    • I samband med det forskningsprojektet Ingrepp genomfördes en Ingreppfestival med avseende på att i ett öppet publikt sammanhang diskutera utgångspunkterna forskningsgruppen formulerat i tabloiden Ingrepp – en tidning om städer, planering och konst, vilket delades ut gratis en vecka innan festivalen. I tidningen hittade man läsning kring städer, stadsutveckling, konst och konstnärlig forskning vilken hade bäring på den utvecklingsprocess som pågick och pågår på Södra Älvstranden i Göteborg. Både tabloiden och festivalen var ett direkt uttryck för gruppen vilja att ta ut diskussionen på gatan och i sitt sammanhang, men också ett sätt att pröva konstens och akademins möjligheter att gripa in och delta i samhälleliga skeenden. Festivalen vilken turnerade runt på fyra platser under fyra dagar längs Södra Älvstranden innefattade allt ifrån inbjudna samtal/debatter/seminarier till workshops kring Södra Älvstrandens framtid till tipspromenader, picnics och olika former av performativa iscensättningar under bar himmel.

  • Transcriptional Dynamics During Human Adipogenesis and Its Link to Adipose Morphology and Distribution

    • White adipose tissue (WAT) can develop into several phenotypes with different pathophysiological impact on type 2 diabetes. To better understand the adipogenic process, the transcriptional events that occur during in vitro differentiation of human adipocytes were investigated and the findings linked to WAT phenotypes. Single-molecule transcriptional profiling provided a detailed map of the expressional changes of genes, enhancers, and long noncoding RNAs, where different types of transcripts share common dynamics during differentiation. Common signatures include early downregulated, transient, and late induced transcripts, all of which are linked to distinct developmental processes during adipogenesis. Enhancers expressed during adipogenesis overlap significantly with genetic variants associated with WAT distribution. Transiently expressed and late induced genes are associated with hypertrophic WAT (few but large fat cells), a phenotype closely linked to insulin resistance and type 2 diabetes. Transcription factors that are expressed early or transiently affect differentiation and adipocyte function and are controlled by several well-known upstream regulators such as glucocorticosteroids, insulin, cAMP, and thyroid hormones. Taken together, our results suggest a complex but highly coordinated regulation of adipogenesis.

  • Comprehensive genomic profiles of small cell lung cancer

    • We have sequenced the genomes of 110 small cell lung cancers (SCLC), one of the deadliest human cancers. In nearly all the tumours analysed we found bi-allelic inactivation of TP53 and RB1, sometimes by complex genomic rearrangements. Two tumours with wild-type RB1 had evidence of chromothripsis leading to overexpression of cyclin D1 (encoded by the CCND1 gene), revealing an alternative mechanism of Rb1 deregulation. Thus, loss of the tumour suppressors TP53 and RB1 is obligatory in SCLC. We discovered somatic genomic rearrangements of TP73 that create an oncogenic version of this gene, TP73Dex2/3. In rare cases, SCLC tumours exhibited kinase gene mutations, providing a possible therapeutic opportunity for individual patients. Finally, we observed inactivating mutations in NOTCH family genes in 25% of human SCLC. Accordingly, activation of Notch signalling in a pre-clinical SCLC mouse model strikingly reduced the number of tumours and extended the survival of the mutant mice. Furthermore, neuroendocrine gene expression was abrogated by Notch activity in SCLC cells. This first comprehensive study of somatic genome alterations in SCLC uncovers several key biological processes and identifies candidate therapeutic targets in this highly lethal form of cancer.

  • Local interactions and protein folding : A model study on the square and triangular lattices

    • We study a simple heteropolymer model containing sequence-independent local interactions on both square and triangular lattices. Sticking to a two-letter code, we investigate the model for varying strength κ of the local interactions; κ=0 corresponds to the well-known HP model [K. F. Lau and K. A. Dill, Macromolecules 22, 3986 (1989)]. By exhaustive enumerations for short chains, we obtain all structures which act as a unique and pronounced energy minimum for at least one sequence. We find that the number of such designable structures depends strongly on κ. Also, we find that the number of designable structures can differ widely for the two lattices at a given κ. This is the case, for example, at κ=0, which implies that the HP model exhibits different behavior on the two lattices. Our findings clearly show that sequence-independent local properties of the chains can play an important rote in the formation of unique minimum energy structures.

  • Monte Carlo procedure for protein design

    • A method for sequence optimization in protein models is presented. The approach, which has inherited its basic philosophy from recent work by Deutsch and Kurosky [Phys. Rev. Lett. 76, 323 (1996)] by maximizing conditional probabilities rather than minimizing energy functions, is based upon a different and very efficient multisequence Monte Carlo scheme. By construction, the method ensures that the designed sequences represent good folders thermodynamically. A bootstrap procedure for the sequence space search is devised making very large chains feasible. The algorithm is successfully explored on the two-dimensional HP model [K. F. Lau and K. A. Dill, Macromolecules 32, 3986 (1989)] with chain lengths N= 16, 18, and 32.

  • Monte Carlo study of the phase structure of compact polymer chains

    • We study the phase behavior of single homopolymers in a simple hydrophobic/hydrophilic off-lattice model with sequence independent local interactions. The specific heat is, not unexpectedly, found to exhibit a pronounced peak well below the collapse temperature, signalling a possible low-temperature phase transition. The system size dependence at this maximum is investigated both with and without the local interactions, using chains with up to 50 monomers. The size dependence is found to be weak. The specific heat itself seems not to diverge. The homopolymer results are compared with those for two nonuniform sequences. Our calculations are performed using the methods of simulated and parallel tempering. The performances of these algorithms are discussed, based on careful tests for a small system.

