    • Genetic discoveries of Alzheimer’s disease are the drivers of our understanding, and together with polygenetic risk stratification can contribute towards planning of feasible and efficient preventive and curative clinical trials. We first perform a large genetic association study by merging all available case-control datasets and by-proxy study results (discovery n = 409,435 and validation size n = 58,190). Here, we add six variants associated with Alzheimer’s disease risk (near APP, CHRNE, PRKD3/NDUFAF7, PLCG2 and two exonic variants in the SHARPIN gene). Assessment of the polygenic risk score and stratifying by APOE reveal a 4 to 5.5 years difference in median age at onset of Alzheimer’s disease patients in APOE ɛ4 carriers. Because of this study, the underlying mechanisms of APP can be studied to refine the amyloid cascade and the polygenic risk score provides a tool to select individuals at high risk of Alzheimer’s disease. © 2021, The Author(s).

    • Characterization of the genetic landscape of Alzheimer’s disease (AD) and related dementias (ADD) provides a unique opportunity for a better understanding of the associated pathophysiological processes. We performed a two-stage genome-wide association study totaling 111,326 clinically diagnosed/‘proxy’ AD cases and 677,663 controls. We found 75 risk loci, of which 42 were new at the time of analysis. Pathway enrichment analyses confirmed the involvement of amyloid/tau pathways and highlighted microglia implication. Gene prioritization in the new loci identified 31 genes that were suggestive of new genetically associated processes, including the tumor necrosis factor alpha pathway through the linear ubiquitin chain assembly complex. We also built a new genetic risk score associated with the risk of future AD/dementia or progression from mild cognitive impairment to AD/dementia. The improvement in prediction led to a 1.6- to 1.9-fold increase in AD risk from the lowest to the highest decile, in addition to effects of age and the APOE ε4 allele.

    • The next years will see the completion of the radioactive ion beam facility SPES (Selective Production of Exotic Species) and the upgrade of the accelerators complex at Istituto Nazionale di Fisica Nucleare – Legnaro National Laboratories (LNL) opening up new possibilities in the fields of nuclear structure, nuclear dynamics, nuclear astrophysics, and applications. The nuclear physics community has organised a workshop to discuss the new physics opportunities that will be possible in the near future by employing state-of-the-art detection systems. A detailed discussion of the outcome from the workshop is presented in this report.

    • Background: Severe acute respiratory syndrome coronavirus 2 (SARSCoV-2) belongs to a subgroup of coronaviruses rampant in bats for centuries. It caused the coronavirus disease 2019 (COVID-19) pandemic. Most patients recover, but a minority of severe cases experience acute respiratory distress or an inflammatory storm devastating many organs that can lead to patient death. The spread of SARS-CoV-2 was facilitated by the increasing intensity of air travel, urban congestion and human contact during the past decades. Until therapies and vaccines are available, tests for virus exposure, confinement and distancing measures have helped curb the pandemic. Vision: The COVID-19 pandemic calls for safeguards and remediation measures through a systemic response. Self-organizing initiatives by scientists and citizens are developing an advanced collective intelligence response to the coronavirus crisis. Their integration forms Olympiads of Solidarity and Health. Their ability to optimize our response to COVID-19 could serve as a model to trigger a global metamorphosis of our societies with far-reaching consequences for attacking fundamental challenges facing humanity in the 21st century. Mission: For COVID-19 and these other challenges, there is no alternative but action. Meeting in Paris in 2003, we set out to "rethink research to understand life and improve health." We have formed an international coalition of academia and industry ecosystems taking a systems medicine approach to understanding COVID-19 by thoroughly characterizing viruses, patients and populations during the pandemic, using openly shared tools. All results will be publicly available with no initial claims for intellectual property rights. This World Alliance for Health and Wellbeing will catalyze the creation of medical and health products such as diagnostic tests, drugs and vaccines that become common goods accessible to all, while seeking further alliances with civil society to bridge with socio-ecological and technological approaches that characterise urban systems, for a collective response to future health emergencies.

