    • The symbiotic fungus Paxillus involutus serves a critical role in maintaining forest ecosystems, which are carbon sinks of global importance. P. involutus produces involutin and other 2,5-diarylcyclopentenone pigments that presumably assist in the oxidative degradation of lignocellulose via Fenton chemistry. Their precise biosynthetic pathways, however, remain obscure. Using a combination of biochemical, genetic, and transcriptomic analyses, in addition to stable-isotope labeling with synthetic precursors, we show that atromentin is the key intermediate. Atromentin is made by tridomain synthetases of high similarity: InvA1, InvA2, and InvA5. An inactive atromentin synthetase, InvA3, gained activity after a domain swap that replaced its native thioesterase domain with that of InvA5. The found degree of multiplex biosynthetic capacity is unprecedented with fungi, and highlights the great importance of the metabolite for the producer.

  • Towards Modelling a Cloud Application's Life Cycle

    • The success of any cloud-based application depends on appropriate decisions being taken at each phase of the life cycle of that application coupled with the stage of the organisation’s business strategy at any given time. Throughout the life cycle of a cloud-based project, various stakeholders are involved. This requires a consistent definition of organizational, legal and governance issues regardless of the role of the stakeholder. We proffer that currently the models and frameworks that offer to support these stakeholders are predominantly IT focused and as such lack a sufficient focus on the business and its operating environment for the decision-makers to make strategic cloud related decisions that benefit their individual business model. We propose an emerging framework that provides a stronger platform on which to base cloud business decisions. We also illustrate the importance of this approach through extrapolating the subject of security from the initial Business Case definition phase, through the Decision Making phase and into the Application Development phase to strengthen the case for a comprehensive Business-based framework for cloud-based application decision-making. We envisage that this emerging framework will then be further developed around all phases of the Application Life Cycle as a means of ensuring consistency.

    • Autism spectrum disorder (ASD) is a highly heritable disorder of complex and heterogeneous aetiology. It is primarily characterized by altered cognitive ability including impaired language and communication skills and fundamental deficits in social reciprocity. Despite some notable successes in neuropsychiatric genetics, overall, the high heritability of ASD (~90%) remains poorly explained by common genetic risk variants. However, recent studies suggest that rare genomic variation, in particular copy number variation, may account for a significant proportion of the genetic basis of ASD. We present a large scale analysis to identify candidate genes which may contain low-frequency recessive variation contributing to ASD while taking into account the potential contribution of population differences to the genetic heterogeneity of ASD. Our strategy, homozygous haplotype (HH) mapping, aims to detect homozygous segments of identical haplotype structure that are shared at a higher frequency amongst ASD patients compared to parental controls. The analysis was performed on 1,402 Autism Genome Project trios genotyped for 1 million single nucleotide polymorphisms (SNPs). We identified 25 known and 1,218 novel ASD candidate genes in the discovery analysis including CADM2, ABHD14A, CHRFAM7A, GRIK2, GRM3, EPHA3, FGF10, KCND2, PDZK1, IMMP2L and FOXP2. Furthermore, 10 of the previously reported ASD genes and 300 of the novel candidates identified in the discovery analysis were replicated in an independent sample of 1,182 trios. Our results demonstrate that regions of HH are significantly enriched for previously reported ASD candidate genes and the observed association is independent of gene size (odds ratio 2.10). Our findings highlight the applicability of HH mapping in complex disorders such as ASD and offer an alternative approach to the analysis of genome-wide association data.

    • Recently, virtual realities and simulations play important roles in the development of automated driving functionalities. By an appropriate abstraction, they help to design, investigate and communicate real traffic scenario complexity. Especially, for edge cases investigations of interactions between vulnerable road users (VRU) and highly automated driving functions, valid virtual models are essential for the quality of results. The aim of this study is to measure, process and integrate real human movement behaviour into a virtual test environment for highly automated vehicle functionalities. The overall system consists of a georeferenced virtual city model and a vehicle dynamics model, including probabilistic sensor descriptions. By motion capture hardware, real humanoid behaviour is applied to a virtual human avatar in the test environment. Through retargeting methods, which enable the independency of avatar and person under test (PuT) dimensions, the virtual avatar diversity is increased. To verify the biomechanical behaviour of the virtual avatars, a qualitative study is performed, which funds on a representative movement sequence. The results confirm the functionality of the used methodology and enable PuT independence control of the virtual avatars in real-time.

