  • Mesoproterozoic dykes in the Timmiarmiit area, Southeast Greenland : evidence for a continuous Gardar dyke swarm across Greenland’s North Atlantic Craton

    • During the Proterozoic, several mafic dykes with variable trends and mineralogies intruded the Archaean basement of Southeast Greenland. Some of the younger ENE-trending dykes are interpreted to represent a prolongation of the Mesoproterozoic Gardar Province, and have been termed Timmiarmiit dykes. Extrapolations of their trends across the inland ice sheet coincide with the northernmost so-called brown dykes (BD’s) which are part of the Gardar Province. Baddeleyite U–Pb ID-TIMS analyses for three ENE-trending Timmiarmiit dykes give ages of 1277 ± 4, 1275 ± 3 and 1268 ± 4 Ma, which are slightly younger than the oldest (BD0 = 1284–1279 ± 3 Ma, Upton 2013) and only dated generation of dykes in the Gardar Province, and thereby indirectly provide a possible age for the two younger dyke generations (BD1 and BD2). The Timmiarmiit–Gardar correlation is strengthened by a rigorous multivariate statistical analysis, on the basis of all major and trace elements. Thus, a coherent ENE-trending trans-Greenlandic dyke swarm is constituted. The major and trace element data of the Timmiarmiit dykes show that they crystallised from comparably evolved mantle-derived magma with minor crustal contamination and indicate a strong contribution of a metasomatised subcontinental lithospheric mantle component in the evolution of melts. This component was probably influenced by supra-subduction zone metasomatism during the Palaeoproterozoic Ketilidian orogeny. The data presented here, in addition to recent plate reconstruction models, give new evidence for a petrogenetic link between rift-related Mesoproterozoic magmatism in North America, South Greenland and Central Scandinavia which possibly formed in response to back-arc basin formation.

    • The Ukrainian Shield comprises the exposed crust of the large Palaeoproterozoic protocraton Volgo-Sarmatia, which together with the Fennoscandian crustal segment constitutes the East European Craton ("Baltica"). Geological and geophysical data indicate that 1.80 to 1.75 Ga mafic dykes related to anorthosite-mangerite-charnockite-granite (AMCG) plutons are widespread within the Ukrainian Shield. We examined their ages, distribution patterns, orientations and compositions in three different crustal blocks (Volyn, Ingul and Azov), and found close spatial relationships with major strike-slip fault systems developed during two phases of extension. The early, 1.80-1.77 Ga, generation of mafic dykes mostly follows NW (330 +/- 20 degrees) and more rarely N-S- or E-W-trending faults corresponding to major NE-SW extension (the Submoshorino phase). These dykes contain olivine dolerites, picrites, camptonites, lamprophyres, kimberlites and other rocks belonging to tholeiitic and subalkaline jotunitic series. The compositions of these dykes differ between the host blocks, but all feature upper mantle geochemical signatures such as high contents of Ni and Cr, and positive values of epsilon Nd-(1800) up to + 2.8. High degrees of REE fractionation indicate deep levels of mantle melting, which is particularly characteristic of the Ingul block as marked by the most extensive and dense mafic dyke swarms. The later, 1.76-1.75 Ga, dyke swarms occur close to the most voluminous AMCG suites of similar age and were emplaced during the second (Korsun) phase of faulting when all the older strike-slip fault zones were reactivated and partly transformed to tensional faults by E-W extension. These dyke swarms mainly trend 030 +/- 20 degrees. They are jotunitic and their isotopic signatures indicate a greater participation of crustal sources in the parent melts. The overall transtensional tectonic setting of the mafic dyking associated with the AMCG magmatism in Volgo-Sarmatia was created by convergent tectonics and postcollisional collapse of the thickened lithosphere, as well as by mantle delamination coupled with the rotation of Volgo-Sarmatia between 1.80 and 1.75 Ga. This agrees with palaeomagnetic reconstruction suggesting rotation(s) of Volgo-Sarmatia during its protracted oblique docking with Fennoscandian terranes and Laurentia as supercontinent Columbia (Nuna) was assembled.   • Expression of ribosomal and actin network proteins and immunochemotherapy resistance in diffuse large B cell lymphoma patients