  • Towards a European health research and innovation cloud (HRIC)

    • The European Union (EU) initiative on the Digital Transformation of Health and Care (Digicare) aims to provide the conditions necessary for building a secure, flexible, and decentralized digital health infrastructure. Creating a European Health Research and Innovation Cloud (HRIC) within this environment should enable data sharing and analysis for health research across the EU, in compliance with data protection legislation while preserving the full trust of the participants. Such a HRIC should learn from and build on existing data infrastructures, integrate best practices, and focus on the concrete needs of the community in terms of technologies, governance, management, regulation, and ethics requirements. Here, we describe the vision and expected benefits of digital data sharing in health research activities and present a roadmap that fosters the opportunities while answering the challenges of implementing a HRIC. For this, we put forward five specific recommendations and action points to ensure that a European HRIC: i) is built on established standards and guidelines, providing cloud technologies through an open and decentralized infrastructure; ii) is developed and certified to the highest standards of interoperability and data security that can be trusted by all stakeholders; iii) is supported by a robust ethical and legal framework that is compliant with the EU General Data Protection Regulation (GDPR); iv) establishes a proper environment for the training of new generations of data and medical scientists; and v) stimulates research and innovation in transnational collaborations through public and private initiatives and partnerships funded by the EU through Horizon 2020 and Horizon Europe.

  • Evaluating Nature-Based Solutions for Water Management in Peri-Urban Areas

    • The term nature-based solutions (NBS) has gained traction in recent years and has been applied in many settings. There are few comprehensive assessment frameworks available that can guide NBS planning and implementation while at the same time capturing the short- and long-term impacts and benefits of the NBS. Here a recently presented framework, which builds on the theory of change and was developed to assess NBS at different phases of the project cycle, was applied to seven diverse case studies. The case studies addressed water quality and quantity issues in peri-urban areas across the global north and south. Framework indicators covering the sustainability dimensions (environmental, social and economic) were assessed at three stages of the framework: context, process and results. The work sought to investigate the following research objectives: (1) Can this framework be robust and yet flexible enough to be applied across a diverse selection of NBS projects that are at different phases of the project cycle and address different kinds of water challenges within varied ecological, social and economic contexts? (2) Is it possible to draw generalisations from a comparative analysis of the application of the framework to the case studies? Results showed that the framework was able to be applied to the case studies; however, their diversity showed that NBS projects designed in one context, for a specific purpose in a specific location, can not necessarily be transferred easily to another location. There were several process-based indicators that were universally significant for the case studies, including expertise, skills and knowledge of the involved actors, roles and responsibilities of involved actors and political support. The result-based indicators were case study-specific when environmental indicators were case study-specific, and important social indicators were environmental identity and recreational values. Overall, the use of the framework benefits the recognition of the implementation’s advances, such as the change in context, the processes in place and the results obtained.

    • Guinea pigs (Cavia spp.) have a long association with humans. From as early as 10,000 years ago they were a wild food source. Later, domesticated Cavia porcellus were dispersed well beyond their native range through pre-Columbian exchange networks and, more recently, widely across the globe. Here we present 46 complete mitogenomes of archaeological guinea pigs from sites in Peru, Bolivia, Colombia, the Caribbean, Belgium and the United States to elucidate their evolutionary history, origins and paths of dispersal. Our results indicate an independent centre of domestication of Cavia in the eastern Colombian Highlands. We identify a Peruvian origin for the initial introduction of domesticated guinea pigs (Cavia porcellus) beyond South America into the Caribbean. We also demonstrate that Peru was the probable source of the earliest known guinea pigs transported, as part of the exotic pet trade, to both Europe and the southeastern United States. Finally, we identify a modern reintroduction of guinea pigs to Puerto Rico, where local inhabitants use them for food. This research demonstrates that the natural and cultural history of guinea pigs is more complex than previously known and has implications for other studies regarding regional to global-scale studies of mammal domestication, translocation, and distribution.

