knowledge, a first example of a dynamically produced GRB demonstrates the role that such bursts may have in probing dense environments and constraining dynamical fractions in gravitational wave populations.

    • Objectives Discussing existential issues is integral to caring for people with acute, progressive, or life-limiting neurological illness, but there is a lack of research examining how nurses approach existential issues with this patient group and their family members. The purpose was to examine the experiential impact of an educational program for nurses designed to facilitate discussions of existential issues with patients and family members in neurological wards. Method Nurses in inpatient and outpatient care at a neurological clinic in Sweden were invited to participate in an education program about discussing existential issues with patients and their family members as related to neurological conditions. The evaluation of the program and of the nurses' view of discussing existential issues was conducted through focus groups before and after participation. The data were analyzed by qualitative content analysis. Results The program gave nurses a deeper understanding of existential issues and how to manage these conversations with patients and their family members. Both internal and external barriers remained after education, with nurses experiencing insecurity and fear, and a sense of being inhibited by the environment. However, they were more aware of the barriers after the education, and it was easier to find strategies to manage the conversations. They demonstrated support for each other in the team both before and after participating in the program. Significance of results The educational program gave nurses strategies for discussing existential issues with patients and family members. The knowledge that internal and external barriers impede communication should compel organizations to work on making conditions more conducive, for example, by supporting nurses to learn strategies to more easily manage conversations about existential issues and by reviewing the physical environment and the context in which they are conducted.

    • Open Networked Learning (ONL) is a collaborative open online course for higher education educators worldwide, focusing on open education, collaborative learning, and course design. The interorganizational course welcomes both institutional and open learners from around 30 nations. Delivered 17 times since 2014 with up to 120 participants in each session, ONL employs problem-based learning to foster digital competencies and strengthen community development. Rooted in the European Framework for Digital Competence of Educators (DigCompEdu), a study involving 48 participants revealed statistically significant, across-the-board competence enhancement post-course, affirming ONL’s efficacy. The article also summarizes our experiences and learnings from participating in and organising the course over the past ten years.

    • Low-lying states in the odd-Z isotopes Ac13289221 and Pa13491225 have been studied using α-particle and αγ-coincidence spectroscopy in the Pa225→Ac221→Fr217 decay chain. Ground-state spin and parity assignments of Iπ = 5/2- are proposed for both Ac221 and Pa225, with the odd proton occupying the ω = 5/2 orbital of the quadrupole-octupole deformed shell model in both nuclei. In Ac221, excited states in the bands based on the ω = 5/2 and ω = 3/2 orbitals have been identified, including proposed parity-doublet states. The results suggest that reflection-asymmetric deformation of the ground state persists in the odd-A members of the isotope chains down to N = 132 for Ac and N = 134 for Pa, before reaching the transitional region at N = 130.

    • Despite the progress to understand inflammatory reactions, mechanisms causing their resolution remain poorly understood. Prostanoids, especially prostaglandin E2 (PGE2), are well-characterized mediators of inflammation. PGE2 is produced in an inducible manner in macrophages (Mϕ) by microsomal PGE2-synthase-1 (mPGES-1), with the notion that it also conveys pro-resolving properties. We aimed to characterize the role of mPGES-1 during resolution of acute, zymosan-induced peritonitis. Experimentally, we applied the mPGES-1 inhibitor compound III (CIII) once the inflammatory response was established and confirmed its potent PGE2-blocking efficacy. mPGES-1 inhibition resulted in an incomplete removal of neutrophils and a concomitant increase in monocytes and Mϕ during the resolution process. The mRNA-seq analysis identified enhanced C-X3-C motif receptor 1 (CX3CR1) expression in resident and infiltrating Mϕ upon mPGES-1 inhibition. Besides elevated Cx3cr1 expression, its ligand CX3CL1 was enriched in the peritoneal lavage of the mice, produced by epithelial cells upon mPGES-1 inhibition. CX3CL1 not only increased adhesion and survival of Mϕ but its neutralization also completely reversed elevated inflammatory cell numbers, thereby normalizing the cellular, peritoneal composition during resolution. Our data suggest that mPGES-1-derived PGE2 contributes to the resolution of inflammation by preventing CX3CL1-mediated retention of activated myeloid cells at sites of injury.

