    • Using the ImmunoChip custom genotyping array, we analyzed 14,498 subjects with multiple sclerosis and 24,091 healthy controls for 161,311 autosomal variants and identified 135 potentially associated regions (P < 1.0 × 10(-4)). In a replication phase, we combined these data with previous genome-wide association study (GWAS) data from an independent 14,802 subjects with multiple sclerosis and 26,703 healthy controls. In these 80,094 individuals of European ancestry, we identified 48 new susceptibility variants (P < 5.0 × 10(-8)), 3 of which we found after conditioning on previously identified variants. Thus, there are now 110 established multiple sclerosis risk variants at 103 discrete loci outside of the major histocompatibility complex. With high-resolution Bayesian fine mapping, we identified five regions where one variant accounted for more than 50% of the posterior probability of association. This study enhances the catalog of multiple sclerosis risk variants and illustrates the value of fine mapping in the resolution of GWAS signals.

  • Effect of grain size on ferroelectric domain and electrical properties of submicron sized sodium potassium niobate ceramics

    • Piezoelectric ceramics of the composition Na0.5K0.5NbO3 (NKN) with grain sizes in the range of 0.2 - 1 mm were fabricated by Spark Plasma Sintering using normal pressure dies and a high pressure cell designed for pressures up to 500 MPa  with the purpose of investigating the effect of grain size on domain structures and electrical properties. Optimized processing conditions enabled ceramics of high densities (>99.5%TD) to be made at T≥850°C. It was found that domain size decreases with decreasing grain size and that non-180° ferroelectric domains walls were still visible in 200 nm sized grains. The room temperature dielectric constant firstly increased with decreasing grain size and then decreased in the low grain size regime. The materials with finer grain size displayed a broad ferro-paraelectric phase transition and a depression of the dielectric maximum at the Curie point. They also displayed an increase in the coercive field and approximately unchanged remnant polarization. The material sintered at 850°C represents a very good candidate for lead-free piezoelectric applications, because of its high piezoelectric constant (d33 = 160±2 pC/N).

    • Piezoelectric ceramics of the composition Na(0.5)K(0.5)NbO(3) (NKN) with grain sizes in the range of 0.2-1 mu m were fabricated by spark plasma sintering. Ferroelectric domain size decreases with decreasing grain size and non-180 degrees ferroelectric domains walls were still visible in 200 nm sized grains. The Curie point of the ceramics was grain size independent. This suggests that the critical grain size for a single domain single grain structure for NKN is <200 nm. Optimized processing conditions enabled ceramics of high densities (>99.5% theoretical density) to be made at T >= 850 degrees C. For the dense ceramics (grain size >= 350 nm), the room temperature dielectric constant and coercive field increased with decreasing grain size. The remnant polarization was grain size independent. The material sintered at 850 degrees C is a very good candidate for lead-free piezoelectric applications because of its high piezoelectric constant (d(33) = 160 +/- 2 pC/N).

    • The majority of piezoelectric ceramics based on Pb(Zr, Ti)O3 (PZT) exhibit hysteretic behavior, which varies depending on various factors, including composition, microstructure and processing conditions. The ternary system 0.05Pb(Mn1/3Sb2/3)O3–0.95Pb(Zr0.52Ti0.48)O3 (5PMS-PZT) has shown attractive piezoelectric properties with superior thermal stability compared to pure Pb(Zr0.52Ti0.48)O3, but the underlying mechanisms of its complex hysteretic behavior have not been fully clarified yet. This work further delves into the hysteretic aspects of 5PMS-PZT ceramics and describes hysteresis features that have not been yet reported for any known piezoelectric system. Four current peaks in I-E loops and S-E loops with unconventional shapes, which have a significant dependence on applied field amplitude, frequency and electrical history have been observed here. This peculiar hysteretic behavior is influenced by convoluted effects of domain switching, electric field-induced structural transitions, point defects and aging/de-aging processes, whose combinations lead to unique hysteresis features.

  • Guidelines for the use and interpretation of assays for monitoring autophagy

    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Interobserver Variation among Pathologists and Refinement of Criteria in Distinguishing Separate Primary Tumors from Intrapulmonary Metastases in Lung

    • Multiple tumor nodules are seen with increasing frequency in clinical practice. On the basis of the 2015 WHO classification of lung tumors, we assessed the reproducibility of the comprehensive histologic assessment to distinguish second primary lung cancers (SPLCs) from intrapulmonary metastases (IPMs), looking for the most distinctive histologic features. An international panel of lung pathologists reviewed a scanned sequential cohort of 126 tumors from 48 patients and recorded an agreed set of histologic features, including tumor typing and predominant pattern of adenocarcinoma, thereby opining whether the case was SPLC, IPM, or a combination thereof. Cohen kappa statistics of 0.60 on overall assessment of SPLC or IPM indicated a good agreement. Likewise, there was good agreement (kappa score 0.64, p < 0.0001) between WHO histologic pattern in individual cases and SPLC or IPM status, but the proportions diversified for histologic pattern and SPLC or IPM status (McNemar test, p < 0.0001). The strongest associations for distinguishing between SPLC and IPM were observed for nuclear pleomorphism, cell size, acinus formation, nucleolar size, mitotic rate, nuclear inclusions, intraalveolar clusters, and necrosis. Conversely, the associations for lymphocytosis, mucin content, lepidic growth, vascular invasion, macrophage response, clear cell change, acute inflammation keratinization, and emperipolesis did not reach significance with tumor extent. Comprehensive histologic assessment is recommended for distinguishing SPLC from IPM with good reproducibility among lung pathologists. In addition to main histologic type and predominant patterns of histologic subtypes, nuclear pleomorphism, cell size, acinus formation, nucleolar size, and mitotic rate strongly correlate with pathologic staging status.

    • The microscopic origin of the grain size effects on the dielectric, piezoelectric, and thermal depoling properties of Aurivillius phase Bi3.15Nd0.85Ti3O12 was investigated. Using atomic force microscopy, domain walls were observed in micrometer grain size ceramics, but gradually disappeared with reducing grain size and were not found in ceramics with 90 nm grain size. In strain-electric field butterfly loops, the strain decreased with decreasing grain size indicating a decreasing contribution of non-180 degrees domain walls switching to the strain. Lattice distortion (a-b)/b decreased with decreasing grain size. The thermal depoling resistance decreased with decreasing grain size, due to increasing internal mechanical stresses.

    • This paper explores the application of quantum nonlocality, a renowned and unique phenomenon acknowledged as a valuable resource. Focusing on an alternative application, we demonstrate its quantum advantage for mobile agents engaged in specific distributed tasks without communication. The research addresses the significant challenge of rendezvous on graphs and introduces a distributed task for mobile agents grounded in the graph domination problem. Through an investigation across various graph scenarios, we showcase the quantum advantage. Additionally, we scrutinize deterministic strategies, highlighting their comparatively lower efficiency compared to quantum strategies. The paper concludes with a numerical analysis, providing further insights into our findings.
