- The study reviews, based on the state-of-the-art findings, some uncertainties associated with wave data and models currently used in design and operation procedures of ship and offshore structures. Although the same basic principles prevail for hydrodynamic loads on ships and offshore structures, actual problems and methods for assessing these loads in the design and operation stage are not the same. Different wave data and models are used for specifying design and operational criteria for these two types of platforms and different uncertainties are related to them. Wave data and models used to define sea state characteristics are discussed and particular attention is given to the associated sources of uncertainties. Some weaknesses of wave input used in design and operational procedures for marine structures are pointed out. Focus is also given on uncertainties related to model tests as tank testing is an important supporting tool for design and operation. Impact of some selected uncertainties on wave description and wave loads is demonstrated by examples. (c) 2016 Elsevier Ltd. All rights reserved.

Identification of targetable lesions in anaplastic thyroid cancer by genome profiling

- Anaplastic thyroid cancer (ATC) is a rare and extremely malignant tumor with no available cure. The genetic landscape of this malignancy has not yet been fully explored. In this study, we performed whole exome sequencing and the RNA-sequencing of fourteen cases of ATC to delineate copy number changes, fusion gene events, and somatic mutations. A high frequency of genomic amplifications was seen, including 29% of cases having amplification of CCNE1 and 9% of CDK6; these events may be targetable by cyclin dependent kinase (CDK) inhibition. Furthermore, 9% harbored amplification of TWIST1, which is also a potentially targetable lesion. A total of 21 fusion genes in five cases were seen, none of which were recurrent. Frequent mutations included TP53 (55%), the TERT promoter (36%), and ATM (27%). Analyses of mutational signatures showed an involvement of processes that are associated with normal aging, defective DNA mismatch repair, activation induced cytidine deaminase (AID)/apolipoprotein B editing complex (APOBEC) activity, failure of DNA double-strand break repair, and tobacco exposure. Taken together, our results shed new light on the tumorigenesis of ATC and show that a relatively large proportion (36%) of ATCs harbor genetic events that make them candidates for novel therapeutic approaches. When considering that ATC today has a mortality rate of close to 100%, this is highly relevant from a clinical perspective.

- Mutation testing has been used to assess test suite coverage, and researchers have proposed adapting the idea for other uses. Safety kernels allow the use of untrusted software components in safety-critical applications: a trusted software safety kernel detects undesired behavior and takes remedial action. We propose to use specification mutation, model checking, and model-based testing to verify safety kernels for component-based, safety-critical computer systems.

Twelve-year follow-up of mandibular incisor stability : Comparison between two bonded lingual orthodontic retainers

- Objective: To compare the long-term outcome 9 years after removal of two different types of fixed retainers used for stabilization of the mandibular anterior segment. Materials and Methods: Sixty-four children who had undergone orthodontic treatment with fixed appliances in both arches were divided into two groups depending on which kind of retainer being used. Twenty-eight of the patients had a canine-to-canine retainer bonded to the canines and 36 had a bonded twistflex retainer 3-3, bonded to each tooth. Measurements were made on study models and lateral head radiographs, before and after treatment, 6 years after treatment, and 12 years after treatment, with a mean of 9.2 years after removal of the retainers. Results: No significant differences were found between the two groups at the long-term follow-up according to Little's Irregularity Index or available space for the mandibular incisors. The overjet and overbite were reduced after treatment in both groups and stayed stable throughout the observation period. Also, no differences in bonding failures between the two retainers were found. Conclusions: Both a canine-to-canine retainer bonded only to the canines and a twistflex retainer 3-3 bonded to each tooth can be recommended. However, neither of the retention types prevented long-term changes of mandibular incisor irregularity or available space for the mandibular incisors after removal of the retainers.

- The aim of this article is, by analysing childrens discourses, to investigate their actions or absence of actions during a domestic violence episode. The empirical data are recorded group therapy sessions and individual interviews with children who have grown up experiencing their fathers violence against their mothers. The analysis shows that the childrens stories contain two aspects of actions: one related to the actions during the ongoing episode, and one the child perceives as possible/desirable for the future. The findings are discussed in the light of Lazarus and Folkmans theory of coping.