    • Cyanobacterial blooms have substantial direct and indirect negative impacts on freshwater ecosystems including releasing toxins, blocking light needed by other organisms, and depleting oxygen. There is growing concern over the potential for climate change to promote cyanobacterial blooms, as the positive effects of increasing lake surface temperature on cyanobacterial growth are well documented in the literature; however, there is increasing evidence that cyanobacterial blooms are also being initiated and persisting in relatively cold-water temperatures (< 15 °C), including ice-covered conditions. In this work, we provide evidence of freshwater cold-water cyanobacterial blooms, review abiotic drivers and physiological adaptations leading to these blooms, offer a typology of these lesser-studied cold-water cyanobacterial blooms, and discuss their occurrence under changing climate conditions.

    • Marine heatwaves have been observed worldwide and are expected to increase in both frequency and intensity due to climate change. Such events may cause ecosystem reconfigurations arising from species range contraction or redistribution, with ecological, economic and social implications. Macrophytes such as the brown seaweed Fucus vesiculosus and the seagrass Zostera marina are foundation species in many coastal ecosystems of the temperate northern hemisphere. Hence, their response to extreme events can potentially determine the fate of associated ecosystems. Macrophyte functioning is intimately linked to the maintenance of photosynthesis, growth and reproduction, and resistance against pathogens, epibionts and grazers. We investigated morphological, physiological, pathological and chemical defence responses of western Baltic Sea F.vesiculosus and Z.marina populations to simulated near-natural marine heatwaves. Along with (a) the control, which constituted no heatwave but natural stochastic temperature variability (0HW), two treatments were applied: (b) two late-spring heatwaves (June, July) followed by a summer heatwave (August; 3HW) and (c) a summer heatwave only (1HW). The 3HW treatment was applied to test whether preconditioning events can modulate the potential sensitivity to the summer heatwave. Despite the variety of responses measured in both species, only Z.marina growth was impaired by the accumulative heat stress imposed by the 3HW treatment. Photosynthetic rate, however, remained high after the last heatwave indicating potential for recovery. Only epibacterial abundance was significantly affected in F.vesiculosus. Hence both macrophytes, and in particular F.vesiculosus, seem to be fairly tolerant to short-term marine heatwaves at least at the intensities applied in this experiment (up to 5°C above mean temperature over a period of 9days). This may partly be due to the fact that F.vesiculosus grows in a highly variable environment, and may have a high phenotypic plasticity. © 2019 John Wiley & Sons Ltd

    • Stutzerimonas balearica (Pseudomonas balearica) has been found principally in oil-polluted environments. The capability of S. balearica to thrive from the degradation of pollutant compounds makes it a species of interest for potential bioremediation applications. However, little has been reported about the diversity of S. balearica. In this study, genome sequences of S. balearica strains from different origins were analyzed, revealing that it is a diverse species with an open pan-genome that will continue revealing new genes and functionalities as the genomes of more strains are sequenced. The nucleotide signatures and intra- and inter-species variation of the 16S rRNA genes of S. balearica were reevaluated. A strategy of screening 16S rRNA gene sequences in public databases enabled the detection of 158 additional strains, of which only 23% were described as S. balearica. The species was detected from a wide range of environments, although mostly from aquatic and polluted environments, predominantly related to petroleum oil. Genomic and phenotypic analyses confirmed that S. balearica possesses varied inherent capabilities for aromatic compounds degradation. This study increases the knowledge of the biology and diversity of S. balearica and will serve as a basis for future work with the species.