  • The bar pattern speed of the Large Magellanic Cloud

    • Context. The internal kinematics of the Large Magellanic Cloud (LMC) have been studied in unprecedented depth thanks to the excellent quality of the Gaia mission data, thus revealing the disc’s non-axisymmetric structure. Aims. We seek to constrain the LMC bar pattern speed using the astrometric and spectroscopic data from the Gaia mission. Methods. We applied three methods to evaluate the bar pattern speed by measuring it via: the Tremaine-Weinberg (TW) method, the Dehnen method, and a bisymmetric velocity (BV) model. These methods provide additional information on the bar properties, such as the corotation radius as well as the bar length and strength. We tested the validity of the methods with numerical simulations. Results. A wide range of pattern speeds are inferred by the TW method, owing to a strong dependency on the orientation of the galaxy frame and the viewing angle of the bar perturbation. The simulated bar pattern speeds (corotation radii, respectively) are well recovered by the Dehnen method (BV model). Applied to the LMC data, the Dehnen method finds a pattern speed of Ωp = −1.0 ± 0.5 km s−1 kpc−1, thus corresponding to a bar that barely rotates and is only slightly counter-rotating with respect to the LMC disc. The BV method gives a LMC bar corotation radius of Rc = 4.20±0.25 kpc, corresponding to a pattern speed of Ωp = 18.5+−11.21 km s−1 kpc−1. Conclusions. It is not possible to determine which global value best represents an LMC bar pattern speed with the TW method, due to the strong variation with the orientation of the reference frame. The non-rotating bar from the Dehnen method would be at odds with the structure and kinematics of the LMC disc. The BV method result is consistent with previous estimates and gives a bar corotation-to-length ratio of 1.8 ± 0.1, suggesting that the LMC is hosting a slow bar.

    • Emerging photovoltaics (PVs) focus on a variety of applications complementing large scale electricity generation. Organic, dye-sensitized, and some perovskite solar cells are considered in building integration, greenhouses, wearable, and indoor applications, thereby motivating research on flexible, transparent, semitransparent, and multi-junction PVs. Nevertheless, it can be very time consuming to find or develop an up-to-date overview of the state-of-the-art performance for these systems and applications. Two important resources for recording research cells efficiencies are the National Renewable Energy Laboratory chart and the efficiency tables compiled biannually by Martin Green and colleagues. Both publications provide an effective coverage over the established technologies, bridging research and industry. An alternative approach is proposed here summarizing the best reports in the diverse research subjects for emerging PVs. Best performance parameters are provided as a function of the photovoltaic bandgap energy for each technology and application, and are put into perspective using, e.g., the Shockley–Queisser limit. In all cases, the reported data correspond to published and/or properly described certified results, with enough details provided for prospective data reproduction. Additionally, the stability test energy yield is included as an analysis parameter among state-of-the-art emerging PVs.

  • Measurement of Oxidative Stress Index in Seminal Plasma Can Predict In Vivo Fertility of Liquid-Stored Porcine Artificial Insemination Semen Doses

    • The study evaluated the relation between the oxidative stress index (OSI) in porcine seminal plasma (n = 76) with sperm resilience and in vivo fertility (farrowing rate and litter size of 3137 inseminated sows) of liquid-stored artificial insemination (AI) semen doses. The OSI was assessed as the ratio of advanced oxidation protein products to Trolox-equivalent antioxidant capacity, both measured using an automated analyzer. Sperm motility (computer-assisted sperm analyzer) and viability (flow cytometry) were evaluated in semen AI-doses at 0 and 72 h of storage at 17 degrees C. Sperm resilience was defined as the difference between storage intervals. Semen AI-doses were hierarchically clustered as having high, medium and low seminal OSI (p < 0.001) with those of low displaying higher resilience (p < 0.01). Boars were hierarchically clustered into two groups (p < 0.001) as having either positive or negative farrowing rate and litter size deviation; the negative one showing higher seminal OSI (p < 0.05). In sum, seminal OSI was negatively related to sperm motility and the in vivo fertility of liquid-stored boar semen AI-doses, with the receiver operating characteristic curve presenting seminal OSI as a good predictive biomarker of in vivo fertility of AI-boars (area under the curve: 0.815, p < 0.05).