  • Multicomponent new particle formation from sulfuric acid, ammonia, and biogenic vapors

    • A major fraction of atmospheric aerosol particles, which affect both air quality and climate, form from gaseous precursors in the atmosphere. Highly oxygenated organic molecules (HOMs), formed by oxidation of biogenic volatile organic compounds, are known to participate in particle formation and growth. However, it is not well understood how they interact with atmospheric pollutants, such as nitrogen oxides (NOx) and sulfur oxides (SOx) from fossil fuel combustion, as well as ammonia (NH3) from livestock and fertilizers. Here, we show how NOx suppresses particle formation, while HOMs, sulfuric acid, and NH3 have a synergistic enhancing effect on particle formation. We postulate a novel mechanism, involving HOMs, sulfuric acid, and ammonia, which is able to closely reproduce observations of particle formation and growth in daytime boreal forest and similar environments. The findings elucidate the complex interactions between biogenic and anthropogenic vapors in the atmospheric aerosol system.

    • The interaction between nitrogen monoxide (NO) and organic peroxy radicals (RO2) greatly impacts the formation of highly oxygenated organic molecules (HOM), the key precursors of secondary organic aerosols. It has been thought that HOM production can be significantly suppressed by NO even at low concentrations. Here, we perform dedicated experiments focusing on HOM formation from monoterpenes at low NO concentrations (0 – 82 pptv). We demonstrate that such low NO can enhance HOM production by modulating the RO2 loss and favoring the formation of alkoxy radicals that can continue to autoxidize through isomerization.These insights suggest that HOM yields from typical boreal forest emissions can vary between 2.5%-6.5%, and HOM formation will not be completely inhibited even at high NO concentrations. Our findings challenge the notion that NO monotonically reduces HOM yields by extending the knowledge of RO2-NO interactions to the low-NO regime. This represents a major advance towards an accurate assessment of HOM budgets, especially in low-NO environments, which prevails in the preindustrial atmosphere, pristine areas, and the upper boundary layer.

    • Autism spectrum disorders (ASDs) are common, heritable neurodevelopmental conditions. The genetic architecture of ASDs is complex, requiring large samples to overcome heterogeneity. Here we broaden coverage and sample size relative to other studies of ASDs by using Affymetrix 10K SNP arrays and 1,168 families with at least two affected individuals, performing the largest linkage scan to date while also analyzing copy number variation in these families. Linkage and copy number variation analyses implicate chromosome 11p12-p13 and neurexins, respectively, among other candidate loci. Neurexins team with previously implicated neuroligins for glutamatergic synaptogenesis, highlighting glutamate-related genes as promising candidates for contributing to ASDs.

    • We propose the AQUILA framework (Adaptive Quality Assurance in Industrial Applications), a concept for digitalization in Industry 4.0 to support the entire industrial manufacturing chain, laying the groundwork for adaptive quality assurance in times of disrupted supply chains and, due to the COVID-19 pandemic, restricted travel possibilities. To that end, our proposed framework allows for the definition and description of industrial processes, quality assurance and testing protocols, and training scenarios in a comprehensive notation based on BPMN, and supports users in task execution, documentation, and evaluation by providing smart glass-based HCI with eye tracking technology, delivering a combination of process documentation, context-sensitive AR visualization, gaze-based interaction schemes, and remote maintenance and assistance functionality.

    • A high level of information security in critical infrastructure IT systems and services has to be preserved when migrating their IT services to the cloud. Often various legislative and security constraints have to be met in line with best practice guidelines and international standards to perform the migration. To support the critical infrastructure providers in migrating their services to the cloud we are developing a process based migration guideline for critical infrastructure providers focusing on information security. First of all we investigate, via questionnaires, how the importance of individual security topics covered in such guidelines differentiates between industry stakeholders and critical infrastructure providers. This supports the selection of relevant security topics and the considered guidelines and standards, which we survey in search for common relevant security topics. Subsequently we present the analysis of the above-mentioned security requirements and how they affect a here developed taxonomy for a process-based security guideline. Furthermore we present potential service migration use cases and how our methodology would affect the migration of secure critical infrastructure services.
