  • Structures of two fimbrial adhesins, AtfE and UcaD, from the uropathogen Proteus mirabilis

    • The important uropathogen Proteus mirabilis encodes a record number of chaperone/usher-pathway adhesive fimbriae. Such fimbriae, which are used for adhesion to cell surfaces/tissues and for biofilm formation, are typically important virulence factors in bacterial pathogenesis. Here, the structures of the receptor-binding domains of the tip-located two-domain adhesins UcaD (1.5 Å resolution) and AtfE (1.58 Å resolution) from two P. mirabilis fimbriae (UCA/NAF and ATF) are presented. The structures of UcaD and AtfE are both similar to the F17G type of tip-located fimbrial receptor-binding domains, and the structures are very similar despite having only limited sequence similarity. These structures represent an important step towards a molecular-level understanding of P. mirabilis fimbrial adhesins and their roles in the complex pathogenesis of urinary-tract infections.

  • In-situ lecithination of dairy powders in spray-drying for confectionery applications

    • Powders are essential ingredients of chocolate. In particular for milk chocolate milk and whey powders are important, together with sucrose, lactose and cocoa solids. During processing to maintain a good flow of the molten chocolate mass, particles with hydrophilic surfaces, such as dairy powders and sugars, are coated with a surface-active compound. Only lecithin and polyglycerol polyricinoleate (PGPR) (at a limited level) are allowed in chocolate, and as these are expensive as little as possible is added, whilst maintaining rheological properties. Conventionally, lecithin is added during conching, and through the intense kneading of the chocolate mass it is distributed throughout the mass. Usually about 0.5% is added, although the level depends upon the composition of the chocolate. Here we present a new approach to lecithination of spray-dried milk and lactose powders, which we call in-situ lecithination. It has been found that the surface of a spray-dried powder is dominated by any surface-active species, and in a competitive situation, the most rapidly adsorbing species dominates. This behaviour is utilised when lecithin is added to the spray-dryer feed, and through the competitive adsorption of surface-active agents during the drying process, it dominates the powder surface composition as measured by X-ray photoelectron spectroscopy (XPS). This is also seen in differences in sedimentation rate when the powders are mixed with cocoa butter to assess the rheological properties of the powder dispersions. The effect was large for lactose powders, but smaller for skim milk powder and whey powder.

  • Circulating Brain-Injury Markers After Surgery for Craniosynostosis

    • Objective: Historically, there have been few quantitative methods for effectively evaluating outcomes after surgery for craniosynostosis. In this prospective study, we assessed a novel approach for detecting possible postsurgery brain injury in patients with craniosynostosis. Methods: We included consecutive patients operated on for sagittal (pi-plasty or craniotomy combined with springs) or metopic (frontal remodeling) synostosis at the Craniofacial Unit at Sahlgrenska University Hospital, Gothenburg, Sweden, from January 2019 to September 2020. Plasma concentrations of the brain-injury biomarkers neurofilament light (NfL), glial fibrillary acidic protein (GFAP), and tau were measured immediately before induction of anesthesia, immediately before and after surgery, and on the first and the third postoperative days using single-molecule array assays. Results: Of the 74 patients included, 44 underwent craniotomy combined with springs for sagittal synostosis, 10 underwent pi-plasty for sagittal synostosis, and 20 underwent frontal remodeling for metopic synostosis. Compared with baseline, GFAP level showed a maximal significant increase at day 1 after frontal remodeling for metopic synostosis and pi-plasty (P = 0.0004 and P = 0.003, respectively). By contrast, craniotomy combined with springs for sagittal synostosis showed no increase in GFAP. For neurofilament light, we found a maximal significant increase at day 3 after surgery for all procedures, with significantly higher levels observed after frontal remodeling and pi-plasty compared with craniotomy combined with springs (P < 0.001). Conclusions: These represent the first results showing significantly increased plasma levels of brain-injury biomarkers after surgery for craniosynostosis. Furthermore, we found that more extensive cranial vault procedures resulted in higher levels of these biomarkers relative to less extensive procedures.

    • On average, airborne aerosol particles cool the Earth's surface directly by absorbing and scattering sunlight and indirectly by influencing cloud reflectivity, life time, thickness or extent. Here we show that over the central Arctic Ocean, where there is frequently a lack of aerosol particles upon which clouds may form, a small increase in aerosol loading may enhance cloudiness thereby likely causing a climatologically significant warming at the ice-covered Arctic surface. Under these low concentration conditions cloud droplets grow to drizzle sizes and fall, even in the absence of collisions and coalescence, thereby diminishing cloud water. Evidence from a case study suggests that interactions between aerosol, clouds and precipitation could be responsible for attaining the observed low aerosol concentrations.