  • Measuring the physical imprints of gas flows in galaxies : I. Accretion rate histories

    • Context. Galaxies are expected to accrete pristine gas from their surroundings to sustain their star formation over cosmic timescales. This mechanism is well established in models and simulations, but evidence from observations is mostly indirect. These gas inflows leave distinct traces in the chemical composition of newborn stars and alter the distribution of stellar abundances compared to what would be expected from a closed-box model of chemical evolution. Aims. The goal of this work is to measure the amount of pristine gas that galaxies accrete during their lifetime, using information on the ages and abundances of their stellar populations and a chemical evolution model. We also aim to determine the efficiency of star formation over time. Methods. We derived star formation histories and metallicity histories for a sample of 8523 galaxies from the MaNGA survey. We use the former to predict the evolution of the metallicity in a closed-box scenario, and estimate for each epoch the gas accretion rate required to match these predictions with the measured stellar metallicity. Results. Using only chemical parameters, we find that the history of gas accretion depends on the mass of galaxies. More massive galaxies accrete more gas and at higher redshifts than less massive galaxies, which accrete their gas over longer periods. We also find that galaxies with a higher star formation rate at z., =., 0 have a more persistent accretion history for a given mass. We characterize the individual accretion histories in terms of two parameters: the total accreted gas mass and the 80 of the accretion history, a measure of when most of the accretion occurred. As expected, there is a strong correlation between the integrated star formation history and the total accreted gas mass, such that more massive galaxies accreted more gas during their lifetime. Currently star-forming galaxies lie above this correlation, so they tend to accrete more gas than average. The relationship between 80, the current stellar mass, and the current specific star formation rate is split such that star-forming galaxies (as now observed) may be found in a population with persistent gas accretion regardless of their stellar mass. The star formation efficiency shows similar correlations: early-type galaxies and higher-mass galaxies had a higher efficiency in the past, and it declined such that they are less efficient in the present. Our analysis of individual galaxies shows that compactness affects the peak star formation efficiency that galaxies reach, and that the slope of the efficiency history of galaxies with current star formation is flat. Conclusions. We show throughout the article that we can obtain information about the processes that regulate the chemical composition of the interstellar medium during the lifetime of a galaxy from the properties of stellar populations. Our results support the hypothesis that a steady and substantial supply of pristine gas is required for persistent star formation in galaxies. Once they lose access to this gas supply, star formation comes to a halt.

  • Searching for the nature of stars with debris disks and planets

    • The nature of the few known solar-mass stars simultaneously containing debris disks and planets remains an open question. A number of works have shown that this property appears to be independent of planetary masses as well as of stellar age, but possible correlations with stellar kinematics and metallicity have not been investigated. In this paper, we show that the majority of known stars containing both debris disks and planets belong to the metal-enriched Galactic thin disk. The few exceptions are stars that seem to be born in the star formation peak occurring in times of thick disk formation (i.e., HD 10700, HD 20794, and HD 40307), that is, between 11 and 8 Gyr. The mass of the dusty disk of these three old stars measured at 70 μm is very small - in fact, it is lower than that of the Kuiper belt of our Solar system by several orders of magnitude. These results are not surprising, as they remain within the values expected for the stellar disk evolution of such primitive stars. In parallel, we found another six thick-disk stars containing only debris disks or planets. These results enable us to establish a correlation between stellar metallicity and the mass of the dust disk modulated by the different formation epochs of the thick and thin Galactic disks.