    • Diffuse large B cell lymphoma (DLBCL) patients with early relapse or refractory disease have a very poor outcome. Immunochemotherapy resistance will probably, also in the era of targeted drugs, remain the major cause of treatment failure. We used proteomic mass spectrometry to analyse the global protein expression of micro-dissected formalin-fixed paraffin-embedded tumour tissues from 97 DLBCL patients: 44 with primary refractory disease or relapse within 1year from diagnosis (REF/REL), and 53 who were progression-free more than 5years after diagnosis (CURED). We identified 2127 proteins: 442 were found in all patients and 102 were differentially expressed. Sixty-five proteins were overexpressed in REF/REL patients, of which 46 were ribosomal proteins (RPs) compared with 2 of the 37 overexpressed proteins in CURED patients (P=76x10(-10)). Twenty of 37 overexpressed proteins in CURED patients were associated with actin regulation, compared with 1 of 65 in REF/REL patients (P=14x10(-9)). Immunohistochemical staining showed higher expression of RPS5 and RPL17 in REF/REL patients while MARCKS-like protein, belonging to the actin network, was more highly expressed in CURED patients. Even though functional studies aimed at individual proteins and protein interactions to evaluate potential clinical effect are needed, our findings suggest new mechanisms behind immunochemotherapy resistance in DLBCL.

    • The complexity of the activated B-cell like (ABC) diffuse large B-cell lymphoma (DLBCL) subtype is probably not only explained by genetic alterations and methods to measure global protein expression could bring new knowledge regarding the pathophysiology. We used quantitative proteomics to analyze the global protein expression of formalin-fixed paraffin-embedded (FFPE) tumor tissues from 202 DLBCL patients. We identified 6430 proteins and 498 were significantly regulated between the germinal center B-cell like (GCB) and non-GCB groups. A number of proteins previously not described to be upregulated in non-GCB or ABC DLBCL was found, e.g. CD64, CD85A, guanylate-binding protein 1 (GBP1), interferon-induced proteins with tetratricopeptide repeat (IFIT)2, and mixed lineage kinase domain-like protein (MLKL) and immunohistochemical staining showed higher expression of GBP1 and MLKL. A cluster analysis revealed that the most prominent cluster contained proteins involved in the tumor microenvironment and regulation of the immune system. Our data suggest that the therapeutic focus should be expanded toward the tumor microenvironment in non-GCB/ABC subtype patients.

  • TBLR1 and CREBBP as potential novel prognostic immunohistochemical biomarkers in diffuse large B-cell lymphoma

    • Recent studies have identified prognostic mutational clusters for diffuse large B-cell lymphoma (DLBCL) patients, both within and outside the original cell-of-origin (COO) classification. For many of these mutations, there is limited information regarding the corresponding protein expression. With the aim to determine the relationship of protein expression and intensity to COO and prognosis, we used digital image analysis to quantitate immunohistochemical staining of CREBBP, IRF8, EZH2, and TBLR1 in 209 DLBCL patients. We found that patients with strong nuclear expression of TBLR1 had inferior progression-free survival (PFS) and overall survival (OS) in univariable analysis and inferior PFS in multivariable analysis. Patients with higher proportion of intermediate to strong nuclear CREBBP expression had a worse PFS and OS in univariable analysis. CREBBP was expressed with stronger intensity in non-GCB patients and the prognostic impact was restricted to this subgroup. These findings suggest that high nuclear protein expression of TBLR1 and CREBBP is negatively associated with prognosis in DLBCL.

    • A new U–Pb baddeleyite age of 1970 ± 3 Ma for the Unoi dolerite sill of the Onega structure of Karelia craton matches other 1.98–1.95 Ga units across the Kola craton (Pechenga) and widely separated parts of the Karelian craton, including the Lake Onega sill area and a extensive NW-trending dolerite dyke swarm. Herein these coeval units are combined into the Pechenga–Onega Large Igneous Province. The sills in the Lake Onega area exhibit similar geochemical patterns, although the Unoi dolerite sills appear less contaminated and less differentiated than the Pudozhgora intrusion, Gabnev sill and Koikary-Svatnavolok and Palieyeozero sills but are similar to other doleritic sills in the northern part of the Onega structure. New AMS data from sills are consistent with emplacement along the same NW–SE trend as the dykes, consistent with the dykes acting as their feeder system. Paleomagnetic data obtained on 1.98–1.95 Ga magmatic rocks result in a new robust paleopole for the Karelian craton and accentuate its variable paleolatitude and paleoorientation during the Paleoproterozoic.