  • Candidatus Neoehrlichia mikurensis in Ticks from Migrating Birds in Sweden

    • Candidatus Neoehrlichia mikurensis (CNM; family Anaplasmataceae) was recently recognized as a potential tick-borne human pathogen. The presence of CNM in mammals, in host-seeking Ixodes ticks and in ticks attached to mammals and birds has been reported recently. We investigated the presence of CNM in ornithophagous ticks from migrating birds. A total of 1,150 ticks (582 nymphs, 548 larvae, 18 undetermined ticks and two adult females) collected from 5,365 birds captured in south-eastern Sweden was screened for CNM by molecular methods. The birds represented 65 different species, of which 35 species were infested with one or more ticks. Based on a combination of morphological and molecular species identification, the majority of the ticks were identified as Ixodes ricinus. Samples were initially screened by real-time PCR targeting the CNM 16S rRNA gene, and confirmed by a second real-time PCR targeting the groEL gene. For positive samples, a 1260 base pair fragment of the 16S rRNA gene was sequenced. Based upon bacterial gene sequence identification, 2.1% (24/1150) of the analysed samples were CNM-positive. Twenty-two out of 24 CNM-positive ticks were molecularly identified as I. ricinus nymphs, and the remaining two were identified as I. ricinus based on morphology. The overall CNM prevalence in I. ricinus nymphs was 4.2%. None of the 548 tested larvae was positive. CNM-positive ticks were collected from 10 different bird species. The highest CNM-prevalences were recorded in nymphs collected from common redpoll (Carduelis flammea, 3/7), thrush nightingale (Luscinia luscinia, 2/29) and dunnock (Prunella modularis, 1/17). The 16S rRNA sequences obtained in this study were all identical to each other and to three previously reported European strains, two of which were obtained from humans. It is concluded that ornithophagous ticks may be infected with CNM and that birds most likely can disperse CNM-infected ticks over large geographical areas.

    • A designer is somebody who points, who designates, and gives directions. Design thereby has a direction into the future. What directions are designers pointing out if design is coupled with posthumanism? Posthumanism has come into being in a landscape of both ideas and design. That which has previously been designed and produced is coming back and it can help us point out harmful inequalities if we sharpen our observational tools and concepts.“An Emerging Posthumanist Design Landscape” is an overflowing designated area for examples and thinking on compositions of design and critical posthumanism. It is a landscape in the making, yet scarred by previous design cultures and histories. As design researchers operating out of Scandinavian academia, we invite readers/travelers to meander through an emerging hybrid landscape and to make a few selected stops at the sites of our own recent design interventions. We articulate concepts, frictions, and opportunities sprouted in a sprawling and increasingly populated landscape of design and posthumanism. Posthumanist thinking questions and recharges fundamental design concepts and methods/approaches, e.g.: Who are the actors of posthumanist design? Where does it take place? What do we design? What materials do we use? How do we work? When does design take place? Why are compositions of design and critical posthumanism important undertakings? The responses to these questions sketch trajectories for further travels and the co-creation of an emerging posthumanist design landscape.

  • After the Revolution : Prototyping Post-Speciesist Futures

    • What could a post-speciesist world be like?Critical Animal Studies activists and scholars have developed convincing counter-arguments to speciesism and animal oppression. These arguments are continuously developed and reshaped through contributions from fields like gender studies, postcolonialism, environmental humanities, and philosophy. This broad range of approaches makes for an diverse and growing body of knowledge on the systematic discrimination, exploitation, and oppression of nonhuman animals, not least regarding the treatment of animals today and in the past. We argue, however, that this knowledge production is significantly more sporadic when it comes to constructive proposals of less speciesist futures. Where are the snapshots from potential futures, and alternative presents, where human-animal relations are radically reconfigured?We suggest that in working towards an anti-speciesist revolution we need to also be able to imagine what living in a post-speciesist society could be like; and explore creative tactics for bringing these material propositions into being.These kinds of speculations and constructions of scenarios involve future-oriented contributions from fields such as the arts, design, literature, architecture, and speculative philosophy. In other words, domains that are engaged with envisioning, prototyping, and rehearsing potential futures and alternative presents. In this paper, we discuss a number of works that in different ways materialise reconfigured relations between humans and other species. Examples include utopian artworks by Hartmut Kievert, Ursula Le Guin’s ecofeminist stories, as well as our own design projects on sketching already existing post-speciesist animal-human encounters and redesigning recreational fishing practices. We discuss what tactics are employed by the creators and how their designerly approaches might help in generating new ideas about possible futures. We also introduce and reflect on tools and practices from the design disciplines, such as sketching, prototyping, and design fiction that can be of use for CAS scholar-activists.Importantly, an affirmative approach of imagining post-speciesist futures does not come without risk. It can be argued that constructive, at times hopeful, projects distract from militating against the currently dim situation that billions of animals face daily. It can also be argued that we are nowhere near attaining a world that can be considered hopeful for most animals on our planet. Shouldn’t we focus on bringing about the revolution before speculating on its aftermath?We argue that research and activism against speciesism ought to be complemented by constructive scenarios for post-speciesist futures. We seek to contribute to the field of Critical Animal Studies by calling for and articulating a stronger speculative and imaginative strand of CAS, without blunting the urgency and critical edge of the field.