  • Complete genome sequences of Streptococcus pyogenes type strain reveal 100%-match between PacBio-solo and Illumina-Oxford Nanopore hybrid assemblies

    • We present the first complete, closed genome sequences of Streptococcus pyogenes strains NCTC 8198(T) and CCUG 4207(T), the type strain of the type species of the genus Streptococcus and an important human pathogen that causes a wide range of infectious diseases. S. pyogenes NCTC 8198(T) and CCUG 4207(T) are derived from deposit of the same strain at two different culture collections. NCTC 8198(T) was sequenced, using a PacBio platform; the genome sequence was assembled de novo, using HGAP. CCUG 4207(T) was sequenced and a de novo hybrid assembly was generated, using SPAdes, combining Illumina and Oxford Nanopore sequence reads. Both strategies yielded closed genome sequences of 1,914,862 bp, identical in length and sequence identity. Combining short-read Illumina and long-read Oxford Nanopore sequence data circumvented the expected error rate of the nanopore sequencing technology, producing a genome sequence indistinguishable to the one determined with PacBio. Sequence analyses revealed five prophage regions, a CRISPR-Cas system, numerous virulence factors and no relevant antibiotic resistance genes. These two complete genome sequences of the type strain of S. pyogenes will effectively serve as valuable taxonomic and genomic references for infectious disease diagnostics, as well as references for future studies and applications within the genus Streptococcus.

    • Formation of specialized pro-resolving lipid mediators (SPMs) such as lipoxins or resolvins usually involves arachidonic acid 5-lipoxygenase (5-LO, ALOX5) and different types of arachidonic acid 12- and 15-lipoxygenating paralogues (15-LO1, ALOX15; 15-LO2, ALOX15B; 12-LO, ALOX12). Typically, SPMs are thought to be formed via consecutive steps of oxidation of polyenoic fatty acids such as arachidonic acid, eicosapentaenoic acid or docosahexaenoic acid. One hallmark of SPM formation is that reported levels of these lipid mediators are much lower than typical pro-inflammatory mediators including the monohydroxylated fatty acid derivatives (e.g., 5-HETE), leukotrienes or certain cyclooxygenase-derived prostaglandins. Thus, reliable detection and quantification of these metabolites is challenging. This paper is aimed at critically evaluating i) the proposed biosynthetic pathways of SPM formation, ii) the current knowledge on SPM receptors and their signaling cascades and iii) the analytical methods used to quantify these pro-resolving mediators in the context of their instability and their low concentrations. Based on current literature it can be concluded that i) there is at most, a low biosynthetic capacity for SPMs in human leukocytes. ii) The identity and the signaling of the proposed G-protein-coupled SPM receptors have not been supported by studies in knock-out mice and remain to be validated. iii) In humans, SPM levels were neither related to dietary supplementation with their omega-3 polyunsaturated fatty acid precursors nor were they formed during the resolution phase of an evoked inflammatory response. iv) The reported low SPM levels cannot be reliably quantified by means of the most commonly reported methodology. Overall, these questions regarding formation, signaling and occurrence of SPMs challenge their role as endogenous mediators of the resolution of inflammation.

