    • A gene trap-type targeting vector was designed to inactivate the beta 1 integrin gene in embryonic stem (ES) cells. Using this vector more than 50% of the ES cell clones acquired a disruption in the beta 1 integrin gene and a single clone was mutated in both alleles. The homozygous mutant did not produce beta 1 integrin mRNA or protein, while alpha 3, alpha 5, and alpha 6 integrin subunits were transcribed but not detectable on the cell surface. Heterozygous mutants showed reduced beta 1 expression and surface localization of alpha/beta 1 heterodimers. The alpha V subunit expression was not impaired on any of the mutants. Homozygous ES cell mutants lacked adhesiveness for laminin and fibronectin but not for vitronectin and showed a reduced association with a fibroblast feeder layer. Furthermore, they did not migrate towards chemoattractants in fibroblast medium. None of these functions were impaired in heterozygous mutants. Scanning electron microscopy revealed that homozygous cells showed fewer cell-cell junctions and had many microvilli not usually found on wild type and heterozygous cells. This profound change in cell shape is not associated with gross alterations in the expression and distribution of cytoskeletal components. Unexpectedly, microinjection into blastocysts demonstrated full integration of homozygous and heterozygous mutants into the inner cell mass. This will allow studies of the consequences of beta 1 integrin deficiency in several in vivo situations.

  • Critical ionization velocity interaction in the CRIT I rocket experiment

    • In the rocket experiment CRIT I, launched from Wallops Island on 13 May 1986, two identical Barium shaped charges were fired from distances of 1.3 km and 3.6 km towards the main experiment payload, which was separated from a sub-payload by a couple of km along the magnetic field. The relevance of earlier proposed mechanisms for electron heating in ionospheric critical velocity experiments is investigated in the light of the CRIT I results. It is concluded that both the "homogeneous" and the "ionizing front" models can be applied, in different parts of the stream. It is also possible that a third, entirely different, mechanism may contribute to the electron heating. This mechanism involves direct energization of electrons in the magnetic-field-aligned component of the DC electric field. © 1989.

    • This paper deals with the quasi-dc electric fields measured in the CRIT I ionospheric release experiment, which was launched from Wallops Island on May 13, 1986. The purpose of the experiment was to study the critical ionization velocity (CIV) mechanism in the ionosphere. Two identical barium shaped charges were fired from distances of 1.99 km and 4.34 km towards a main payload, which made full three-dimensional measurements of the electric field inside the streams. There was also a subpayload separated from the main payload by a couple of kilometers along the magnetic field. The relevance of earlier proposed mechanisms for electron heating in CIV is investigated in the light of the CRIT I results. It is concluded that both the “homogeneous” and the “ionizing front” models probably apply, but in different parts of the stream. It is also possible that electrons are directly accelerated by a magnetic-field-aligned component of the electric field; the quasi-dc electric field observed within the streams had a large magnetic-field-aligned component, persisting on the time scale of the passage of the streams. The coupling between the ambient ionosphere and the ionized barium stream in CRIT I was more complicated than is usually assumed in CIV theories, with strong magnetic-field-aligned electric fields and probably current limitation as important processes. One interpretation of the quasi-dc electric field data is that the internal electric fields of the streams were not greatly modified by magnetic-field-aligned currents, i.e., a state was established where the transverse currents were to a first approximation divergence-free. It is argued that this interpretation can explain both a reversal of the strong explosion-directed electric field in burst 1 and the absence of such a reversal in burst 2.

  • Extending thrombolysis to 4·5-9 h and wake-up stroke using perfusion imaging: a systematic review and meta-analysis of individual patient data.

    • Stroke thrombolysis with alteplase is currently recommended 0-4·5 h after stroke onset. We aimed to determine whether perfusion imaging can identify patients with salvageable brain tissue with symptoms 4·5 h or more from stroke onset or with symptoms on waking who might benefit from thrombolysis.In this systematic review and meta-analysis of individual patient data, we searched PubMed for randomised trials published in English between Jan 1, 2006, and March 1, 2019. We also reviewed the reference list of a previous systematic review of thrombolysis and searched ClinicalTrials.gov for interventional studies of ischaemic stroke. Studies of alteplase versus placebo in patients (aged ≥18 years) with ischaemic stroke treated more than 4·5 h after onset, or with wake-up stroke, who were imaged with perfusion-diffusion MRI or CT perfusion were eligible for inclusion. The primary outcome was excellent functional outcome (modified Rankin Scale [mRS] score 0-1) at 3 months, adjusted for baseline age and clinical severity. Safety outcomes were death and symptomatic intracerebral haemorrhage. We calculated odds ratios, adjusted for baseline age and National Institutes of Health Stroke Scale score, using mixed-effects logistic regression models. This study is registered with PROSPERO, number CRD42019128036.We identified three trials that met eligibility criteria: EXTEND, ECASS4-EXTEND, and EPITHET. Of the 414 patients included in the three trials, 213 (51%) were assigned to receive alteplase and 201 (49%) were assigned to receive placebo. Overall, 211 patients in the alteplase group and 199 patients in the placebo group had mRS assessment data at 3 months and thus were included in the analysis of the primary outcome. 76 (36%) of 211 patients in the alteplase group and 58 (29%) of 199 patients in the placebo group had achieved excellent functional outcome at 3 months (adjusted odds ratio [OR] 1·86, 95% CI 1·15-2·99, p=0·011). Symptomatic intracerebral haemorrhage was more common in the alteplase group than the placebo group (ten [5%] of 213 patients vs one [<1%] of 201 patients in the placebo group; adjusted OR 9·7, 95% CI 1·23-76·55, p=0·031). 29 (14%) of 213 patients in the alteplase group and 18 (9%) of 201 patients in the placebo group died (adjusted OR 1·55, 0·81-2·96, p=0·66).Patients with ischaemic stroke 4·5-9 h from stroke onset or wake-up stroke with salvageable brain tissue who were treated with alteplase achieved better functional outcomes than did patients given placebo. The rate of symptomatic intracerebral haemorrhage was higher with alteplase, but this increase did not negate the overall net benefit of thrombolysis.None.

  • Surface optical reflectance combined with x-ray techniques during gas-surface interactions

    • High energy surface x-ray diffraction (HESXRD), x-ray reflectivity (XRR), mass spectrometry (MS) and surface optical reflectance (SOR) have been combined to simultaneously obtain sub-second information on the surface structure and morphology from a Pd(100) model catalyst during in situ oxidation at elevated temperatures and pressures resulting in Pd bulk oxide formation. The results show a strong correlation between the HESXRD and SOR signal intensities during the experiment, enabling phase determination and a time-resolved thickness estimation of the oxide by HESXRD, complemented by XRR measurements. The experiments show a remarkable sensitivity of the SOR to changes in the surface phase and morphology, in particular to the initial stages of oxidation/reduction. The data imply that SOR can detect the formation of an ultrathin PdO surface oxide layer of only 2-3 angstrom thickness.