    • Boron carbide in its rhombohedral form (r-BxC), commonly denoted B4C or B13C2, is a well-known hard material, but it is also a potential semiconductor material. We deposited r-BxC by chemical vapor deposition between 1100 degrees C and 1500 degrees C from triethylboron in H-2 on 4H-SiC(0001) and 4H-SiC(0001). We show, using ToF-ERDA, that pure B4C was grown at 1300 degrees C, furthermore, using XRD that graphite forms above 1400 degrees C. The films deposited above 1300 degrees C on 4H-SiC(0001) were found to be epitaxial, with the epitaxial relationships B4C(0001)[1010]||4H-SiC(0001)[1010] obtained from pole figure measurements. In contrast, the films deposited on 4H-SiC(0001) were polycrystalline. We suggest that the difference in growth mode is explained by the difference in the ability of the different surfaces of 4H-SiC to act as carbon sources in the initial stages of the film growth.

  • QTLs for the elongation of axile and lateral roots of maize in response to low water potential

    • Changes in root architecture and the maintenance of root growth in drying soil are key traits for the adaptation of maize (Zea mays L.) to drought environments. The goal of this study was to map quantitative trait loci (QTLs) for root growth and its response to dehydration in a population of 208 recombinant inbred lines from the International Maize and Wheat Improvement Center (CIMMYT). The parents, Ac7643 and Ac7729/TZSRW, are known to be drought-tolerant and drought-sensitive, respectively. Roots were grown in pouches under well-watered conditions or at low water potential induced by the osmolyte polyethylene glycol (PEG 8000). Axile root length (L (Ax)) increased linearly, while lateral root length (L (Lat)) increased exponentially over time. Thirteen QTLs were identified for six seedling traits: elongation rates of axile roots (ER(Ax)), the rate constant of lateral root elongation (k (Lat)), the final respective lengths (L (Ax) and L (Lat)), and the ratios k (Lat)/ER(Ax) and L (Lat)/L (Ax.) While QTLs for lateral root traits were constitutively expressed, most QTLs for axile root traits responded to water stress. For axile roots, common QTLs existed for ER(Ax) and L (Ax). Quantitative trait loci for the elongation rates of axile roots responded more clearly to water stress compared to root length. Two major QTLs were detected: a QTL for general vigor in bin 2.02, affecting most of the traits, and a QTL for the constitutive increase in k (Lat) and k (Lat)/ER(Ax) in bins 6.04-6.05. The latter co-located with a major QTL for the anthesis-silking interval (ASI) reported in published field experiments, suggesting an involvement of root morphology in drought tolerance. Rapid seedling tests are feasible for elucidating the genetic response of root growth to low water potential. Some loci may even have pleiotropic effects on yield-related traits under drought stress.

    • This work combined gene and protein expression, gas chromatography-flame ionization detector, and hydrophilic interaction liquid chromatography-tandem mass spectrometry to compare lipid metabolism changes in undifferentiated/proliferating vs. functionally differentiated mammary epithelial cells (MECs) and to study their correlation to breast cancer survival. Sixty-eight genes involved in lipid metabolism were changed in MEC differentiation. Differentiated cells showed induction of Elovl6 (2-fold), Scd1 (4-fold), and Fads2 (2-fold), which correlated with increased levels of C16:1 n-7 and C18:1 n-9 (1.5-fold), C20:3 n-6 (2.5-fold), and C20:4 n-6 (6-fold) fatty acids (FAs) and more phospholipids (PLs) containing these species. Further, increased expression (2- to 3-fold) of genes in phosphatidylethanolamine (PE) de novo biosynthesis resulted in a 20% PE increase. Proliferating/undifferentiated cells showed higher C16:0 (1.7-fold) and C18:2 n-6 (4.2-fold) levels and more PLs containing C16:0 FAs [PC(16:0/16:1), PG(16:0/18:2), PG(16:0/18:1), and SM(16:0/18:0)]. Kaplan-Meier analysis of data from 3455 patients with breast cancer disclosed a positive correlation for 59% of genes expressed in differentiated MECs with better survival. PE biosynthesis and FA oxidation correlated with better prognosis in patients with breast cancer, including the basal-like subtype. Therefore, genes involved in mammary gland FA and PL metabolism and their resulting molecular species reflect the cellular proliferative ability and differentiation state and deserve further studies as potential markers of breast cancer progression