    • Healthcare workers (HCWs) are at the frontline of the Coronavirus Disease 2019 (COVID-19) pandemic response, yet there is a paucity of literature on their knowledge, attitudes and practices (KAP) in relation to the pandemic. Community Health Workers (CHWs) in Mozambique are known locally as agentes polivalentes elementares (APEs). While technical guidance surrounding COVID-19 is available to support APEs, communicating this information has been challenging due to restrictions on travel, face-to-face group meetings and training, imposed from May to August 2020. A digital health platform, upSCALE, that already supports 1,213 APEs and 299 supervisors across three provinces, is being used to support APEs on effective COVID-19 management by delivering COVID-19 sensitive SMS messages, training modules and a COVID-19 KAP survey. The KAP survey, conducted from June 2020 to August 2020, consisted of 10 questions. Of 1,065 active upSCALE APEs, 28% completed the survey. Results indicate that only a small proportion of APEs listed the correct COVID-19 symptoms, transmission routes and appropriate prevention measures (n = (25%), n = (16%) and n = (39%), respectively) specifically included in national health education materials. Misconceptions were mainly related to transmission routes, high risk individuals and asymptomatic patients. 84% said they followed all government prevention guidelines. The results from the KAP survey were used to support the rapid development and deployment of targeted COVID-19 awareness and education materials for the APEs. A follow-up KAP survey is planned for November 2020. Adapting the existing upSCALE platform enabled a better understanding, in real time, of the KAP of APEs around COVID-19 management. Subsequently, supporting delivery of tailored messages and education, vital for ensuring a successful COVID-19 response.

  • Galactoseismology in cosmological simulations Vertical perturbations by dark matter, satellite galaxies, and gas

    • Context. Complex models recently became available for studying the dynamics of disk galaxies such as the Milky Way (MW). These models include the global dynamics from dwarf satellite galaxies, dark matter halo structure, gas infall, and stellar disks in a cosmological context. Aims. We use a MW model from a suite of high-resolution hydrodynamical cosmological simulations named GARROTXA to establish the relationship between the vertical disturbances seen in its galactic disk and multiple perturbations from the dark matter halo, satellites, and gas. Methods. We calculated the bending modes in the galactic disk in the last 6 Gyr of evolution. We computed the vertical acceleration exerted by dark matter and gas in order to quantify the impact of these components on the disk, and compared this with the bending behavior with Fourier analysis. Results. We find complex bending patterns at different radii and times, such as an inner retrograde mode with high frequency and an outer slower retrograde mode excited at different times. The amplitudes of these bending modes are highest during the early stages of formation of the thin disk (20 km s−1) and reach up to 8.5 km s−1 in the late disk evolution. We find that the infall of satellite galaxies leads to a tilt of the disk, and produces strong anisotropic gas accretion with a misalignment of 8◦ with subsequent star formation events and supernovae, creating significant vertical accelerations on the disk plane. The misalignment between the disk and the inner stellar and dark matter triaxial structure, which formed during the ancient assembly of the galaxy, also leads to a strong vertical acceleration of the stars. We also find dark matter subhalos that temporally coincide with the appearance of bending waves in certain periods. Conclusions. We conclude that several agents trigger the bending of the stellar disk and its phase spirals in this simulation, including satellite galaxies, dark subhalos, misaligned gaseous structures, and the inner dark matter profile. These phenomena coexist and influence each other, sometimes making it challenging to establish direct causality.

  • Kinematic analysis of the Large Magellanic Cloud using Gaia DR3

    • Context. The high quality of the Gaia mission data has allowed for studies of the internal kinematics of the Large Magellanic Cloud (LMC) to be undertaken in unprecedented detail, providing insights into the non-axisymmetric structure of its disc. Recent works by the Gaia Collaboration have already made use of the excellent proper motions of Gaia DR2 and Gaia EDR3 for a first analysis of this sort, but these were based on limited strategies aimed at distinguishing the LMC stars from the Milky Way foreground that did not use all the available information. In addition, these studies could not access the third component of the stellar motion, namely, the line-of-sight velocity - which has now become available via Gaia DR3 for a significant number of stars. Aims. Our aim is twofold: 1) to define and validate an improved, more efficient and adjustable selection strategy to distinguish the LMC stars from the Milky Way foreground; 2) to check the possible biases that assumed parameters or sample contamination from the Milky Way can introduce in analyses of the internal kinematics of the LMC based on Gaia data. Methods. Our selection was based on a supervised neural network classifier, using as much as of the Gaia DR3 data as possible. Based on this classifier, we selected three samples of candidate LMC stars with different degrees of completeness and purity. We validated these classification results using different test samples and we compared them with the results from the selection strategy used in the Gaia Collaboration papers, based only on the proper motions. We analysed the resulting velocity profiles and maps for the different LMC samples and we checked how these results change when we use the line-of-sight velocities that are available for a subset of stars. Results. We show that the contamination in the samples from Milky Way stars basically affects the results for the outskirts of the LMC. We also show that the analysis formalism used in absence of line-of-sight velocities does not bias the results for the kinematics in the inner disc. Here, for the first time, we performed a kinematic analysis of the LMC using samples with the full three dimensional (3D) velocity information from Gaia DR3. Conclusions. The detailed 2D and 3D kinematic analysis of the LMC internal dynamics demonstrate that: 1) the dynamics in the inner disc is mainly bar dominated; 2) the kinematics on the spiral arm overdensity seems to be dominated by an inward motion and a rotation that is faster than that of the disc in the part of the arm attached to the bar; 3) the contamination of Milky Way stars seem to dominate the outer parts of the disc and mainly affects old evolutionary phases; and 4) uncertainties on the assumed disc morphological parameters and line-of-sight velocity of the LMC can (in some cases) have significant effects on the results of the analysis.