    • Background: The heaviest T-z = 0 doubly-magic nucleus, Sn-100, and the neighboring nuclei offer unique opportunities to investigate the properties of nuclear interaction. For instance, the structure of light-Sn nuclei has been shown to be affected by the delicate balance between nuclear-interaction components, such as pairing and quadrupole correlations. From Cd to Te, many common features and phenomena have been observed experimentally along the isotopic chains, leading to theoretical studies devoted to a more general and comprehensive study of the region. In this context, having only two proton holes in the Z = 50 shell, the Cd isotopes are expected to present properties similar to those found in the Sn isotopic chain.Purpose: The aim of this work was to measure lifetimes of excited states in neutron-deficient nuclei in the vicinity of Sn-100.Methods: The neutron-deficient nuclei in the N approximate to Z approximate to 50 region were populated using a multinucleon transfer reaction with a Cd-106 beam and a Mo-92 target. The beamlike products were identified by the VAMOS++ spectrometer, while the gamma rays were detected using the AGATA array. Lifetimes of excited states were determined using the recoil distance Doppler-shift method, employing the Cologne differential plunger.Results: Lifetimes of low-lying states were measured in the even-mass Cd-102-(108) isotopes. In particular, multiple states with excitation energy up to MeV, belonging to various bands, were populated in approximate to 3 Cd-106 via inelastic scattering. The transition strengths corresponding to the measured lifetimes were compared with those resulting from state-of-the-art beyond-mean-field calculations using the symmetry-conserving configuration-mixing approach.Conclusions: Despite the similarities in the electromagnetic properties of the low-lying states, there is a fundamental structural difference between the ground-state bands in the Z = 48 and Z = 50 isotopes. The comparison between experimental and theoretical results revealed a rotational character of the Cd nuclei, which have prolate-deformed ground states with beta(2) approximate to 0.2. At this deformation Z = 48 becomes a closed-shell configuration, which is favored with respect to the spherical one.

    • Taste is essential for the interaction of animals with their food and has co-evolved with diet. Humans have peopled a large range of environments and present a wide range of diets, but little is known about the diversity and evolution of human taste perception. We measured taste recognition thresholds across populations differing in lifestyles (hunter gatherers and farmers from Central Africa, nomad herders, and farmers from Central Asia). We also generated genome-wide genotype data and performed association studies and selection scans in order to link the phenotypic variation in taste sensitivity with genetic variation. We found that hunter gatherers have lower overall sensitivity as well as lower sensitivity to quinine and fructose than their farming neighbors. In parallel, there is strong population divergence in genes associated with tongue morphogenesis and genes involved in the transduction pathway of taste signals in the African populations. We find signals of recent selection in bitter taste-receptor genes for all four populations. Enrichment analysis on association scans for the various tastes confirmed already documented associations and revealed novel GO terms that are good candidates for being involved in taste perception. Our framework permitted us to gain insight into the genetic basis of taste sensitivity variation across populations and lifestyles.

    • The 3.3 million km marine ecosystem around the North Pole, defined as the Central Arctic Ocean (CAO), is a blind spot on the map of the world's fish stocks. The CAO essentially comprises the permanently ice-covered deep basins and ridges outside the continental shelves, and is only accessible by ice-breakers. Traditional trawling for assessing fish stocks is impossible under the thick pack ice, and coherent hydroacoustic surveys are unachievable due to ice-breaking noise. Consequently, nothing is known about the existence of any pelagic fish stocks in the CAO, although juveniles of Boreogadus saida richly occur at the surface associated with the sea ice and ice-associated Arctogadus glacialis has been reported as well. We here present a first indication of a possible mesopelagic fish stock in the CAO. We had the opportunity to analyse a geophysical hydroacoustic data set with 13 time windows of usable acoustic data over a transect from 84.4 °N in the Nansen Basin, across the North Pole (90.0 °N), to 82.4 °N in the Canada Basin. We discovered a deep scattering layer (DSL), suggesting the presence of zooplankton and fish, at 300–600 m of depth in the Atlantic water layer of the CAO. Maximum possible fish abundance and biomass was very low; values of ca. 2,000 individuals km and ca. 50 kg km were calculated for the DSL in the North-Pole area according to a model assuming that all acoustic backscatter represents 15-cm long B. saida and/or A. glacialis. The true abundance and biomass of fish is even lower than this, but cannot be quantified from this dataset due to possible backscatter originating from pneumatophores of physonect siphonophores that are known to occur in the area. Further studies on the DSL of the CAO should include sampling and identification of the backscattering organisms. From our study we can conclude that if the central Arctic DSL contains fish, their biomass is currently too low for any sustainable fishery.
