    • A new generation of high-resolution hypernuclear gamma-spectroscopy experiments with high-purity germanium detectors (HPGe) are presently designed at the FINUDA spectrometer at DA Phi NE, the Frascati phi-factory, and at (P) over bar ANDA, the (p) over barp hadron spectrometer at the future FAIR facility. Both, the FINUDA and (P) over bar ANDA spectrometers are built around the target region covering a large solid angle. To maximise the detection efficiency the HPGe detectors have to be located near the target, and therefore they have to be operated in strong magnetic fields (B approximate to I T). The performance of HPGe detectors in such an environment has not been well investigated so far. In the present work VEGA and EUROBALL Cluster HPGe detectors were tested in the field provided by the ALADiN magnet at GSI. No significant degradation of the energy resolution was found, and a change in the rise time distribution of the pulses from preamplifiers was observed. A correlation between rise time and pulse height was observed and is used to correct the measured energy, recovering the energy resolution almost completely. Moreover, no problems in the electronics due to the magnetic field were observed.

  • The identification of discontinuous epitope in the human cystatin C – Monoclonal antibody HCC3 complex

    • Human cystatin C (hCC) is a cysteine proteinase inhibitor involved in pathophysiological processes of dimerization and amyloid formation. These processes are directly associated with a number of neurodegenerative disorders such as Alzheimer disease or hereditary cystatin C amyloid angiopathy (HCCAA). One of the ideas on how to prevent amyloid formation is to use immunotherapy. HCC3 is one of a group of antibodies binding to hCC and reducing the in vitro formation of cystatin C dimers. Therefore, identification of the binding sites in the hCC-HCC3 complex may facilitate a search of effective drugs against HCCAA as well as understanding the mechanisms of neurodegenerative disorders. In this work we present epitope identification of the hCC-HCC3 complex using methods such as affinity chromatography, epitope excision and extraction MS approach, enzyme-linked immunosorbent assay and hydrogen-deuterium exchange mass spectrometry (HDX MS). Comprehensive analysis of the obtained results allowed us to identify the epitope sequence with the key fragment covering hCC L1 loop and two potential epitopic fragments – α-helical part, hCC (17–28) and β4 strand in C-terminal part of hCC. The presence of the L1 loop in the epitope sequence accounts for the significant reduction of hCC dimer formation in the presence of HCC3 antibody. Significance of the study: Deciphering the mechanism of the cystatin C aggregation process and detailed analysis of the interactions between hCC, or its pathogenic variant, and monoclonal antibodies, potentially constituting aggregation inhibitors, might be of great value as there still is a complete lack of any kind of efficient therapy for young people with the pathogenic mutation of hCC.

    • The use of High Purity Germanium detectors (HPGe) has been planned in some future experiments of hadronic physics. The crystals will be located close to large spectrometers where the magnetic fringing field will not be negligible and their performances might change. Moreover high precision is required in these experiments. The contribution of magnetic field presence and long term measurements is unique. In this paper the results of systematic measurements of the resolution, stability and efficiency of a crystal operating inside a magnetic field of 0.8 T, using radioactive sources in the energy range from 0.08 to 1.33 MeV, are reported. The measurements have been repeated during several months in order to test if any permanent damage occurred. The resolution at 1.117 and 1.332 MeV gamma-rays from a Co-60 source has been measured at different magnetic fields in the range of 0-0.8 T and the results are compared with the previous data.

  • Properties of the signal mode in the polariton optical parametric oscillator regime

    • Theoretical analyses of the polariton optical parametric oscillator (OPO) regime often rely on a mean-field approach based on the complex Gross-Pitaevskii equations in a three-mode approximation, where only three momentum states, the signal, pump, and idler, are assumed to be significantly occupied. This approximation, however, lacks a constraint to uniquely determine the signal and idler momenta. In contrast, multimode numerical simulations and experiments show a unique momentum structure for the OPO states. In this work we show that an estimate for the signal momentum chosen by the system can be found from a simple analysis of the pump-only configuration. We use this estimate to investigate how the chosen signal momentum depends on the properties of the drive.

    • The original guidelines drawn up for the management of the neuronopathic forms of Gaucher disease were felt to be in need of revision; in particular, the role of high-dose enzyme replacement therapy (120 IU/kg of body weight every 2 weeks) in stabilizing neurological disease. The existing published evidence was analysed; it was concluded that it did not support the role of high-dose ERT, although this might be required to treat severe visceral disease.