  • Ovarian Follicle Growth during Lactation Determines the Reproductive Performance of Weaned Sows

    • Simple Summary In this field study, the ovaries of weaned (n= 191, experiment 1) and lactating (n= 40, experiment 2) sows were transrectally scanned to measure the diameter of the follicles. Both the weaned and lactating sows showed great variability in the diameter of the ovarian follicles, indicating that the variability at weaning already existed during early lactation and was carried over to weaning. Sows with small follicles at weaning showed low reproductive performance and were more frequent among those with fewer farrowings and those weaned in summer-autumn. Factors causing variability in ovarian follicle size among weaned sows are not well known. This field study aimed to disclose influencing factors and evaluate if the differences at weaning were established during lactation. Ovaries were scanned using transrectal ultrasound. The first experiment was conducted over a year with 191 randomly chosen sows that were hierarchically grouped (p <0.001) according to ovarian follicle diameter reached at weaning: Small (0.20-0.30 cm;n= 37), medium (0.31-0.39 cm;n= 75), and large (0.40-1.00 cm;n= 69). Sows with small follicles showed a higher incidence of post-weaning anestrus (p< 0.01), longer wean-to-estrus/ovulation intervals (p <0.01) and farrowing smaller litters (p< 0.05). Ovaries with small follicles were more common among sows weaned in summer-autumn than in winter-spring (p <0.01) and among sows of lower parity (1-3) (p <0.05). In the second experiment, with 40 sows randomly chosen at farrowing, the ovaries were scanned at 7, 14, and 21 d post-partum. Sows showed great variability in ovarian follicular size during lactation with a consistent relationship between the three measurement times (r = 0.84,p <0.01). Follicle size was smaller in sows nursing in summer-autumn than in winter-spring (p <0.05). In conclusion, early lactation dictates the great variability in ovarian follicular diameter at weaning shown by sows. Sows with smaller follicles at weaning had longer intervals for estrus and ovulation and smaller litters at farrowing and they were in greater numbers among sows weaned during the summer and fall and among those with fewer previous farrowing.

  • Weaned Sows with Small Ovarian Follicles Respond Poorly to the GnRH Agonist Buserelin

    • Simple Summary This study evaluated the influence of mean ovarian follicle size and the season of weaning on the effectiveness of administering the GnRH agonist buserelin to synchronize ovulation in weaned sows. The results from 352 sows demonstrated that sows with small follicles (<0.5 cm in diameter) at treatment are poor responders, a condition more frequent among sows weaned in summer-autumn than in those weaned in winter-spring. The GnRH agonist buserelin (GnRH), used to synchronize ovulation in weaned sows, attains only 70-80% effectivity, owing to several reasons of ovarian origin. This study evaluated in particular whether mean ovarian follicle size at treatment and the season of weaning are among those influencing GnRH responsiveness. The experiment was carried out in a temperate-region farm with 352 sows of 1-6 parities weaned either in winter-spring (WS, 174 sows) or in summer-autumn (SA, 178 sows). The sows were randomized into two groups: GnRH (10 mu g of buserelin acetate at 86 h after weaning, 172 sows) and control (180 sows). The ovaries were transrectally scanned from weaning to ovulation and the sows clustered according to their mean follicular size at treatment time: small (<0.5 cm in diameter), medium (0.5 to 0.64 cm) and large (0.65 to 1.09 cm). In total, 88.33% of the GnRH-treated sows ovulated, with 82% of them within the expected time window (120-132 h after weaning). In contrast, 95.45% of the unresponsive sows had small follicles at the time of treatment and were mostly weaned in SA (20.45%) than in WS (4.76%). In conclusion, the conspicuous presence of sows having small ovarian follicles at treatment time compromises the efficiency of the GnRH agonist buserelin to synchronize ovulation in weaned sows, which occurs more frequently in summer-autumn weaning.

