  • Neonatal invasive candidiasis in low- and middle-income countries: Data from the NeoOBS study

    • Neonatal invasive candidiasis (NIC) has significant morbidity and mortality. Reports have shown a different profile of those neonates affected with NIC and of fluconazole-resistant Candida spp. isolates in low- and middle-income countries (LMICs) compared to high-income countries (HICs). We describe the epidemiology, Candida spp. distribution, treatment, and outcomes of neonates with NIC from LMICs enrolled in a global, prospective, longitudinal, observational cohort study (NeoOBS) of hospitalized infants <60 days postnatal age with sepsis (August 2018–February 2021). A total of 127 neonates from 14 hospitals in 8 countries with Candida spp. isolated from blood culture were included. Median gestational age of affected neonates was 30 weeks (IQR: 28–34), and median birth weight was 1270 gr (interquartile range [IQR]: 990–1692). Only a minority had high-risk criteria, such as being born <28 weeks, 19% (24/127), or birth weight <1000 gr, 27% (34/127). The most common Candida species were C. albicans (n = 45, 35%), C. parapsilosis (n = 38, 30%), and Candida auris (n = 18, 14%). The majority of C. albicans isolates were fluconazole susceptible, whereas 59% of C. parapsilosis isolates were fluconazole-resistant. Amphotericin B was the most common antifungal used [74% (78/105)], followed by fluconazole [22% (23/105)]. Death by day 28 post-enrollment was 22% (28/127). To our knowledge, this is the largest multi-country cohort of NIC in LMICs. Most of the neonates would not have been considered at high risk for NIC in HICs. A substantial proportion of isolates was resistant to first choice fluconazole. Understanding the burden of NIC in LMIC is essential to guide future research and treatment guidelines.

    • Annealing dependence of the lattice parameter, resistivity, magnetoresistance and thermopower have been studied on Nd0.87Sr0.33MnO3 thin films deposited on LaAlO3 and alumina substrates by pulsed laser ablation. Upon annealing at 800 degrees C and 1000 degrees C the lattice constant of the LaAlO3 film tends toward that of the bulk target due to reduction in oxygen vacancies. This results in a metal-insulator transition at temperatures which increase with progressive annealing along with a decrease in the observed low temperature MR. Using a magnon scattering model we estimate the e(g) bandwidth of the film annealed at 1000 degrees C and show that the magnon contribution to the resistivity is suppressed in a highly oxygen deficient film and gains prominence only upon subsequent annealing. We also show that upon annealing, the polaron concentration and the spin cluster size increases in the paramagnetic phase, using an adiabatic polaron hopping model which takes into account an exchange dependent activation energy above the resistivity peak.

  • Cyber security analysis of state estimators in electric power systems

    • In this paper, we analyze the cyber security of state estimators in Supervisory Control and Data Acquisition (SCADA) systems operating in power grids. Safe and reliable operation of these critical infrastructure systems is a major concern in our society. In current state estimation algorithms there are bad data detection (BDD) schemes to detect random outliers in the measurement data. Such schemes are based on high measurement redundancy. Although such methods may detect a set of very basic cyber attacks, they may fail in the presence of a more intelligent attacker. We explore the latter by considering scenarios where deception attacks are performed, sending false information to the control center. Similar attacks have been studied before for linear state estimators, assuming the attacker has perfect model knowledge. Here we instead assume the attacker only possesses a perturbed model. Such a model may correspond to a partial model of the true system, or even an out-dated model. We characterize the attacker by a set of objectives, and propose policies to synthesize stealthy deceptions attacks, both in the case of linear and nonlinear estimators. We show that the more accurate model the attacker has access to, the larger deception attack he can perform undetected. Specifically, we quantify trade-offs between model accuracy and possible attack impact for different BDD schemes. The developed tools can be used to further strengthen and protect the critical state-estimation component in SCADA systems.

    • We study hybrid systems from a global geometric perspective as piecewise smooth dynamical systems. Based on an earlier work, we define the notion of the hybrifold as a single piece-wise smooth state space reflecting the dynamics of the original system. Structural stability for hybrid systems is introduced and analyzed in this framework. In particular, it is shown that a Zeno state is locally structurally stable and that a standard equilibrium on the boundary of a domain implies structural instability.

    • Soft materials are usually defined as materials made of mesoscopic entities, often self-organised, sensitive to thermal fluctuations and to weak perturbations. Archetypal examples are colloids, polymers, amphiphiles, liquid crystals, foams. The importance of soft materials in everyday commodity products, as well as in technological applications, is enormous, and controlling or improving their properties is the focus of many efforts. From a fundamental perspective, the possibility of manipulating soft material properties, by tuning interactions between constituents and by applying external perturbations, gives rise to an almost unlimited variety in physical properties. Together with the relative ease to observe and characterise them, this renders soft matter systems powerful model systems to investigate statistical physics phenomena, many of them relevant as well to hard condensed matter systems. Understanding the emerging properties from mesoscale constituents still poses enormous challenges, which have stimulated a wealth of new experimental approaches, including the synthesis of new systems with, e.g. tailored self-assembling properties, or novel experimental techniques in imaging, scattering or rheology. Theoretical and numerical methods, and coarse-grained models, have become central to predict physical properties of soft materials, while computational approaches that also use machine learning tools are playing a progressively major role in many investigations. This Roadmap intends to give a broad overview of recent and possible future activities in the field of soft materials, with experts covering various developments and challenges in material synthesis and characterisation, instrumental, simulation and theoretical methods as well as general concepts.

    • Genome-scale network reconstructions have helped uncover the molecular basis of metabolism. Here we present Recon3D, a computational resource that includes three-dimensional (3D) metabolite and protein structure data and enables integrated analyses of metabolic functions in humans. We use Recon3D to functionally characterize mutations associated with disease, and identify metabolic response signatures that are caused by exposure to certain drugs. Recon3D represents the most comprehensive human metabolic network model to date, accounting for 3,288 open reading frames (representing 17% of functionally annotated human genes), 13,543 metabolic reactions involving 4,140 unique metabolites, and 12,890 protein structures. These data provide a unique resource for investigating molecular mechanisms of human metabolism. Recon3D is available at http://vmh.life.

    • The actions of an autonomous vehicle on the road affect and are affected by those of other drivers, whether overtaking, negotiating a merge, or avoiding an accident. This mutual dependence, best captured by dynamic game theory, creates a strong coupling between the vehicle's planning and its predictions of other drivers' behavior, and constitutes an open problem with direct implications on the safety and viability of autonomous driving technology. Unfortunately, dynamic games are too computationally demanding to meet the real-time constraints of autonomous driving in its continuous state and action space. In this paper, we introduce a novel game-theoretic trajectory planning algorithm for autonomous driving, that enables real-time performance by hierarchically decomposing the underlying dynamic game into a long-horizon "strategic" game with simplified dynamics and full information structure, and a short-horizon "tactical" game with full dynamics and a simplified information structure. The value of the strategic game is used to guide the tactical planning, implicitly extending the planning horizon, pushing the local trajectory optimization closer to global solutions, and, most importantly, quantitatively accounting for the autonomous vehicle and the human driver's ability and incentives to influence each other. In addition, our approach admits non-deterministic models of human decision-making, rather than relying on perfectly rational predictions. Our results showcase richer, safer, and more effective autonomous behavior in comparison to existing techniques.
