    • The rat provides valuable and sometimes unique models of human complex diseases. To fully exploit the rat models in biomedical research, it is important to have access to detailed knowledge of the rat genome organization as well as its relation to the human genome. Rat Chromosome 10 (RNO10) harbors several important cancer-related genes. Deletions in the proximal part of RNO10 were repeatedly found in a rat model for endometrial cancer. To identify functional and positional candidate genes in the affected region, we used radiation hybrid (RH) mapping and single- and dual-color fluorescence in situ hybridization (FISH) techniques to construct a detailed chromosomal map of the proximal part of RNO10. The regional localization of 14 genes, most of them cancer-related ( Grin2a, Gspt1, Crebbp, Gfer, Tsc2, Tpsb1, Il9r, Il4, Irf1, Csf2, Sparc, Tp53, Thra1, Gh1), and of five microsatellite markers ( D10Mit10, D10Rat42, D10Rat50, D10Rat72, and D10Rat165) was determined on RNO10. For a fifteenth gene, Ppm1b, which had previously been assigned to RNO10, the map position was corrected to RNO6q12-q13.

    • Using FISH and RH mapping a chromosomal map of rat chromosome 10 (RNO10) was constructed. Our mapping data were complemented by other published data and the final map was compared to maps of mouse and human chromosomes. RNO10 contained segments homologous to mouse chromosomes (MMU) 11, 16 and 17, with evolutionary breakpoints between the three segments situated in the proximal part of RNO10. Near one of these breakpoints (between MMU17 and 11) we found evidence for an inversion ancestral to the mouse that was not ancestral to the condition in the rat. Within each of the chromosome segments identified, the gene order appeared to be largely conserved. This conservation was particularly clear in the long MMU11-homologous segment. RNO10 also contained segments homologous to three human chromosomes (HSA5, 16, 17). However, within each segment of conserved synteny were signs of more extensive rearrangements. At least 13 different evolutionary breakpoints were indicated in the rat-human comparison. In contrast to what was found between rat and mouse, the rat-human evolutionary breaks were distributed along the entire length of RNO10.

  • Measurements of memory effect and resolution for an Al2O3 coated plastic scintillator used for radioxenon detection

    • A cylindrical plastic scintillator cell, used for radioxenon monitoring within the verification regime of the Comprehensive Nuclear-Test-Ban Treaty, has been coated with 425 nm Al2O3 using low temperature Atomic Layer Deposition, and its performance has been evaluated. The motivation is to reduce the memory effect caused by radioxenon diffusing into the plastic scintillator material during measurements, resulting in an elevated detection limit. Measurements of the coated detector show a resolution comparable to uncoated detectors, and a memory effect reduction of a factor of 1000. If these results are reproducible, and the quality of the detector is maintained for a longer period of time, the Al2O3 coating method is believed to be viable solution to the memory effect problem in question.

    • Denna slutrapport redogör för resultatet av projektet: AutoMon Del I—Automatisk trafikövervakning genom AI i svenska farvatten (TRV2019/95873). Projektägare är Linköpings universitet och genomförts tillsammans med Sjöfartsverket, SAAB, VTI, och Luftfartsverket. Vi kommer i denna rapport redogöra för resultaten samt diskutera framtida utmaningar med konceptet AutoMon. Syftet med AutoMon är att undersöka och belysa problemområdet automatiserad trafikövervakning genom att implementera en AI-demonstrator för svenska farvatten, initialt med fokus på att detektera anomalier i trafiken, och där hanteringen (dvs. granskning och lösning) av problemsituationer lämnas över till en mänsklig operatör. Förekomsten av fartygsolyckor och -incidenter, med potentiellt stora konsekvenser för individ och miljö - utanför etablerade VTS-områden - visar på behovet av utökad övervakning av sjötrafiken i svenska farvatten för att möjliggöra proaktiva åtgärder för att minimera olycksrisker till sjöss. Forskningsprojektet undersöker hur en hög nivå av automation kan användas för att upptäcka och hantera trafikavvikelser på svenskt vatten samt ge möjlighet till s.k. situational awareness (lägesbild) över hela dess yta, samtidigt som stöd för centralt eller distribuerat beslutsfattande ges. Runt om i världen förekommer även manipulation av AIS-systemet och dess positionsangivelser; ett problem som sannolikt kommer att öka även i Sverige. Båda problemställningarna är globala, vilket ger hög relevans. Lösningar har högt värde där digitalisering och artificiell intelligens kan användas som medel för ökad sjösäkerhet (safety och cyber security).

    • BACKGROUND: Fluid resuscitation after uncontrolled hemorrhage might promote rebleeding and irreversible shock. Tranexamic acid is a procoagulant drug that limits blood loss after surgery of the hip, knee, and heart. We hypothesized that pretreatment with tranexamic acid reduces the rebleeding in uncontrolled hemorrhage and thereby allows safe administration of crystalloid fluid resuscitation.METHODS: A 120-minute intravenous infusion of 100 mL/kg of Ringer's solution was given to 24 pigs (mean weight, 20 kg) 10 minutes after lacerating the infrarenal aorta. The animals were randomized to receive an intravenous injection of 15 mg/kg of tranexamic acid or placebo just before starting the resuscitation. Rebleeding events were monitored by two ultrasonic probes positioned proximal and distal to the laceration.RESULTS: Tranexamic acid had no effect on the number of rebleeding events, bled volume, or mortality. The initial bleeding stopped within 4 minutes after the injury. The five animals that died suffered from 4.4 rebleeding events on average, which tripled the total blood loss, whereas the survivors had only 1.3 such events during fluid resuscitation (p < 0.02). At autopsy, death was associated with a larger total hemorrhage; the blood recovered from the abdomen weighed 1.4 kg (median) in nonsurvivors and 0.6 kg in survivors (p < 0.001), with the difference being attributable to rebleeding.CONCLUSION: Rebleeding events increased the amount of blood lost and the mortality in uncontrolled aortic hemorrhage. Tranexamic acid offered no benefit.