  • Tumor Infiltrating Neutrophils Are Enriched in Basal-Type Urothelial Bladder Cancer

    • Background: Urothelial bladder cancers (UBCs) are distinct in two main molecular subtypes, namely basal and luminal type. Subtypes are also diverse in term of immune contexture, providing a rationale for patient selection to immunotherapy. Methods: By digital microscopy analysis of a muscle-invasive BC (MIBC) cohort, we explored the density and clinical significance of CD66b+ tumor-associated-neutrophils (TAN) and CD3+ T cells. Bioinformatics analysis of UBC datasets and gene expression analysis of UBC cell lines were additionally performed. Results: Basal type BC contained a significantly higher density of CD66b+ TAN compared to the luminal type. This finding was validated on TCGA, GSE32894 and GSE124305 datasets by computing a neutrophil signature. Of note, basal-type MIBC display a significantly higher level of chemokines (CKs) attracting neutrophils. Moreover, pro-inflammatory stimuli significantly up-regulate CXCL1, CXCL2 and CXCL8 in 5637 and RT4 UBC cell lines and induce neutrophil chemotaxis. In term of survival, a high density of T cells and TAN was significantly associated to a better outcome, with TAN density showing a more limited statistical power and following a non-linear predicting model. Conclusions: TAN are recruited in basal type MIBC by pro-inflammatory CKs. This finding establishes a groundwork for a better understanding of the UBC immunity and its relevance.

These developments span a wide range of topics in computational chemistry and are presented in thematic sections: electronic structure theory, electronic spectroscopy simulations, analytic gradients and molecular structure optimizations, ab initio molecular dynamics, and other new features. This report offers an overview of the chemical phenomena and processes OpenMolcas can address, while showing that OpenMolcas is an attractive platform for state-of-the-art atomistic computer simulations.

    • Recent efforts to achieve social, economic, and environmental goals related to sustainability emphasize the importance of nature-based solutions (NBS), as grey infrastructure alone is insufficient to address current challenges. The majority of frameworks proposed in the literature fail to address the full potential of NBS, neglecting long-term results, unintended consequences, co-benefits, and their contribution to achieving global environmental agreements, such as the Agenda 2030, especially for water management in a peri-urban context. Here we present an innovative framework that can be applied to both NBS project planning and evaluation for several water-based challenges, giving practitioners and researchers a tool not only to evaluate ongoing projects but also to guide new ones. The framework considers three main stages of a NBS project: (1) context assessment, (2) NBS implementation and adaptation process, and (3) NBS results. This tool has the potential to be used to evaluate whether NBS projects are aligned with sustainability dimensions through a set of adaptable sustainability indicators. The framework can also highlight how the NBS targets are related to the sustainable development goals (SGD) and contribute to catalyzing the 2030 Agenda. The framework is an important tool for water management and other NBS types.

