  • Long-lived plasma cells in mice and men
  • 2018
  • Ingår i: Frontiers in Immunology. - : Frontiers Media SA. - 1664-3224. ; 9
  • Tidskriftsartikel (refereegranskat)abstract
    • Even though more than 30 years have passed since the eradication of smallpox, high titers of smallpox-specific antibodies are still detected in the blood of subjects vaccinated in childhood. In fact, smallpox-specific antibody levels are maintained in serum for more than 70 years. The generation of life-long immunity against infectious diseases such as smallpox and measles has been thoroughly documented. Although the mechanisms behind high persisting antibody titers in the absence of the causative agent are still unclear, long lived plasma cells (LLPCs) play an important role. Most of the current knowledge on LLPCs is based on experiments performed in mouse models, although the amount of data derived from human studies is increasing. As the results from mouse models are often directly extrapolated to humans, it is important to keep in mind that there are differences. These are not only the obvious such as the life span but there are also anatomical differences, for instance the adiposity of the bone marrow (BM) where LLPCs reside. Whether these differences have an effect on the function of the immune system, and in particular on LLPCs, are still unknown. In this review, we will briefly discuss current knowledge of LLPCs, comparing mice and humans. © 2007 - 2018 Frontiers Media S.A.

  • Scientific (wo)manpower - gender and the composition and earnings of PhDs in Sweden
  • 2013
  • Ingår i: International Journal of Manpower. - 0143-7720. ; 34:6, s. 658-673
  • Tidskriftsartikel (refereegranskat)abstract
    • Purpose - The purpose of this paper is to investigate to what extent male and female PhDs choose academic vs non-academic employment. Further, it analyses gender earnings differences in the academic and non-academic labour markets. Design/methodology/approach - Rich Swedish cross-sectional register data on the stock of PhDs in 2004 is used for investigating differences between male and female PhDs in earnings levels and in the choice of labour market (in academia or outside academia). Findings - No significant gender differences prevail for the probability of becoming academically employed. On average, women earn 16 per cent less than men, and the academically employed earn 26 per cent less than PhDs outside academia. In the non-academic labour market, women earn approximately 25 per cent less than men, and this gender gap stays constant over time. Within academia, men and women start at similar earnings levels, but higher returns to graduate experience for men causes the gender earnings differences to increase over time to approximately 18 per cent by ten years after graduation. Practical implications - Provides background information relevant for efforts to make efficient use of female scientific human capital. Originality/value - The study is the first to investigate career-choice and earnings of Swedish PhDs. Further, the study is the first to investigate both the academic and the non-academic labour markets.

  • Impacts of moose (Alces alces) at different simulated densities on eco-morphological groups of soil mesofauna
  • 2010
  • Ingår i: Entomologica Fennica. - 0785-8760. ; 21, s. 168-180
  • Tidskriftsartikel (refereegranskat)abstract
    • We experimentally tested the impact of browsing, defecation and urination corresponding to four different levels of moose population density on abundance and number of eco-morphological groups of soil mesofauna in a Swedish boreal forest. The study was carried out in three fenced exclosures representing different levels of productivity (from nutrient-rich to nutrient-poor). Moose impact on soil communities was evaluated by analysing abundance, richness and diversity of mesofaunal groups, and by means of a multi-taxa index based on ecomorphological groups, the Biological Soil Quality index (QBS). There was a negative impact of high moose densities on the abundance and richness of soil mesofauna. Furthermore, low-moderate moose densities had a positive impact on abundance and number of eco-morphological groups. We conclude that moose can have a large impact on soil-living mesofauna, and therefore probably soil decomposition processes and biological soil quality, in young boreal forest stands.

  • PAPD5-mediated 3' adenylation and subsequent degradation of miR-21 is disrupted in proliferative disease.
  • 2014
  • Ingår i: Proceedings of the National Academy of Sciences. - : Proceedings of the National Academy of Sciences. - 1091-6490 .- 0027-8424. ; 111:31, s. 11467-11472
  • Tidskriftsartikel (refereegranskat)abstract
    • Next-generation sequencing experiments have shown that microRNAs (miRNAs) are expressed in many different isoforms (isomiRs), whose biological relevance is often unclear. We found that mature miR-21, the most widely researched miRNA because of its importance in human disease, is produced in two prevalent isomiR forms that differ by 1 nt at their 3' end, and moreover that the 3' end of miR-21 is posttranscriptionally adenylated by the noncanonical poly(A) polymerase PAPD5. PAPD5 knockdown caused an increase in the miR-21 expression level, suggesting that PAPD5-mediated adenylation of miR-21 leads to its degradation. Exoribonuclease knockdown experiments followed by small-RNA sequencing suggested that PARN degrades miR-21 in the 3'-to-5' direction. In accordance with this model, microarray expression profiling demonstrated that PAPD5 knockdown results in a down-regulation of miR-21 target mRNAs. We found that disruption of the miR-21 adenylation and degradation pathway is a general feature in tumors across a wide range of tissues, as evidenced by data from The Cancer Genome Atlas, as well as in the noncancerous proliferative disease psoriasis. We conclude that PAPD5 and PARN mediate degradation of oncogenic miRNA miR-21 through a tailing and trimming process, and that this pathway is disrupted in cancer and other proliferative diseases.