  • Nonpharmaceutical interventions reduce the incidence and mortality of COVID-19: A study based on the survey from the International COVID-19 Research Network (ICRN)

    • The recently emerged novel coronavirus, "severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2)," caused a highly contagious disease called coronavirus disease 2019 (COVID-19). It has severely damaged the worlds most developed countries and has turned into a major threat for low- and middle-income countries. Since its emergence in late 2019, medical interventions have been substantial, and most countries relied on public health measures collectively known as nonpharmaceutical interventions (NPIs). We aimed to centralize the accumulative knowledge of NPIs against COVID-19 for each country under one worldwide consortium. International COVID-19 Research Network collaborators developed a cross-sectional online survey to assess the implications of NPIs and sanitary supply on the incidence and mortality of COVID-19. The survey was conducted between January 1 and February 1, 2021, and participants from 92 countries/territories completed it. The association between NPIs, sanitation supplies, and incidence and mortality were examined by multivariate regression, with the log-transformed value of population as an offset value. The majority of countries/territories applied several preventive strategies, including social distancing (100.0%), quarantine (100.0%), isolation (98.9%), and school closure (97.8%). Individual-level preventive measures such as personal hygiene (100.0%) and wearing facial masks (94.6% at hospitals; 93.5% at mass transportation; 91.3% in mass gathering facilities) were also frequently applied. Quarantine at a designated place was negatively associated with incidence and mortality compared to home quarantine. Isolation at a designated place was also associated with reduced mortality compared to home isolation. Recommendations to use sanitizer for personal hygiene reduced incidence compared to the recommendation to use soap. Deprivation of masks was associated with increased incidence. Higher incidence and mortality were found in countries/territories with higher economic levels. Mask deprivation was pervasive regardless of economic level. NPIs against COVID-19 such as using sanitizer, quarantine, and isolation can decrease the incidence and mortality of COVID-19.

    • Cells are constantly exposed to various chemical and physical stimuli. While much has been learned about the biochemical factors that regulate secretory trafficking from the endoplasmic reticulum (ER), much less is known about whether and how this trafficking is subject to regulation by mechanical signals. Here, we show that subjecting cells to mechanical strain both induces the formation of ER exit sites (ERES) and accelerates ER-to-Golgi trafficking. We found that cells with impaired ERES function were less capable of expanding their surface area when placed under mechanical stress and were more prone to develop plasma membrane defects when subjected to stretching. Thus, coupling of ERES function to mechanotransduction appears to confer resistance of cells to mechanical stress. Furthermore, we show that the coupling of mechanotransduction to ERES formation was mediated via a previously unappreciated ER-localized pool of the small GTPase Rac1. Mechanistically, we show that Rac1 interacts with the small GTPase Sar1 to drive budding of COPII carriers and stimulates ER-to-Golgi transport. This interaction therefore represents an unprecedented link between mechanical strain and export from the ER.

  • Awake Proning as an Adjunctive Therapy for Refractory Hypoxemia in Non-Intubated Patients with COVID-19 Acute Respiratory Failure: Guidance from an International Group of Healthcare Workers

    • Non-intubated patients with acute respiratory failure due to COVID-19 could benefit from awake proning. Awake proning is an attractive intervention in settings with limited resources, as it comes with no additional costs. However, awake proning remains poorly used probably because of unfamiliarity and uncertainties regarding potential benefits and practical application. To summarize evidence for benefit and to develop a set of pragmatic recommendations for awake proning in patients with COVID-19 pneumonia, focusing on settings where resources are limited, international healthcare professionals from high and low- and middle-income countries (LMICs) with known expertise in awake proning were invited to contribute expert advice. A growing number of observational studies describe the effects of awake proning in patients with COVID-19 pneumonia in whom hypoxemia is refractory to simple measures of supplementary oxygen. Awake proning improves oxygenation in most patients, usually within minutes, and reduces dyspnea and work of breathing. The effects are maintained for up to 1 hour after turning back to supine, and mostly disappear after 6–12 hours. In available studies, awake proning was not associated with a reduction in the rate of intubation for invasive ventilation. Awake proning comes with little complications if properly implemented and monitored. Pragmatic recommendations including indications and contraindications were formulated and adjusted for resource-limited settings. Awake proning, an adjunctive treatment for hypoxemia refractory to supplemental oxygen, seems safe in non-intubated patients with COVID-19 acute respiratory failure. We provide pragmatic recommendations including indications and contraindications for the use of awake proning in LMICs.
